Probiotic Properties of Lactobacillus strains Isolated from Kimchi (김치로부터 분리된 Lactobacillus strains의 probiotic 특성)

  • The objective of this study was to evaluate the safety and functional properties of four potential probiotic strains isolated from Kimchi, traditional Korean fermented vegetables. Based on being higher tolerance to bile salts and showing higher acid resistance or hydrophobic properties, one Lactobacillus arizonensis strain (BCNU 9032) and three L. brevis strains (BCNU 9037, BCNU 9098 and BCNU 9101) were selected in the screening experiment. All strains can survived up to 99% after 3h culture in pH 2.5 and resistant to 1% bile salts. These strains also showed good antimicrobial activities against a number of food borne pathogens, especially against Escherichia coli and Shigella sonnei. The ability to lower cholesterol levels of L. arizonensis BCNU 9032 and L. brevis 9037 were demonstrated by bile salt hydrolytic activity and cholesterol assimilation tests. Moreover, L. brevis BCNU 9098 and BCNU 9101 showed higher adherence to Caco-2 cells (12.76 and 11.86%, respectively) than Lactobacillus rhamnosus GG, a commercial probiotic strain used worldwide. The results suggest that these strains could be used as probiotics.

Production of Indigoid Pigments by Persolvent Fermentation with Pseudomonas putida BCNU 106 (Pseudomonas sp. BCNU 106의 persolvent fermentation에 의한 인디고이드계 색소 생산)

  • Pseudomonas sp. BCNU 106 isolated from industrial wastewater was able to produce indigo from indole by utilizing various organic solvents. BCNU 106 produced indigo effectively when grown in the presence of a large volume of p-xylene, propylbenzene, and mesitylene and a high level of indole. The present study demonstrated that the maximal yield was achieved with 20% (v/w) p-xylene and 4 g/l indole. Under these conditions, the indigo yield and the transformation efficiency of indole were 315.5 mg/l and 97%, respectively. The results suggest that Pseudomonas sp. BCNU 106 might be a potential candidate for industrially important indigo production.

Characterization of an Antimicrobial Substance-producing Pseudomonas sp. BCNU 2001 (항생물질을 생산하는 Pseudomonas sp. BCNU 2001 균주의 특성)

  • Strain BCNU 2001 was isolated from soil samples collected from Tea-baek Mountain area. The biochemical characteristics and 16S ribosomal RNA gene sequences of the isolate revealed that the strain belonged to the Pseudomonas aeruginosa. The supernatants had an antimicrobial effect on various kind of bacteria and fungi. Especially BCNU 2001 was able to greatly inhibit the growth of Micrococcus luteus, Proteus mirabilis, Proteus vulgaris, and Aspergillus niger, and its inhibition zone was measured as 18.5 mm against Micrococcus luteus, 19.0mm against Proteus mirabilis, 17.0mm against Proteus vulgaris, and 13.5 mm against Aspergillus niger, respectively. Hexane and dichloromethane extracts of BCNU 2001 exhibited significant activity against bacteria, and dichloromethane and ethylacetate extracts showed significant activity against fungi. Pseudomonas strain BCNU 2001 was also determined to have antimicrobial peptide against various microorganisms including Gram positive bacteria, Gram negative bacteria and fungi. The obtained results may provide preliminary support for the usefulness of Pseudomonas strain BCNU 2001.

Biodegradation of Aromatic Hydrocarbons by Toluene-tolerant Pseudomonas sp. BCNU 154 (Toluene내성세균 Pseudomonas sp. BCNU 154을 이용한 방향족화합물의 분해)

  • The biodegradative potentialities of a toluene-tolerant Pseudomonas sp. BCNU 154, isolated from waste water, were investigated. Among 16 aromatic substrates tested, cumene, cyclohexane, ethylbenzene, p-xylene, m-xyene, toluene and diphenylether were metabolized. Pseudomonas sp. BCNU 154 degraded aerobically toluene, ethylbenzene, p-xylene and cumene. With toluene competitive degradation occurred after 12 hours, but with p-xylene and cumene, and with ethylbenzene, 90 and 75% degradation occurred after 12 hours of incubation, respectively.

고농도 Indole 내성 Pseudomonas savastanoi BCNU 106에 의한 Indigo 및 Indirubin의 생산

  • Since the biosynthetic production of indigo and indirubin normally reflects a difficult process including the toxicity of indole to microorganisms, only several bacterial strains have been exploited to produce indigo and indirubin from indole or its derivatives. P. savastanoi BCNU 106, which was a gram negative bacterium, was isolated and tolerant to 10% (v/v) toluene. The indole tolerance level of P. savastanoi BCNU 106 was as high as 160 mg/ml when toluene or p-xylene was added to the medium to 20% by volume. P. savastanoi BCNU 106 grown in a two-phase culture system containing indole(100 mg/ml) and P-xylene (0.2 ml/ml) produced P-xylene-soluble pigments, blue indigo and purple indirubin. Of the conditious tried, the production of indigo and indirubin was found only when P. savastanoi BCNU 106 was grown in the two-phase system overlaid with the organic solvents with appropriate polarity. This study may illustrate that the isolated extremophile P. savastanoi could be used in the microbial conversion process of the industrial potentials.

Characterization of Organic Solvent Stable Lipase from Pseudomonas sp. BCNU 106 (Pseudomonas sp. BCNU 106이 생산하는 유기용매 내성 리파아제의 특성)

  • A crude extracellular lipase from solvent-tolerant bacterium Pseudomonas sp. BCNU 106 was highly stable in the broad pH range of 4-10 and at temperature of 37℃. Crude lipase of BCNU 106 exhibited enhanced stability in 25% organic solvents such as xylene (121.85%), hexane (120.35%), octane (120.41 %), toluene (118.14%), chloroform (103.66%) and dodecane (102.94%) and showed excellent stability comparable with the commercial immobilized enzyme. In addition, the stability of BCNU 106 lipase retained above 110% of its enzyme activity in the presence of Cu2+, Hg2+, Zn2+ and Mn2+, whereas Fe2+ strongly inhibited its stability. The detergents including tween 80, triton X-100 and SDS were positive signals for lipase stability. Because of its stability in multiple organic solvents, cations and surfactants, the Pseudomonas sp. BCNU 106 lipase could be considered as a potential biocatalyst in the industrial chemical processes without using immobilization.

  • An organic solvent-tolerant lipase of Pseudomonas sp. BCNU 154 that was isolated from wastewater in the industrial complex region had optimal activity at $37^{\circ}C$ and pH 8. This crude extracellular lipase from BCNU 154 exhibited maximum stability in toluene, retaining about 6.01 U/ml (117.53%) activity for 2 h. $Ca^{2+}$, $Mg^{2+}$, $NH_4{^+}$, and $Na^+$ ions and triton X-100 activated the enzymes, whereas $Ba^{2+}$, $Hg^{2+}$, and $Zn^{2+}$ ions inhibited their activity. Pseudomonas sp. BCNU 154 lipase revealed stable activity comparable to that of the commercial immobilized Novozym 435. Thus, this organic solvent-tolerant lipase could have potential as a whole cell biocatalyst in industrial chemical processes without the use of immobilization.

  • In the screening process of organic solvent tolerant bacteria showing good growth in media containing several kinds of organic solvents, one strain was isolated and identified as Bacillus sp. BCNU 5006. The strain was able to tolerate many organic solvents including benzene, toluene, xylene, octane, dodecane, butanol and ethylbenzene. Likewise, it could also utilize these solvents as the sole source of carbon with significant enzyme production. The lipolytic enzyme stability of Bacillus sp. BCNU 5006 was studied in the presence of several kinds of solvents at a 25% (v/v) concentration. The highest enzyme stability was observed in the presence of octane (107%), followed by ethylbenzene (88%), decane (86%), and chloroform (85%). Especially, BCNU 5006 lipase was determined to be more stable than immobilized enzyme (Novozyme 435) in the presence of octane, chloroform and xylene. This organic solvent tolerant Bacillus sp. BCNU 5006 could be expected as a potential bioremediation agent and biocatalyst for biodegradation and provide on organic-solvent-based enzymatic synthetic method in industrial chemical processes.

Antifungal Activity of Bacillus sp. BCNU 2003 against the Human Pathogenic Fungi (인체 병원성 진균에 대한 Bacillus sp. BCNU 2003의 항진균 효과)

  • An antifungal antibiotic-producing strain, BCNU 2003, was isolated from forest soil in Korea. The morphological and physiological characters, and 16S rRNA sequences analysis of strain BCNU 2003 identified this strain as Bacillus genus. The Bacillus sp. BCNU 2003 showed strong antifungal activities against Aspergillus niger, Trichophyton mentagrophytes and Trichophyton rubrum with inhibition ranging from 62.05 to 63.49% by using dual culture technique. Bacillus sp. BCNU 2003 produced a maximum level of antifungal substances under aerobic incubation at 28oC and pH 6.5-7.2 for 6 days in LB broth. Ethyl acetate extract of the cultured broth showed strong antifungal activity and a broad antifungal spectrum against various pathogenic fungi. The minimum inhibitory concentration (MIC) values for its active extracts ranged between 0.0625 mg/ml and 1 mg/ml. In addition, Bacillus sp. BCNU 2003 was determined to have the ability to produce enzymes such as amylase, protease, gelatinase and catalase.
