Studies on the Immuno Modulating Acitivity of Fermented Artemisiae Argyi Folium Extract (애엽(艾葉) 발효 추출물의 면역활성에 관한 연구)

  • Objectives : This research aimed to study the cytotoxicity and immuno modulating activity of fermented Artemisia argyi Lev. et Vant.(Compositae). Methods : Effect of fermented Artemisiae Argyi Folium extracts, which were fermented by Sacchromyces cerevisiae STV89(AFS), on cell viability, generation of ROS within cells, generation of NO and the level of cytokines($TNF-{\alpha}$ and IL-6) was measured using mouse macrophage RAW 264.7 cell. Results : 1. Result of MTT assay conducted to verify the cytotoxicity of fermented Artemisiae argyi folium extract illustrated that, when fermented Artemisiae argyi folium extract was processed for each concentration, there was no excessive induction of cytoxicity in the RAW 264.7 cell. 2. Fermented Artemisiae Argyi Folium extract increased the generation of H2O2 within RAW 264.7 cell as well as significantly increased inhibition of generation of H2O2 in macrophage induced by LPS. 3. Fermented Artemisiae Argyi Folium extract inhibited generation of NO in RAW 264.7 cell, and significantly inhibited increase in generation of NO of macrophage induced by LPS. 4. Fermented Artemisiae Argyi Folium extract, AFS has significantly reduced the increase in the generation of $TNF-{\alpha}$ above 10 ${\mu}g/mL$. 5. Fermented Artemisiae Argyi Folium extract, AFS has significantly reduced the increase in generation of IL-6 above 50 ${\mu}g/mL$. Conclusions : AFS fermented extract produced from Artemisiae Argyi Flium, have increased generation of ROS and reduced generation of NO in RAW 264.7 cell without excessively inducing cytotoxicity of RAW 264.7 cell. In addition, they displayed significant immuno modulating activities including inhibition of generation of $TNF-{\alpha}$ and IL-6 in macrophage, induced by LPS.

Whitening Activities of Extracts of Seomaeyakssuk (Artemisia argyi H.) (섬애약쑥 (Artemisia argyi H.) 추출물의 미백활성)

  • This study investigated the whitening activity using seomaeyakssuk (Artemisia argyi H.) extract. Seomaeyakssuk was extracted from hot DW (AAD) and 70% ethanol (AAE). And confirmed safety through assessment of cytotoxicity. Also, whitening activities were measured through changes in the levels of extracellular melanin, melanin synthesis, cellular tyrosinase activity and transcription factor. The results confirmed that significant cytotoxicity does not appear in the concentration range of 50, 100, and 200 ㎍/㎖ of both extracts of this study. The production of extracellular melanin was slowed by AAD 45.0% and AAE 1.3% at 200 ㎍/㎖ concentration. Also, production of intracellular melanin was decreased AAD 37.2% and AAE 24.6%. In the case of intra cellular tyrosinase activity was reduced to AAD 49.2% and AAE 35.6% at 200 ㎍/㎖ concentration. The mRNA expression of tyrosinase, TRP-1 and TRP-2 significantly decreased by AAD 63.0%/AAE 58.0%, AAD 60.0%/AAE 56.0% and AAD 59.0%/AAE 53.0%, respectively, following the 200 ㎍/㎖ sample treatment when compared to the control. Both extracts showed efficient changes of production of whitening-related factor and transcription factor. But AAD was found to have a higher inhibitory effect than AAE. In other words, seomaeyakssuk was showed significant biological activities showing whitening without cytotoxicity. These results will be provided as fundamental data for further development of the new material of functional cosmetics to the results above.

Anti-amnesic and Neuroprotective Effects of Artemisia argyi H. (Seomae mugwort) Extracts (섬애쑥 추출물의 뇌 신경세포 보호효과에 의한 학습 및 기억능력 개선 효과)

  • The anti-amnesic effect of Artemisia argyi H against trimethyltin (TMT)-induced learning and memory impairment and its neuroprotective effect against $H_2O_2$-inducedoxidative stress were investigated. Cognitive behavior was examined by Y-maze and passive avoidance test for 4 weeks, which showed improved cognitive functions in mice treated with the extract. In vitro neuroprotective effects against $H_2O_2$-induced oxidative stress were examined using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium-bromide and lactate dehydrogenase (LDH) assays. A. argyi H. extract showed protective effects against $H_2O_2$-induced neurotoxicity; moreover, LDH release into the medium was inhibited. Finally, high-performance liquid chromatography (HPLC) analysis showed that eupatilin and jaceosidin were the major phenolic compounds in A. argyi H. extract. These results suggest that A. argyi H. could be a good source of functional substances to prevent neurodegenerative diseases.

Effect of ARTEMISIAE ARGI FOLIUM Acupuncture Solution on Raw 264.7 Cells Treated by Toxicants (애엽(艾葉) 약침액(藥鍼液)이 에탄올 등에 의한 마우스 대식세포의 활성변화에 미치는 영향)

  • Objectives : The leaves of Artemisia argyi L. have been used for the treatment of bleeding-related diseases in traditional korean medicine. But the immunological activities with macrophage have not been sufficiently reported. This study is to investigate the immunological bioactivities of the herbal acupuncture solution obtained from leaves of Artemisia argyi L. (AAAS). Methods & Results : Against Nicotine and Acetaldehyde, AAAS increased significantly the production of hydrogen peroxide (H2O2) within mouse macrophage Raw 264.7 cells above the concentration of 10 ${\mu}g/m{\ell}$. AAAS increased significantly the production of nitric oxide (NO) in Raw 264. 7 cells above the concentration of 100 ${\mu}g/m{\ell}$ against EtOH. And AAAS increased significantly the production of nitric oxide (NO) in Raw 264. 7 cells above the concentration of 200 ${\mu}g/m{\ell}$ against Nicotine, Acetaminophen, and Acetaldehyde. Conclusions : These results suggest that AAAS could be thought to have the immunological activities related with the production of hydrogen peroxide and NO in macrophage.

Flavonoids from the arial parts of Artemisia agryi and their antioxidant capacity through GSH recovery effect (황해쑥(Artemisia agryi)으로부터 flavonoid 화합물들의 분리 동정과 세포 내 GSH 회복능을 통한 항산화 활성 평가)

  • The arial parts of Artemisia argyi were extracted with methanol : water (70:30), and the concentrates was partitioned into EtOAc (ethyl acetate), n-BuOH (normal butanol), and H2O (water) fractions. The repeated silica gel and ODS (octadecyl silica gel) column chromatographies for EtOAc and n-BuOH fractions led to isolation of four flavonoids without any ambiguity based on intensive interpretation of several spectroscopic data including nuclear magnetic resonance, and mass spectrometry. The chemical structure of the isolated compounds revealed to (2S)-naringenin (1), 3-methylkaempferol (2), 3,3'-dimethylquercetin (3), and 3,3',4'-trimethylquercetin (4). These four compounds were first isolated from A. argyi through this study. In this study, four compounds isolated from A. argyi showed an increase in glutathione mean and a decrease in glutathione heterogeneity so that the compounds uniformly raised the intracellular glutathione (GSH) level. Based on these results, it is considered that it can be used as a functional pharmacological material.

Quality characteristics of fermented vinegar prepared with Seomaeyaksuk (Artemisia argyi H.) extract (섬애약쑥 주정 추출물로 제조한 발효식초의 품질특성)

  • This study was carried out to investigate the quality characteristics of vinegars containing jaceosidin and eupatilin using Artemisia argyi H. ethanol extract (AEE). 10% malt extract (ME) and water extract of Artemisia argyi H. (AWE) were also prepared for vinegar production. Three kinds of materials were mixed in the same amount to prepare vinegar as follows; CO (ME, water, 18% edible ethanol), SE (ME, water, and AEE), SW (ME, AWE, and 18% edible ethanol) and SM (ME, AWE, AEE). All samples were fermented by Acetobacter pasteurianus A8 at $30^{\circ}C$ for 25 days and analyzed at 10, 15, 20 and 25 days. The pH decreased significantly during the fermentation. pH was lower in SE and SM than CO and SW. The acidity increased significantly during the fermentation, and was highest in SM (4.44%) at 25 days of fermentation. The concentration of acetic acid was higher than other organic acids for all vinegars. Jaceosidin and eupatilin were not detected in both CO and SW, but both were detected in the SE and SM. At 25 days of fermentation, jaceosidin and eupatilin concentrations in SE and SM were 6.49-6.88 mg/kg and 2.23-2.24 mg/kg, respectively. From these results, we confirmed that production of vinegar containing jaceosidin, eupatilin and phenolic compounds can be prepared by using Artemisia argyi H. edible ethanol extract.

Nutritional Chemical Composition in the Different Parts of Artemisia argyi H. (섬애쑥(Artemisia argyi H.)의 부위별 영양화학성분)

  • The chemical components in different parts of Artemisia argyi was investigated to provide industrial possibilities as functional foods The analysis result of proximate composition in leaves, stems and roots of Artenisia argyi was substantially as follows. The crude protein contents were 19.87, 6.14 and 5.68%, the crude lipid contents were 4.56, 1.30 and 1.20%, the crude fiber contents were 16.80, 29.70 and 29.45%, respectively. The major mineral components in Artemisia argyi were potassium, calcium and magnesium. Contents of potassium and calcium in leaves were 4,270.24 and 617.64 mg/100 g, respectively, they were more than double the contents of root. Sucrose and glucose as main free sugars were detected in the leaves and roots. However, glucose and fructose were identified in the stem. Total amino acids showed 17 amino acids. Contents of total amino acid in the leaves was the highest as 4,864.11mg/100g, and the stems and roots showed 1,953.99 and 1,601.73mg/100g, respectively. The major amino acids in the leaves and stems were proline(963.91 and 407.52mg/100g) and aspartic acid(577.38 and 299.17mg/100g), respectively. Glutamic acid(206.34mg/100g) and arginine(193.23mg/100g) were main amino acids in the roots. The major fatty acids in all parts were linoleic acid($C_{18:2}$), behenic acid($C_{22:0}$), and palmitic acid($C_{16:0}$). Eupatilin(35.0mg/100g) and jaceosidin (107.63mg/100g) as physiological compounds contents were higher in leaves than other parts.

Physicochemical Characteristics of Ethanol Extract from Artemisia Argyi H. Using Different Preparation Methods (전처리 방법을 달리한 섬애약쑥 에탄올 추출물의 이화학적 특성)

  • This study was performed to determine the ethanol ratios (30%, 50%, 70%, and 90%) of extraction solvent of Seomaeyakssuk (Artemisia Argyi H.) prepared using different methods (drying at room temperature [D], aging at $60^{\circ}C$ for 7 days after drying [AD], and roasting for 30 min at $160^{\circ}C$ after drying [RD]). The extract yield of the D extracts was lower than that of the AD and RD extracts, but the ethanol concentration of extract solvent did not affect. The L, a, and b values of the D extracts were highest, whereas those of the AD extracts were lowest. No clear trend was observed in the ethanol ratios. The soluble solids, total phenol, total flavonoid, jaceosidin, and eupatilin contents of each extract varied significantly, with RD > AD > D. The soluble solids significantly increased by ethanol ratio of extraction solvent, but other phytochemicals contents of 50% and 70% ethanol extracts were higher than others without affecting the processing methods. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was highest (77.71%) in the 70% ethanol extract obtained from RD. 2,2-azinbis-(3-ethylbenzo-thiazoline-6-sulphonate(ABTS) radical scavenging activity was significantly higher in 30-70% ethanol extract than 90% ethanol extract from RD. The results suggest that the contents of active ingredients and radical scavenging activity of ethanol extracts from Seomaeyakssuk were highest in the RD extract using 50-70% ethanol.

Variation of Chamazulene Contents in Local Collections of Mugwort (Artemisia spp.) (수집종 쑥의 청색색소 카마줄렌 함량 변이)

  • This study was carried out to select chamazulene containing species and to know whether chamazulene content varies depend on individual plants within a population or not. Among five collected mugworts, chamazulene was detected only in Artemisia indica. Chamazulene content in the leaves was higher than that in stems at vegetative stage. Camazulene in inflorescence was not higher than that in leaves even at reproductive stage. Chamazulene was higher in the leaves of the upper part of the plant than those of the middle and lower of the plant at vegetative stage. Chamazulene contents of 50 individuals within a population were varied from 0.01% to 0.11% with mean of 0.045%. About 36 individuals, 72 percent of total individuals examined, had more than 0.03% of chamazulene. There was no difference in chamazulene contents between parents and their clones.

  • 황해쑥(Artemisia argyi H.Lev. & Vaniot)은 우수한 항염증 활성을 지닌 것으로 다양하게 보고 되었다. 그러나, 대표적인 염증 질환 중 하나인 골관절염에 미치는 영향은 현재까지 알려져 있지 않다. 따라서, 본 연구에서는 염증 유발 연골 세포 및 골관절염 유발 동물 모델에 미치는 황해쑥 효과에 대해 조사하였다. 첫째, interleukin 1 beta를 투여한 관절 연골 세포에 황해쑥 물 추출물을 투여한 후 metalloporeinase (MMP) -3 및 MMP-13의 발현을 mRNA 및 단백질 수준에서 분석 하였다. 또한, 내측 반월상 연골의 불안정화에 의해 유도 된 골관절염 마우스 모델을 사용하여 황해쑥 물 추출물의 골관절염 억제 효과를 분석 하였다. 세포 실험에서, 본 황해쑥은 MMP-3와 MMP-13의 mRNA 및 단백질 발현을 유의하게 억제하였다. 또한. 황해쑥 물 추출물을 투여한 실험 동물의 관절 조직을 Safranin O 염색을 실시하여 분석한 결과 연골 하 골판 두께의 감소 및 활막 염증 개선 효과가 관찰 되었다. HPLC를 이용한 성분 분석 결과, 황해쑥 물 추출물은 항염증 및 항관절염 활성을 가진 jaceosidin과 eupatilin을 함유하는 것으로 나타났다. 본 연구결과로부터 황해쑥은 골관절염의 치료 또는 예방에 유망한 소재로 개발될 수 있음을 시사한다.
