Therapeutic effects of dihydroartemisinin and transferrin against glioblastoma

  • BACKGROUND/OBJECFTIVES: Artemisinin, a natural product isolated from Gaeddongssuk (artemisia annua L.) and its main active derivative, dihydroartemisinin (DHA), have long been used as antimalarial drugs. Recent studies reported that artemisinin is efficacious for curing diseases, including cancers, and for improving the immune system. Many researchers have shown the therapeutic effects of artemisinin on tumors such as breast cancer, liver cancer and kidney cancer, but there is still insufficient data regarding glioblastoma (GBM). Glioblastoma accounts for 12-15% of brain cancer, and the median survival is less than a year, despite medical treatments such as surgery, radiation therapy, and chemotherapy. In this study, we investigated the anti-cancer effects of DHA and transferrin against glioblastoma (glioblastoma multiforme, GBM). MATERIALS/METHODS: This study was performed through in vitro experiments using C6 cells. The toxicity dependence of DHA and transferrin (TF) on time and concentration was analyzed by MTT assay and cell cycle assay. Observations of cellular morphology were recorded with an optical microscope and color digital camera. The anti-cancer mechanism of DHA and TF against GBM were studied by flow cytometry with Annexin V and caspase 3/7. RESULTS: MTT assay revealed that TF enhanced the cytotoxicity of DHA against C6 cells. An Annexin V immune-precipitation assay showed that the percentages of apoptosis of cells treated with TF, DHA alone, DHA in combination with TF, and the control group were $7.15{\pm}4.15%$, $34.3{\pm}5.15%$, $66.42{\pm}5.98%$, and $1.2{\pm}0.15%$, respectively. The results of the Annexin V assay were consistent with those of the MTT assay. DHA induced apoptosis in C6 cells through DNA damage, and TF enhanced the effects of DHA. CONCLUSION: The results of this study demonstrated that DHA, the derivative of the active ingredient in Gaeddongssuk, is effective against GBM, apparently via inhibition of cancer cell proliferation by a pharmacological effect. The role of transferrin as an allosteric activator in the GBM therapeutic efficacy of DHA was also confirmed.

  • In the present study, we investigated anti-inflammatory activity of artemisinic acid in HaCaT cells and RAW264.7 cells. Artemisinic acid showed inhibitory activity on macrophage-derived chemokines (MDC) expression, a factor related with atopic dermatitis (AD), in interferon (IFN)-${\gamma}$ and tumor necrosis factor (TNF)-${\alpha}$-stimulated HaCaT cells. In the study on action mechanism, pretreated artemisinic acid reduced the phosphorylation of STAT1 and p38 and the degradation of $I{\kappa}B$ by IFN-${\gamma}$ and TNF-${\alpha}$ stimulations. However, artemisinic acid didn't show the inhibitory activity on LPS-induced inflammatory mediators (NO, $PGE_2$, IL-6) in RAW264.7 cell. These results indicate that artemisinic acid inhibits IFN-${\gamma}$ and TNF-${\alpha}$-induced MDC expression through inhibition of signal factors, STAT1, NF-${\kappa}B$, and p38, in HaCaT keratinocytes.

Effect of Gaeddongssuk (Artemisia annua L.) Powder on Quality and Shelf Stability of Emulsion Sausages during Refrigerated Storage

  • The objective of this study was to evaluate effects of Gaeddongssuk powder (GP) on quality characteristics and shelf stability of emulsion sausages during storage. Proximate composition properties showed no significant differences in all treatment (p>0.05). Control showed the highest cooking loss while the treatment with GP showed decreased cooking loss depending on increasing GP content (p<0.05). Apparent viscosity of batter was increased as the amount of GP increased, whereas hardness of emulsion sausages was decreased with increasing GP level. In sensory evaluation, emulsion sausage with 0.1% GP resulted in the highest score in overall acceptability. The pH values of all treatments decreased at the early storage stage, followed by gradual increase. The lightness and redness of treatments were decreased when the level of GP was increased. However, the yellowness of sausages with GP were higher than that of control (p<0.05). The addition of GP inhibited lipid oxidation of emulsion sausages during storage depending on its level. The aerobic bacteria population and VBN was unaffected by addition of GP during the storage (p>0.05). Therefore, Gaeddongssuk powder up to 0.1% has a potential as a natural antioxidant for meat products because it can inhibit lipid oxidation of sausages without decreasing their sensory properties.

온도 조건에 따른 개똥쑥(Artemisa annua L.) 종자의 발아특성 (The Germination Characteristics of Seeds by Temperature Conditions in Artemisa annua L.)

  • 본 실험은 다양한 생리활성이 보고되어 약용으로 널리 쓰이는 개똥쑥(Artemisia annua L.) 종자의 온도에 따른 발아 특성을 조사하기 위해 진행하였다. 미세온도구배 발아기를 이용하여 낮과 밤의 온도를 각각 5 ~ 35℃ 범위에서 낮과 밤의 시간을 12시간으로 고정하고, 낮 온도가 밤 온도보다 크거나 같은 조건을 설정하여 총 27개의 온도조건으로 개똥쑥 종자의 최종 발아율 및 발아율과의 관계를 분석하였다. 실험결과, 개똥쑥 종자는 실험에 사용한 모든 온도조건에서 발아가 가능한 것으로 나타났으며, 25/10℃(낮/밤) 조건에서 90%로 가장 높게 조사되었다. 또한, 발아율 조사결과를 통해 일평균온도뿐만 아니라 일교차온도도 발아율에 영향을 미치는 것으로 판단되어 일평균온도와 일교차온도로 나누어 발아율과의 관계를 분석하였다. 온도조건과 발아율과의 연속적인 발아특성을 분석하기 위해 다중회귀분석과 비선형 회귀분석을 이용하여 온도 조건과 상대적 발아율의 관계를 수식으로 표현하였다. 일평균온도를 기준으로 발아율과의 관계를 분석한 결과, 5 ~ 35℃의 모든 일평균 온도범위에서 유의성이 나타났으며 5, 7.5, 32.5, 35℃는 상대적인 음의 영향력을, 나머지 조건에서는 상대적인 양의 영향력을 가진 것으로 분석되었다. 일교차온도를 기준으로 발아율과의 관계를 분석한 결과, 0 ~ 25℃의 모든 일교차 온도범위에서 양의 영향력을 가진 것으로 분석되었다. 일평균온도는 19.3℃에 가까울수록, 일교차온도는 14.9℃에 가까울수록 발아에 대한 영향력이 큰 것으로 조사되었다. 각 수식을 통해 도출된 수치화된 온도에 따른 개똥쑥 종자의 일일누적 온도 영향력을 temperature score(TS)로 설정하였다. 본 연구에서 도출된 수식을 통해 누적 TS를 계산한 결과, 14.9 TS가 누적되었을 때 발아율이 85% 이상으로 나타날 것으로 예측되었다.

약초 추출액의 닭콕시듐 Eimeria tenella 대한 항콕시듐 효과 (Anticoccidial Effect of Herb Extracts against Eimeria tenella)

  • Ionophorous antibiotics have been used popularly for the protection of avian coccidiosis, though Halo-fuginone which is derived from an extract of the Dichroa febrifuga, was developed as an antimalarial and anticoccidial agent. The antibiotics are regarded as the causes of residues in the avian products, therefore the author has tried to find out more safe herbal materials for the control of avian coccidiosis. Thus, the extracts of 5 kinds of herbs, roots of Pulsatilla koreana Nakai and Sophora flavescens Aiton, nuts of Quisqualis indica Linne, whole herbs of Artemisia annua Linne and Polygonum aviculare Linne, were investigated on the efficacy against E tenella. Survival rates, bloody diarrhea, lesion scores, body-weight gains and feed conversion rate were investigated at the 1st and the 2nd week after infection. The bloody diarrhea in the groups treated with P koreana, A annua, Q indica and P aviculare were milder than those in the groups treated with S flavescens and infected control. The lesion score in the groups treat.4 with herb extracts(2.00$\pm$0.82~2.65$\pm$0.89) were lower than those in the control group (2.80$\pm$0.7l). But, there was not significant in all infected groups. The body weight gai in the groups treated with P koreana, S flavescens, A annua and Q indica(291.74$\pm$15.76~303.43$\pm$51.90) were higher than that in the control group (283.71$\pm$16.53) but there was not significant. In a conclusion, analyzing the data of the survival rates, bloody diarrheal symptoms, lesion scores, body weight gains and oocyst excretions, the extract of S flavescens, P koreana, A annua and Q indica were effective. The further research on the above herbal materials will have been carried out by the author and the other researchers by means of the chemical analysis of the components.

자생식물 Essential Oil 5 종의 항 아토피피부염 활성 연구 (A Study on the Activities of Five Natural Plant Essential Oils on Atopic Dermatitis)

  • 본 연구는 개똥쑥(Artemisia annua L., AA), 유자(Citrus junos Sieb. ex TANAKA, CJ), 산국화(Chrysanthemum boreale Makino, CB), 잣나무(Pinus koraiensis, PK), 금강송(Pinus densiflora for. erecta, PD) 총 5 종의 자생식물 essential oil의 항 아토피 효능을 확인하기 위한 실험이다. 항균 효과를 확인하기 위해 자생식물 essential oil 5 종을 Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Candida albicans 총 4 종류의 균에 처리했다. 항염증 효과를 확인하기 위해 TNF-α와 IFN-γ (TI)를 처리한 HaCaT 세포에 5 종의 자생식물 essential oil을 처리했다. AA, CJ, CB, PK, PD은 10 mg/mL 농도에서 Candida albicans에 대한 항균 효과를 나타냈다. AA (1 ㎍/mL), CB (1 ㎍/mL), PK (0.1, 1 ㎍/mL)에서 thymus and activation-regulated chemokine (TARC) 생성량이 억제되었고, AA 및 PK (1 ㎍/mL)에서 macrophage-derived chemokine (MDC) 생성량이 억제되었으며, AA (0.1, 1 ㎍/mL), PK (0.1, 1 ㎍/mL)에서 IL-6 생성량이 억제되었다. AA, CB, PK essential oil의 항균 및 항염증 효과가 확인되었으며, 이에 아토피 피부염 완화에 기여할 수 있음을 기대한다.

Cloning and Expression of a Farnesyl Diphosphate Synthase in Centella asiatica (L.) Urban

  • A cDNA encoding farnesyl diphosphate synthase (FPS; EC2.5.1.1/EC2.5.1.10) was isolated from Centella asiacita (L.) Urban, using degenerate primers based on two highly conserved domains. A full-length cDNA clone was subsequently isolated by rapid amplification of cDNA ends (RACE) PCR. The sequence of the CaFPS (C. asiatica farnesyl diphosphate synthase) cDNA contains an open reading frame of 1029 nucleotides encoding 343 amino acids with a molecular mass of 39.6 kDa. The deduced CaFPS amino acid sequence exhibits 84, 79, and 72%, identity to the FPSs of Artemisia annua, Arabidopsis thaliana, and Oryza sativa, respectively. Southern blot analysis suggested that the C. asiatica genome contains only one FPS gene. An artificially expressed soluble form of the CaFPS was identified by SDS-PAGE. It had high specific activity and produced farnesyl diphosphate as the major isoprenoid.

Lactobacillus plantarum DK119로 발효한 약용식물 당침액의 특성 (Characteristics of sugar extracts of medicinal plants fermented with Lactobacillus plantarum DK119)

  • 본 연구에서는 약초 당침 추출액의 발효특성을 분석하고 관능적 특성을 개선할 수 있는 적정 발효조건을 도출하여 유산균 발효 약초음료 개발에 응용하고자 하였다. 설탕을 이용하여 1년 숙성, 추출한 개똥쑥, 어성초 및 삼백초 당침액($50{\pm}5^{\circ}Bx$)을 10, 15 및 $20^{\circ}Bx$로 희석한 후 김치로부터 분리한 L. plantarum DK119 ($5{\times}10^9CFU/mL$)를 2% (v/v) 접종하고 $37^{\circ}C$에서 4일 동안 발효하면서 유산균 수, pH, 적정산도 및 환원당 함량 변화를 분석한 결과 개똥쑥은 $20^{\circ}Bx$, 어성초는 $10^{\circ}Bx$, 그리고 삼백초는 $15^{\circ}Bx$가 발효 가능한 농도로 나타났다. 즉, 희석농도와 접종 유산균 수가 유사한 조건에서도 사용한 약초 종류에 따라 발효특성에 차이가 있음을 알 수 있다. 발효 기간 중 개똥쑥, 어성초 및 삼백초 희석액의 총폴리페놀 함량은 모든 희석액에서 변화가 없는 반면 DPPH radical 제거능과 환원력(FRAP)은 발효 2일에 유의적 증가를 보여 발효 중 항산화 활성에 영향을 미치는 발효 산물이 생성되는 것으로 추정된다. 관능검사 결과, 발효 전 당침액에서 나는 특유의 불쾌한 약초 맛이 발효 1일부터 감소하고 종합적 기호도가 발효 2-3일에 높아져 L. plantarum DK119를 이용한 유산균 발효는 약초 당침액의 항산화 활성을 증가시키고 관능적 품질특성을 개선시켜 약초 추출물을 이용한 건강 음료 개발에 응용 가능한 것으로 사료된다.

반응표면분석을 이용한 흑마늘과 개똥쑥 혼합음료 개발을 위한 추출조건의 최적화 (Optimization of Extraction Conditions for Mixing Beverage Development of Black Garlic and Gaeddongssuk by Response Surface Methodology)

  • 흑마늘과 개똥쑥을 반응표면분석법에 따른 중심합성계획에 의하여 $110{\sim}130^{\circ}C$와 2~4 시간 범위의 11가지 조건(A~K)에서 각각 열수추출하여 총 페놀, 플라보노이드 함량 및 항산화 활성이 최대화되는 추출조건을 모니터링 하였다. 항산화 활성의 정도에 따라 흑마늘 추출조건 3종($130^{\circ}C$, 4 hr; $120^{\circ}C$, 3 hr; $130^{\circ}C$, 3 hr)과 개똥쑥의 최적 추출조건 1종($120^{\circ}C$, 3 hr)을 선정하였다. 흑마늘 추출물에 대한 개똥쑥 추출물의 혼합비율을 1:0.5, 1:1, 1:1.5 및 1:2(w/w)로 조절하여 총 페놀, 플라보노이드 함량 및 항산화 활성을 측정한 결과, $120^{\circ}C$, 3시간에서 추출된 흑마늘 추출물과 개똥쑥 추출물의 혼합시 활성이 우수하였으며 최적의 혼합비율은 흑마늘과 개똥쑥 추출물에서 1:1.5(w/w)인 것으로 판단된다.
