The CD28-B7 Family in Anti-Tumor Immunity: Emerging Concepts in Cancer Immunotherapy

  • The interactions between B7 molecules and CD28-family receptors are crucial in the regulation of adaptive cellular immunity. In cancer, the aberrant expression of co-inhibitory B7 molecules has been attributed to reduced anti-tumor immunity and cancer immune evasion, prompting the development of cancer therapeutics that can restore T cell function. Murine tumor models have provided significant support for the targeting of multiple immune checkpoints involving CTLA-4, PD-1, ICOS, B7-H3 and B7-H4 during tumor growth, and clinical studies investigating the therapeutic effects of CTLA-4 and PD-1 blockade have shown exceptionally promising results in patients with advanced melanoma and other cancers. The expression pattern of co-inhibitory B7 ligands in the tumor microenvironment has also been largely correlated with poor patient prognosis, and recent evidence suggests that the presence of several B7 molecules may predict the responsiveness of immunotherapies that rely on pre-existing tumor-associated immune responses. While monotherapies blocking T cell co-inhibition have beneficial effects in reducing tumor burden, combinatorial immunotherapy targeting multiple immune checkpoints involved in various stages of the anti-tumor response has led to the most substantial impact on tumor reduction. In this review, we will examine the contributions of B7- and CD28-family members in the context of cancer development, and discuss the implications of current human findings in cancer immunotherapy.

PBSA 분해효소 유전자의 분석 (Analysis of Gene Encoding the PBSA Degradation Enzyme)

  • 우리나라 토양으로부터 분리된 (주와 김 2009) poly (butylene succinate-co-butylene adipate; PBSA) 분해균 Burkholderia cepacia PBSA-7, Bacillus licheniformis PBSA-8 및 Burkholderia sp. PBSA-9에서 PBSA 분해효소를 암호화하는 유전자를 분석하였다. PCR을 수행하여 B.cepacia PBSA-7과 Burkholderia sp. PBSA-9는 약 1.5 Kb, B. licheniformis PBSA-8은 약 600 bp의 lipase 유전자(lip A) 절편을 가지는 것을 확인하였다. 세 균주 모두에서 유전자의 염기서열 내 lipase box인 Gly-X1-Ser-X2-Gly와 Ala-X1-Ser-X2-Gly가 존재하였다. B. cepacia PBSA-7의 PBSA 분해효소 유전자는 family I-1 lipases와 36~40%, family I-2 lipases와는 82~92%의 높은 유전적 상동성을 보였다. 또한 B. licheniformis PBSA-8의 PBSA 분해효소 유전자는 subfamily 1-4 lipases와 64~65%의 유전적 상동성을 나타내었으나, subfamily 1-5에 속하는 lipase들과는 거의 유전적 상동성이 없는 것으로 나타났다. Burkholderia sp. PBSA-9의 PBSA 분해효소 유전자도 family I-1 lipases와 35~37%, family I-2 lipases와는 83~94%로 높은 유전적 상동성을 보여 B. cepacia PBSA-7의 PBSA 분해효소 유전자와 유사한 결과를 보였다.

Emerging Co-signaling Networks in T Cell Immune Regulation

  • Co-signaling molecules are surface glycoproteins that positively or negatively regulate the T cell response to antigen. Co-signaling ligands and receptors crosstalk between the surfaces of antigen-presenting cells (APCs) and T cells, and modulate the ultimate magnitude and quality of T cell receptor (TCR) signaling. In the past 10 years, the field of co-signaling research has been advanced by the understanding of underlying mechanisms of the immune modulation led by newly identified co-signaling molecules and the successful preclinical and clinical trials targeting co-inhibitory molecules called immune checkpoints in the treatment of autoimmune diseases and cancers. In this review, we briefly describe the characteristics of well-known B7 co-signaling family members regarding the expression, functions and therapeutic implications and to introduce newly identified B7 members such as B7-H5, B7-H6, and B7-H7.

기혼직장여성의 우울증에 미치는 영향요인: 여성가족패널 조사 7차년도(2017-2018) 자료 활용 (Factors Influencing Depressive Symptoms of Married Working Women: The Korean Longitudinal Survey of Women and Family 2017-2018)

  • Objectives: The aim of this study was to examine factors influencing depressive symptoms among married working women using the dataset of the Korean Longitudinal Survey of Women and Family (KLoWF 7th). There were 1,030 subjects. Methods: A multiple regression model was used to study the factors influencing depressive symptoms among married working women. Results: The authors found a negative relationship between satisfaction with spouse household-labor (b=-0.606, p=0.022) and depressive symptoms among married working women, a negative relationship with spouse (b=-0.237, p<0.001) and a negative spousal perception of working (b=-0.709, p=0.045), a positive relationship with excessive working hours (b=0.397, p=0.027), a positive relationship with temporal oppression on workload (b=0.422, p=0.002), and a positive relationship between workplace discrimination (b=0.053, p=0.046) and depressive symptoms among married working women. Conclusions: This study suggests that family life and working environments are important factors for depressive symptoms in married women workers. The findings of this study will be helpful to policymakers to design plans to decrease depressive symptoms among married working women.

노인 우울에 영향을 미치는 요인: 건강요인과 가족요인의 상대적 영향력을 중심으로 (Relative Effects of Health and Family Factors on Geriatric Depression)

  • Over the past few decades, the proportion of elderly people with depression in Korea has been rapidly increasing. The purpose of this study is to explore relative effects of health and family factors on geriatric depression. The data sources are from Korean Longitudinal Study of Aging(2008). The Analysis sample consists of 4,040 cases. Analysis results show that there is a significant variability in geriatric depression according to sociodemographic characteristics, health characteristics, and family characteristics. Hierarchical logistic regression analyses are employed to assess the causal relationship between health characteristics and geriatric depression. Model II showed that ADL(exp(B)=1.732), having difficulties due to sight deterioration(exp(B)=1.398), having difficulties due to masticatory force deterioration(exp(B)=1.414), having difficulties due to pain(exp(B)=2.435), and subjective health status(exp(B)=2.010) are reported as predictors of geriatric depression. Namely, the probability of having geriatric depression of the above predictors has been 1.7 times, 1.4 times, 1.4 times, 2.4 times. 2.0 times higher than normal figures. Among health characteristics, pain is the most prevalent factor. Model III showed that as for family characteristics, coresiding with children(exp(B)=1.312) and frequency of contacting with children by phones, letters, or emails(exp(B)=1.477) are reported as predictors of geriatric depression. Key findings are that health factors have larger effects than family factors on geriatric depression.

담자균 Phanerochaete chrysosporium으로부터 유래한 Glycoside Hydrolase Family 74 유전자 클로닝과 전사산물 분석 (Molecular Cloning of Glycoside Hydrolase Family 74 Genes and Analysis of Transcript Products from the Basidiomycete Phanerochaete chrysosporium)

  • 셀룰로오스의 가수분해 기작을 구명하기 위하여 Phanerochaete chrysosporium으로부터 74A (PcGHF74A) 유전자를 클로닝한 결과 2162 bp의 염기서열에 해당하는 721개의 아미노산을 가지고 있으며, 다른 사상균에서 유래한 GHF74와 70~77%의 상동성을 나타냈다. Phanerochaete chrysosporium GHF74B (PcGHF74B)는 family 1에 속하는 Cellulose Binding Module (CBM)을 가지고 있으며 셀룰로오스 배양계에서 다양한 전사산물이 존재하였다. PcGHF74B 전사산물에서 나타난 splice variants를 조사하기 위해서 annotation data와 sequence data로부터 primer를 설계하여 RT-PCR분석을 수행하였으며 그 결과 다양한 배양조건에서 splice variants가 존재함을 확인하였다. 첫 번째는 annotation data와 다르게 11번째 intron을 포함하고 있어 full length로 추정되어지는 것으로 2562 bp에 stop codon이 존재했으며, 두 번째는 7번째 exon 1187 bp에 stop codon을 가지고 있으며 12개의 exon으로 구성되어 있다. 세 번째는 10개의 exon과 9개의 intron을 포함하고 있으며 7번째 exon에 stop codon이 존재했다. Splice variants로서 intron에 나타난 stop codon으로 인해 활성단백질의 합성이 일어나지 않을 것이며 비활성 단백질을 생성하거나 원래의 GHF74의 기능이 아닌 다른 새로운 기능을 갖는 단백질을 생성할 수 있을 것으로 사료된다.

소양면 지역사회 환경기초조사 (A Basie Community Health Survey in Rural Korea (Soyang-Myun))

  • 1. Introduction Community medicine with the concept of comprehensive medical care and an ideal medical care delivery system not only for an individual or family but for the whole community has emerged. In April 1970, the Presbyterian Medical Center started a hospital based community health service project in order to improve the health of the people in rural areas. Prior to commencing a comprehensive medical care system, a family survey was needed. The major objective of this survey was to obtain information concerning the people and their environment so as to be able to plan and implement a comprehensive medical care program in Soyang-Myun. 2. Survey Method An interview using a family record form was carried out for each household. This family record form was designed to get information about demography, family planning, environmental sanitation and vital statistics. Prior to beginning, the members of the survey team were trained in interviewing techniques for three days. The team consisted of a public health nurse, four nurse-aides, a sanitarian and four health extension workers who are working in our project, The survey was carried out during the period November 1971 to March 1972. 3. Project area 1) Population of Soyang-Myun was 11,668; male, 5,962 and female, 5,706. Bacillus licheniformis WL-12의 cellulase 유전자 클로닝과 발현 (Cloning and Expression of A Bacillus licheniformis Cellulase Gene)

  • 가정에서 제조된 된장으로부터 cellulase 생산균으로 분리된 고온성 WL-12는 형태적 특성, 생화학적 성질 및 16S rRNA의 염기서열에 근거하여 Bacillus licheniformis로 동정되었다. B. licheniformis WL-12의 cellulase 유전자를 클로닝하여 그 염기서열을 결정한 결과 cellulase 유전자(celA)는 517 아미노산으로 구성된 단백질을 코드하며 1,551 뉴클레오티드로 이루어졌다. 아미노산 잔기배열을 분석한 결과 WL-12의 cellulase는 활성영역과 cellulose 결합영역으로 구성되어 있었으며, glycosyl hydrolase (GH) family 5에 속하는 B. licheniformis, B. subtilis와 B. amyloliquefaciens의 cellulase와 높은 상동성을 보였다. 클론된 celA를 발현용 vector에 도입하여 B. subtilis에서 발현시켜 cellulase 최대생산성이 7.0 units/ml에 이르렀다.

Identification of Differentially Expressed Genes by TCDD in Human Bronchial Cells: Toxicogenomic Markers for Dioxin Exposure

  • Differentially expressed genes by 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) were identified in order to evaluate them as dioxin-sensitive markers and crucial signaling molecules to understand dioxin-induced toxic mechanisms in human bronchial cells. Gene expression profiling was analyzed by cDNA microarray and ten genes were selected for further study. They were cytochrome P450, family 1, subfamily B, polypeptide 1 (CYP1B1), S100 calcium binding protein A8 (calgranulin A), S100 calcium binding protein A9 (calgranulin B), aldehyde dehydrogenase 1 family, member A3 (ALDH6) and peroxiredoxin 5 (PRDX5) in up-regulated group. Among them, CYP1B1 was used as a hallmark for dioxin and sharply increased by TCDD exposure. Down-regulated genes were IK cytokine, interferon-induced protein with tetratricopeptide repeats 1 (IFIT1), nuclease sensitive element binding protein 1 (NSEP1), protein tyrosine phosphatase type VI A, member 1 (PTP4A1), ras oncogene family 32 (RAB32). Although up-regulated 4 genes in microarray were coincided with northern hybridization, down-regulated 5 genes showed U-shaped expression pattern which is sharply decreased at lower doses and gradually increased at higher doses. These results introduce some of TCDD-responsive genes can be sensitive markers against TCDD exposure and used as signaling cues to understand toxicity initiated by TCDD inhalation in pulmonary tissues.
