Effects of Selected Inoculants on Chemical Compositions and Fermentation Indices of Rye Silage Harvested at Dough Stage

  • This study was carried out to estimate the effect of selected inoculants on chemical compositions and fermentation characteristics of rye silage. Rye was harvested at dough stage and divided into 5 treatments, following: No additives (CON); L. plantarum R48-27 (LP27); L. buchneri R4-26 (LB26); Mixture of LP27 and LB26 at 1:1 ratio (MIX); and L. buchneri (LB). The rye forage was ensiled into 10 L bucket silo for 100 days. The contents of NDF and ADF were lowest (P<0.05) in LB26. The pH in LB26, MIX, and LB were lower (P<0.05) than CON and LP27. Lactate content in LB was higher (P<0.05) than the others, while acetate content in LB26 and LB were higher (P<0.05) than that in CON and LP27. Lactate to acetate ratio was highest (P<0.05) in LB, but lowest in LB26. Lactic acid bacteria (LAB) count in LB was higher (P<0.05) than that in CON, while yeast count in CON was lower than in all silages applied inoculants. In conclusion, silages inoculated with LB26 could improve potentially the aerobic stability caused by increases of acetate and propionate concentrations.

  • Asthma is a complex inflammatory disease of the lung characterized by variable airflow obstruction, airway hyperresponsiveness, airway inflammation, and reduction of respiratory function. Its prevalence and incidence are increasing because of the effect of various environmental and lifestyle risk factors. Steroid inhalation, long-acting agonists, and other synthetic drugs are used for the treatment of this disease. However, they have some side effects and show unsatisfied result and response after treatment. Therefore, many researchers have focused on the development of natural product-related treatment for asthma to suppress the side effects and unsatisfied results. Seaweeds contain various bioactive compounds with anti-inflammatory, antibacterial, and anti-oxidant activities. Thus, we investigated the asthma treatment-related literature using marine algae via the Google scholar search engine. Consequently, the literature is rarely investigated, but is increasing steadily. The literature was performed as a comparison study with an ovalbumin-induced group or drug-treated group, and investigated the antiasthma activity of algae ethanol extract. Although many researchers have studied marine algae-derived therapeutic agents for asthma, the amount of literature is rare compared with those of herbal medicine-derived therapeutic agents. Conclusively, we suggest that many researchers should investigate and develop algae-derived therapeutic agents for asthma treatment.

The Effects of Natural Antioxidants on Protein Oxidation, Lipid Oxidation, Color, and Sensory Attributes of Beef Patties during Cold Storage at 4℃

  • This study aimed to investigate the use of golden thread extract (GTE), clove extract (CE), and commercially available nitrite for retarding lipid and protein oxidation and for maintaining color stability and sensory attributes in beef patties stored at $4^{\circ}C$. GTE, CE, and nitrite treatment samples were found to be efficient in retarding lipid oxidation as all three treatments resulted in low thiobarbituric acid reactive substance (TBARS) content (p<0.05). By using GTE, CE, and nitrite into beef patties, protein oxidation was not developed. Incorporation of GTE and CE into beef patties maintained color stability by protecting against the decrease of $L^*$, $a^*$, $b^*$, chroma, and hue angle values and exhibited significant influence on sensory characteristics, including color and odor of beef patties (p<0.05). Compared to commercially available nitrite, GTE and CE were more effective as antioxidants for inhibiting lipid oxidation, and preserving color stability of fresh beef patties. The study indicated that GTE and CE could be utilized efficiently to extend the shelf life of beef patties.

Protective Effect of Isoliquiritigenin against Ethanol-Induced Hepatic Steatosis by Regulating the SIRT1-AMPK Pathway

  • Ethanol-induced fat accumulation, the earliest and most common response of the liver to ethanol exposure, may be involved in the pathogenesis of liver diseases. Isoliquiritigenin (ISL), an important constituent of Glycyrrhizae Radix, is a chalcone derivative that exhibits antioxidant, anti-inflammatory, and phytoestrogenic activities. However, the effect of ISL treatment on lipid accumulation in hepatocytes and alcoholic hepatitis remains unclear. Therefore, we evaluated the effect and underlying mechanism of ISL on ethanol-induced hepatic steatosis by treating AML-12 cells with 200 mM ethanol and/or ISL ($0{\sim}50{\mu}M$) for 72 hr. Lipid accumulation was assayed by oil red O staining, and the expression of sirtuin1 (SIRT1), sterol regulatory element-binding protein-1c (SREBP-1c), AMP-activated protein kinase (AMPK), and peroxisome proliferator-activated receptor alpha ($PPAR{\alpha}$) was studied by western blotting. Our results indicated that ISL treatment upregulated SIRT1 expression and downregulated SREBP-1c expression in ethanol-treated cells. Similarly, oil red O staining revealed a decrease in ethanol-induced fat accumulation upon co-treatment of ethanol-treated cells with 10, 20, and $50{\mu}M$ of ISL. These findings suggest that ISL can reduce ethanol induced-hepatic lipogenesis by activating the SIRT1-AMPK pathway and thus improve lipid metabolism in alcoholic fatty livers.

연마지를 이용한 촉각적 시각에 관한 연구 (A Study on Tactile Visuality Using Sandpapers)

  • Human senses have been studied widely. Many studied are being actively conducted to analyze correlations between two or more different senses. Among them, research on tactile visuality where a subject can indirectly experience visual sensation through tactile elements is also progressing in various fields. In this study, we analyzed color information by having subjects perceive sandpapers with different roughness through tactile sense, then having them choose a color that is reminiscent of the tactile sensation. Unlike existing tactile-visuality studies allowing the perception of indirect visual sense by using tactile sense, this study shows that it is possible to analyze with quantitative value by representing visual image perceived by tactile sense as a color. This study will contribute to emotional evaluation research that combines two or more of the senses felt by humans, and especially is considered to be useful as basic data when conducting research about tactile visuality or auditory tactile sense.

Sprague-Dawley계 흰쥐에서 현지초 부탄올 분획물의 항비만 및 항고지혈증 효과 (Anti-obesity and Anti-hyperlipidemic Effects of Butanol Soluble Fraction from Methanol Extract of Geranium thunbergii in Sprague-Dawley Rats)

  • The purpose of this study was to investigate anti-obesity and anti-hyperlipidemic effects of extracts from Geranium thunbergii (GT) in high-fat-diet-induced obese rats. Animals were randomly divided into four groups [normal diet, high fat diet, MeOH extract of GT (GTM), and BuOH fraction of GT (GTB)] and GT samples were treated with dose of 100 mg/kg for 8 weeks. It was observed that GTB-treated group significantly reduced body weight gain, food intake, epididymal fat weight, and triglyceride level in serum and liver compared to control group. The rats fed GTB also decreased contents of thiobarbituric acid-reactive substances (TBARS), hydroxyl radical, and xanthin oxidase (XO) increased by high fat diet. Furthermore, the anti-oxidant enzymes such as superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and catalase were increased by GTB treatment. The experimental results indicate that GTB has anti-obesity and anti-hyperlipidemic effects, as well as radical scavenging activity.

Properties of Doenjang (Soybean Paste) Prepared with Different Types of Salts

  • Samples of doenjang (a fermented soybean paste) were prepared with different types of salts; purified salt (PS), 3-year-aged solar salt (SS3), 1-year-aged solar salt (SS1), and bamboo salt (BS, 3rd processing product). For starter doenjang samples, selected starters comprising two bacilli, one yeast, and one fungus were inoculated, whereas for non-starter doenjang samples, microorganisms present in rice straw were inoculated after enrichment. The doenjang samples were fermented for 13 weeks at 25℃. During the fermentation period, SS and BS doenjang samples showed higher bacilli counts as well as much lower yeast counts than PS doenjang. At 13 weeks, yeast counts of starter doenjang samples were 7.75, 5.69, 6.08, and 4.74 log CFU/g for PS, SS3, SS1, and BS doenjang, respectively. For non-starter doenjang samples, counts were 7.17, 5.05, 5.92, and 4.54 log CFU/g for PS, SS3, SS1, and BS doenjang, respectively. SS and BS promoted growth of bacilli but inhibited growth of yeasts compared with PS. Debaryomyces hansenii was the dominant yeast in PS doenjang, whereas Candida guilliermondii and Pichia sorbitophila were dominant in SS and BS doenjang. In the sensory evaluation, SS and BS doenjang scored better than PS doenjang. In conclusion, SS and BS seem better than PS for production of high-quality doenjang.

Effect of Microbial and Chemical Combo Additives on Nutritive Value and Fermentation Characteristic of Whole Crop Barley Silage

  • This study was conducted to assess the effects of microbial and chemical combo additives on nutritive values, fermentation indices and aerobic stability of whole crop barley silage. Barley forage (Youngyang) was harvested at about 30% dry matter (DM) by treatments, chopped to 5 cm length and treated with distilled water only (CON), Lactobacillus plantarum (INO), propionic acid (PRO) or an equal mixture of INO and PRO (MIX). Barley forages were ensiled in 4 replications for 0, 2, 7, and 100 days. On 100 days of ensiling, MIX silage had higher (p<0.05) in vitro DM digestibility than CON silage, but lower (p<0.05) acid detergent fiber concentration. The pH in all treated silages was lower (p<0.05) than CON silage. The MIX silage had higher (p<0.05) lactate concentration and lactate to acetate ratio than in CON, but lower (p<0.05) yeast count. Aerobic stability in CON, PRO, and MIX silages were higher (p<0.05) than in INO silage. It is concluded that microbial and chemical combo additives using L. plantarum and propionic acid could efficiently improve nutritive values of barley silage in terms of increased in vitro DM digestibility compared to other treatments. In addition, all treatments except CON reduced yeast count which is the initiate microorganism of aerobic spoilage.

Improvement of Fibrinolytic Activity of Bacillus subtilis 168 by Integration of a Fibrinolytic Gene into the Chromosome

  • Fibrinolytic enzyme genes (aprE2, aprE176, and aprE179) were introduced into the Bacillus subtilis 168 chromosome without any antibiotic resistance gene. An integration vector, pDG1662, was used to deliver the genes into the amyE site of B. subtilis 168. Integrants, SJ3-5nc, SJ176nc, and SJ179nc, were obtained after two successive homologous recombinations. The integration of each fibrinolytic gene into the middle of the amyE site was confirmed by phenotypes (Amy-, SpecS) and colony PCR results for these strains. The fibrinolytic activities of the integrants were higher than that of B. subtilis 168 by at least 3.2-fold when grown in LB broth. Cheonggukjang was prepared by inoculating each of B. subtilis 168, SJ3-5nc, SJ176nc, and SJ179nc, and the fibrinolytic activity of cheonggukjang was 4.6 ± 0.7, 10.8 ± 0.9, 7.0 ± 0.6, and 8.0 ± 0.2 (U/g of cheonggukjang), respectively at 72 h. These results showed that construction of B. subtilis strains with enhanced fibrinolytic activities is possible by integration of a strong fibrinolytic gene via a marker-free manner.

Properties of a Bacteriocin Produced by Bacillus subtilis EMD4 Isolated from Ganjang (Soy Sauce)

  • A Bacillus species, EMD4, with strong antibacterial activity was isolated from ganjang (soy sauce) and identified as B. subtilis. B. subtilis EMD4 strongly inhibited the growth of B. cereus ATCC14579 and B. thuringiensis ATCC33679. The antibacterial activity was stable at pH 3-9 but inactive at pH 10 and above. The activity was fully retained after 15 min at 80℃ but reduced by 50% after 15 min at 90℃. The activity was completely destroyed by proteinase K and protease treatment, indicating its proteinaceous nature. The bacteriocin (BacEMD4) was partially purified from culture supernatant by ammonium sulfate precipitation, and Q-Sepharose and Sephadex G-50 column chromatographies. The specific activity was increased from 769.2 AU/mg protein to 8,347.8 AU/mg protein and the final yield was 12.6%. The size of BacEMD4 was determined to be 3.5 kDa by Tricine SDS-PAGE. The N-terminal amino acid sequence was similar with that of Subtilosin A. Nucleotide sequencing of the cloned gene confirmed that BacEMD4 was Subtilosin A. BacEMD4 showed bactericidal activity against B. cereus ATCC14579.
