BCP, Mg-BCP, Si-BCP 분말을 공침법(co-precipitation process)을 이용하여 합성하였다. 제조된 분말을 X-선 회절 분석(XRD), 적외선분광분석(FTIR)을 이용하여 특성을 분석하였다. Ca/P 몰 비율이 1.602인 칼슘 결손 아파타이트 공침물을 제조하여 $1000^{\circ}C$ 열처리 과정을 통하여 HAp와 ${\beta}$-TCP 상이 혼재된 BCP, Mg-BCP, Si-BCP을 합성할 수 있었다. 제조된 분말의 생체활성 거동을 평가하기 위하여 Hanks' Balanced Salt Solution(HBSS)에 침적시켜 시간에 따라 형상의 변화 및 결정상을 분석한 결과, BCP 분말에 비하여 금속 이온이 치환된 BCP 분말에서 빠른 생체활성을 관찰할 수 있었다. MTT assay를 통한 세포 성장률 평가에서모든 분말에서 시간 경과에 따라 독성을 나타내지 않았으며, 세포의 활성이 증가하였다.
The electrospinning process was established as a promising method to fabricate nano and micro-textured scaffolds for tissue engineering applications. A BCP-loaded PCL micro-textured scaffold thus can be a viable option. The biocompatibility as well as the mechanical properties of such scaffold materials should be optimized for this purpose. In this study, a composite scaffold of poly ($\varepsilon$-caprolactone) (PCL)-biphase calcium phosphate (BCP) was successfully fabricated by electrospinning. EDS and XRD data show successful loading of BCP nano particles in the PCL fibers. Morphological characterization of fibers shows that with a higher loaded BCP content the fiber surface was rougher and the diameter was approximately 1 to 7 ${\mu}m$. Tensile modulus and ultimate tensile stress reached their highest values in the PCL- 10 wt% BCP composite. When content of nano ceramic particles was low, they were dispersed in the fibers as reinforcements for the polymer matrix. However, at a high content of ceramic particles, the particles tend to agglomerate and lead to decreasing tensile modulus and ultimate stress of the PCL-BCP composite mats. Therefore, the use of nano BCP content for distribution in fiber polymer using BCP for reinforcement is limited. Tensile strain decreased with increasing content of BCP loading. From in vitro study using MG-63 osteoblast cells and L-929 fibroblast like cells, it was confirmed that electrospun PCL-BCP composite mats were biocompatible and that spreading behavior was good. As BCP content increased, the area of cell spreading on the surface of the mats also increased. Cells showed the best adherence on the surface of composite mats at 50 wt% BCP for both L-929 fibroblast-like cells and MG-63 osteoblast cell. PCL- BCP composites are a promising material for application in bone scaffolds.
In this study, we investigated primary biocompatibility and osteogenic gene expression of porous granular BCP bone substitutes with or without strontium (Sr) doping. In vitro biocompatibility was investigated on fibroblasts like L929 cells and osteoblasts like MG-63 cells using a cell viability assay (MTT) and one cell morphological observation by SEM, respectively. MTT results showed a cell viability percent of L929 fibroblasts, which was higher in Sr-BCP granules (98-101%) than in the non-doped granules (92-96%, p < 0.05). Osteoblasts like MG-63 cells were also found to proliferate better on Sr-doped BCP granules (01-111%) than on the non-doped ones (92-99%, p < 0.05) using an MTT assay. As compared with pure BCP granules, SEM images of MG-63 cells grown on sample surfaces confirmed that cellular spreading, adhesion and proliferation were facilitated by Sr doping on BCP. Active filopodial growth of MG-63 cells was also observed on Sr-doped BCP granules. The cells on Sr-doped BCP granules were well attached and spread out. Gene expression of osteonectin, osteopontin and osteoprotegrin were also evaluated using reverse transcriptase polymerase chain reaction (RT-PCR), which showed that the mRNA phenotypes of these genes were well maintained and expressed in Sr-doped BCP granules. These results suggest that Sr doping in a porous BCP granule can potentially enhance the biocompatibility and bone ingrowth capability of BCP biomaterials.
이전의 연구에서, bacterioferritin comigratory protein (BCP)을 인코딩하는 Schizosaccharomyces pombe의 구조유전자를 shuttle vector인 pRS316에 클로닝하여 BCP 과잉발현 플라즈미드인 pBCP10을 제조한 바 있다. 본 연구에서는, 플라즈미드 pBCP10을 사용하여 고온 스트레스에 대한 BCP의 열저항적 성질을 평가하였다. 대수기의 초기까지 성장시킨 S. pombe 세포의 배양 온도를 $30^{\circ}C$에서 $37^{\circ}C$나 $42^{\circ}C$로 전이시키는 경우, pBCP10 함유 S. pombe 세포가 벡터 대조 세포보다 $37^{\circ}C$와 $42^{\circ}C$ 모두에서 유의하기 더 잘 성장하였다. 높은 배양 온도로 전이한 뒤 6시간에서, pBCP10 함유 S. pombe 세포가 벡터 대조 세포보다 낮은 활성산소종(ROS)과 일산화질소(NO)의 지표로 측정된 아질산염(nitrite) 함량을 갖고 있음이 확인되었다. 온도 전이 뒤에, 총 글루타치온(total glutathione) 함량과 총 수퍼옥사이드 디스뮤타제(superoxide dismutase) 활성은 대응되는 벡터 대조 세포보다 pBCP10 함유 S. pombe 세포에서 현저하게 높다는 사실도 확인되었다. 종합하면, S. pombe BCP는 열저항적 역할을 보유하는 데, 활성산소종과 일산화질소에 대한 하강시키는 활성과 총 글루타치온과 수퍼옥사이드 디스뮤타제 등의 항산화 성분들을 상승시키는 활성, 즉 종합적으로 열안정성을 유지하는 활성에 근거하는 것으로 추정되었다

A highly porous Biphasic Calcium Phosphate (BCP) scaffold was fabricated by the sponge replica method with a microwave sintering technique. The BCP scaffold had interconnected pores ranging from $80\;{\mu}m$ to $1000\;{\mu}m$, which were similar to natural cancellous bone. To enhance the mechanical properties of the porous scaffold, infiltration of polycaprolactone (PCL) was employed. The microstructure of the BCP scaffold was optimized using various volume percentages of polymethylmethacrylate (PMMA) for the infiltration process. PCL successfully infiltrated into the hollow space of the strut formed after the removal of the polymer sponge throughout the degassing and high pressure steps. The microstructure and material properties of the BCP scaffold (i.e., pore size, morphology of infiltrated and coated PCL, compressive strength, and porosity) were evaluated. When a 30 vol% of PMMA was used, the PCL-BCP scaffold showed the highest compressive strength. The compressive strength values of the BCP and PCL-BCP scaffolds were approximately 1.3 and 2MPa, respectively. After the PCL infiltration process, the porosity of the PCL-BCP scaffold decreased slightly to 86%, whereas that of the BCP scaffold was 86%. The number of pores in the $10\;{\mu}m$ to $20\;{\mu}m$ rage, which represent the pore channel inside of the strut, significantly decreased. The in-vitro study confirmed that the PCL-infiltrated BCP scaffold showed comparable cell viability without any cytotoxic behavior.
Purpose: The role of hard-type crosslinked hyaluronic acid (HA) with particulate bone substitutes in bone regeneration for combined inlay-onlay grafts has not been fully investigated. We aimed to evaluate the effect of hard-type crosslinked HA used with bone substitute in terms of new bone formation and space maintenance. Methods: A 15-mm-diameter round defect was formed in the calvaria of 30 New Zealand White rabbits. All animals were randomly assigned to 1 of 3 groups: the control group (spontaneous healing without material, n=10), the biphasic calcium phosphate (BCP) graft group (BCP, n=10), and the BCP graft with HA group (BCP/HA, n=10). The animals were evaluated 4 and 12 weeks after surgery. Half of the animals from each group were sacrificed at 4 and 12 weeks after surgery. Samples were evaluated using micro-computed tomography, histology, and histomorphometry. Results: The BCP group showed higher bone volume/tissue volume (BV/TV) values than the control and BCP/HA groups at both 4 and 12 weeks. The BCP and BCP/HA groups showed higher bone surface/tissue volume (BS/TV) values than the control group at both 4 and 12 weeks. The BCP group showed higher BS/TV values than the control and BCP/HA groups at both 4 and 12 weeks. No statistically significant difference in newly formed bone was found among the 3 groups at 4 weeks. The BCP group showed significantly higher new bone formation than the BCP/HA group at 12 weeks. Conclusions: Hard-type crosslinked HA did not show a positive effect on new bone formation and space maintenance. The negative effect of hard-type crosslinked HA may be due to the physical properties of HA that impede osteogenic potential.
In the structure of ITO/N,N'-diphenyl-N,N' bis (3-methylphenyl)-1,1'-biphenyl-4,4'-diamine(TPD) /2,9-Dimethy 1-4,7-diphenyl-1,10-phenanthroline (BCP)/tris (8-hydroxyquinoline)aluminum$(Alq_3)$/Al device, we studied the efficiency improvement of organic light-emitting diodes due to thickness variation of BCP materials used for a electron breaking layer. The thickness of TPD and $Alq_3$ was manufactured 40 nm, 60 nm, respectively under a base pressure of $5\times10^{-6}$Torr using a thermal evaporation. The TPD and $Alq_3$ layer were evaporated to be at a deposition rate of 2.0 A/s. The BCP was evaporated to be at a deposition of 1.0 A/s. When the thickness of BCP increased from 5 to 30 nm, we found that the luminous efficiency and the external quantum efficiency is superior to the others when the thickness of BCP is 20 nm. Compared to the ones from the devices made without BCP, the luminous efficiency and the external quantum efficiency was improved by 57 %, 70%, respectively.
Purpose: The aim of this study was to compare the clinical outcome of open flap debridement (OFD) with a biphasic calcium phosphate (BCP) graft to that of OFD without BCP graft for the treatment of intrabony periodontal defects (IBDs). Methods: The study included 25 subjects that had at least one intrabony defect of 2- or 3-wall morphology and an intrabony component ${\geq}4$ mm as detected radiographically. Subjects were randomly assigned to treatment with (BCP group, n=14) or without BCP (OFD group, n=11). Clinical parameters were recorded at baseline and 6 months after surgery and included the plaque index, gingival index, probing depth (PD), clinical attachment level (CAL), and gingival recession (REC). A stringent plaque control regimen was enforced for all of the patients during the 6-month observation period. Results: In all of the treatment groups, significant PD reductions and CAL gains occurred during the study period (P<0.01). At 6 months, patients in the BCP group exhibited a mean PD reduction of $3.7{\pm}1.2$ mm and a mean CAL gain of $3.0{\pm}1.1$ mm compared to the baseline. Corresponding values for the patients treated with OFD were $2.5{\pm}0.8$ mm and $1.4{\pm}1.0$ mm, respectively. Compared to OFD group, the additional CAL gain was significantly greater in the patients in BCP group (P=0.028). The additional PD reduction was significant for the BCP group (P=0.048). The REC showed a significant increase in both groups, and the amount of recession was significantly smaller in the BCP group than OFD group (P=0.023). In radiographic evaluation, the height of the bone fill in the BCP group was significantly greater than OFD group. Conclusions: The clinical benefits of BCP found in this study indicate that BCP may be an appropriate alternative to conventional graft materials.
본 연구의 목적은 지난 2004년에 공표된 바젤II 규정에 의거 각국 은행들에 대한 체계적인 위험관리가 강화되는 상황에서 최근 국내 S은행에서 은행권 최초로 기존의 DRP(Disaster Recovery Planning)차원을 넘어선 전행 차원의 BCP프로세스를 구축하게 되었다. 여타 은행들도 대형 재난 및 바젤II 규정등에 대비해 금명간 체계적인 BCP시스템을 갖추게 될 것인바, 이러한 BCP시스템의 구축에 따른 효과로 실제 재해 상황에서 이러한 시스템이 은행의 안정적인 업무수행을 보장하는데 그 주된 목적이 있겠지만, BCP 프로세스에 포함된 내용으로서 BIA(Business Impact Analysis)를 통한 RTO(Recovery Time Objectives), 그리고 BCP 프로세스의 추진으로 은행의 여러 이해관계자들에게 미치는 영향등을 고려해 은행원들의 평시 업무수행절차나 프로세스등의 개선등을 통해 은행원들의 직무만족도에 어느 정도 영향을 미치는지 살펴보고자 한다.
