Disposable Electrochemical Immunosensors for the Detection of Herbicide (제초제 검출을 위한 전기화학적 일회용 면역센서)

  • A disposable electrochemical immunosensor system has been developed for the detection of herbicide in aqueous samples. Disposable screen printed carbon electrodes(SPCE) were used as basic electrodes and an enzyme, horseradish peroxidase (HRP), and anti-herbicide antibodies was immobilised on to the working electrode of SPCE by using avidin-biotin coupling reactions. An herbicide-glucose oxidase conjugates have been used for the competitive immunoreaction with sample herbicides. The enzymatic reaction between the conjugated glucose oxidase and glucose added generates hydrogen peroxide, which was reduced by the peroxidase immobilised. The latter process caused an electrical current change, due to direct re-reduction of peroxidase by a direct electron transfer mechanism, which was measured to determine the herbicides in the sample. The optimal operational condition was found to be: $20\;{\mu}gl-1$ deglycosylated avidin loading to the working electrode and working potential +50 mV vs. Ag/AgCl. The total assay time was 15 min after sample addition. The detection limits for herbicides, atrazine and simazine, were found to be 3 ppb and 10 ppb, respectively.

  • The objective of this study is to the integrated evaluation of the present condition and satisfaction of visitors of rooftop greening area using quantitative and qualitative method. The thirteen green roofs were selected to investigate environmental variables as like building structure, soil, water, atmosphere and climate environment and questionnaire survey for investigate the satisfaction of visitors was conducted. The results of this study are as follows : remove of the hazardous inflow of species, install rain water recycling facility, install outdoor unit of air condition where not to affect plants in green roof, install safety facilities in the case of making resting place, plant variable vegetation and so on. The result of questionnaire survey are that visitors want to make more comfortable resting space. The more bigger the resting space and privacy, the more satisfaction of visitor became high. In conclusion, green roofs implementation should be considered reduction of temperature and delay of runoff, insulation effect and ecological restoration as well as rooftop greening is focused on the development of resting space nowadays.

The Effect of Mixed Amino Acids on Nitrate Uptake and Nitrate Assimilation in Leafy Radish

  • The objective of the present work was to determine the corresponding uptake and assimilation of ${NO_3}^-$ in roots and shoots of leafy radish by applying of mixed amino acids (MAA). The amino acids used in this experiment were alanine (Ala), ${\beta}-alanine\;({\beta}-Ala)$, aspartic acid (Asp), asparagines (Asn), glutamic acid (Glu), glutamine (Gln), and glycine (Gly). Leafy radish was grown by conventional fertilization with macro- and micronutrients under controlled conditions. The 15-day-old seedlings were treated 0, 0.3 and 3.0 mM of MAA containing 5 mM ${NO_3}^-$ in growth medium. Nitrate uptake was determined by following ${NO_3}^-$ depletion from the uptake solution. The activity of the enzymes related to the process of ${NO_3}^-$ reduction (NR: nitrate reductase; NiR: nitrite reductase; GS: glutamine synthetase) and the content of ${NO_2}^-\;and\;{ND_3}^-$ were analyzed in shoots and roots. The results of this study showed that ${NO_3}^-$ uptake was inhibited 38% with treatment of 0.3 mM of MAA. However, there was more than three times increase of N03- uptake in 3.0 mM MAA. In addition, the enzymatic activities were positively affected by the high MAA rate. Finally, the ${NO_3}^-$ content was increased slightly both in shoots and roots of leafy radish by MAA treatments.

Development of Saccharomyces cerevisiae Reductase YOL151W Mutants Suitable for Chiral Alcohol Synthesis Using an NADH Cofactor Regeneration System

  • The aldo-keto reductases catalyze reduction reactions using various aliphatic and aromatic aldehydes/ketones. Most reductases require NADPH exclusively as their cofactors. However, NADPH is much more expensive and unstable than NADH. In this study, we attempted to change the five amino acid residues that interact with the 2'-phosphate group of the adenosine ribose of NADPH. These residues were selected based on a docking model of the YOL151W reductase and were substituted with other amino acids to develop NADH-utilizing enzymes. Ten mutants were constructed by site-directed mutagenesis and expressed in Escherichia coli. Among them, four mutants showed higher reductase activities than wild-type when using the NADH cofactor. Analysis of the kinetic parameters for the wild type and mutants indicated that the $k_{cat}/K_{m}$ value of the Asn9Glu mutant toward NADH increased 3-fold. A docking model was used to show that the carboxyl group of Glu 9 of the mutant formed an additional hydrogen bond with the 2'-hydroxyl group of adenosine ribose. The Asn9Glu mutant was able to produce (R)-ethyl-4-chloro-3-hydroxyl butanoate rapidly when using the NADH regeneration system.

Simple Preparation of Diaphorase/Polysiloxane Viologen Polymer Modified Electrode for Sensing NAD and NADH

  • Nicotinamide adenine dinucleotide, $NAD^+$, and its reduced form, NADH, play important roles as coenzymes in many enzymatic reactions. Electrochemical methods for $NAD^+$ or NADH detection or generation are drawn attention because it can provide the simple and low cost platform with fairly good sensitivity. In this study, the polysiloxane viologen polymer/diaphorase/hydrophilic polyurethane (PSV/DI/HPU) modified electrodes were simply prepared and demonstrated for bio-electrocatalytic $NAD^+$ sensors. The electrodes were co-immobilized with diaphorase and polysiloxane viologen polymer as an electron mediator followed by the overcoating with HPU membrane. The mixture of the enzyme and the electron mediator was well stabilized within HPU membrane and exhibited good reversibility and stability. The sensitivity was 0.2 $nA{\cdot}{\mu}M^{-1}$ and the detection limit was 28 ${\mu}M$ with a response time of 50 s ($t_{90%}$). The capability for NADH sensor was also observed on the PSV/DI/HPU electrode.

Inhibition of Hydrogen Formation with Calcium Hydroxide on Zinc Electrode of Film-type Manganese Battery

  • A manganese dioxide (MnO2) layer and zinc (Zn) layer are used as the cathode and the anode to develop filmtype manganese battery, in which a stack of a MnO2 layer, gel electrolyte, and Zn layer are sandwiched between two plastic layers. This paper describes the chemical equation of swelling control upon the film-type manganese battery. We examined the reduction of hydrogen formation, by using calcium hydroxide Ca(OH)2 as an additive in the electrolyte of film-type manganese battery. The phenomena or an effect of reduced hydrogen gas was proven by cyclic voltammogram, X-ray photoelectron spectra (XPS), and volume of hydrogen formation. The amount of H2 gas generation in the presence of Ca2+ ion was reduced from 4.81 to 4.15 cc/g-zinc (14%), and the corrosion of zinc electrode in the electrolyte was strongly inhibited as time passed.

  • Although the demand for histopathological examinations has been increasing, medical accidents in management of specimens also have been increasing because most of the examinations are processed manually which can cause careless handing, confusing information and mismatching during the procedure. In the future, histopatological examination will be used frequently for handing incurable diseases and verification of new drug. Thus, efficient and error-free management system for handling personalized medical history and test results is infallibly necessary. In this paper, I have proposed an integrated printing system for informatization of histopathological examination that support the u-Healthcare environment based on RFID in near future. The proposed system supports systematization of whole examination process and information of pathological samples. This system will contribute to reduction of costs, improvement of operational efficiency, and mostly fundamental prevention of medical accidents.

A Study on Characteristic Factors of Tubular Membranes in a Membrane coupled Activated Sludge Process (Tubular형 막을 이용한 활성슬러지법에 있어서 여과특성에 미치는 인자에 관한 연구)

  • We elucidated factors such as membrane pore size, activity of the activated sludge and MLSS. affecting filtering characteristics or filtering resistance in an activated sludge bio-process using tubular type membranes. We used a commercial membrane module with $0.5m^2$ of effective membrane area and a bundle of 7 tubes(13mm of ID and 2m long) in the experiments. As a result, the flux was maintained in the range of $35{\sim}105l/m^2$ hr at 5,000~20,000mg/l of MLSS concentration, and there seemed to be a correlation between the flux and the MLSS. Also the greater the pores were, the higher the flux was in some comparison experiments using UF and MF membranes, but the water permeated through both of the membranes showed almost the same quality. However, the MF membrane showed a great reduction in filtering resistance initially, but went back to a stable stage afterwards. The activity of the activated sludge in the aeration tank was also found to be considered as a factor showing permeation characteristics of the membrane.

Towards a Strategic Approaches in Alternative Tests for Pesticide Safety

  • Pesticides have provided significant benefits including plant disease control and increased crop yields since people developed and utilized them. However, pesticide use is associated with many adverse effects, which necessitate precise toxicological tests and risk assessment. Most of these methods are based on animal studies, but considerations of animal welfare and ethics require the development of alternative methods for the evaluation of pesticide toxicity. Although the usage of laboratory animals is inevitable in scientific evaluation and alternative approaches have limitations in the whole coverage, continuous effort is necessary to minimize animal use and to develop reliable alternative tests for pesticide evaluation. This review discusses alternative approaches for pesticide toxicity tests and hazard evaluation that have been used in peer-reviewed reports and could be applied in future studies based on the critical animal research principles of reduction, replacement, and refinement.

Antimicrobial Fiber Products Treated with Silica Hybrid Ag Nanoparticles

  • Silica hybrid silver nanoparticles showing the strong antimicrobial activity, in which nano-silver is bound to silica molecules, has been synthesized using ${\gamma}-irradiation$ at room temperature. The present study relates to an antimicrobial composition for coating fiber products comprising silica hybrid silver nanoparticles. In this study, we describe antimicrobial fiber products coated with the silica hybrid silver nanoparticles and a method of antimicrobially treating fiber products by coating the fiber products with the silica hybrid silver nanoparticles. The antimicrobial fiber products exhibited excellent antimicrobial effects. In detailed practice, when the present composition comprising nanosized silica-silver was applied to a cloth (fabric) in a concentration of $6.4mg\;yard^{-1}$, the viable cell number decreased to less than 10 cells before and after laundering, resulting in a reduction of 99.9% or greater in the viable cell number. The present composition displays long-lasting potent disinfecting effects on bacteria. Also, we investigated the toxicity of silica hybrid silver nanoparticles in rats. The skin of rats was treated with a 30 ppm nanoparticles solution ($2ml\;Kg^{-1}$) for 8 days. No toxicity was detected in the treatment. These results suggest that the fiber products coated with the silica hybrid silver nanoparticles can be used to inhibit the growth of various microorganisms.
