Enhanced Anthocyanin Accumulation by UV-B and JA Treatment in Cell Suspension Culture System of Grope (Vitis vinifera L.)

  • Effects of jasmonic acid treatment, UV-B and white light treatment on the anthocyanin biosynthesis and cell growth were investigated using the cell suspension culture system of grape (Vitis vinifera L.). Cell growth was not affected by white light irradiation, while it was remarkably suppressed by UV-B irradiation from 8 to 32 h. Anthocyanin accumulation dramatically increased after 16 h from irradiation of UV-B. Simultaneous treatment of jasmonic acid and UV-B increased anthocyanin accumulation by 10-fold. The cell division was restored when anthocyanin was abundantly accumulated after 32 h from UV-B irradiation. Optimum concentration of jasmonic acid was found to be 5 uM for maximum accumulation of anthocyanin. Application of jasmonic acid to grape suspension cells rapidly induced the expression of CHS gene after 2 h from treatment and showed maximum level at 32 h. Simultaneous treatment of jasmonic acid and light also induced CHS gene expression after 2 h, but the maximum level of CHS transcript was observed at 16 h with white light and 8 h with UV-B exposure. The synergistical effects could be explained by the defense mechanism that UV irradiation is mediated in part by alterations in JA and its signaling pathway.

Action Spectra of Apoptosis Induction and Reproductive Cell Death in L5178Y cells in UV-B Region

  • It is important to determine the action spectrum of UV-B radiation contained in the sunlight to estimate the risk of skin cancer. We have investigated action spectra for induction of apoptosis and reproductive cell death in L5178Y cells using the Okazaki Large Spectrograph at NIBB. L5178Y cells were exposed to light at different wavelengths in UV-B or UV-A region. Frequencies of apoptosis induction and reproductive cell death were determined by counting cells with chromatin condensation, and by the colony formation assay, respectively. The measured sensitivity spectra for the two end-points were in very good agreement. Sensitivity decreased steeply with increase of wavelength in UV-B region and remains nearly constant in UV-A region. The action spectra were also slightly steeper than that for the minimum erythematic dose (MED), but very similar to the light absorption spectrum of DNA in UV-B region. On the other hand, the spectra for both endpoints were similar to MED spectrum but not DNA spectrum in the UV-A region. Also different time-course and morphological difference of apoptosis were found between UV-B (long time, fragmentation) and UV-A (short time, shrinkage) region. These results suggest that DNA damage induced by UV-B light triggers apoptosis and reproductive cell death, but other damaged targets (membrane, protein and so on) trigger these effects in UV-A region.

  • To identify nongenotoxic carcinogen determined as negative by ICH guideline-recommended standard genotoxicity test battery; Ames test, chromosome aberration assay, mouse lymphoma $tk^{+/-}$ assay, in vivo micronucleus assay, we picked bisphenol A as a model compound. In this study, we applied in vitro BalB/c 3T3 cell transformation assay and Syrian hamster embryo (SHE) cell transfarmation assay. Bisphenol A was treated upto $769.2 ug/m{\ell}$ in BalB/c 3T3 cells and upto $125 ug/m{\ell}$ in SHE cells. bisphenol A didn't induced morphological transformation both with one stage treatment protocol and with two stage treatment protocol. But, treated far 48 hr, Bisphenol A induced morphological transformation significantly in SHE cells.

Presence of Foxp3-expressing CD19(+)CD5(+) B Cells in Human Peripheral Blood Mononuclear Cells: Human CD19(+)CD5(+)Foxp3(+) Regulatory B Cell (Breg)

  • Foxp3 is a transcript factor for regulatory T cell development. Interestingly, Foxp3-expressing cells were identified in B cells, especially in CD19(+)CD5(+) B cells, while those were not examined in CD19(+)CD5(-) B cells. Foxp3-expressing CD5(+) B cells in this study were identified in human PBMCs and were found to consist of $8.5{\pm}3.5%$ of CD19(+)CD5(+) B cells. CD19(+)CD5(+)Foxp3(+) B cells showed spontaneous apoptosis. Rare CD19(+)CD5(+) Foxp3(+) regulatory B cell (Breg) population was unveiled in human peripheral blood mononuclear cells and suggested as possible regulatory B cells (Breg) as regulatory T cells (Treg). The immunologic and the clinical relevant of Breg needs to be further investigated.

In Vitro Evolution of Lipase B from Candida antarctica Using Surface Display in Hansenula polymorpha

  • Lipase B from Candida antarctica (CalB) displayed on the cell surface of H. polymorpha has been functionally improved for catalytic activity by molecular evolution. CalB was displayed on the cell surface by fusing to a cell-wall anchor motif (CwpF). A library of CalB mutants was constructed by in vivo recombination in H. polymorpha. Several mutants with increased whole-cell CalB activity were acquired from screening seven thousand transformants. The two independent mutants CalB 10 and CalB 14 showed an approximately 5 times greater whole-cell activity than the wild-type. When these mutants were made as a soluble form, CalB 10 showed 6 times greater activity and CalB 14 showed an 11 times greater activity compared with the wild-type. Sequence analyses of mutant CALB genes revealed amino acid substitutions of $Leu^{278}Pro$ in CalB10 and $Leu^{278}Pro/Leu^{219}Gln$ in CalB14. The substituted $Pro^{278}$ in both mutants was located near the proline site of the ${\alpha}$10 helix. This mutation was assumed to induce a conformational change in the ${\alpha}$10 helix and increased the $k_{cat}$ value of mutant CalB approximately 6 times. Site-directed mutagenized CalB, LQ ($Leu^{219}Gln$) was secreted into the culture supernatant at an amount of approximately 3 times more without an increase in the CalB transcript level, compared with the wild-type.

B-ISDN UNI에서 폭주를 예방하기 위한 B-NT의 트래픽 흐름 제어 (Traffic Flow Control of B-NT for Prevention of Congestion in B-ISDN UNI)

  • 본 논문에서는 망 노드에서의 폭주를 B-ISDN UNI에서 예방하기 위해 B-NT에서 일시적인 셀 저장 및 선택적 셀 폐기를 수행하는 트래픽 흐름 제어를 제안하였다. 제안된 구조는 B-NT 시스템에서 T 접속을 향하는 출력 셀 흐름을 감소 또는 억제시키도록 구성하고, 인접 망 노드의 상태를 정상, 준폭주, 폭주의 세상태로 정의하였다. 준폭주 상태에서는 손실에 민감한 트래픽은 일시적으로 저장되어 출력 셀 흐름의 속도를 저하시키고, 폭주 상태에서는 손실에 민감한 트래픽의 셀 저장과 더불어 지연에 민감한 트래픽의 셀이 선택적으로 폐기되어 최대한 출력 트래픽 흐름을 억제시킨다. 입력 셀 흐름과 망 노드 상태의 변화를 IBP와 3 상태 마르코프 체인으로 모델링하여 B-NT 시스템에서 제안된 구조의 성능 분석을 위한 시뮬레이션을 수행하여 적당한 버퍼 사이즈를 구하고 망 노드의 상태에 따른 제안한 구조의 성능 변화를 조사한 결과 망 노드에서의 폭주 정도가 극심한 경우에는 제안한 방법으로 제어가 거의 불가능하지만, 그 외의 경우에는 2,000셀 이상의 버퍼 사이즈로 제어가 가능함을 알 수 있었다.

Lack of Increased P15INK4B Protein Expression in Basal Cell Carcinomas

  • Background: The basal cell carcinoma (BCC) is the most common non-melanoma skin cancer (NMSK). BCC might develop because of the faulty cell cycle arrest. $p15^{INK4b}$ is a tumor suppressor gene, involved in cell cycle arrest and inactivated in most human cancers. The role of $p15^{INK4b}$ protein expression in the genesis of BCC is as yet unknown. In a previous study we showed the absence of $p15^{INK4b}$ expression in the majority of tissue microarray cores of cutaneous squamous cell carcinoma (SCCs), another type of non-melanoma skin cancer, indicating that $p15^{INK4b}$ could possibly be involved in the pathogenesis of cutaneous SCC. The aim of this study was to investigate $p15^{INK4b}$ protein expression in BCCs. Materials and Method: Protein expression of $p15^{INK4b}$ in 35 cases of BCC tissue arrays and 19 cases of normal human skin tissue was studied using an immunohistochemical approach. Results: The expression of $p15^{INK4b}$ was not significantly different in the BCC cases as compared with normal human skin (p=0.356; p>0.05). In addition, there were no significant relationship between clinicopathologic variables of patients (age and sex) and $p15^{INK4b}$ protein expression. Conclusions: Our finding may indicate that $p15^{INK4b}$ protein expression does not play a role in the genesis of BCC.

Enterococcus faecalis 2B4-1 세포벽 성분 중 Polysaccharide Fraction의 종양세포 증식억제 효과 (Growth Inhibition of Polysaccharide Fraction in Cell Wall Components from Enterococcus faecalis 2B4-1 against Tumor Cell Lines)

  • This study was developed to evaluate the growth inhibition effects of cell wall components of Enterococcus faecalis 2B4-1 obtained from feces of neonates against tumor cell lines. Polysaccharide fraction (PS) shown sensitive growth inhibition effect in the cell wall components was isolated and characterized. In growth inhibition effects, residue fractin of whole cell was shown sensitive level of percent survival about 30% when administrated at ehe concentration of 100${\mu}$g/ml, and that was more effective than that of supernatant fraction against the tumor cell lines, SNU-1, 3LL, FARROW and HEC-1-B. Sensitive growth inhibition effects against SNU-1, FARROW and HEC-1-B were performed by whole cell (WC) fraction from Ent. faecalis 2B4-1. Cytoplasm fractin (CP) of WC was shown non-inhibition effect, however, the other part of WC, precipitate of disrupted cell (PD), was sensitive against the tumor cell line mentioned above. Followed by separation to peptidoglycan fraction (PG) and polysaccharide fraction (PS) were all sensitive which the latter was shown more sensitive percent survival than the former. Composed sugars of polysaccharide fraction were determined to D-glucose, L-rhamnose and D-glucosamine, and the rate fo composition was calculated to about 1:1:1 by the data of elemental analysis, IR, TLC and HPLC.

운동 강도와 에너지소비량에 따른 단기간의 유산소운동이 면역세포에 미치는 영향 (Effects on Immune Cell of Short-term Aerobic Exercise by Exercise Intensity and Expenditures Calorie)

  • 본 연구는 적정 운동 강도 중 VO2max 50%와 70%의 운동 강도로 운동을 실시할 때, 각각 300kcal, 600kcal를 소비하는 시점에서 유발되는 면역세포의 변화를 분석함으로써 인체 면역 세포의 긍정적 변화와 면역력 강화를 위한 적정 운동방법을 규명하고자 남자학생 8명을 대상으로 GXT로 사전검사를 실시하여 최대운동검사를 통하여 측정된 피험자 개개인의 VO2max를 기준으로 각각 50%, 70%에 해당되는 산소섭취량과 경사도 및 속도를 산출하고, 산소섭취량(ml/min/kg)의 METs 및 칼로리 소비량을 대입, 전체 300kcal와 600kcal가 소모되는 운동시간을 산출하였다. 유산소운동에 따른 T, B, NK cell의 변화는 림프구 전체에서 T, B, NK 림프구가 차지하는 상대적 비율이 운동 강도에서는 유의한 차이가 없었고, T cell에서 에너지소비량(p<.01), 상호작용(p<.05)효과에서, B cell은 에너지소비량(p<.01)에서, NK cell은 에너지소비량(p<.001), 상호작용(p<.05)효과에서 통계적으로 유의한 것으로 나타났다.

Plumbagin from Plumbago Zeylanica L Induces Apoptosis in Human Non-small Cell Lung Cancer Cell Lines through NF-κB Inactivation

  • Objective: To detect effects of plumbagin on proliferation and apoptosis in non-small cell lung cancer cell lines, and investigate the underlying mechanisms. Materials and Methods: Human non-small cell lung cancer cell lines A549, H292 and H460 were treated with various concentrations of plumbagin. Cell proliferation rates was determined using both cell counting kit-8 (CCK-8) and clonogenic assays. Apoptosis was detected by annexin V/propidium iodide double-labeled flow cytometry and TUNEL assay. The levels of reactive oxygen species (ROS) were detected by flow cytometry. Activity of NF-${\kappa}B$ was examined by electrophoretic mobility shift assay (EMSA) and luciferase reporter assay. Western blotting was used to assess the expression of both NF-${\kappa}B$ regulated apoptotic-related gene and activation of p65 and $I{\kappa}B{\kappa}$. Results: Plumbagin dose-dependently inhibited proliferation of the lung cancer cells. The IC50 values of plumbagin in A549, H292, and H460 cells were 10.3 ${\mu}mol/L$, 7.3 ${\mu}mol/L$, and 6.1 ${\mu}mol/L$ for 12 hours, respectively. The compound concentration-dependently induced apoptosis of the three cell lines. Treatment with plumbagin increased the intracellular level of ROS, and inhibited the activation of NK-${\kappa}B$. In addition to inhibition of NF-${\kappa}B$/p65 nuclear translocation, the compound also suppressed the degradation of $I{\kappa}B{\kappa}$. ROS scavenger NAC highly reversed the effect of plumbagin on apoptosis and inactivation of NK-${\kappa}B$ in H460 cell line. Treatment with plumbagin also increased the activity of caspase-9 and caspase-3, downregulated the expression of Bcl-2, upregulated the expression of Bax, Bak, and CytC. Conclusions: Plumbagin inhibits cell growth and induces apoptosis in human lung cancer cells through an NF-${\kappa}B$-regulated mitochondrial-mediated pathway, involving activation of ROS.
