  • Prunus serrulata var. spontanea(Rosaceae) is a large sized tree widely distributed throughout Korea. The red fruits are edible and are used in traditional folk medicine against heart failure from beriberi. dropsy. mastitis, an emmenagogue, and toothache. Also. the bark of P. serrulata var. spontanea has been used for detoxification. and relaxation and as an antitussive in traditional Korea medicine. (omitted)

  • (-)-${\alpha}$-narcotine( 1 R.9S) is one of the major bases in Papaver somniferum L.. the sourse plant for opium. while (-)-${\beta}$-narcotine(1R.9R) is a synthetic phthalideisoquinoline alkaloid. Although some advanced methods for the preparation of $\alpha$-narcotine have been developed using modified Bischler-Napieralski cyclization. the facile synthesis of $\beta$-narcotine has not further been attempted. supposingly because of its no clinical efficacy contrary to $\alpha$-narcotine having an antitussive effect. Relief Effect of Medicinal Herb Mixture HAE Series on Respiratory Symptoms (호흡기 증상에 대한 복합제 HAE의 완화 효과)

  • HAE series (HAE 01 - 06), are the mixtures of medicinal plants; Lonicerae Folium et Caulis (Lonicera japonica), Scutellariae Radix (Scutellaria baicalensis), Adenophorae Radix (Adenophora triphylla var. japonica), Polygonati Oddorati Rhizoma (Polygonatum odoratum var. pluriflorum), Platycodi Radix (Platycodon grandiflorum), Liriopis seu Ophiopogonis Tuber (Liriope platyphylla). HAE 06 showed the significant antitussive effect in HAE series. HAE 04, 05, and 06 increased the effect of expectorant measured by secretion of phenol red in rats. All tested HAE series were observed to have the anti-oxidant activity determined by the scavenging activity. HAE 01, 03 and 06 were significantly reduced the production of nitric oxide (NO) and tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) increased by LPS. HAE 06 was selected based on the results and done further experiments. HAE 06 inhibited the number of coughing induced by citric acid in dose dependent manner. HAE 06 effectively increased the effect of expectorant. HAE 06 exhibited anti-inflammatory effects by inhibiting the production of NO and TNF-${\alpha}$. In addition, HAE 06 significantly suppressed inducible Cyclooxygenase-2 (COX-2). Especially HAE 06 has the highest effect in alleviating the respiratory symptoms. Moreover, the enhancement antioxidant activity and reduction inflammation partly were attributed to it. HAE 06 may become the promising candidate for the treatment of respiratory symptom in the future.

  • Alnus sibirica (AS) geographically distributes in Korea, Japan, Northeast China and Russia. The bark of this plant had been used for antipyretic, expectorant, anti-phlogistic, antitussive, anti-asthmatic and as a health tea for alcoholism. Recently, we studied various biological activities of AS and the isolated diarylheptanoid. In present study, we conducted fermentation of AS (FAS) and isolated two diarylheptanoid (hirsutenone and muricarpone B). Moreover, we established the validation and contents determinations of the two compounds by HPLC on FAS.

  • The mutagenic activities of the pyrolysates(300, 600, 750 and 85$0^{\circ}C$) of extracts from four antitussive or expectorant medicinal plants(Daturae Folios, Scopoliae Rhizoma, Acori Rhisoma, and Schigandrae Fructus) were studied in the Ames Salmonella/microsomes test system. The pyrolyzate of Schizandrae Fructus did not show the mutagenicity to tester strain TA98 of Salmonella typhinurium. However, those of other medicinal Plants, Daturae Folium, Scopoliae Rhigoma and Acori Rhizoma, exhibited the mutagenic activity to this strain and the mutagenicity of them were decreased gradually to 70% of the initial activity according to time course after the preparation of the samples with DMSO.

  • The root-bark of the mulberry tree has been used as a medicine with antitussive, antiphlogistic, diuretic etc. in Korea. Among many kinds of mulberry trees, Morus Lhou has been widely cultivated in our country. In this experiment, three substances were obtained from the root-bark of Morus Lhou removed its cork layer by column chromatography of methanol extract. One of the substances was supposed as ${\alpha}$-amyrin acetate, another was alyceride, the other was confirmed as ${\beta}$-sistosterol by chemical and physical methods.

  • Dextromethorphan (Romilar. DEX) is a synthetic analogue of codeine. is not classified as a narcotic and is used only for its antitussive effects in Korea. The daily intake by adults range up to 120mg. Usually in the case of traffic accident. alcohol concentration of blood is analyzed in Korea. but drug tests (medicine, narcotic. alcohol) are submitted in Australia.   • The abuse of zipeprol, an antitussive agent, was found to be most prevalent among young people in Korea. Because abusers take large doses of this drug for its hallucinogenic effects, fatalities from zipeprol overdose abuse have been on the rise since 1991. Since 1991, a total of 69 zipeprol-related deaths have occurred throughout the nation. A demographic study shows that in ninety six percent of cases involving zipeprol alone, the victims were in their teens and twenties. The mate/female ratio in zipeprol related death was 3.5:1. Most of these zipeprol-associated deaths occurred in the larger cities of Seoul and Inchon. The blood concentration of zipeprol ranged from 0.8 to $38.3{\mu}g/mL $in single drug involved deaths, while zipeprol varied from to 35.3 $0.1{\mu}g/mL $in zipeprol and dextromethorphan victims.

  • This study was undertaken to investigate effects of 'Oyo-Tang' on the respiratory system. The results of this study were summarized as follows; 1. Antihistamine and relaxed dialating actions were recognized on the extracted ileum and serial broncheal samples in guinea-pigs. 2. Mild hypertensive action was recognized on the carotid artery in rabbits. 3. Antitussive action was noted on mechanically irritated coughs in dogs and cats. In connection with the results of these studies, effects based on the Oriental medical references were consistent with the actual experimental results. It was considered that effective application of 'Oyo-Tang' for the treatment of cough and asthma could be justified.

The Effect of Petasites japonicus Extract on Hepatotoxicity in Rats

  • Petasites japonicus (Compositae) is a perennial herb which has been used as treatment of antitussive, expectorant, sedatives, paralysis, diurectics in folk remedies. The pharmacological studies of this natural drug have not yet estabilished. So, we examined anti-lipid peroxidative effects and liver protective effect on $CCl_4$ induced lipid peroxidation and hepatotoxicity in rats. In vivo liver lipid, MeOH Ex. revealed significant increase of anti-lipid peroxidative effects according to concentration dependently. In chemical parameters obtained from serum analysis, MeOH Ex. as a increase of medicine concentration (0.25 g/kg, 0.5 g/kg, 1.0 g/kg), GOT, GPT, AlP decreased. In 0.5 g/kg GPT administered group, there was relative in GOT, AlP. Cholesterol decreased in 1.0 g/kg and 1.0 g/kg administered group, BUN decreased relatively in 1.0 g/kg administered group.
