항산화 비타민과 미네랄 보충이 고혈압 노인의 혈압과 혈중 지질에 미치는 효과 (Effects of Antioxidant Vitamins & Minerals Supplementation on Blood Pressure and Lipids in the Elderly with Hypertension)

  • Purpose: The Purpose of this study was to evaluate the effects of antioxidant vitamins and minerals supplementation on blood pressure and lipids in the elderly with hypertension. Methods: This study adopted a unequivalent control group pretest-posttest design. For this study, 79 subjects with hypertension were selected among the home visiting clients of public health centers. The experimental group received 12-weeks' treatment with antioxidant vitamin A, C, E, carotene, magnesium and selenium and the control group did not take any antioxidant vitamin and mineral. Results: There were statistical differences in systolic blood pressure and diastolic blood pressure between the experimental group and the control group after the intervention. In the experimental group, systolic blood pressure decreased from 145.4mmHg before the intervention to 135.7mmHg after the intervention, and diastolic blood pressure decreased from 88.3mmHg before the intervention to 81.9mmHg after the intervention. However, there were no statistical differences in total cholesterol and triglyceride between the experimental group and the control group after the intervention. Conclusions: A short-term supplementation with antioxidant vitamins & minerals can reduce blood pressure in the elderly with hypertension. The continuous effects of the supplementation and its beneficial effects on the prevention of cardiovascular diseases still need to be studied further.

Effect of Quercetin on the Activity and mRNA Expression of Antioxidant Enzymes and Physiological Responses in Olive Flounder (Paralichthys olivaceus) Exposed to Cadmium

  • We investigated the antioxidant efficacy of quercetin (0% Diet 1, 0.25% Diet 2, and 0.5% Diet 3) pretreatment for 30 and 60 days in response to cadmium (Cd) toxicity in the olive flounder, and measured the plasma lysozyme activity to understand the immune effects of quercetin. The lysozyme activity with Diets 2 and 3 was higher than with Diet 1. Based on this result, to examine the immune ability and antioxidant role of quercetin, we exposed olive flounder fed quercetin to Cd and then measured the expression and activity of antioxidant enzymes (superoxide dismutase (SOD) and catalase (CAT)) and lipid peroxidation (LPO). With Diets 2 and 3, the expression and activity of antioxidant enzymes and the $H_2O_2$ concentration were lower than with Diet 1. In addition, the LPO levels were lower than with Diet 1, which protected the cell membrane. Therefore, quercetin removed the reactive oxygen species (ROS) produced by Cd, indicating that quercetin has antioxidant ability. In addition to its antioxidant ability, quercetin has immune effects.

  • This study was undertaken in order to investigate the effects of Dioscorea daemona stem on the inflammatory reactions and antioxidant system in vivo. The methanolic extract of Dioscorea daemona stem, administered by gavage at the dose of 0.2 $\mu\textrm{g}$/kg, showed anti-inflammatory and anti-allergic (typeIV) effects in different test models. We also showed that plasma antioxidant enzyme activities, lipid peroxidation and HDL cholesterol levels are affected by administration (0.2 $\mu\textrm{g}$/kg body weight, for 3 weeks) of Dioscorea daemona stem extract in rats.   • Boiled-water extract (BWE) of Hizikia fusiformis has antioxidant effects. To investigate antioxidant effects of BWE, 1,1-dephenyl-2-picrylhydrazyl (DPPH) and thiobarbituric acid (TBA) methods were used. BWE exhibited strong radical-scavenging activity (87.3% at final conc. $2000\;{\mu}g$ BWE/ml in an assay mixture) on DPPH and good inhibitory activity (64% at final conc. $1000\;{\mu}g$ BWE/ml in an assay mixture) on lipid peroxidation. Raw 264.7 cells treated with lipopolysaccharide (LPS) showed 26.7-fold increase in nitric oxide (NO) ($48.9\;{\mu}M$) compared with control group ($1.83\;{\mu}M$). When treated with BWE and LPS, NO production was inhibited by BWE dose-dependently.

비자 추출물의 식품부패균에 대한 항균효과 및 항산화활성 (Antimicrobial Effects on Food-Borne Pathogens and the Antioxidant Activity of Torreya Nucifera Extract)

  • This study investigates antimicrobial effects of food-borne pathogens and the antioxidant activity of Torreya nucifera extract. The growth of food-borne pathogens including Staphylococcus aureus, Pseudomonas aeruginosa, and Candida albicans was inhibited by the extract. The antimicrobial activity of the extract was highest for Staphylococcus aureus among seven gram-positive bacteria and for Pseudomonas aeruginosa among six gram-negative bacteria. The extract exhibited slightly lower DPPH radical-scavenging activity, but its ABTS radical-scavenging activity was higher than that of ${\alpha}$-tocopherol. The results demonstrate the extract's antimicrobial effects on food-borne pathogens as well as potent antioxidant capacity and suggest that Torreya nucifera may be used as a natural antibacterial agent and an effective antioxidant in food.

가시오갈피 추출물의 항산화효과 (Antioxidant Effects of the Extracts of Acanthopanax senticosus)

  • Antioxidant properties of the extracts of Acanthopanax senticosus were investigated. The dried roots, stems or leaves were extracted with hot water or ethanol each. The ethanol extracts exhibited higher potency than aqueous extracts in scavenging free radicals and in inhibiting microsomal lipid peroxidation: the aqueous extracts of stems showed higher anti-oxidant effects than the root extracts. Copper-mediated LDL oxidation was also protected by the ethanol exlracts: antioxidant effects of the extracts tested were stronger than ascorbic acid, but not butylated hydroxytoluene. The activity of angiotensin converting enzyme was effectively suppressed by the aqueous extracts of the stems. However, in vivo antioxidant properties of the ethanol extracts of the stems did not seem to be significant, judged from the lipid peroxide values of serum and liver in normal mice. Thus, the ethanol extracts of the stems were shown to be more potent for protecting biological systems against various oxidant stresses in vitro, but not in vivo.

  • The beneficial effects of fruits, vegetables, and beverages on human health have been attributed to their antioxidant activities. Therefore, antioxidant activity of food products is recognized as one of the important parameters in determining their functional values. Until now, antioxidant activity has been measured by various chemical and biological methods; however, many factors confound the reliability and reproducibility of measurements of antioxidant activity of food. In vitro methods may provide a useful indication of antioxidant activity but their results may not translate to the human biological system, while in vivo tests are difficult to carry out due to the intricate processes of uptake, cellular transportation, and metabolism of individual antioxidant components. Therefore, as long as these limitations exist, our best option is to measure the antioxidant activity in food directly. This review briefly summarizes currently available methods for the measurement of antioxidant activity in food and examines their respective validity.

포공영, 금은화, 익모초 혼합물의 항산화 및 항염증 효과에 관한 연구 (The Study on Antioxidant and Anti-inflammatory Effects of Taraxacum platycarpum H. Dahlstedt, Lonicera japonica Thunberg and Leonurus japonicus Houtt. Complex)

  • Objectives: This study was designed to experiment with the antioxidant and anti-inflammatory effects of Taraxacum platycarpum H. Dahlstedt, Lonicera japonica Thunberg, and Leonurus japonicus Houtt. complex (TLL) in LPS-induced RAW264.7 cell. Methods: The antioxidant activity of TLL was measured by FRAP assay, DPPH radical scavenging activity, ABTS radical scavenging activity. Total polyphenol and flavonoid contents of TLL were measured by using standard methods. The anti-inflammatory effects of TLL were measured by NO production, biomarker production (PGE2, IL-1β, IL-6, TNF-α), mRNA expression level (iNOS, COX-2, IL-1β, IL-6, TNF-α) and protein expression level (ERK, JNK, p38). Results: Total polyphenol and flavonoid contents in TLL were 58.03±1.02 mg of Gallic acid equivalents (GAE)/g and 16.58±0.60 mg of Quercetin equivalents (QE)/g respectively. In FRAP assay, DPPH and ABTS radical scavenging activity, a concentration-dependent increase in TLL was observed. To explore antioxidant and anti-inflammatory effects of TLL, RAW 264.7 cells were treated with TLL and LPS for 24 hours. Cell viability of RAW 264.7 cells were measured by adding EZ-Cytox, It was remarkably increased at 50, 100, 200 ㎍/㎖ concentrations of TLL. NO, ROS, iNOS, IL-1β, IL-6, TNF-α, ERK, JNK and p38 were remarkably decreased at 50, 100, 200 ㎍/㎖ concentrations of TLL compared to the control group. PGE2 and COX-2 were remarkably decreased at 100, 200 ㎍/㎖ concentrations Conclusion: These results suggest that TLL complex has antioxidant and anti-inflammatory effects.

초식동물 쓸개즙 추출물의 천연항산화 성분; 생물학적인 기능 및 특성규명 (Natural Antioxidant Activity of Ethanol Extracted from Bovine Bile ; Biological Effects and Characterization)

  • 본 연구에서는 Bovine bile에서 항산화 물질분획을 분리하고 이에 따른 항산화 효소들의 여러 활성효과를 측정하였으며 GC/FID에 의한 항산화성 물질의 패턴을 확인하였다. 항산화력은 Butylated hydroxy toluene(BHT)에 비해 Bovine bile의 EtOAc추출물(BBE)이 51.2%로 나타났고 xanthine oxidase를 측정 48.3%의 저해효과를 보였으며 glutatione-S-transferase의 활성은 85.7%의 활성이 나타났다. MTT방법에 의한 세포생존율의 조사에서는 6mM glutathione 투여에서 95%라는 생존율과 비교하여 20mg, 60mg의 BBE를 처리했을 때 56%와 65%, 140mg처리에서는 78%의 세포생존율이 나타났다. Bovine bile EtOAc추출물을 TLC로 분석한 결과 Rf값이 0.72에서 항산화 활성이 나타났고 이 분획을 methylation과 trimethysilyl(TMS) 유도체화를 시켜서 Gas Chromatography(FID)와 mass spectrum을 통한 성분 분석을 하고 bile acid의 구성분과 비교하여 계통분석을 실시한 결과 Coprostan-3ol로 추정되었다.

The Study of Antioxidant Effects in Melanins Extracted from Various Tissues of Animals

  • This study was carried out to investigate the antioxidant effects of melanin extracts, including silkie fowl skin melanin (SS-melanin), silkie fowl comb melanin (SC-melanin), sepia ink sac melanin (SE-melanin), octopus ink sac melanin (OC-melanin) and synthetic melanin (SY-melanin). The results showed that with the addition of melanin extracts, linoleic acid peroxide significantly, decreased (p<0.05) with the increase in the irradiative time of UV and that OC-melanin had the highest efficiency on antioxidant activity (p<0.05). Melanin extract had reducing power and chelating power to $Fe^{2+}$, which increased with the increase in the different melanin concentration. Therefore, it could be concluded that the antioxidant action of melanin extracts did not come from one single function, but is a result of many characteristic functions.
