흰쥐 조직에서의 카드뮴 결합 고분자량 단백질 [Cd-BP(II)]의 유도 (Induction of Cd-binding High Molecular Weight Protein [Cd-BP(II)] in Rat Tissues)

  • The study was carried out on the biochemical characters of Cd-BP(II) after isolation and purification of the protein from the liver of rat ip injection with Cd. A continued study has been doing whether Cd-BP(II) could be induced by Cd or by the other metals such as Zn and Cu. Antisera were made against the antigen of Cd-BP(II) from New Zealand white rabbits. We carried out g-globulin purification, then Ouchterlony test and gel immunodiffusion test. Cd-BP(II) was also found in normal tissues of rat. It was induced up to a considerable level by Cd, whose induced level was higher than that Cu or Zn treatment. The level of induction by Cu or Zn pretreatment plus Cd treatment was lower than that by single treatment of Cu or Zn. Such a result was presumably related to the Cd toxicity.

A Major DNA Marker Mining of BMS941 Microsatellite Locus in Hanwoo Chromosome 17

  • We describe tests for detecting and locating quantitative traits loci (QTL) for traits in Hanwoo. Lod scores and a permutation test have been described. From results of a permutation test to detect QTL, we select major DNA markers of BMS941 microsatellite locus in Hanwoo chromosome 17 for further analysis. K-means clustering analysis applied to four traits and eight DNA markers in BMS941 resulted in three cluster groups. We conclude that the major DNA markers of BMS941 microsatellite locus in Hanwoo chromosome 17 are markers 80bp, 85bp 90bp and 105bp.

  • Benzo(a)pyrene(BP)의 monooxygenase(AHH)에 의해서 생성된 반응성 대사물질들의 in vitro DNA와의 결합 및 BP 대사에 관여하는 효소들의 활성도에 미치는 인삼 물추출물의 영향을 조사하였으며, DNA-BP metabolite adduct들은 Sephadex LH-20 column으로 chromatography하여 5개의 major peak 들을 얻었다. 이 peak 들을 극성이 큰 순서대로 A부터 E까지 임의로 정하고 5개의 peak들을 7,8-diol-9,10-oxide(A), 7,8-oxide(B). 4,5-oxide(C), 9-HO-BP(D & E) adduct들로 잠정적으로 확인하였다. Peak A, C, D 그리고 E는 각각 대조군의 30, 15, 20 그리고 30%로 감소되었으며 peak B는 의미있는 변화를 보이지 않았다. DNA-BP 결합 억제와 관련하여 in vitro와 in vivo 투여시의 경향이 유사하여 EH의 활성도만 BP투여 대조군보다 38%정도 의미있게 유도되었다.

MC1R gene의 PCR-RFLP를 이용한 한우.젖소고기 감별 (Analysis of Melanocortin receptor 1 (MC1R) gene differential test for beef species between Hanwoo and Holstein using polmerase chain reaction -restriction fragment length polymorphism (PCR-RFLP))

  • The objective of this study was to differentiate the beef species between Hanwoo and Holstein from a total of 1,081 beef samples using PCR-RFLP of MC1R gene. When a PCR product of 403 bp specific band amplified from bovine MC1R gene sequence was digested with restriction enzyme MspA1I, Hanwoo type showed 2 bands, 220 bp and 183 bp size bands. Holstein type, however, showed three bands, 220 bp, 138 bp and 45 bp size band, respectively. The results of the differential test for beef species were as following; 7 samples (0.64%) were determined to Holstein type, of which 4 were submitted from administrative authorities, other 3 from self-collection planing, and none from civilian clients including school.

  • 스택 체결압에 의해 접촉되는 분리판(BP)과 탄소펠트전극(CFE) 사이의 전기적 접촉저항은 상대적으로 낮은 바나듐 레독스 흐름전지(VRFB) 스택의 체결압 때문에 스택 효율에 큰 여향을 미친다. 본 연구에서는 이러한 접촉저항을 줄이고 셀 성능을 향상시키기 위해 국부 가열 접합 공정을 통해 폴리에틸렌(PE) 복합재료-CFE 하이브리드 BP 구조를 개발하였다. 탄소섬유 복합재료 BP의 PE 매트릭스를 국부적으로 녹여 CFE의 탄소 섬유와 BP의 탄소 섬유의 직접 접촉 구조를 만들어 전기 접촉 저항을 감소시겼다. PE 복합재료-CFE 하이브리드 BP의 성능을 평가하기 위해 면적비저항(ASR)과 기체투과도를 측정하였다. 또한 스택 신뢰성을 측정하기 위해 내산성 시험을 수행하였다. 최종적으로, 개발된 PE 복합재료-CFE 하이브리드 BP와 기존의 BP의 성능을 비교 분석하기 위하여 VFRB 단위셀 충/방전 시험을 수행하였다.

  • We selected, in previous research, a major DNA Marker 235bp of ILSTS035 microsatellite locus in progeny test Hanwoo chromosome 6. We apply a major DNA Marker 235bp to perormance valuation Hanwoo chomosome 6. We use bootstrap BCa method and calculate confidence interval. A major DNA Marker 235bp is verified that it does not have environmental effect but affects primely economic trait factor.

치어기 조피볼락 Sebastes schlegeli 사료내 생균제 첨가효과 (Effects of Dietary Probiotics Supplementation on Juvenile Korean Rockfish Sebastes schlegeli)

  • 본 연구는 치어기 조피볼락 Sebastes schlegeli 사료내 생균제 첨가가 넙치의 성장 및 면역반응에 미치는 영향을 평가하였다. 실험사료는 주 단백질원으로 북양어분(white fish meal), 콘글루텐밀(corn gluten meal), 탈피대두박(dehulled soybean meal)을 사용하였으며, 탄수화물원으로는 밀가루(wheat flour)를, 지질원으로는 고도불포화지방산(n-3 HUFA)이 다량 함유된 오징어간유(squid liver oil)를 사용하였다. 생균제의 첨가효과를 확인하기 위하여 B. polyfermenticus(BP), Bacillus licheniformis(BL) 및 복합종균(B. polyfermenticus+Saccharomyces cerevisiae; BP+SC)을 실험사료 내에 각각 $1.0{\times}10^7$ CFU/kg diet 수준으로 첨가하였다. 2주간의 예비사육 후, 평균무게 $12.0{\pm}0.1g(mean{\pm}SD)$인 조피볼락을 500 L 원형수조에 각 실험구 당 각각 20마리씩 3반복으로 무작위로 배치하였고, 실험사료는 1일 2회 어체중의 $1.7{\pm}0.6%$(오전 10시, 오후 4시)씩 12주간 공급 하였다. 증체율, 사료효율, 일간성장율 및 단백질 전환효율에 있어서 BP+SC를 공급한 실험구가 대조구(control)에 비하여 유의하게 높게 나타났지만(P<0.05), BP, BL 및 BP+SC를 공급한 실험구간에는 유의한 차이가 나타나지 않았다. 생존율에 있어서는 모든 실험구간에 있어 유의한 차이가 나타나지 않았다. 전어체 지방함량에 있어서 대조구가 다른 모든 실험구에 비해 높게 나타났으며, 전어체 단백질, 회분 및 수분함량에 있어서는 모든실험구간에 유의한 차이가 없었다. 혈액분석에 있어서 혈장내 glucose의 함량은 BP+SC를 공급한 실험구가 대조구에 비해 유의적으로 낮게 나타났으나, BP, BL 및 BP+SC를 공급한 실험구간에는 유의적인 차이가 나타나지 않았다. Respiratory burst activity(NBT assay)에 있어서 BP+SC와 BL을 공급한 실험구가 대조구에 비해 유의적으로 낮게 나타났으며, BP, BL 및 BP+SC를 공급한 실험구간에는 유의적인 차이가 나타나지 않았다. 혈청의 lysozyme 활성에 있어서는 BP와 BL을 공급한 실험구가 대조구에 비해 높게 나타났으며, BP, BL 및 BP+SC를 공급한 실험구간에는 유의적인 차이가 나타나지 않았다. 공격실험 결과, 폐사는 Edwardsiella tarda를 접종한지 1일 후부터 시작하여 11일째 종료 하였다. 생균제를 투여한 모든 실험구가 대조구에 비해 초기폐사율이 낮게 나타났다. 공격실험 후 8일째부터 종료시 까지는 BP+SC를 공급한 실험구가 대조구에 비해 유의적으로 낮게 나타났으며, BP, BL및 BP+SC를 공급한 실험구간에는 유의한 차이가 나타나지 않았다. 상기 결과를 토대로, 조피볼락 사료내 B. polyfermenticus, B. licheniformis 및 B. polyfermenticus 와 S. cerevisiae의 혼합첨가는 조피볼락의 성장 및 사료효율 증진과 항산화능 및 특정질병저항성에 좋은 효과를 나타낼 수 있을 것으로 사료된다.

Comparison Task-Oriented Training according to the Applicable Blocked Practice and Random Practice: Chronic Hemiplegic Patients

  • Purpose: The purpose of this study was to compare the blocked practice and random practice of task-oriented training in patients with chronic stroke to determine the effect of lower extremity muscle activity and balance ability. Methods: The thirty participants were randomly assigned to either the block practice group (BP) group or the random practice group (RP) and received the training three times per week, 30 minutes per day, for six weeks. Surface electromyography was used for measurement of lower extremity muscle activity. Static balance was to measured the stability index (SI) and weight distribution index (WDI) using the Tetrax. The four square step test (FSST) was used to measure dynamic balance. The paired t-test was used for determination of differences before and after intervention, and the independent t-test was used for determination of differences between groups. Results: Lower extremity muscle activity, RA and GCM was improved in the RP group after intervention and between groups. TA was significantly improved in the RP group compared with the BP group. In comparison of before and after interventions, SI was reduced in BP and RP. WDI in OS was reduced in comparison of BP and RP before and after intervention. CS was reduced in BP and RP. The OS and CS was improved in RP compared with BP. In comparison of before and after intervention, FSST was improved in BP and RP. Conclusion: Task-oriented training methods using random practice was found to be effective in promoting lower extremity muscle activity and balance ability in chronic stroke patients.
