Study on Atractylodes Rhizomes (I) : Pharmacognosy and Chemical Identification of Atractylodes Species (출류(朮類)에 관한 연구( I ) : 한국(韓國)과 일본(日本)의 창출(蒼朮)과 백출(白朮) 의 생약학적(生藥學的) 연구)

  • Traditional Chinese herbal drugs, namely, Atractylodes Rhizome and Atractylodes Rhizome White were identified by means of chemical analysis such as TLC and GC/MS. In Korea, the old-grown and the newly-grown rhizomes of Atractylodes japonica Koidzumi (Compositae) are used as the Atractylodes Rhizome and the Atractylodes Rhizome White, respectively. Both rhizomes contain the atractylon asa major spot on TLC. The content of atractylon in a newly-grown rhizome of A. japonica (Atractylodes Rhizome White called in Korea) appears to contain much larger quantity in comparison with that of an old-growon one (Atractylodes Rhizome). The TLC spot pattern analysis showed that rhizomes of A. japonica and A. macrocephala contain the atractylon as a major indicator spot, but no atractylodin (extreamly small amount). However, rhizomes of A. lancea De Candolle and A. koreana Kitamura contain the atractylodin as a major indicator spot.

DNA Sequencing and Phylogenetic Analysis of the 18S rRNA Gene of Atractylodes japonica Koidz and Analysis of Atractylon (삽주의 18S rRNA 유전자의 염기서열 결정, 계통분류학적 분석 및 atractylon 분석)

  • The region containing 18S rRNA gene, ITS 1 and part of the 5.8S rRNA gene of the Atractylodes japonica Koidz was amplified by PCR and the product cloned in a pBluescript SK II plasmid. DNA sequence of the cloned DNA was determined and submitted to the GenBank (accession number EU678363). Phylogenetic analysis of the ITS 1 DNA showed close similarity with the other plant species of the family Compositae. The extract of the plant materials of five different members of the family Compositae was analyzed by HPLC to detect atractylon. Extract of the A. japonica Koidz showed presence of significant amount of atractylon. However, noticeable amount of atractylon was not detected by the same analyses from the extracts of the other plants belonging to the family Compositae including Artemisia capillaris, Chrysantemum zawadskii, Eclipta prostrata or Taraxacum platycarpum.

  • From the rhizome of Atractylodes japonica Koidzumi (Compositae) which is the original plant of oriental medicine 'Cang-Zhu', four essential oil compounds were isolated. Three of them were identified as atractylon, hydroxyatractylon and $5{\alpha}H,\;10{\beta}-selina-4(14),\;7(11)-diene-8-one$, which were already known as the constitutents of Atractylodes Rhizoma. The fourth is a novel polyacetylene type compound, and the structure is postulated as 1, 4-diacetoxytetradeca-6, 12-diene-8, 10-diyne by the spectral data.

  • Traditional herbal drugs of Korea, namely, Atractylodes Rhizome(蒼朮) and Atractylodes Rhizome White(白朮) which are derived from and old-grown rhizome and a newly-grown rhizome of Atractylodes japonica Koidz.(Compositae), respectively, were investigated their potential liver-protective activities against hepatotoxicity induced by $CCl_4$ intoxication in mice. Each herbal drug was extracted separately with chloroform, methanol and butanol. No extract of Atractylodes Rhizome showed a significant reduction of the duration of hypnosis produced by hexobarital after $CCl_4$ intoxication in mice. In addition, the the treatments of Atractylodes Rhizome extracts produced no decrease of aspartate aminotransferase(EC 2.6.1.1) activity in serum. However, the methanol extract of Atractylodes Rhizome White exhibited a marked protection from hepatotoxicity induced by $CCl_4$ intoxication in mice. It produced significant reductions of the duration of hypnosis and serum enzyme activity, but no other extract showed liver-protective activity against $CCl_4-induced$ hepatotoxicity.

  • Methanol extracts of the roots of Atractylodes japonica (alba) (Compositae) and Gentiana scabra (Gentianaceae) were further fractionated with n-butanol, chloroform, n-hexane and water. Then each fraction was p.o. administered to an animal model of hepatitis caused by $CCl_4$ intoxication. Measurements of sleeping time of mice induced by hexobarbital administration indicated that n-butanol and chloroform fractions of Atractylodes japonica (alba), and chloroform and n-hexane fractions of Gentiana scabra showed significant liver protective activities. Measurements of the activities of serum transaminase, S-GOT and S-GPT appeared to support these results.

  • The rhizome extract of Atractylodes japonica Koidzumi(Compositae) exhibited a particular inhibition on the proliferation of cultured human tumor cell lines, in vitro. Thus, the intensive phytichemical investigation of the MeOH extract of Atractylodes japonica have been conducted by the way of activity-guided purification. The repeated column chromatographic separation of the n-hexane soluble part of extract resulted in the isolation of four sesquiterpenes (1-4) and a polyacetylene component (5). Chemical structures of them were identified as atractylon (1), atractylenolide Ⅰ(2), atractylenolide Ⅲ(3), eudesma-4(15),7(11)-dien-8-one (4) and 1,3-diacetyl-atractylodiol (5) by spectroscopic means. Among the isolates, compound 2-4 were shown to give moderate inhibitory effect in a dose dependent manner on the proliferation of cultured human tumor cell lines such as A549 (non small cell lung), SK-OV-3 (ovary), SK-MEL-2 (melanoma), XF498 (central nerve system) and HCT 15(colon), respectively.

Antioxidant and Anti-inflammatory Activities of Atractylodes japonica According to Extract Methods (백출 용매추출 방법에 따른 항산화 활성 및 항염증 효과)

  • Atractylodes japonica has been widely used in a traditional Korean herbal medicine exerting various pharmacological activities such as diauretic action, asriction, anti-allergy, neuroprotective activity, anti-cancer, immunomodulation and gastrointestinal protective effect. This study was to investigate the antioxidant, nitric oxide and inflammatory cytokines production of A. japonica extract by water and 70% ethanol. DPPH and ABTS free radical scavenging activity were increased in a dose-dependent manners with both extracts and there was no difference with extract solvents. 70% ethanol extract of A. japonica showed a very strong inhibitory effect on NO production. Both extracts of A. japonica significantly reduce the expression of iNOS and COX-2 proteins involved in NO prodction. A. japonica extract by water and 70% ethanol inhibited LPS-induced proinflammatory cytokines such as IL-6 and IL-1b. In this study, 70% ethanol extract of A. japonica significantly suppresses LPS-induced NO and inflammatory cytokine production. Therefore it can be widely used to treat and improve inflammatory diseases.

Studies of Taxonomic Origins of Atractylodis Rhizoma Alba and Atractylodis Rhizoma (백출과 창출의 기원에 대한 식물분류학적 연구)

  • The plant origines of Atractylodis Rhizoma Alba and Atractylodis Rhizoma are genus Atractylodes*Compositae). Their origines in pharmacopoeia are slightly different among Korea, Japan, North Korea and China. The species of Atractylodis Rhizoma Alba are Atrctylodes japonica $K_{OIDZ}$. A. ovata $T_{HUNB}$.. Atractylodis Rhizoma are A. lancea DC., A. chinensis DC. and A. koreana $K_{ITM}$. But the texonomic genealogy of Atractylodis Rhizoma Alba is different from that of Atractylodis Rhizoma. The herbal origines of Atractylodis Rhizoma Alba and Atractylodis Rhizoma were not different at the early days. Since Tao-Hong-Jing(陶弘景) distinguished between Atractylodis Rhizoma Alba and Atractylodis Rhizoma at the very beginning, theri medicinal usages are different, Atractylodis Rhizoma have been used to remove pathogenic dampness in the digestive organs and Atractylodis Rhizoma Alba have been used to invigorate the digestive system and replenish qi. In additions, the figures and historical records of herbal appearance are different between Atractylodis Rhizoma Alba and Atractylodis Rhizoma. The diagnostics of them are in accord with the texonomic genealogies.

  • The investigation was made about distribution and ecological characteristics of Rhododendron mucronulatum var. mucronulatum, R. dauricum and R. schlippenbachii on Mt. Changbai in China. The Ericaceae plants are known to be highly ornamental and easy to cultivate. The results of investigations are like followings. The plant species diversity was the highest in habitats of R. mucronulatum var. mucronulatum, followed by those of R. schlippenbachii and R. dauricum. R. mucronulatum var. mucronulatum community was found in the northwest exposure areas of 300m in altitude. Corylus heterophylla, Carex humilis var. nana, Atractylodes japonica species also grew in the R. mucronulatum var. mucronulatum community. R. dauricum communities were widely distributed in the west exposure areas of 1226-1860m above sea level. R. schlippenbachii populations were found in areas of under 300m above sea level, which has dry soil. But humidity of air was relatively high in the place. Artemisia japonica, Lespedeza bicolor and Dactylis glomerata were found around the habitats of R. schlippenbachii. Phalanx type of plants were dominant near the habitats of Ericaceae plants as 61%. Further, there appeared higher plant life totalling to 72 taxa comprising 54 species, 16 varieties and 2 forma with 31 families and 59 genuses at the whole survey area. Among these, Compositae plants excellent in resistance to environment was surveyed the most, accounting for 30.0%.
