마이크로파 처리 유무에 따른 우엉뿌리의 항비만 효과 비교 연구 (The comparative study on the anti-obesity effect of Arctium lappa L. roots with and without microwave processing)

  • Objective : The present study comparatively analyzed the anti-obesity effect of Arctium lappa L. roots with and without microwave processing. Methods : Four HFD groups except for the Normal group (n=8) were allocated: Control, microwave-processed dried Arctium lappa L. roots (MAL) extract 400 mg/kg/d (MAL400), MAL extract 800 mg/kg/d (MAL800), and the dried Arctium lappa L. roots (DAL) extract 800 mg/kg/d (DAL800). The efficacy of MAL and DAL was confirmed in terms of the serum biochemical index, protein expressions related to the synthesis of triglyceride (TG) and total cholesterol (TC) and 𝛽-oxidation, and histopathological staining. Results : Both MAL and DAL treatments significantly reduced final body weight and body weight gain. MAL800 treatment significantly reduced serum TG, TC, low-density lipoprotein cholesterol, and leptin levels, but the serum high-density lipoprotein cholesterol and adiponectin concentrations were dramatically increased. In particular, leptin in the MAL800 group was reduced by 14.1% compared with the DAL800 group. Moreover, the MAL800 treatment showed an effect superior to the DAL800 treatment in the reduction of serum TC, the sirtuin 1 (Sirt1) activation, and the inhibition of 3-hydroxy-3-methyl glutaryl coenzyme A reductase (HMGCR) gene expression. In particular, unlike the DAL800 treatment, MAL treatment significantly led to the activation of peroxisome proliferator-activated receptor alpha (PPAR𝛼). Subsequently, PPAR𝛼 meaningfully regulated downstream proteins associated with 𝛽-oxidation. Conclusion : These findings suggest that Arctium lappa L. roots with microwave processing effectively ameliorate obesity through the regulation of leptin and TC and the promotion of 𝛽-oxidation compared with Arctium lappa L. roots without microwave processing.

발효 우엉 에탄올 추출물의 항산화 및 항비만 활성 (Antioxidant and Anti-Obesity Activity of Ethanol Extracts from Fermented Arctium lappa L.)

  • This study was conducted to investigate the antioxidant and anti-obesity activity of ethanol extracts from fermented Arctium lappa L. Arctium lappa fermented twice with Phellinus linteus ($2^{nd}$ FA) showed more DPPH radical scavenging activity than non-fermented Arctium lappa (NFA), and Arctium lappa fermented once ($1^{st}$ FA) at a concentration of 62.5 ppm. The $2^{nd}$ FA showed the highest level of ABTS radical scavenging activity at concentration range of 31.5~125 ppm. The ABTS free radical scavenging activities of $1^{st}$ FA and $2^{nd}$ FA were similar to that of BHA, a synthetic antioxidant, at a concentration of 250 ppm. The total polyphenol content of $2^{nd}$ FA was higher than those of NFA and $1^{st}$ FA. The flavonoid content was significantly increased in $1^{st}$ FA and $2^{nd}$ FA than NFA. During adipocyte differentiation, lipid accumulation in 3T3-L1 cells was significantly decreased in the order of NFA, $1^{st}$ FA, and $2^{nd}$ FA at all concentrations. In conclusion, the antioxidant and anti-obesity activities of A. lappa were increased depending on the degree of fermentation. It is suggested that fermented Arctium lappa, especially $2^{nd}$ FA, could be used as a natural ingredient for functional foods and medicine.

영남지역 재래종 우엉(Arctium lappa L.)의 주요특성 (Major Characteristics of Burdock(Arctium lappa L.) Native to Yeong-Narn Region)

  • 영남지역에서 재배되고 있는 우엉 (Arctium lappa) 재래종 18계통을 수집하여 기초적인 자료를 얻고자 본 시험을 수행하였다. 종자의 천립중은 2.2∼4.6g, 충실률은 26.7∼91.7%로 계통간 차이가 심하였다. 온도에 따른 발아율은 25℃에서 가장 높았으며 수집 계통간 저온 발아율은 최고 42.2%(문경)였고 최저 0.3%(유천)였다. '유천' 및 '칠곡Ⅱ' 가 당년에 10%이상 추대되었고 나머지는 추대율이 미미하거나 추대가 되지 않았다. 근장은 62∼77㎝정도였으며 지근수는 최고 5.5개(고령)에서 최저0.3개(성주)로 계통간 차이가 심하였다. 뿌리에서 Tannin함량 및 잎에서의 쓴맛 정도도 계통간 차이가 심하였는데 Tannin 함량은 '청도 I ' 이 14.2㎎/100g으로 가장 많았고 쓴맛은 '경산' , '고령' , '경주', '구미' 및 '함안' 계통들이 전혀나지 않았다.

우엉 에탄올 추출물의 항산화활성과 항돌연변이 효과 (Antioxidative and Antimutagenic Effects of $Arctium$ $lappa$ Ethanol Extract)

  • The antioxidant activities of the ethanol extract of Arctium lappa were assessed by measuring the 1,1-diphenyl-2-picrylhydrazyl( DPPH) radical scavenging effect, inhibition of $Fe^{2+}$-induced lipid peroxidation, inhibition of malondialdehyde(MDA)-bovine serum albumin(BSA) conjugation reaction and antimutagenic capacities using the Ames test. The DPPH radical scavenging activity and inhibition of $Fe^{2+}$-induced lipid peroxidation of the $Arctium$ $lappa$ ethanol extract significantly increased in a dose-dependent manner. In the radical scavenging assay using DPPH, the $IC_{50}$ of the Arctium lappa extract was 296 ${\mu}g$/assay(1.29 mg of dry sample). In addition, the $IC_{50}$ in the inhibition of $Fe^{2+}$-induced lipid peroxidation was 1,759 ${\mu}g$/assay(7.65 mg of dry sample). This extract also significantly inhibited the MDA-BSA conjugation reaction with an $IC_{50}$ of 57.58 mg/assay(250 mg of dry sample). However, no inhibitory effects against the direct and indirect mutagenicities in $Salmonella$ Typhimurium TA98 and TA100 were observed. Based on these results, the ethanol extract of $Arctium$ $lappa$ was shown to display considerable antioxidative activities.

우엉(Arctium lappa L.) 뿌리 Polyphenol Oxidase의 부분정제 및 특성 (Purification and Characterization of Polyphenol Oxidase from Burdock (Arctium lappa L.))

  • 우엉 뿌리(Arctium lappa L.)에서 polyphenol oxidase가 DEAE-Cellulose 음이온 수지, 황산암모늄 침전법, Phenyl-sepharose CL-4B 친화크로마토그래피 그리고 Sephadex G-100 겔여과크로마토그래피 등의 과정으로 정제되었며 정제효소의 효소학적 특성을 검토하였다. 정제된 효소의 분자량은 약 30 kDa의 단일폴리펩티드 사슬로 구성되어 있었다. 효소반응의 최적 pH와 온도는 각각 7.0과 35$\circC$이었으며, pH 2와 6 사이의 산성조건과, pH 8과 10 사이의 알카리 조건에서는 활성이 감소되거나 상실되었다. 온도에 대한 영향을 살펴본 결과, 40$\circC$까지 비교적 안정한 활성을 나타내고 있었으나, 50$\circC$에서 30분간 정치 시 효소활성을 50%이상 감소하는 것으로 나타내었다. 이 효소는 catechol과 L-DOPA에 대하여 높은 효소활성을 나타내었으며, chlorogenic acid과 catechol에 대한 Km 값은 각각 16.18 mM과 23.11 mM이었다. Ascorbic acid와 L-cysteine은 우엉 뿌리 polyphenol oxidase의 효소활성을 감소시켰으며, 금속이온의 경우 $Cu^{2+}$가 효소의 활성을 감소시키는 인자로 나타났다.

우엉 뿌리 추출물의 항산화 활성 및 세포 독성 평가 (Antioxidant Activity and Cytotoxicity on cell of Arctium lappa L. root extract)

  • 본 연구는 우엉 뿌리 추출물의 항산화 활성 및 피부에 대한 안전성을 평가하기 위하여 총 폴리페놀 함량, 총 플라보노이드 함량, DPPH radical 소거 활성을 통하여 항산화 활성을 살펴보고, B16F10 melanoma 세포에 대한 세포 독성 및 자외선 A에 대한 피부 세포 보호 효과를 확인하였다. 또한 화장품 소재로서의 활용을 검증하기 위하여 1차 피부 첩포 테스트를 실시하였다. 본 실험 결과 우엉뿌리 추출물의 함량이 증가됨에 따라 높은 폴리페놀과 플라보노이드의 함량이 확인되었으며, DPPH radical 소거 활성을 확인하였다. B16F10세포에 대한 세포 독성을 확인한 결과 B16F10 melanoma 세포에 대해 독성이 낮고, 자외선 A에 대해 80% 이상의 세포 보호 효과를 확인하였다. 또한 1차 피부 첩포 테스트를 통해 우엉 뿌리 추출물이 피부에 자극이 거의 없음을 확인하였다. 이러한 결과를 통하여 우엉뿌리 추출물이 항산화 활성과 자외선에 대한 피부 보호 효과가 뛰어나고 피부 세포에 대한 독성이 낮으며, 피부에 대한 안전성이 확인됨에 따라 화장품 소재로서의 가능성을 확인하였다.

Evaluation of Pharmacognostical Characters and Comparative Morphoanatomical Study of Saussurea costus (Falc.) Lipchitz and Arctium lappa L. Roots

  • Saussurea costus (Falc.) Lipchitz syn S. lappa C. B. Clarke (commonly known as 'Kuth') belonging to the family Asteraceae is a well known medicinal plant which finds wide usage in different indigenous systems of medicine of India, China, Korea & Tibet. In different folk medicines the roots of S. costus are used to treat various disorders like ulcer, stomachache, malaria, leprosy, dysentery and toothache. However due to over exploitation, it has become endangered and has become the concern of different governmental bodies in India. The increasing demand of this endangered Himalayan species has resulted in a situation where it is often substituted, knowingly or unknowingly, by other morphologically similar species. Arctium lappa, belonging to the same family, is one such plant that has often been found to be present in the market samples of 'Kuth'. The present study was thus carried out and morphoanatomical characters, physicochemical as well as chemical parameters were developed for proper identification of roots of S. costus and its differenciation from A. lappa as well as authentication of the commercial market samples. The detailed morphoanatomical studies revealed that roots of S. costus can be distinguished from A. lappa on the basis of some important microscopial characters eg. the schizogenous resin ducts observed in roots of S. costus, were absent in A. lappa.. Besides, the HPTLC fingerprint profile showed a distinct band at Rf. 0.72 in S. costus, which was totally absent in A. lappa and a band at $R_f$ 0.64 in A. lappa which was absent in S. costus Chlorogenic acid, used as a chemical marker for HPTLC analysis, was estimated to be 0.077% in S. costus as compared to 0.107% in A. lappa. Thus these detailed pharmacognostical parameters can be successfully used to distinguish between roots of S. costus and A. lappa.

전통 식재료의 항산화 효과 (Antioxidant Effect of Traditional food Ingredient)

  • 본 연구는 전통 식재료로 많이 사용되는 10종의 약용식물 및 채소류 추출물과 10종 혼합추출물의 항산화 효과를 살펴보기 위해서 디자인되었다. 세포 생존률은 MTS assay로, 항산화효과는 DPPH free radical 소거능, Riboflavin 유래 Superoxide 억제활성(SQA)에 의한 항산화활성을 측정하였다. 그 결과 세포생존률은 10종 약용식물 및 채소류 추출물과 10종 혼합추출물 모두 0.5mg/mL에서 대조군과 유사한 생존율을 보였다. DPPH free radical 소거능은 Cinnamomum cassia Blume, Eugenia caryophyllata Thunb. Arctium lappa, Total extract가 우수하였고, Riboflavin 유래 Superoxide 억제활성(SQA)은 Cinnamomum cassia Blume, Arctium lappa, Prunus mume Sieb. et Zucc., 우수하였으나, Total extract가 가장 우수한 항산화 효과를 보였다. 전통식재료로 사용되는 약용식물 및 채소 추출물과 혼합추출물의 항산화 효과를 비교한 결과, 단독으로 사용하는 것보다 혼합물로 사용시 항산화능이 증가되었다. 이는 항산화 기능성 소재로 활용이 가능한 것으로 사료되며, 향후 항산화 소재 개발 시 가치가 있을 것으로 사료된다.

Yield and Free Sugar Contents of Burdock (Arctium lappa L.) depending on Nitrogen Levels

  • Excessive nitrogen fertilization influences crop yields and quality as well as environmental pollution. In this study, yields, nitrogen use efficiency and free sugar contents of burdock (Arctium lappa L.) were evaluated at different levels of nitrogen fertilization. Nitrogen fertilizer was applied at 5 levels (0, 50, 100, 150, 200%) based on the conventional fertilization ($N=230kg\;ha^{-1}$), and phosphate and potassium fertilizer were treated by conventional P and K fertilization ($P_2O_5-K_2O=140-210kg\;ha^{-1}$) in all plots. The root yields of burdock were the highest in N 100~150% treatment plots. Nitrogen use efficiency and nitrogen recovery decreased from over N 150% treatment. Nitrogen uptake of root was greater than that of shoot in N 50~200% treatments. Fructose contents in root were inversely proportional to the level of nitrogen fertilization. As considering nitrogen recovery and root quality, economical burdock yield was obtained in N $230kg\;ha^{-1}$.

혈관내피세포에서 우방자(牛蒡子) 에탄올 추출물의 항염증 효과 (Anti-Inflammatory Effect of Ethanol Extract from the Seeds of Arctium Lappa L. in Vascular Endothelial Cells)

  • Objectives: The seeds from Arctium lappa have been considered for its various pharmacological properties, which include anti-carcinogenic, anti-inflammatory, anti-diabetic, and anti-viral activities. Methods: In the present study, we investigated the anti-inflammatory effect of the ethanol extract from the seeds of Arctium lappa L (EAL) on cytokine-induced vascular inflammation in human umbilical vein endothelial cells (HUVEC). Results: Pretreatment with EAL significantly decreased tumor necrosis factor alpha ($TNF-{\alpha}$)-induced cell adhesion molecules expression such as intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), and endothelial-selectin (E-selectin) in a dose-dependent manner. Cell adhesion assay showed that pretreatment with EAL suppressed HUVEC-monocyte adhesion by $TNF-{\alpha}$ over $1{\mu}g/ml$ concentration. We investigated the involvement of nuclear transcription factor kappa-B ($NF-{\kappa}B$) in $TNF-{\alpha}$-induced vascular inflammation. $NF-{\kappa}B$ p65 nuclear expression was induced by $TNF-{\alpha}$, however, pretreatment with EAL was attenuated that nuclear translocation. In cytoplasm, EAL was also attenuated $TNF-{\alpha}$-induced decrease of inhibitor of ${\kappa}B-{\alpha}$ ($I{\kappa}B-{\alpha}$) expression. Moreover, EAL significantly decreased $TNF-{\alpha}$-induced production of intracellular reactive oxygen species (ROS). Conclusions: Taken together, our findings suggest that seeds of Arctium lappa L could be a therapeutic herb for prevention of cardiovascular diseases throughout the inhibition of vascular endothelial inflammation.
