  • In this study, a series of hydroxyaptite (HAp) are produced on Ti dental implant using electrochemical deposition. Based on the preliminary analysis of the coating structure, composition and morphology. In vitro studies were performed with MC3T3-E1 cell to investigate the effect of biological change on different surface conditions.

Fabrication and Characterization of BCP Nano Particle Loaded PCL Fiber and Their Biocompatibility

  • The electrospinning process was established as a promising method to fabricate nano and micro-textured scaffolds for tissue engineering applications. A BCP-loaded PCL micro-textured scaffold thus can be a viable option. The biocompatibility as well as the mechanical properties of such scaffold materials should be optimized for this purpose. In this study, a composite scaffold of poly ($\varepsilon$-caprolactone) (PCL)-biphase calcium phosphate (BCP) was successfully fabricated by electrospinning. EDS and XRD data show successful loading of BCP nano particles in the PCL fibers. Morphological characterization of fibers shows that with a higher loaded BCP content the fiber surface was rougher and the diameter was approximately 1 to 7 ${\mu}m$. Tensile modulus and ultimate tensile stress reached their highest values in the PCL- 10 wt% BCP composite. When content of nano ceramic particles was low, they were dispersed in the fibers as reinforcements for the polymer matrix. However, at a high content of ceramic particles, the particles tend to agglomerate and lead to decreasing tensile modulus and ultimate stress of the PCL-BCP composite mats. Therefore, the use of nano BCP content for distribution in fiber polymer using BCP for reinforcement is limited. Tensile strain decreased with increasing content of BCP loading. From in vitro study using MG-63 osteoblast cells and L-929 fibroblast like cells, it was confirmed that electrospun PCL-BCP composite mats were biocompatible and that spreading behavior was good. As BCP content increased, the area of cell spreading on the surface of the mats also increased. Cells showed the best adherence on the surface of composite mats at 50 wt% BCP for both L-929 fibroblast-like cells and MG-63 osteoblast cell. PCL- BCP composites are a promising material for application in bone scaffolds.

  • Si -substituted hydroxyapatite has been prepared to obtain biomaterials having an improved biocompatibility. From FT-IR, XRD, and ICP analyses, it was confirmed that the single-phase of hydroxyapatite substituted by Si has formed. Si- substituted hydroxyapatite of up to 2 wt% for Si keeps its original structures intact for the sintering temperatures of up to 1200$^{\circ}C$. However, it is observed that the ion substitutions by the amount higher than the above ratios for the hydroxyapatite leads to destabilize original structures of the hydroxyapatite and to produce tricalcium phosphate and calcium phosphate silicate phases when the samples were sintered at 1l00$^{\circ}C$ or higher.

  • Objectives: The purpose of the present in vitro study was to evaluate the biocompatibility of mineral trioxide aggregate (MTA) mixed with glass ionomer cement (GIC), and to compare it with that of MTA, GIC, IRM and SuperEBA. Materials and Methods: Experimental groups were divided into 3 groups such as 1 : 1, 2 : 1, and 1 : 2 groups depending on the mixing ratios of MTA powder and GIC powder. Instead of distilled water, GIC liquid was mixed with the powder. This study was carried out using MG-63 cells derived from human osteosarcoma. They were incubated for 1 day on the surfaces of disc samples and examined by scanning electron microscopy. To evaluate the cytotoxicity of test materials quantitatively, XTT assay was used. The cells were exposed to the extracts and incubated. Cell viability was recorded by measuring the optical density of each test well in reference to controls. Results: The SEM revealed that elongated, dense, and almost confluent cells were observed in the cultures of MTA mixed with GIC, MTA and GIC. On the contrary, cells on the surface of IRM or SuperEBA were round in shape. In XTT assay, cell viability of MTA mixed with GIC group was similar to that of MTA or GIC at all time points. IRM and SuperEBA showed significantly lower cell viability than other groups at all time points (p < 0.05). Conclusions: In this research MTA mixed with GIC showed similar cellular responses as MTA and GIC. It suggests that MTA mixed with GIC has good biocompatibility like MTA and GIC.

Fabrication of Porous β-TCP Bone Graft Substitutes Using PMMA Powder and their Biocompatibility Study (PMMA를 이용한 다공질 β-TCP 골충진제 제조 및 생체적합성 평가)

  • Porous ${\beta}-tricalcium$ phosphate $({\beta}-TCP)$ bioceramic was fabricated by pressureless sintering using commercial HAp and different volume percentages of PMMA powders (30-60 vol.%). The range of spherical pore size was about $200-250\;{\mu}m$ in diameter. By increasing the PMMA content, the number of pores and their morphology were dramatically changed as well as decreased the material properties. In case of using 60 vol.% PMMA content, network-type pores were found, due to the necking of the PMMA powders. The values of relative density, elastic modulus, bending strength and hardness of the 60 vol.% PMMA content sample, sintered at $1500^{\circ}C$, were about 46%, 22.2 GPa, 5MPa and 182 Hv respectively. Human osteoblast-like MG-63 cells and osteoclast-like Raw 264.7 cells were well grown and fully covered all of the porous ${\beta}-TCP$ bodies sintered at $1500^{\circ}C$.

Effect of surface-treatments on flexibility and guided bone regeneration of titanium barrier membrane

  • Titanium barrier membranes are prepared to investigate the effect of surface-treatments, such as machining, electropolishing, anodizing, and electropolishing + TiN coating, on the biocompatibility and physical properties of the membranes. The surface roughness (Ra) of the membrane decreases from machining ($0.37{\pm}0.09{\mu}m$), TiN coating ($0.22{\pm}0.09{\mu}m$), electropolishing ($0.20{\pm}0.03{\mu}m$), to anodizing ($0.15{\pm}0.03{\mu}m$). The highest ductility (24.50 %) is observed for the electropolished Ti membrane. No evidence of causing cell lysis or toxicity is found for the membranes regardless of the surface-treatments. Cell adhesion results of L-929 and MG-63 show that the machined Ti membrane exhibits the highest cell adhesion while the electropolished membrane is the best membrane for the L-929 cell proliferation after 7 days. However, no appreciable difference in MG-63 cell proliferation among variously surface-treated membranes is detected, suggesting that the electropolished Ti membrane is likely to be the best membrane due to the synergic combination of tailored flexibility and excellent fibroblast proliferation.

Bone regeneration of the fluoridated hydroxyapatite and the bio-glass in the rabbit cranium defect model (가토 두개골 결손 모델에서 Bioglass가 첨가된 불소화 수산화인회석의 골재생능력)

  • Introduction: Hydroxyapatite ($Ca_{10}(PO_4)_6(OH)_2$, HA) is the main inorganic phase of human hard tissue that is used widely as the repair material for bones. When HA is applied to a bony defect, however, it can be encapsulated with fibrous tissue and float in the implanted area due to a lack of consolidation. Bioceramics as allogenic graft materials are added to HA to improve the rate and bone healing capacity. Fluoridated hydroxyapatite ($Ca_{10}(PO_4)_6(OH,F)_2$, FHA), where F- partially replaces the OH- in hydroxyapatite, is considered a good alternative material for bone repair owing to its solubility and biocompatibility. Materials and Methods: This study was designed to determine the bone healing capacity of FHA newly produced as a nanoscale fiber in the laboratory. HA and FHA with bioglass was implanted in a rabbit cranium defect and the specimen was analysed histologically. Results: 1. At 4 weeks, fibrous connective tissue and little bone formation was observed around the materials of the experimental group I implanted HA and bioglass. Newly formed bone was observed around the materials in the experimental group II implanted FHA and bioglass. 2. At 8 weeks, the amount of newly formed and matured bone was higher in experimental group II than in experimental group I and the control group. Conclusion: These results suggest that FHA and bioglass is a relatively favorable bone substitute with biocompatibility and better bone healing capacity than pure HA and bioglass.

Biological assessment of a new ready-to-use hydraulic sealer

  • Objectives: This study compared the cytotoxicity, biocompatibility, and tenascin immunolabeling of a new ready-to-use hydraulic sealer (Bio-C Sealer) with MTA-Fillapex and white MTA-Angelus. Materials and Methods: L929 fibroblasts were cultivated and exposed to undiluted and diluted material extracts. Polyethylene tubes with or without (the control) the materials were implanted into the dorsa of rats. At 7 days and 30 days, the rats were euthanized, and the specimens were prepared for analysis; inflammation and immunolabeling were measured, and statistical analysis was performed (p < 0.05). Results: MTA-Fillapex exhibited greater cytotoxicity than the other materials at all time points (p < 0.05). The undiluted Bio-C Sealer exhibited greater cytocompatibility at 6 and 48 hours than white MTA-Angelus, with higher cell viability than in the control (p < 0.05). White MTA-Angelus displayed higher cell viability than the control at 24 hours, and the one-half dilution displayed similar results at both 6 and 48 hours (p < 0.05). At 7 days and 30 days, the groups exhibited moderate inflammation with thick fibrous capsules and mild inflammation with thin fibrous capsules, respectively (p > 0.05). At 7 days, moderate to strong immunolabeling was observed (p > 0.05). After 30 days, the control and MTA-Fillapex groups exhibited strong immunolabeling, the white MTA-Angelus group exhibited moderate immunolabeling (p > 0.05), and the Bio-C Sealer group exhibited low-to-moderate immunolabeling, differing significantly from the control (p < 0.05). Conclusions: Bio-C Sealer and white MTA-Angelus exhibited greater cytocompatibility than MTA-Fillapex; all materials displayed adequate biocompatibility and induced tenascin immunolabeling.

  • Silk polymer has been focused recently on medical applications as a novel biomaterials. The factors for biomaterials were considered and reviewed recent research on surgical suture, wound dressing, tympanic regeneration patch, guided bone regeneration membrane, bone implant and ligament. Silk polymer is good mechanical properties and biocompatibility. Therefore it might be used as one of promising materials for medical application.

  • The use of nanovesicles (NVs) has contributed to nanotechnology in the development of new concept medicine to compete with diseases of deleterious and infectious to human health. Due to their properties of size, morphology, and biocompatibility NVs have great impact on public health especially in the development of new therapeutic and prophylaxis approaches in addition to the device for biosensors to diagnose human diseases. Recent data also strongly suggest that NVs are regarded as innovative materials in developing for vaccines and diagnostic tools. In this review, we focus on the basic concepts and recent applications of NVs to utilize or engineer them as therapeutic materials.
