mRNA Expression of Bax, Bcl-2, p53, Cathepsin B, Caspase-3 and Caspase-9 in the HepG2 Cell Line Following Induction by a Novel Monoclonal Ab Hep88 mAb: Cross-Talk for Paraptosis and Apoptosis

  • Monoclonal antibodies with specific antigens have been widely used as targeted therapy for cancer. Hep88 mAb is a monoclonal antibody which shows specific binding with anti-cancer effects against the HepG2 cell line. However, its mechanisms of action are still not completely understood. We examined cell cycling and apoptosis by flow cytometry and mRNA expression of factors involved in apoptosis and paraptosis in Hep88 mAb-treated HepG2 cells by real-time PCR. The cell-cycle analysis demonstrated that growth-inhibitory activity was associated with G2/M cell cycle arrest. Hep88 mAb induced a significant increase in apoptotic cell populations in a dose- and time-dependent manner. The mRNA expression results also suggested that the process triggered by Hep88 mAb involved up-regulation of tumor suppressor p53, pro-apoptotic Bax, Cathepsin B, Caspase-3 and Caspase-9, with a decrease of anti-apoptotic Bcl-2 - thus confirming paraptosis and apoptosis programmed cell death. These findings represent new insights into the molecular mechanisms underlying the anti-cancer properties of Hep88 mAb in liver cancer cells.

Seroepidemiology of Hepatitis A and Hepatitis B in Korean Children (학동기 아동에서 A형 및 B형 간염의 혈청학적 역학조사 -1998년 현재 서울 일부지역을 대상으로-)

  • Purpose: The aim of this study is to investigate the seroepidemiology of hepatitis A and hepatitis B in Korean children. Methods: 801 children aged 6 to 12 years were enrolled. IgG antibody to hepatitis A virus (anti-HAV), hepatitis B surface antigen (HBsAg), antibody to HBsAg (anti-HBs), and IgG antibody to HBc (anti-HBc) were measured by radioimmunoassay. Results: Of the 801 children, the seroprevalence of anti-HAV was 0%. Three (0.4%) were seropositive for HBsAg. The seroprevalence of anti-HBs was 61.8%. Two (0.4%) of 495 anti-HBs positive subjects were positvie for anti-HBc. Of the 801 children, 658 (82.1%) had received hepatitis B vaccination. Conclusions: Since natural anti-HAV is not acquired in the childhood, the outbreak of hepatitis A is possible in adolescents and young adults. The decrease in seroprevalence of HBsAg and the increase in seroprevalence of anti-HBs might result from hepatitis B vaccination program in childhood.

  • The present study was conducted to characterize maternal antibody status which haemagglutination inhibition(HI) titers against canine parvovirus(CPV) in the 15 puppies delivered from 3 dams. The range of HI titers of 5 puppies delivered from a mother dog(A) with HI titer of 1 : 1,024 were 1 : 16~1 : 64 at 1 day old before suckling, 1 : 512~1 : 1,024 at 2 days old after suckling, 1 : 512~1 : 2,048 at 1 week old, 1 : 256~l : 1,024 at 2 weeks old, 1 : 128~l : 512 at 3 weeks old, 1 : 128~l : 256 at 4 weeks old, 1 : 32~1 : 128 at 5 weeks old, 1 : 16~1 : 64 at 6 weeks old, 1 : 16~1 : 64 at 7 weeks old, and 1 : 16~l : 32 at 8 weeks old. After vaccination with DHPPL to canine parvovirus in 60 days and 80 days old puppies, 1 : 8~l : 32 at 9 weeks old, 1 :16~1 : 128 at 10 weeks old, 1 : 32~1 : 256 at 11 weeks old, 1 : 16~1 : 256 at 12 weeks old, 1 : 128~1 : 256 at 13 weeks old, 1 : 64~l : 512 at 14 weeks old, and 1 : 128~1 : 512 at 15 weeks old. The HI titers of 7 puppies delivered from mother dog(C) with Hl titer 1 : 1,024 were 1 : 512~1 : 1,024 at 2 days old after suckling, 1 : 256~1 : 1,024 at 1 week old, 1 : 256~l : 1,024 at 2 weeks old, 1 : 64~1 : 512 at 3 weeks old, 1 : 64~1 : 512 at 4weeks old, 1 : 8~l : 64 at 5 weeks old, 1 : 8~1 : 64 at 6weeks old, 1 : 8~1 : 32 at 7 weeks old, and < : 8~1 : 8 at 8 weeks old. Antibody to CPV was transferred mainly from mother to progeny through the colostrum and the transferred maternal antibody was in proportion to the HI titer of the mother As the HI titer of maternal antibody in puppies was low, puppies have a rapid immune response and a massive rise in HI titer to vaccination against CPV compared with puppies haying high level of maternal antibody.

The knowledge and attitudes towards hepatitis B among paramedic students (응급구조학과 대학생들의 B형간염에 대한 지식 및 태도)

  • Purpose: The purpose of this study was to investigate the knowledge and attitudes towards hepatitis B among paramedic students. Methods: A self-reported questionnaire was completed by 395 paramedic students in Daejeon and Chungnam from May 10 to June 10, 2016. The questionnaire consisted of general characteristics of the subjects (9 items), knowledge of hepatitis B (20 items), and attitudes towards hepatitis B (7 items). Data were analyzed by t test, ANOVA, post hoc Duncan test, and Pearson's correlation analysis using SPSS v. 23.0. Results: The knowledge of hepatitis B was 10.52(${\pm}5.18$) out of 20 points and was significantly correlated with grade, vaccination, or antibody-positive irerspective of education. The positive attitude toward hepatitis B was 3.15(${\pm}.43$) and there was a significant correlation with family history irrespective of education. There was a statistically positive correlation between the knowledge and attitudes towards hepatitis B (r=.104, p=.039). Conclusion: Precise knowledge delivery and full understanding of hepatitis B should be incorporated into the education of paramedic students. Active infection control by emergency medical technicians should be maintained by hepatitis B prevention practices.

Expression of cytochrome P-450(CYP)2B1/2 in lymphocytes of workers exposed to toluene and xylene (Toluene과 Xylene 노출 근로자의 림포사이트에서 Cytochrome P-450(CYP)2B1/2의 발현)

  • In order to develop the methods for exposure assessment and find susceptibility markers for the workers who are exposed to low doses of toluene, xylene and other chemical in petroleum industries, we investigated the application of P-450 expression in human lymphocytes utilizing mouse monoclonal anti-rat CYP2B1/2, the levels of toluene and xylene in air and their metabolite levels in urine with the levels of expressed CYP2B1/2 proteins. The general characteristics such as age, smoking and drinking habit were no significant difference between the control and exposed workers, but the working durations and working hours were significantly different. Workers in exposed group were exposed to the mean of 2.1 ppm (range, 0.00-4.54) of toluene and 0.3 ppm (rang, 0.00-1.23) of xylene. The mean concentration of urinary hippuric acid was low and less than 1/5 of the biological exposure index recommended by the Ministry of Employment and Labor Korea. Methyl hippuric acid in urine was not detected in control and exposed workers. Also, there were no significant differences in the levels of the urinary metabolites between the control and exposed group. When chemiluminescence dot blottings were carried out utilizing mouse monoclonal antibody against CYP2B1/2, the strong density dots corresponding to a mouse monoclonal antibody was observed in the human lymphocytes from the exposed workers. These results suggested that the chemiluminescence dot blot assay for CYP of lymphocytes should be valuable for identifying CYP expression as biomarkers in the workers exposed to toluene and xylene.

Severe Anemia Due to Parvovirus Infection Following Treatment with Rituximab in a Pediatric Kidney Transplant Recipient : Anemia after Treatment of Rituximab in Kidney Recipient Patient

  • Rituximab (RTX), a monoclonal antibody against the B-cell marker CD20, is commonly used as a treatment for antibody-mediated diseases or B-lymphocyte-mediated diseases. Destruction of B cells may reverse the disease course in many conditions; however, patients who are treated with RTX cannot respond appropriately to de novo infection due to lack of B lymphocytes. Here, we report one such case. A 7-year-old renal allograft recipient presented with severe anemia due to parvovirus infection after RTX treatment. The patient had focal segmental glomerulosclerosis and had received cadaveric kidney transplantation 6 months previously. She was treated with high-dose steroid for acute rejection and RTX for Epstein Barr Virus infection 3 months previously. At presentation, her hemoglobin level was 5.4 g/dL and leukocyte and platelet counts were normal. She had microcytic normochromic anemia and high viral load of parvovirus B19(70,578 copies/mL). Intravenous immunoglobulin ($200mg/kg{\cdot}d$) treatment controlled the progression of anemia and parvovirus infection. De novo parvovirus infection during the B lymphocyte-depletion period may have precipitated the severe anemia in this case. Close monitoring of infection is required after RTX therapy.

Effect of a Gluten Free Diet on Hepatitis B Surface Antibody Concentration in Previously Immunized Pediatric Celiac Patients

  • Purpose: To evaluate the effect of gluten-free diet (GFD) on hepatitis B surface antibody (HBsAb) concentrations among previously immunized pediatric celiac disease (CD) subjects. Methods: We retrospectively evaluated pediatric CD subjects in serological remission who were previously immunized for hepatitis B virus as infants. The temporal relationship between HBsAb concentration, the amount of time on a GFD, and age were evaluated. Results: Overall, 373 CD subjects were analyzed: 156 with HBsAb sampled prior to GFD initiation and 217 after initiation of a GFD and in serological remission. Median age at HBsAb concentration measurement for those before and after GFD initiation was 5.3 years (interquartile range [IQR], 3.1-9.2 years) and 7.6 years (IQR, 5.4-10.9 years), respectively (p<0.001). There was no sex difference between the groups. The median time of HBsAb measurement was 2 months (IQR, 0-5.7 months) before and 12.8 months (IQR, 5.3-30.3 months) after initiation of GFD. The HBsAb concentration was low in 79 (50.6%) and 121 (55.7%) subjects before and after GFD initiation, respectively (p=0.350). Age was inversely associated with low HBsAb concentrations. Neither being on a GFD nor sex was associated with low HBsAb concentrations. Conclusion: Adherence to a GFD does not affect HBsAb concentration in children with CD. Age is inversely associated with HBsAb concentration.

Primary Structure of the Human VkII Regions Elicited by Haemophilus influenzae Type b Polysaccharide Vaccines; The J Gene Usage Is Restricted in Child Antibodies Using the A2 Gene

  • The Haemophilus influenzae type b (Hib) has been a major cause of bacterial meningitis in children who are less than two years old. The variable (V) region repertoire of adult Caucasian antibodies (Abs) to Hib polysaccharide (PS) has been characterized well. The majority of adult antibodies against Hib uses VL that is derived from the Vk gene A2 and have arginine at the N region. In order to explore the possibility those antibody responses to Hib-PS is variable in various age groups, we examined the VL regions of the antibodies to Hib-PS in Korean adults and children. We immunized Korean adults (n = 8) and children (n = 39) with Hib tetanus conjugated vaccines, isolated RNAs from the peripheral lymphocytes, and amplified the A2-derived VL regions by RT-PCR. The PCR products were subcloned and sequenced. Forty-seven out of 54 independent clones from children used the $J{\kappa}2$, or $J{\kappa}3$ gene in preference. The adults, however, used all of the $J{\kappa}$ genes evenly. With respect to the amino acid sequences of variable regions, adult $A2-J{\kappa}$ recombinants have a germline sequence. But, the 76th codon (AGC) of child $A2-J{\kappa}2$ recombinants was substituted with CGC (arginine) in most cases (88 %) and 77 percent of child clones using the $A2-J{\kappa}3$ genes have isoleucine-109 at the junction of $J{\kappa}-C{\kappa}$ instead of threonine that is found in a germline sequence. These results suggest that the mechanism of antibody production in young children is different from that of adults.

  • This research chose three kinds of sleeping bags with different fillings (A: interfill polyester, B: brushed polyester, C: down) and put them in two environmental conditions (Env. I : $25{\pm}1^{\circ}C$, $50{\pm}10%$; Env. II : $18{\pm}1^{\circ}C$, $70{\pm}10%$). The measured Skin temperature, Temperature and Humidity in sleeping bag, Rectal temperature and Psychological reaction. The results of experiment were as follows: 1. The mean skin temperature of Sleeping Bag B was the closest to the most comfortable value in the two environmental conditions. 2. Temperature and humidity in sleeping bags, in both environmental conditions, contained the value nearest to the most comfortable one in Sleeping Bag B. 3. Rectal temperature in both environments had a slight change in the range of antibody temperature in all the subjects of sleeping bags. 4. As subjective psychological reaction, both thermal and humid sensation approached the most comfortable zone in Sleeping Bag B.
