Effects of Tomato Extracts on Detergent-Induced Dry Skin in Rats (토마토추출물의 흰쥐 건성피부에 미치는 효과)

  • Dry skin, called xerosis as medical term, is one of the most common skin problems. Many epidemiological studies show that the consumption of foods containing lycopene plays an important role in protecting the epithelial tissue. In this study, water extracts of tomato (WET) containing lycopene were fed and applied to evaluate the effects on dry skin induced by kitchen detergent in rats. These effects were identified by protein analysis and histological changes such as inflammatory erythematic skin as well as acanthosis. The visual scoring for skin observation showed the value such as 4 indicating fiery red with edema after detergent application to skin for 3 wk. However, WET feeding and application to skin showed the decreased values, from 0.7 to 1.0. In addition, it was noteworthy that the epidermis of dry skin show apparent acanthosis with abnormally accentuated keratinization and parakeratosis. However, acanthosis was reversed by feeding and application of WET to dry skin. In order to analyze the effects of WET on dry skin induced by detergent, protein analysis was carried out. The increased amount of protein in dry skin after WET feeding and application would be suggested as one of biochemical mechanisms for recovering the damaged skin. Thus, it would be recommended that water-extracted tomato is a new ingredient in skin regeneration from dry skin induced by detergent.

  • Cellulose, chitin, chitosan and hyaluronic acid are well known as polysaccharides. These polysaccharides have many effects on cell growth and differentiation. Cell activation increases with increasing the polysaccharides concentration. In this study, gelatin scaffold containing microorganism fermented cellulose, citrus gel were prepared by using irradiation technique. Physical properties of the scaffolds were investigated as a function of the concentrations of gelatin and citrus gel and the cell attachment, cell morphology and inflammation of the scaffolds also were characterized for regeneration of skin tissue.

2-O-digalloyl-1,3,4,6-tetra-O-galloyl-β-D-glucose isolated from Galla Rhois suppresses osteoclast differentiation and function by inhibiting NF-κB signaling

  • Natural compounds isolated from medicinal herbs and plants have immense significance in maintaining bone health. Hydrolysable tannins have been shown to possess a variety of medicinal properties including antiviral, anticancer, and anti-osteoclastogenic activities. As a part of a study on the discovery of alternative agent against skeletal diseases, we isolated a hydrolysable tannin, 2-O-digalloyl-1,3,4,6-tetra-O-galloyl-${\beta}$-D-glucose (DTOGG), from Galla Rhois and examined the effect on osteoclast formation and function. We found that DTOGG significantly inhibited receptor activator of nuclear factor-${\kappa}B$ ligand (RANKL)-induced osteoclast differentiation by downregulating the expression of the key regulator in osteoclastogenesis as well as osteoclast-related genes. Analysis of RANKL/RANK signaling revealed that DTOGG impaired activation of $I{\kappa}B{\alpha}$ and p65 in the nuclear factor kappa-lightchain-enhancer of activated B cells (NF-${\kappa}B$) signaling pathway. Furthermore, DTOGG reduced bone resorbing activity of osteoclasts, compared to the vehicle-treated control. These results suggest that DTOGG could be a useful natural compound to manage osteoclast-mediated skeletal diseases.

  • To protect the plant against several soil-borne pathogens, we are currently constructing disease-resistant transgenic root stock for the growth of cucurbitaceae vegetable plants, watermelon and gourd. We made a watermelon cDNA library from Cladosporium cucumerinum-Infected leaves for substractive hybriazation and differential screening. We isolated the several pathogen inducible cDNA clones, such as caffeoyl-CoA-methyltransferase, LAA induced protein, receptor-like kinase homolog, hydroxyproline-rich glycoprotein, catalase, calmodulin binding protein, mitochondrial ATPase beta subunit, methyl tRNA synthetase and WRKY transcription factors. We previously obtained CaMADS in pepper and galactinol synthase ( CsGolS) in cucumber that were confirmed to be related with disease-resistance. CaMADS and CsGolS2 were transformed into the inbred line 'GO701-2' gourd, the inbred line '6-2-2' watermelon and the Kong-dye watermelon by Agrobacterium tumerfaciens LBA4404. Plant growth regulators (zeatin, BAP and IAA) were used for shoot regeneration and root induction for optimal condition. Putative transgenic plants were selected in medium containing 100mg/L kanamycin and integration of the CaMADS and CsGO/S2 into the genomic DNA were demonstrated by the PCR analysis. We isolated major soil-borne pathogens, such as Monosporascus cannonballus, Didymella bryoniae, Cladosporium cuvumerinum from the cultivation area of watermelon or root stock, and successfully established artificial inoculation method for each pathogen. THE EFFECT OF RESORBABLE MEMBRANE ON BONE REGENERATION IN CALVARIAL DEFECTS OF RATS (백서의 두개골 결손부에서 탈단백우골 이식 시 흡수성악의 효과)

  • Purpose : This research evaluates the effect of the use of absorbable membrane barrier with deproteinized bovine bone (Bio-$Oss^{(R)}$, Switzerland) on bone healing in surgically created critical-sized defects in rat calvaria. Materials and Methods : Two standardized transosseous circular calvarial defects (5 mm in diameter) are made in each calvarium of 30 rats. These rats are divided into negative control group(n=15), positive control group(n=15) and two experimental groups(n=15). In the negative control group, defects are only filled with blood clots. In the positive control group, defects are filled with autogenous bone obtained from calvarium; in the experimental group 1, defects are filled with deproteinized bovine bone; and in the experimental group 2, defects are filled with deproteinized bovine bone with absorbable membrane. At the postoperative 1 week, 3 weeks. and 6 weeks, clinical. histologic and histomorphometric evaluations of the defects are performed. Results : 1. The grafted bone without membrane in the calvarial bone defect was scattered but, the grafted bone with membrane was stable. 2. $BioMesh^{(R)}$ membrane was absorbed beginning at 3 weeks, and was absorbed considerably at 6 weeks while maintaining the structural form of the membrane. 3. The use of membrane blocked soft tissue invasion. 4. In histomorphometric analysis. it showed the greatest amount of new bone formation in the positive control group. The amount of new bone formation was greater in the experimental group 2 than experimental group 1. At 6 weeks. the amount of new bone formation was greater in the positive control group than experimental group l(p<0.005). Conclusion : These results suggest that membrane increase the stability of grafted bone and protects from soft tissue invasion, and the use of the membrane may promote new bone formation in deproteinized bovine bone graft area.

Effects of Different Natural Extracts and Plant Growth Regulators on Plant Regeneration and Callus Induction from Pseudobulbs Explants through in vitro Seed Germination of Endangered Orchid Bulbophyllum auricomum Lindl.   • Bulbophyllum auricomum Lindl. is a rare orchid and has flowers with an attractive fragrance. The present study investigated the tissue culture method for micropropagation. Capsules derived from artificial self-pollination were obtained for the best seed germination in MS basal medium. Plant growth regulators (1.0 mg·L-1 of BAP and 2.0 mg·L-1 of NAA) were affected by callus induction from subcultured pseudobulb explants. For the callus subculture, different natural plant extracts were tested in 11 treatment media. Among them, MS medium with 150 mL·L-1 of coconut water was generally effective in fresh weight (1.75 ± 0.08) and (3.01 ± 0.20) of callus proliferation and PLBs induction at 1 and 2 months, respectively, followed by an MS combination of 30 g·L-1 of banana and 20 g·L-1 of potato extract. The results of a comparative study of different MS mediums containing plant growth regulators with a natural extract combination and MS medium supplemented with natural extract only showed that MS medium supplemented with a combination of natural extracts (150 mL·L-1 of coconut water) and plant growth regulators (2.0 mg·L-1 of BAP and 1.0 mg·L-1 of NAA) obtained the highest shoot regeneration (3.37 ± 0.17) and (6.41 ± 0.68) after 1 month and 2 months of culturing, respectively.

Analysis of Satisfaction on Alley Garden's Components through Urban Regeneration - Focused on Bisan 2·3-dong in Daegu Metropolitan City - (도시재생사업에 따른 골목정원 구성요소의 만족도 분석 - 대구광역시 비산 2·3동을 대상으로 -)

  • This study analyzed the opinions of residents for desirable urban regeneration and suggested an improvement plan for alley environments. This study conducted a questionnaire survey of the residents living in Alley Garden of Bisan 2,3-dong, Daegu Metropolitan City. In the analysis of the importance and satisfaction of Alley Garden components, items related to a safe, cleanly environment such as 'Lighting facility installation', 'Sewage and waste disposal' and 'CCTV installation' had a high level of importance. It was also found that items improved by the Residential Environment Improvement Project and Alley Garden such as 'Lighting facility installation', 'Quantity of herbaceous flowers' and 'Kinds of herbaceous flowers' had a high level of satisfaction. The IPA results showed that items such as 'Empty house maintenance', 'Rest facilities such as benches and pergolas', 'Space for resident interaction' and 'Public parking lot' had a high level of importance, but had a low level of satisfaction, which suggests that they should be improved by priority. As a result of factor analysis, Alley Garden components were classified into four factors: 'Safety and cleanliness', 'Greenness', 'Exchange and convenience facility' and 'Aesthetics renewal'. Based on this classification, a regression analysis was conducted regarding the effects of the four factors on overall satisfaction. Results showed that all four factors had a significant influence on the overall satisfaction and that 'Aesthetics renewal' and 'Safety and cleanliness', respectively showing levels of significance at 0.274 and 0.235, were highly influential to overall satisfaction. Therefore, it was concluded that spaces for resident interaction and rest facilities should be preferentially installed to improve the environment of alleys. For the improvement of the overall satisfaction of alley environments, it was also concluded that residents should be encouraged to engage in activities such as sculpture installation and mural drawing, along with the introduction of safety bells and crime prevention environment design and the implementation of alley beautification projects.

  • Purpose: Choukroun's platelet-rich-fibrin (PRF) is composed of platelets, white blood cells and fibrin matrix. It does not induce enough bone formation by itself but it can improve bone formation with calcium. Silk fibroin does not cause inflammatory reactions because it is bio-compatible and degradable. The purpose of this study was to exam the bone formation when a combination of Choukroun's PRF and silk fibroin was used. Methods: In this study, cell reactions to silk powder with differing molecular weights was first tested to select the appropriate silk powder. Then we applied these bone graft materials on defects of skull and in a peri-implant bony defect model in New Zealand rabbits. The results between the experimental and control s (non-grafted) group were analyzed. Results: The small sized silk fibroin powder showed increased cellular proliferation for bone-regeneration. There was no statistically significant difference between the experimental group and the control group at 6 weeks, but more new bone formation was observed in the combination graft group at 12 weeks (P<0.05). And in the dental implant model, the combination bone graft group showed much improved torque test results, which was statistically significant. Histomorphometric analysis showed more regenerated cortical bone and a higher mean bone to implant in the experimental group. Both were statistically significant. Conclusion: The combination graft of Choukroun's platelet-rich-fibrin (PRF) and silk fibroin powder can successfully restore the bony defects in a skull defected model and a peri-implant bony defects model.

Adhesion Behavior of Chondrocyte and Osteoblast on Surface-Modified Biodegradable PLLA Films and Scaffolds (표면개질된 생분해성 PLLA 필름 및 지지체의 연골세포와 조골세포 점착거동)

  • Surface-modified poly(L-lactic acid) (PLLA) films and scaffolds were treated with plasma discharge in oxygen gas and subsequently subjected to $in$ $situ$ grafting of acrylic acid (AA) in order to increase the cell compatibility. The surface of AA-grafted PLLA was converted to hydroxyapatite (HA)-deposited PLLA in stimulated body fluid (SBF). After the samples were immersed in phosphate-buffered saline (PBS), fetal bovine serum (FBS), normal saline, or cell medium, the water contact angles were significantly reduced on the surface of HA-deposited PLLA. Chondrocyte and osteoblast showed a higher attachment and cell proliferation on HA-deposited surfaces and in particular, it was confirmed that chondrocyte was considerably influenced by HA. However, osteoblast showed better cell proliferation on the surfaces immersed in FBS, cell medium or HA-deposited surface. In addition, the cell proliferation in 3D scaffolds was much higher than that on film type, irrespective of chondrocyte and osteoblast. Therefore, such surface-modified PLLAs are expected to be useful as organic-inorganic hybrid scaffolds in the regeneration of cartilage and bone.

Effect of Electron-beam Irradiaton on the Artificial Bone Substitutes Composed of Hydroxyapatite and Tricalcium Phosphate Mixtures with Type I Collagen (수산화인회석과 인산삼칼슘 및 1형 콜라젠 혼합골의 전자빔 조사 효과)

  • Purpose: The aim of this study is to evaluate the effect and potential of electron beam (E-beam) irradiation treatment to the synthetic bony mixtures composed of hydroxyapatite (HA; Bongros$^{(R)}$, Bio@ Co., Korea) and tricalcium phosphate (${\beta}$-TCP, Sigma-Aldrich Co., USA), mixed at various ratios and of type I collagen (Rat tail, BD Biosciences Co., Sweden) as an organic matrix. Methods: We used 1.0~2.0 MeV linear accelerator and 2.0 MeV superconductive linear accelerator (power 100 KW, pressure 115 kPa, temperature $-30{\sim}120^{\circ}C$, sensor sensitivity 0.1~1.2 mV/kPa, generating power sensitivity 44.75 mV/kPa, supply voltage $5{\pm}0.25$ V) with different irradiation dose, such as 1, 30 and 60 kGy. Structural changes in this synthetic bone material were studied in vitro, by scanning electron microscopy (SEM), elementary analysis and field emission scanning electron microscope (FE-SEM), attenuated total reflection (ATR), and electron spectroscopy for chemical analysis (ESCA). Results: The large particular size of HA was changed after E-beam irradiation, to which small particle of TCP was engaged with organic collagen components in SEM findings. Conclusion: The important new in vitro data to be applicable as the substitutes of artificial bone materials in dental and medical fields will be able to be summarized.
