  • In archaeology, oxygen isotope analysis using marine shells has been used to reconstruct past environment and determine the season of shellfish-gathering activities in the past. Modern oysters(Crassostrea gigas) from Solsum ao the mouth of Chonsu Bay were anayzed to examine the potential of the species for determining seasonality by oxygin isotope analysis. As a result, it appears that the species can be used for this purpose.

Equol Induces Mitochondria-Dependent Apoptosis in Human Gastric Cancer Cells via the Sustained Activation of ERK1/2 Pathway

  • The cancer chemo-preventive effects of equol have been demonstrated for a wide variety of experimental tumours. In a previous study, we found that equol inhibited proliferation and induced apoptotic death of human gastric cancer MGC-803 cells. However, the mechanisms underlying equol-mediated apoptosis have not been well understood. In the present study, the dual AO (acridine orange)/EB (ethidium bromide) fluorescent assay, the comet assay, MTS, western blotting and flow cytometric assays were performed to further investigate the pro-apoptotic effect of equol and its associated mechanisms in MGC-803 cells. The results demonstrated that equol induced an apoptotic nuclear morphology revealed by AO/EB staining, the presence of a comet tail, the cleavage of caspase-3 and PARP and the depletion of cIAP1, indicating its pro-apoptotic effect. In addition, equol-induced apoptosis involves the mitochondria-dependent cell-death pathway, evidenced by the depolarization of the mitochondrial membrane potential, the cleavage of caspase-9 and the depletion of Bcl-xL and full-length Bid. Moreover, treating MGC-803 cells with equol induced the sustained activation of extracellular signal-regulated kinase (ERK), and inhibiting ERK by U0126, a MEK/ERK pathway inhibitor, significantly attenuated the equol-induced cell apoptosis. These results suggest that equol induces mitochondria-dependent apoptosis in human gastric cancer MGC-803 cells via the sustained activation of the ERK1/2 pathway. Therefore, equol may be a novel candidate for the chemoprevention and therapy of gastric cancer.

Evaluation of Plasma NT-proBNP Concentration in Dogs with Chronic Mitral Valve Insufficiency (개의 만성 이첨판 폐쇄부전증 환자군에서 혈장 NT-proBNP 농도 평가연구)

  • This study aimed to evaluate the plasma concentration of NT-proBNP in dogs with different stages of heart failure by chronic mitral valve insufficiency (CMVI). Fifty small-breed dogs with CMVI and 7 healthy control dogs without cardiac disease and critical systemic diseases were included in the study population. As a preliminary study, we compared the plasma concentrations of N-terminal pro-brain natriuretic peptide (NT-proBNP) and the echocardiographic parameters between dogs of the International Small Animal Cardiac Health Council (ISACHC) classes. Then, we evaluated the associations between NT-proBNP and echocardiographic parameters. Plasma NT-proBNP levels showed a significant difference among the ISACHC groups. In the comparison between echocardiographic parameters and NT-proBNP, NT-proBNP were found to be associated with left atrium/aorta (LA/AO), early diastolic transmitral flow (E) velocity, late diastolic transmitral flow (A) velocity, end diastolic volume index (EDVI). Our study found plasma NT-proBNP might be useful to predict the disease progression in dogs with CMVI.

  • Curvature of Crystalline lens changes by Accommdation's change. When Accommdation gives force vertically to Crystalline lens that is elastic body, length increases for vertex direction. Density distribution and form of Crystalline lens that receive force lean to posterior surface, horizontal force of anterior surface direction is bigger more than horizontal force of posterior surface direction. But, if Accommdation begins to grow more than threshold value, expansity reaches in limit on anterior surface. This time, horizontal force of posterior surface direction is great mored more than horizontal force of anterior surface direction, thickness of posterior surface direction increases because is more than anterior surface direction. Anterior and posterior relationship thickness change difference accomplish the 2-nd funtional line(${\Delta}=B_1D+B_2D^2$) about Accommdation. Thickness (${\Delta}t_a$, ${\Delta}t_p$) difference change curved line of anterior pole-border and border-posterior pole by Accommdation is expressed as following. $${\Delta}t_a=t_a-t_{ao}=t_{max}+t_0{\exp}(-A/B)-t_{ao}$$ $${\Delta}t_p=t_p-t_{po}=t_{min}+t_0{\exp}(A/B)-t_{po}$$ The Parameter value that save in human's Crystalline lens obtain $t_{min}=1.1.06$, $t_0=-0.33$, B=9.32 in anterior, and $t_{max}=1.97$, $t_0=0.10$, B=7.96 etc. in posterior. Vertex curvature radius' change is as following Crystalline lens' anterior and posterior by Accommation $$R=R_0+R_1{\exp}(D/k)$$ The Parameter value that save in human's Crystalline lens obtain $R_{min}=5.55$, $R_1=6.87$, k=4.65 in anterior, and $R_{max}=-68.6$, $R_1=76.7$, k=308.5 in posterior, respectively.

Synergistic Effect of Flavonoids from Artocarpus heterophyllus Heartwoods on Anticancer Activity of Cisplatin Against H460 and MCF-7 Cell Lines

  • Artocarpus heterophyllus has been used as traditional medicine. This plant is one of the sources of flavonoid. Flavonoid compounds possessed a wide range of biological properties including anticancer. This study was performed to investigate the cytotoxic effect of flavonoids from A. heterophyllus on H460 and MCF-7 cell lines. The interaction of flavonoids and cisplatin against tested cancer cells was also evaluated. MTT assay was used to determine the cytotoxic effect of flavonoid. Isobologram analysis was selected to evaluate the synergistic effect between flavonoid and cisplatin, their interaction was then confirmed using AO/PI staining method. Amongst of flavonoid compounds, artocarpin exhibited strong cytotoxic effect on both MCF-7 and H460 cell lines with IC50 values of 12.53 ㎍/mL (28.73 μM) and 9.77 ㎍/mL (22.40 μM), respectively. This compound enhanced anticancer activity of cisplatin against H460 and MCF-7. The combination produced a synergistic effect on H460 and MCF-7 cell lines with a combination index (CI) values of 0.2 and 0.18, respectively. The AO/PI stained demonstrated that the combination of artocarpin and cisplatin caused morphological changes that indicated apoptosis. Moreover, artocarpanone also significantly increased cytotoxic effect of cisplatin compared to its single concentration with CI below than 1. This result suggested the potency of flavonoid named artocarpin to enhance the anticancer activity of cisplatin on H460 and MCF-7 cell lines.

  • A dynamic fatigue characteristic of dental implant system has been evaluated with applying single axial compressive shear loading based on the ISO 14801 standard. For the advanced dynamic fatigue test, multi-directional force and motion needed to be accompanied for more information of mechanical properties as based on mastication in oral environment. In this study, we have prepared loading and motion protocol for the multi-directional fatigue test of dental implant system with single (Apical/Occlusal; AO), and additional mastication motion (Lingual/Facial; LF, Mesial/Distal; MD). As following the prepared protocol (with modification of ISO 14801), fatigue test was conducted to verify the worst case results for the development of highly stabilized dental implant system. Mechanical testing was performed using an universal testing machine (MTS Bionix 858, MN, USA) for static compression and single directional loading fatigue, while the multi-directional loading was performed with joint simulator (ADL-Force 5, MA, USA) under load control. Basically, all mechanical test was performed according to the ISO 14801:2016 standard. Static compression test was performed to identify the maximum fracture force with loading speed of 1.0 mm/min. A dynamic fatigue test was performed with 40 % value of maximum fracture force and 5 Hz loading frequency. A single directional fatigue test was performed with only apical/occlusal (AO) force application, while multi directional fatigue tests were applied $2^{\circ}$ of facial/lingual (FL) or mesial/distal (MD) movement. Fatigue failure cycles were entirely different between applying single-directional loading and multi-directional loading. As a comparison of these loading factor, the failure cycle was around 5 times lower than single-directional loading while applied multi-directional loading. Also, the displacement change with accumulated multi-directional fatigue cycles was higher than that of single directional cycles.

Antioxidant Activities of Extracts from Fruiting Bodies of Mushrooms (버섯추출물의 항산화활성에 관한 연구)

  • Antioxidant activities of 80% ethanol extracts of 63 species of mushroom fruiting bodies were investigated. The ethanol extracts from Daedalea dickinsii, Armillariella mellea, and Fomitella fraxinea showed markedly inhibition on lipid peroxidation in rat liver microsome. The extracts from Daedaleopsis tricolor, Trametes suaveolens, Armillariella mellea, Trichaptium abietinum, Daedalea dickinsii, Fomitella fraxinea, Tylophilus neofelleus, Boletellus obscurecoccineus, and Xerocomus subtomentosus significantly inhibited the hepatic aldehyde oxidase activity, and the extracts from Daedaleopsis tricolor, Armillariella mellea, Daedalea dickinsii, and Fomitella fraxinea slightly stimulated the hepatic SOD activity. These results suggest that Daedalea dickinsii, Armillariella mellea, and Fomitella fraxinea contain the bioactive substances for natural antioxidant and may be useful for development of antioxidant from mushrooms.

Assessment of 6-Month Lead Prediction Skill of the GloSea5 Hindcast Experiment (GloSea5 모형의 6개월 장기 기후 예측성 검증)

  • This study explores the 6-month lead prediction skill of several climate indices that influence on East Asian climate in the GloSea5 hindcast experiment. Such indices include Nino3.4, Indian Ocean Diploe (IOD), Arctic Oscillation (AO), various summer and winter Asian monsoon indices. The model's prediction skill of these indices is evaluated by computing the anomaly correlation coefficient (ACC) and mean squared skill score (MSSS) for ensemble mean values over the period of 1996~2009. In general, climate indices that have low seasonal variability are predicted well. For example, in terms of ACC, Nino3.4 index is predicted well at least 6 months in advance. The IOD index is also well predicted in late summer and autumn. This contrasts with the prediction skill of AO index which shows essentially no skill beyond a few months except in February and August. Both summer and winter Asian monsoon indices are also poorly predicted. An exception is the Western North Pacific Monsoon (WNPM) index that exhibits a prediction skill up to 4- to 6-month lead time. However, when MSSS is considered, most climate indices, except Nino3.4 index, show a negligible prediction skill, indicating that conditional bias is significant in the model. These results are only weakly sensitive to the number of ensemble members.

The Analysis of the Treatment Outcomes of Proximal Humeral Fractures with Locking Plates

  • Background: The aim of this study was to assess the clinical outcomes after treatment of proximal humeral fractures with locking plates, and to determine which factors influence the clinical and radiological outcomes. Methods: Fifty six patients who were treated with locking plates for proximal humeral fractures and had been followed for more than 1 year were enrolled in this study. We performed functional evaluation using the Constant score and analyzed radiographic results. The following factors that may potentially influence the clinical outcomes were assessed: age, gender, type of fracture, presence of medial metaphyseal comminution, bone mineral density, anatomical reduction, restoration of medial mechanical support, and postoperative complications. Results: The mean Constant score was 70.1 points at the final follow-up. Female gender, 4-part fractures, AO type-C fractures, and fractures with medial metaphyseal comminution were associated with a poor clinical outcome. On the other hand, restoration of medial mechanical support and accurate anatomical reduction had a positive influence on clinical outcomes. Postoperative complications resulted in 3 patients (intra-articular screw perforation: 1 patient, varus deformity with screw loosening: 1 patient, nonunion: 1 patient). Conclusions: When treating proximal humeral fractures with locking plate fixation, following factors: a female gender, Neer type 4-part fracture, AO type C fracture, and medial metaphyseal comminution are important risk factors that surgeons should take into consideration. Factors that contribute to better clinical outcomes of operative treatment for humeral fractures are accurate anatomical reduction and restoration of medial mechanical support.

PARALLEL IMAGE RECONSTRUCTION FOR NEW VACUUM SOLAR TELESCOPE

  • Many advanced ground-based solar telescopes improve the spatial resolution of observation images using an adaptive optics (AO) system. As any AO correction remains only partial, it is necessary to use post-processing image reconstruction techniques such as speckle masking or shift-and-add (SAA) to reconstruct a high-spatial-resolution image from atmospherically degraded solar images. In the New Vacuum Solar Telescope (NVST), the spatial resolution in solar images is improved by frame selection and SAA. In order to overcome the burden of massive speckle data processing, we investigate the possibility of using the speckle reconstruction program in a real-time application at the telescope site. The code has been written in the C programming language and optimized for parallel processing in a multi-processor environment. We analyze the scalability of the code to identify possible bottlenecks, and we conclude that the presented code is capable of being run in real-time reconstruction applications at NVST and future large aperture solar telescopes if care is taken that the multi-processor environment has low latencies between the computation nodes.
