A Linkage Study of Chromosome 5 D5S39(p105-153Ra), D5S76(p105-599Ha), and $D_2$, $D_3$ Receptor Gene($DRD_2$, $DRD_3$) in Schizophrenics in Korean Pedigree (한국인 정신분열병 가계의 염색체 5번 D5S39(p105-153Ra), D5S76(p105-599Ha) 및 도파민 $D_2$, $D_3$ 수용체 유전자좌간의 연관관계 연구)

  • The author examined the relationship of two markers, D5S39(p105-153Ra), D5S76(p105-599Ha) of chromosome 5 and $D_2$, $D_3$ receptor genes in a Korean schizophrenic pedigree using polymerase chain reaction(PCR). The results were as follows : 1) On D5S39 locus, 5 different alleles(224/226 bp : 4 cases, 218/226 bp : 3 cases, 222/226 bp : 3 cases, 218/230 bp : 1 case, 222/224 bp : 1 case) were produced. 2) On D5S76 locus, 5 different alleles(102/112 bp : 4 cases, 94/112 bp : 3 cases, 108/112 bp 3 cases, 94/94 bp : 1 case, 102/108 bp 1 case) were produced. 3) On $D_2$ receptor gene, 3 different alleles($A_1A_2$ : 8 cases, $A_1A_1$ : 2 cases, $A_2A_2$ : cases) were produced. 4) On $D_3$ receptor gene, 2 different alleles(1/2 : 7 cases, 1/1 : 5 cases) were produced. The author had not find any specific alleles on all four loci in all pedigree nor any specific alleles in the schizophrenic patients. Though the author has not found absolute relationship between the four loci and the onset of schizophrenia, there still remains the possibilities if the more detailed and elaborated pedigree studies are done.

Genetic Variation of Cytochrome P450 Genes in Garlic Cultivars (마늘유래 Cytochrome P450 유전자의 변이 분석)

  • Wound inducible P450-Esg cDNA, one of cytochrome P450 gene family, was isolated from shoot of Euiseong garlic cultivar. P450-Esg cDNA possesses highly conserved heme-binding domain in the nucleotide sequence, and 1,419 bp of open reading frame (ORF) coding of 473 amino acids. Based on the nucleotide sequence analysis of P450-Esg homologous from twelve garlic cultivars, two domains, one domain between 472 to 510 bp, and the other between 1,210 to 1,249 bp from start codon (ATG), showed various nucleotide polymorphism among cultivars. Sequence of heme-binding domain in P450-Esg homologous, which is located at the domain between 1,210 to 1,240 bp from start codon, showed various nucleotide polymorphism as well as amino acid sequence polymorphism among twelve garlic cultivars. Anther domain, between 472 to 510 bp from start codon, showed exactly same amino acid sequence in the twelve garlic cultivars, but there were various single nucleotide polymorphism to the cultivars.

Nucleotide Sequence and Secondary Structure of 16S rRNA from Sphingomonas chungbukensis DJ77 (Sphingomonas chungbukensis DJ77의 16S rRNA 염기서열과 이차구조)

  • A 16S ribosomal RNA gene from S. chungbukensis DJ77 has been sequenced. This sequence had a length of 1,502 bp and was extended for 29 bp at 5' and for 37 bp at 3' from the partial sequence (1,435 bp) registered in 2000 year. Besides, 1 bp was newly added near to the 3' end. We made the secondary structure of the 16S rRNA based on E. coli model and found four specific regions. We found constant and variable regions in genus Sphingomonas as the result of multiple alignment of 16S rRNA gene sequences from Sphingomonas spp. and S. chungbukensis DJ77. We found a stem loop structure in S. chungbukensis DJ77, which was only discovered in C. jejuni to date. It showed the structural agreement despite the difference of the sequences from the both organisms. Finally, S. chungbukensis DJ77 belonged to cluster II (Sphingobium) group, after the classification using phylogenetic analysis and nucleotide signature analysis.

PEP-1-FK506BP12 inhibits matrix metalloproteinase expression in human articular chondrocytes and in a mouse carrageenan-induced arthritis model

  • The 12 kDa FK506-binding protein (FK506BP12), an immunosuppressor, modulates T cell activation via calcineurin inhibition. In this study, we investigated the ability of PEP-1-FK506BP12, consisting of FK506BP12 fused to the protein transduction domain PEP-1 peptide, to suppress catabolic responses in primary human chondrocytes and in a mouse carrageenan-induced paw arthritis model. Western blotting and immunofluorescence analysis showed that PEP-1-FK506BP12 efficiently penetrated chondrocytes and cartilage explants. In interleukin-1β (IL-1β)-treated chondrocytes, PEP-1-FK506BP12 significantly suppressed the expression of catabolic enzymes, including matrix metalloproteinases (MMPs)-1, -3, and -13 in addition to cyclooxygenase-2, at both the mRNA and protein levels, whereas FK506BP12 alone did not. In addition, PEP-1-FK506BP12 decreased IL-1β-induced phosphorylation of the mitogen-activated protein kinase (MAPK) complex (p38, JNK, and ERK) and the inhibitor kappa B alpha. In the mouse model of carrageenan-induced paw arthritis, PEP-1-FK506BP12 suppressed both carrageenan-induced MMP-13 production and paw inflammation. Analysis of Red Pepper (Capsicum annuum) Genome (고추의 게놈 분석)

  • The genome of red pepper was investigated by thermal denaturation, reassociation kinetics and measurement of nuclear volume for its base composition, spectrum of kinetic components and genome size. Base composition was estimated to be 37% (G+C) based on melting temperature. The reassociation of 300 nt fragments analyzed by hydroxyapatite chromatography revealed the presence of three kinetic components differing in fraction of genome, kinetic complexity and number of copies as follows; 4.8% (fast) with $5.6{\times}10^{4}\;bp$ and 10,754, 26% (intermediate) with $1.9{\times}10^{6}\;bp$ and 177, and 65% (slow) with $8.48{\times}10^{8}\;bp$ and 1. These measurements demonstrate that the genome of red pepper has a 1C DNA content of $1.25{\times}10^{9}\;bp$, which is about 33% of $4.05{\times}10^{9}\;bp$ calculated from nuclear volume of $62.4\;\mu\textrm{m}^3/1C$..

Quality Characteristics of Sulgidduk Added by Different Amount of Bamboo Leaf Flour (대잎 분말 첨가량을 달리한 설기떡의 품질 특성)

  • The purpose of this study is analyze the sensory and mechanical characteristics, moisture content and color values of bamboo leaf flour sulgidduk added by different amount of bamboo leaf flour; 0%(BP-0 group), 1%(BP-1 group), 2%(BP-2 group), 3%(BP-3 group), 4%(BP-4 group). As a result of analyzed the texture of sulgidduk bamboo leaf flour, its hardness, adhesiveness, gumminess, chewiness and cohesiveness increased while except for the springiness wasn't significantly differences. The moisture content of sulgidduk with bamboo leaf flour ranged from 37% to 39%. L-value of the color decreased with increasing bamboo leaf flour, but a-value and b-value of the color increased with increasing bamboo leaf flour. According to the sensory evaluation BF3 had the highest in color and moistness. Based on results, it is considered that the most desirable ratio of bamboo leaf flour was BF3. The results of the study support the benefits of bamboo leaf flour supplementation in sulgidduk in the aspects of taste and functionality. The degree of further increase of sulgidduk with bamboo leaf flour without causing the adverse quality effects remains for study.

  • For the classification of B. burgdorferi sensu lato strains, PCR-restriction fragment length polymorphism (RFLP) analysis was performed. PCR was carried out with B. burgdorferi sensu lato specific primer set (BB uni set), and amplicons of 470-bp DNA were digested with Alu 1. The Alu I restriction polymorphism of the amplicons provided a useful tool for identifying B. burgdorferi sensu late strains. Both amplicons from B. burgdorferi sensu stricto and B. garinii except HPI strain showed identical RFLP pattern (50 bp, 70 bp, and 150 bp), but amplicons from B. afzelii and B. garinii showed two types of subgroups, respectively. The result of PCR-RFLP using extracted DNAs from ticks was similar to those patterns of B. burgdorferi species including B. afzelii.

  • Mac-2BP is a ligand of the galectin family that has been suggested to affect tumor proliferation and metastasis formation. We assessed Mac-2BP expression at the transcriptional and translational levels to evaluate nerve growth factor (NGF)-induced Mac-2BP expression. A time kinetic analysis using reverse transcription-polymerase chain reaction showed that NGF-induced Mac-2BP transcript levels were 4-5 times higher than in controls. Mac-2BP enzyme-linked immunosorbent assay and immuno-fluorescence staining showed a 2-3-fold increase in intracellular and secreted Mac-2BP as a result of NGF stimulation. This increase was regulated by Akt activation and NF-${\kappa}B$ binding. p65 and p50-NF-${\kappa}B$ are major transcriptional factors in the Mac-2BP promoter region, and were shown to be regulated in accordance with the Akt activation states. Collectively, these results suggest that NGF induces Mac-2BP expression via the PI3K/Akt/NF-${\kappa}B$ pathway.

  • This paper is study on real-time hand gesture recognition system based on vision for intelligent robot control. We are proposed a recognition system using PCA and BP algorithm. Recognition of hand gestures consists of two steps which are preprocessing step using PCA algorithm and classification step using BP algorithm. The PCA algorithm is a technique used to reduce multidimensional data sets to lower dimensions for effective analysis. In our simulation, the PCA is applied to calculate feature projection vectors for the image of a given hand. The BP algorithm is capable of doing parallel distributed processing and expedite processing since it take parallel structure. The BP algorithm recognized in real time hand gestures by self learning of trained eigen hand gesture. The proposed PCA and BP algorithm show improvement on the recognition compared to PCA algorithm.

Effects of Cereal and Red Ginseng Flour on Blood Glucose and Lipid Level in Streptozotocin - Induced Diabetic Rats (곡물류와 홍삼분말 급여가 당뇨성 흰쥐의 혈당 및 지질대사에 미치는 영향)

  • This study was conducted to examine the effects of feeding diets containing bioorganic power (BP) flour, wheat flour and rice flour on blood glucose and lipid level in streptozotocin-induced diabetic rats. BP flour was composed with the mixture of defatted flour (16 g soybean, 12 g wheat, 8 g barley), 2 g red ginseng and 2 g whole wheat flour per 100 g diet. Experimental groups were divided into non-diabetic normal and 4 diabetic groups containing control, 40% BP, 41.6% wheat and 41.6% rice flours with basal diet. BP group was significantly increased body weight gain and decreased plasma glucose compared with the diabetic control, wheat and rice flour groups (p<0.05). Food efficiency ratio of diabetic rats were significantly lower than that of the normal rats. The concentrations of total cholesterol and triglyceride in plasma and atherogenic index were significantly decreased in BP, wheat and rice diabetic groups than diabetic control (p<0.05). The concentration of total cholesterol and triglyceride in liver was significantly lower in BP group as compared with the diabetic control, wheat and rice flour groups (p<0.05). In conclusion, the results indicated that BP flour feeding improved plasma glucose level, total cholesterol and triglyceride in diabetic rats.
