함초 추출액을 첨가한 브라운 소스의 품질 특성 (Quality Characteristics of Salicornia herbacea L. Extract Added Brown Sauce)

  • This study performed physicochemical characteristics and sensory tests on brown sauce with different Salicornia herbacea L. (saltwort) content to find the best brown sauce recipe with saltwort. Moisture content increased with greater saltwort extract content. There was significant (p<0.001) pH difference with saltwort content. There was also significant (p<0.001) sweetness difference in the samples and sweetness increased with greater saltwort content. Viscosity measured at $20^{\circ}C$ and $60^{\circ}C$ was significantly (p<0.001) lower with greater saltwort content. The brightness of brown sauce containing saltwort extracts significantly (p<0.001) increased with greater saltwort content and its redness and yellowness usually decreased. SPS4 with the greatest saltwort content showed the highest value of 4.25% and DPPH radical removal increased significantly (p<0.001) with greater saltwort content. In the sensory test, the color value was lowest at 4.82 with BSL1 and the flavor value was lowest at 5.00 with BSL0, the control group. The taste and the flavor values were highest with BSL2 with 2% saltwort content. Overall acceptance was highest with BSL3, the brown sauce with 3% saltwort content, at 6.09. As a result, it was concluded that the brown sauce with 3% saltwort content was most suitable. Therefore, this study concluded that saltwort with various functions could be used for food and may be used to replace salt and have other functions in brown sauce, a popular sauce consumed in Korea.

  • 식용대두유(食用大豆油)에 0.02% BHT, 0.01% citric acid 및 0.1 ppm $CuSo_4\;5H_2O$를 단독 또는 혼합 첨가하여 암소저장조건하(暗所貯藏條件下)$(40{\pm}0.5^{\circ}C)$ 및 일사광선조사조건하(日射光線照射條件下)에서 저장(貯藏)하여 과산화물가(過酸化物價)와 유리지방산가(遊離脂肪酸價)의 변화(變化)를 측정하여 다음과 같은 결과(結果)를 얻었다. 1. 일반적(一般的)으로 일사광선조사조건(日射光線照射條件)에서는 암소저장조건(暗所貯藏條件)에서 보다 과산화물가(過酸化物價)와 유리지방산가(遊離脂肪酸價)가 빠른 속도로 상승하였다. 2. $CuSO_4{\cdot}5H_2O$를 첨가한 시료구(試料區)에서는 일사광선조건(日射光線條件)과 암소저허조건(暗所貯虛條件)에서 과산화물(過酸化物)과 유리지방산(遊離脂肪酸)을 강(强)하게 형성(形成)하였다. 3. BHT에 대한 citric acid의 상승효과는 암소저장조건(暗所貯藏條件)과 일사광선조건(日射光線條件)에서 모두 강(强)하게 나타났다.

First record of a marine microalgal species, Micractinium singularis (Trebouxiophyceae) isolated from Janghang Harbor, Korea

  • A eukaryotic microalga was isolated from seawater in Janghang Harbor, Korea and its morphological, molecular, and physiological characteristics were investigated. Due to its simple morphology, no distinctive characters were found by morphological observation, such as light microscope or scanning/transmission electron microscopy (S/TEM). However, molecular phylogenetic evidence inferred from the concatenated small subunit (SSU) 18S rRNA and internal transcribed spacer (ITS) sequence data indicated that the isolate belonged to the newly described Micractinium singularis. Furthermore, it was clustered with Antarctic Micractinium strains and it also showed a psychrotolerant property, surviving at temperatures as low as 5℃. However, its optimal growth temperatures range from 15℃ to 25℃, indicating that this microalga is a mesophile. Additionally, gas chromatography-mass spectrometry (GC/MS) analysis showed that the isolate was rich in nutritionally important omega-3 polyunsaturated fatty acid, and high-performance liquid chromatography analysis (HPLC) revealed that the high-value antioxidant lutein was biosynthesized as an accessory pigment by this microalga, with glucose as the major monosaccharide. Therefore, in this study, a Korean marine M. singularis species was discovered, characterized, and described. It was subsequently added to the national culture collections.

동결 건조한 가지 분말을 첨가한 설기떡의 품질특성 (Quality Characteristics of Sulgidduk Prepared with Added Freeze Dried-eggplant Powder)

  • The results of testing general components, physical property and physical function after having made steamed rice cake by adding eggplant, which caters to consumers' tastes are as follows. It showed that when eggplant powder content increased, the moisture content of steamed rice cake with eggplant was decreased. It also showed that the lightness value (L) of steamed rice cake with eggplant powder was the highest as 82.75 in the control group. When eggplant powder content increased, the redness (a) and yellowness (b) were increased. From the results of having measured the antioxidant potential over the steamed rice cake with freezing-drying eggplant powder by DPPH radical elimination, we figured out that it was the lowest as 48.30% in the control group. As more eggplant powder was added, its elimination increased accordingly. As per hardness of steamed rice cake with eggplant, the added group with 7% was revealed as the highest at 0.757. It showed that adhesiveness was the lowest as 30.233 in the control group, and springiness and cohesiveness were on the rise as freezing-drying eggplant powder was increased. Furthermore, softness and chewiness were shown to be high in the added group with eggplant powder of 3, 5%. The evaluation over the overall preference was the highest in 5% added group. Judging from this, adding 3~5% eggplant powder against non-glutinous rice powder is considered an optimal quantity in making steamed rice cake with eggplant.

  • A protein isolate was prepared from black soybean (Glycine max L. Merrill) that possessed higher antioxidant activity than ordinary white soy protein isolates. The isolate was partially hydrolyzed by alcalase to reduce the allergenicity of black soybean. Alcalase remarkably reduced the molecular mass of the major soybean allergens that have molecular weights of 53, 38, and 24 kDa. Hydrolytic breakdown occurred more effectively in Gly m Bd 30K than in Gly m Bd 60K or Gly m Bd 28K. Alcalase hydrolysis increased the solubility and hydrophobicity of the black soybean protein isolate. The foaming activity and stability of black soybean proteins were highly increased by the partial hydrolysis.

Characterization of Physicochemical Properties of Ferulic Acid

  • Ferulic acid (3-methoxy, 4-hydroxy cinnamic acid) is a flavoid component possessing antioxidant property. The compound is currently under development as a new drug candidate for the treatment of the dementia. The objective of this preformulation study was to determine the physicochemical properties of ferulic acid. The n-octanol to water partition coefficients of ferulic acid were 0.375 and 0.489 at the pHs of 3 and 10, respectively. Accelerated stability study for ferulic acid indicated that the t 90 value for the drug was estimated to be 459 days at $25^{\circ}C$. Ferulic acid was also found to be unstable under the relative humidity of more than 76%, probably because of the hygroscopic nature of the drug. In order to study compatibility of ferulic acid with typical excipients, potential change in differential scanning calorimetry spectrum was studied in 1: 1 binary mixtures of ferulic acid and typical pharmaceutical excipients (e.g., Aerosil, Avicel, CMC, Eudragit, lactose, PEG, PVP, starch and talc). Avicel, CMC, PVP and starch were found to be incompatible with ferulic acid, indicating the addition of these excipients may complicate the manufacturing of the formulation for the drug. Particle size distribution of ferulic acid powder was in the size range of 10-190 $\mu$m with the mean particle size of 61 $\mu$m. The flowability of ferulic acid was apparently inadequate, indicating the granulation may be necessary for the processing of the drug to solid dosage forms. Two polymorphic forms were obtained by recrystallization from various solvents used in formulation. New polymorphic form of ferulic acid, Form II, was obtained by recrystallization from 1,4-dioxane. The equilibrium solubility for Form I was approximately twice of that for Form II. The dissolution rate of Form II was higher than that of Form I in the early phase (<6 min). Therefore, these physicochemical information has to be taken in the consideration for the formulation of ferulic acid.

장기간 발효 김치인 묵은지에서 분리한 Lactobacillus sakei JK-17의 기능성 조사 (Functional Characterization of Lactobacillus sakei JK-17 Isolated from Long-term Fermented Kimchi, Muk Eun Ji)

  • The purpose of this work was to investigate the several functional characteristics of Lactobacillus sakei JK-17 isolated from long-term fermented kimchi, Muk Eun Ji. Initially, phylogenetic analysis using 16S rRNA sequencing was performed to identify the isolate JK-17, and the strain could be assigned to Lactobacillus sakei and designated as L. sakei JK-17. The strain was registered in GenBank as [JX841311]. The changes of bacterial growth and residual organic acids were monitored and HPLC was used to measure quantitatively two organic acids, lactic acid and acetic acid, produced in the culture during 84 hours of incubation. During the incubation period, several functional characteristics of L. sakei JK-17 were examined. L. sakei JK-17 culture depleted nitrite concentration 94.75%. Antioxidant activity of cultural supernatants of L. sakei JK-17 was approx. 53.8%, and ${\beta}$-galactosidase activities were 0.243 units/mL at pH 7.0 and 0.387 units/mL at pH 4.1, respectively. The antibacterial activities against food-poisoning causing bacteria were examined with 20-fold concentrated culture supernatants from L. sakei JK-17 and the antibacterial effects were clearly observed against all bacteria tested in this work.

  • The Korean daily intake of vegetable oils has increased about 2.5-fold from 17 g/day to 46 g/day for the last several decades. Perilla (Perilla frutescens var. frutescens) has been cultivated in Korea for a long time as a dietary oil seed which has the highest content of ${\alpha}$-linolenic acid, accounting for nearly 60%. It is known that the main role of ALA is as a precursor to the longer-chain ${\omega}-3$, eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA), the metabolic products of ${\alpha}$-linolenic acid (ALA, ${\omega}-3$). Dietary ${\omega}-3$ fatty acids reduce inflammation and the risk of chronic diseases such as heart disease, cancer, and arthritis, but they also may act as functional components for cognitive and behavioral function. Thus, ${\alpha}$-linolenic acid is one of the essential nutrients in modern dietary patterns in which much linoleic acid is consumed. Nevertheless, perilla oil, rich in ${\alpha}$-linolenic acid, can be easily oxidized, giving rise to controversies with respect to shelf life, the deterioration of the product's commercial value, and further related toxicity. Recent research using genetic modifications has tried to develop new plant oil seeds that balance the ratio of ${\omega}-6/{\omega}-3$ fatty acids. Such trials could be a strategy for improving an easily oxidizable property of perilla oil due to high ${\alpha}$-linolenic acid. Alternatively, appropriate application of antioxidant to the oil can be considerable.

  • 기능성 소재로서의 버찌분말의 이용가능성을 알아보기 위해 버찌분말을 0, 3, 5, 7 및 10% 첨가한 머핀을 제조하여 품질특성을 조사하였다. 머핀의 물성은 버찌분말의 첨가에 따라 경도(hardness)는 증가하였고 검성(gumminess) 및 씹힘성(chewiness)은 감소하였다. 색도는 첨가량이 증가할수록 빵껍질(crust)과 빵속(crumb)에서 명도와 황색도가 감소하고 적색도는 증가하였다. 또한 버찌 분말의 농도가 증가함에 따라 항산화 활성이 증가하였다. 관능검사 결과 버찌분말첨가 머핀에서 색, 향, 맛, 부드러움, 촉촉함, 전체적인 기호도면에서 무첨가구에 비해 전반적으로 높게 평가되었으며, 전체적인 기호도는 5% 첨가구에서 가장 높은 점수를 나타내었다. 이러한 결과로 볼 때 머핀의 품질 및 관능 특성, 기능성을 고려한 버찌분말 첨가량은 5% 첨가구가 최적 조건일것으로 판단되었다.

김치 유래 GABA 생성 Lactobacillus plantarum의 발효 및 항상화 특성 (Fermented Property and Antioxidative Effect of GABA Producing Lactobacillus plantarum from Kimchi)

  • GABA는 식물, 동물 및 미생물 등 자연계에 다양하게 존재하고 있으며, 항고혈압, 이뇨, 진정, 수면유도 및 항스트레스 등 다양한 효과가 있는 것으로 알려져 있어서 식품, 의약품 소재로 활용되고 있다. 본 연구에서는 김치로 부터 분리한 GABA 형성 유산균을 옥수수 수염 추출물에 적용하여 발효 특성과 발효물의 항산화 효과에 대해 분석하였다. 신규 L. plantarum LAB459의 분리와 동정은 생화학적 특성, 당자화성 및 16s rRNA 염기 서열 분석을 통해 확인되었다. 그리고 TLC와 HPLC 분석을 통해 분리된 L. plantarum LAB459가 GABA 생성능이 있는 것으로 나타났다. 그리고 탈지유가 포함된 옥수수 수염 열수 추출물에 대해 발효를 수행한 결과, 동결 건조된 발효물에 약 1 ㎍/mg 수준의 GABA가 형성된 것을 확인하였다. 또한 발효물에 대한 플라보노이드와 항산화 분석은 옥수수 수염 열수 추출물에 비해 더 높은 것으로 나타났다. 따라서 본 연구에서 분리된 L. plantarum LAB459는 다양한 식품 발효의 스타터 또는 식품 소재와 의약품 소재로 적용이 가능할 것으로 판단된다.
