사료급이(oral feeding)에 의한 vaccination을 통한 흰반점바이러스(WSSV)에 대한 재조합단백질 rVP19+28의 백신효능의 확인 (Vaccination of Shrimp (Litopenaeus vannamei) against White Spot Syndrome Virus (WSSV) by Oral Vaccination of Recombinant Fusion Protein, rVP19+28)

  • 본 연구는 WSSV의 주요 구조단백질인 VP19와 VP28을 모두 포함하는 VP19+28 fusion protein을 제조하여, Litopenaeus vannamei에서 WSSV에 대한 백신으로서의 효능을 평가하고자 수행하였다. VP19와 VP28 유전자를 fusion하여 제작한 VP19+28 유전자를 pET-28a(+) vector에 삽입하고 단일단백질로서 제작된 VP19+28 유전자를 E. coli BL21 (DE3)에서 발현시켰다. 백신실험을 위해 새우에게 2주 동안 실험용 사료를 급이하였으며, 그 후 바이러스액($1{\times}10^2$배로 희석한 WSSV)을 이용하여 새우에게 주사 감염에 의해 in vivo 공격실험(challenge test)을 수행하였다. 실험결과, vaccination을 하지 않은 새우들은 감염 후 11일째에 100%의 누적폐사율을 보였으며, host control로써 E. coli BL21을 사용하여 vaccination한 새우들은 감염 후 17일째에 100%의 누적폐사율을 보였다. rVP19, rVP28, rVP19+28을 이용하여 vaccination한 새우들의 경우 감염 후 21일째에 각각 66.7%, 41.7%, 41.7%의 누적폐사율을 보였다. 이상의 결과를 통해 rVP28과 rVP19+28이 WSSV에 대해 높은 백신효능을 가짐을 확인하였다. 또한 감염 후 21일째에 fusion protein rVP19+28과 rVP28의 누적폐사율은 동일하였지만 공격실험기간 동안 폐사율이 rVP19+28을 투여 한 실험군이 낮게 나타나는 것을 보아 WSSV에 대한 새우의 방어효능은 rVP19+28이 더 높음을 나타내는 것이다.

모형 그물에 대한 어군행동의 수직 모델링에 관한 연구 - 모형 그물이 없는 수조에서의 무지개송어의 유영특성 - (A Study on the Numerical Modeling of the Fish Behavior to the Model Net - Swimming Characteristics of Rainbow Trout, Salmo Gairdnerii in the Water Tank Without Model Net -)

  • 모형 그물에 대한 어군행동의 수치모델링에 관한 연구의 일환으로서 어군행동의 모델링을 위하여 어군행동을 나타내는 운동방정식을 구성하는 각종 힘들의 parameter를 추정하기 위한 기초자료를 제공하고자, 모형 그물을 설치하지 않은 상태에서의 유속 변화에 따른 무지개송어의 유영행동을 화성처리하여 각 개체 위치좌표로부터 계산된 유영속도, 유영깊이, 수조의 변과 개체 사이의 거리, 개체상호간의 최근접거리, 어구의 3차원적 구조 등의 유역특성을 조사 분석한 결과는 다음과 같다. 유영궤적으로부터 무지개송어의 유영행동을 살펴보면, 정수상태에서는 수종 중앙부에서 유영하는 빈도가 높았으며, 수조 벽면으로부터 상당히 떨어져서 유회운동을 하는 것으로 나타났다. 정수상태에서의 평균 유영속도는 21.6~23.6cm/sec(2.5~2.9BL cm/sec)로서 개체수가 많을수록 체장당 유영속도가 빠른 것으로 나타났으며, 유속의 변화에 따른 어군의 평균유영속도는 유속이 증가할수록 유영속도도 증가하여 유속이 25cm/sec인 경우에는 유영속도가 30.6 cm/secd(3.8BLcm/sec)에 달하였다. 어군의 평균 유영깊이는 17~38cm로서 주로 중층의 깊이에서 유영하였으며, 유속이 빨라질수록 유영 깊이가 깊어지고 유영 깊이의 변화가 적어졌다. 수조의 벽과 개체 사이의 평균 거리는 17.6~21.4cm였으며, 유속이 빨라질수록 수조의 벽과 개체사이의 거리가 다소 멀어지는 경향을 나타내었다. 개체상호간의 최근거리는 개체수가 많을수록 가까워지며, 유속의 변화에 따른 개체상호간의 최근접거리 평균치는 3.0~5.9cm(0.4~0.7BL cm)로 나타났다. 어군의 3차원적 구조는 군을 형성하는 개체수가 증가함에 따라 군의 형상이 길이, 폭 및 깊이의 모든 방향으로 넓어지며, 유속이 빨라질수록 길이방향과 폭 방향의 크기가 깊이 방향에 비해 상대적으로 커져 각 방향의 평균 상태비는 2.8 : 2.7 : 1이었다.

Synergistic Action Modes of Arabinan Degradation by Exo- and Endo-Arabinosyl Hydrolases

  • Two recombinant arabinosyl hydrolases, α-L-arabinofuranosidase from Geobacillus sp. KCTC 3012 (GAFase) and endo-(1,5)-α-L-arabinanase from Bacillus licheniformis DSM13 (BlABNase), were overexpressed in Escherichia coli, and their synergistic modes of action against sugar beet (branched) arabinan were investigated. Whereas GAFase hydrolyzed 35.9% of L-arabinose residues from sugar beet (branched) arabinan, endo-action of BlABNase released only 0.5% of L-arabinose owing to its extremely low accessibility towards branched arabinan. Interestingly, the simultaneous treatment of GAFase and BlABNase could liberate approximately 91.2% of L-arabinose from arabinan, which was significantly higher than any single exo-enzyme treatment (35.9%) or even stepwise exo- after endo-enzyme treatment (75.5%). Based on their unique modes of action, both exo- and endo-arabinosyl hydrolases can work in concert to catalyze the hydrolysis of arabinan to L-arabinose. At the early stage in arabinan degradation, exo-acting GAFase could remove the terminal arabinose branches to generate debranched arabinan, which could be successively hydrolyzed into arabinooligosaccharides via the endo-action of BlABNase. At the final stage, the simultaneous actions of exo- and endo-hydrolases could synergistically accelerate the L-arabinose production with high conversion yield.

Inbred 마우스 수정란을 이용한 형질전환마우스 생산에 있어서의 효과적인 수정란 이식 방법에 관한 연구 (Studies on the Efficient Embryo Transfer Methods using Inbred Embryos in Generation of Transgenic Mice)

  • 본 연구의 목적은 inbred 마우스 (C57BL/6J)의 수정란을 이용하여 형질전환마우스를 생산할 때, 수정란이식의 효율성을 증진시키기 위한 것이다. C57BL/6J 및 BCF1 마우스로부터 과배란처리 방법에 의해 수정란을 얻고, DNA를 1 세포기 수정란에 미세 주입한 다음, 1세포기 또는 2 세포기의 수정란을 가임신된 마우스의 한쪽 또는 양쪽 난관에 각각 이식하였다. 1세포기의 수정란을 0.75 d.p.c. 가임신된 마우스의 한쪽 난관에 이식했을 때, 임신율이 C57BL/6J는 68.8$\pm$7.83%, BCF1은 48.3$\pm$14.22% 이었고, 이식한 수정란 당 산자의 발달율은 C57BL/6J가 11.9$\pm$5.51%, BCF1은 10.5$\pm$8.03%로 성적이 저조하였다. 그러나, 2세포기의 수정란을 0.5 d.p.c. 가임신된 마우스의 양쪽 난관에 이식했을 때, 임신율이 C57BL/6J는 94.4$\pm$9.64%, 13CFl은 100$\pm$0% 이었고, 이식한 수정란 당 산자의 발달율은 C57BL/6J가 22.1 $\pm$0.4%, BCF1은 21.8$\pm$0.38%였다. 따라서 C57BL/6J 마우스의 2세포기 수정란을 0.5 d.p.c. 가임신된 마우스의 양쪽 난관에 이식하는 것이, BCF1마우스와 유사한 성적을 얻어 경쟁력이 있는 것으로 판단되었다. 이러한 결과에 영향을 미치는 인자가 여러 가지 있을 것으로 판단되지만, C57BL/6J 마우스의 2세포기 수정란을 0.5 d.p.c.가임신된 마우스의 양쪽 난관에 이식하는 방법이 다른 방법보다 형질전환마우스를 생산하는데 효율성이 더 높은 것으로 본 실험에서 확인되었다.

침구대성에 수록된 금침혈 연구 (A Study of Acupuncture Contraindications in Zhenjiudacheng)

  • Objectives : This research aimed to verify the validities of contraindicated acupoints in acupuncture treatment in Zhenjiudacheng. Methods : We investigated contraindicated acupoints when performing needling in Zhenjiudacheng, then verified them in today's medical knowledge and clinical studies. Results : In Zhenjiudacheng, 39 acupoints - LU2, LI4, LI13, LI14, LI16, ST1, ST9, ST12, ST17, ST25, ST30, ST42, SP6, SP11, SI18, BL8, BL9, BL56, KI11, TE7, TE8, TE19, TE20, GB3, GB18, GB21, GV10, GV11, GV17, GV22, GV24, CV1, CV4, CV5, CV8, CV9, CV15, CV17 and EX-HN11 - were described to need a careful approach in acupuncture treatment and 21 acupoints among them had contraindications. Most of the contraindications can be explained based on anatomical knowledge and results of clinical studies, and acupuncture treatment at the 39 contraindicated acupoints can induce injury of tissues or organs near the acupoints. Conclusions : The contraindicated acupoints in acupuncture treatment in Zhenjiudacheng seem to have been classified based on the adverse events experienced by doctors at that time. Therefore careful acupuncture treatment at these acupoints is needed to avoid injury to tissue or organs.

후계(後谿), 위중(委中), 후계배위중(後谿配委中) 침자(鍼刺)가 백서(白鼠)의 신경병리성(神經病理性) 동통억제(疼痛抑制) 및 c-Fos 단백(蛋白) 발현(發顯)에 미치는 영향(影響) (Effects of Acupuncture at SI3, BL40, SI3·BL40 on Neuropathic pain control and c-Fos protein expression in rats)

  • Objective: We have studied to know effects of acupuncture at SI3, BL40, SI3 BL40 on mechnical allodynia, cold allodynia and c-fos protein expression in a model of neuropathic pain of rat. Methods: A model of neuropathic pain was made by injuring tibial nerve and sural nerve while common peroneal nerve was maintained. after 2 weeks, we performed behavioral tests for 7 days to try out mechnical allodynia using von frey filament and cold allodynia using acetone, which are calculated by counting withdrawal response on foot. Rat brains removed and sliced on 8th days. Serial sections were immunohistochemically reacted with polyclonal c-fos antibody. The numbers of c-Fos protein immunoreactive neurons in the central gray were examined using scion image program. Results: Mechanical allodynia in the SI3, BL40, SI3 BL40 groups were diminished compared with the control group. Cold allodynia in the SI3, BL40, SI3 BL40 groups were diminished compared with the control group. c-Fos protein expression on the central gray in the SI3 group were lower than that of the control group. Conclusions: We have noticed that acupuncture at SI3, BL40, SI3 BL40 diminished mechanical allodynia and cold allodynia in a model of neuropathic pain compared with the control group. c- Fos protein expression in the central gray of that group was also decreased compared with the control group. pain control using acupunture was accumulated as time goes by. This study can be used as a basic resource on a study and a treatment of pain.

Rainbow Power Therapy의 경항통에 대한 효과 (The Effects of Rainbow Power Therapy on Neck Pain Patients)

  • Objective : This study was designed to estimate the effects of Rainbow Power therapy on neck pain patients by using Visual Analogue Scale(VAS) and pressure algometer. Methods : Rainbow Power therapy group consisted of 18 patients and acupuncture therapy group consisted of 7 patients. The degree of improvement of neck pain was evaluated by VAS and pain pressure threshold(at the points of Jianjing GB21 and Jianwaishu SI14) before treatment, before 3rd treatment and before 5th treatment. Rainbow Power therapy was performed at the points of Jianjing GB21, Zhongfu LU1, Yangxi LI5, Ganshu BL18, Weishu BL21 and Shenshu, BL23. Each points were stimulated with RP-UM103(Rainbow Power therapy instrument) for 20 seconds. The points of acupuncture therapy were Jeonggeun, Jeongjong, Sangbaekn and Hegu LI4 and acupuncture was maintained for 15 minutes. After above therapy dry cupping was performed at the points of Jianjing GB21, Jianwaishu SI14, Gaohuang BL43, Tianzong, SI11 and Bingfeng SI12 for 5 minutes. Results : There was no significant difference between the two groups in VAS, pain threshold of Jianjing GB21 and Jianwaishu SI14 following treatment. In the two groups VAS was significantly decreased. In Rainbow Power therapy group the pain threshold of the two points was significantly increased. But there was no significant increase in acupuncture therapy groups about the pain threshold of Jianwaishu SI14. Conclusions : The effectiveness of Rainbow Power therapy on neck pain was shown through VAS and pressure algometer. These imply that Rainbow Power therapy may be useful for neck pain. Further study is needed about Rainbow Power therapy.

방사선에 의한 Apoptosis에서 Fas/Fas L의 역할 (The Role of Fas/FasL in Radiation Induced Apoptosis in vivo)

  • 목적: 변이를 보이는 lpr 마우스와 Fas ligand 변이를 보이는 gld 마우스를 이용하여 in vivo에서 Fas와 Fas ligand의 발현이 전리 방사선에 의해 유도되는 apoptosis에서 어떤 역할을 하는지 조사하고자 하였다. 대상 및 방법: Fas의 변이를 보이는 $C57BL/6J-Fas^{lpr}$ 마우스와 대조군인 C57BL/6J 마우스, Fas ligand 변이를 보이는 $C3H/HeJ-Fas^{gld}$ 마우스와 대조군인 C3H/HeJ 마우스를 대상으로 하였다. 마우스는 8주령 웅성으로서 전신 방사선 조사하여 일정 시간 후 비장을 적출하였다. 조직을 hematoxylin-eosin 염색하여 apoptosis 유도 수준을 비교 분석하였다. 또한 apoptosis 조절 물질인 p53, Bcl-2, Bax, Bcl-X_L,\;Bcl-X_S$에 대하여 Western Western blotting을 시행하고 발현수준을 densitometry로 분석하여 관련된 기전을 연구하였다. 결과: 2.5 Gydh k10 Gy 조사시에 $C57BL/6J-Fas^{lpr}$ 마우스와 $C3H/HeJ-Fas^{gld}$ 마우스에서 대조군 비하여 방사선에 의한 apoptosis가 유의하게 감소되는 것으로 나타났다(p<0.05). C57BL/6J 마우스와, C3H/HeJ 마우스에서 10 Gy 방사선 조사 후 Bax가 8시간 째에 각각 3배, 3.3배의 증가를 보였으나 $C57BL/6J-Fas^{lpr}$ 마우스와, $C3H/HeJ-Fas^{gld}$ 마우스에서는 뚜렷한 발현증가가 관찰되지 않았다. 결과: Fas의 변이가 있는 lpr 마우스와 Fas ligand의 변이가 있는 gld 마우스에서 방사선에 의한 apoptosis가 대조군 보다 현저하게 낮으며 이는 방사선에 의한 Bax의 유도가 미약한 것과 연관된 것으로 나타났다. 방사선에 의한 apoptosis 유도에 Fas의 역할이 매우 중요한 것으로 보인다.

개에서 추간판 질환에 대한 침술치료 증례 (Application of Traditional Acupuncture on Canine Intervertebral Disc Disease)

  • A four-year-old female Pekingese dog was presented with intermittent ambulatory hindlimb lameness and severe back pain. The course of the disease was acute and static and 5 days passed by until presentation. In neurological examination, severe back pain and slightly decreased proprioception in right hindlimb were shown. In radiographic examination, radiopacity was increased at T10-11 intervertebral disc space. According to the history taking, neurological examination and radiographic examination, it was tentatively diagnosed as intervertebral disc disease (IVDD). Conservative treatment was performed including cage rest, medication with prednisolone and traditional acupuncture. Acupoints were GV-6, GV-7, BL-18, BL-19. BL-40 and GB-34. Dry needlings rested for 20 minutes. Just after the first acupuncture treatment back pain was significantly decreased. With additional acupuncture treatment, all the clinical signs were not observed. It is considered that conservative therapy including traditional acupuncture, medication and cage rest might be the first choice of therapeutic method for Grade I or II IVDD in dogs.

Occurrence and Biological Control of Postharvest Decay in Onion Caused by Fungi

  • Postharvest decay of onion bulbs was examined by inspecting the commercial packages in the market or in storage. Bulb rot incidence was unexpectedly high, and onion bulbs with 1st quality grade were rotten most severely by 51%, followed by 32% for 2nd and 21% for 3rd grades. This indicates that larger bulbs had higher incidences of bulb rots. Major pathogens associated with basal and neck rots were Fusarium oxysporum and Aspergillus sp. or Botrytis allii, respectively, of which basal rot was most prevalent and damaging during storage. Among the epiphytic microorgani는 from onion plants, several Bacillus and Paenibacillus spp. and previously selected Pseudomonas putida and Trichoderma harzianum had inhibitory efficacy against bulb rot pathogens. Among these B. amyloliquefaciens BL-3, Paenibacillus polymyxa BL-4, and P. putida Cha 94 were highly inhibitory to conidial germination of F. oxysporum and B. allii. P. putida Cha 94, B. amyloliquefaciens BL-3, P. polymyxa BL-4, and T. harzianum TM were applied in the rhizoplane of onion at transplanting. Initially antagonist populations decreased rapidly during the first one month. However, among these antagonists, rhizoplane population densities of BL-3, Cha 94, and TM were consistently high thereafter, maintaining about 10$^4$-10$^{5}$ cells or spores per gram of onion root up to harvest time. The other bacterial antagonist BL-4 survived only for two months. TM was the most effective biocontrol agent against basal rot, with the number of rotten bulbs recorded at 4%, while that of the control was 16%. Cha 94 was effective for the first 20 days, but basal rot increased thereafter and had about the same control efficacy as that of BL-3 and BL-4. When the antagonists were applied to the topping areas of onion bulbs at harvest, TM was the most effective in protecting the stored onion bulbs from neck rotting. The second effective antagonist was BL-3. TM and BL-3 completely suppressed the neck rot in another test, suggesting that biocontrol of postharvest decay of onion using these microorganisms either at the time of transplanting or at harvesting may be promising.
