The antimutagenic activity of resveratrol on the mutagenicity induced by Trp P-1 (3-amino-1,4-dime-thyl-5H-pyrido{4,3-b}indole) was studied using the Ames test with Salmonella typhimurium TA98 and 100. Trp P-1 showed strong mutagenecity in S. typhimurium TA98, but was higly decreased mutagenecity in S. typhimurium TA100. This result suggests that the mutagenecity of Trp P-1 can be mainly induced by the DNA lesions causing frame shift. Resveratrol itself did not show antibacterial effect upon 300 $\mu\textrm{g}$/assay. Resveratrol showed the strongest inhibitory effect with dose dependent manner on the mutagenicity induced by Trp P-1. The inhibition rates of resveratrol at concentration of 2, 10, 25, 50, 100, 300 $\mu\textrm{g}$/assay were 13%, 37%, 52%, 65%, 81%, 89%, respectively.
By the 505 chromotest which utilized Escherichia bolt PQ 37, Korean medicinal plants had been screened to Investigate the antimutagenic effect to aflatoxin B1(AFBl). Ikmocho(IMC, Leonurus sibiricus L.) was extracted with hot water. The extract was not found to be mutagenic in the Salmonella mutation test with or without metabolic activation, and the extract was showed to possess the antimutagenic properties towards AFB1-induced metation. The mutagenicity of AFB1 was inhibited by methanol soluble fracstion (IMC-MS) in dose-dependent. However, water-soluble fraction exhibited comutagenic activity. The greatest inhibitory effect of IMC-MS on AFB1 mutagenicity occurred when IMC-MS was first incubated, AFB1 followed by a second incubation with the cells and 59 mixture. Also lower inhibition was occurred when S9 mixtures were first incubated, with IMC-MS followed by a second incubation with AFBI. The results of the sequential incubation study support the probability that one mechanism of inhibition could involve the formation of chemical complex between IMC-MS and AFB1 rather than deactivation of S9 enzyme.
To determine whether the omega 3 family, linolenic acis(LnA) is effective to inhibit carcinogens/mutagens-induced mutagenesis, we employed the Ames test using Salmonella typhimurium strain of TA100 and the SOS chromotest using Escherichia coli PQ37 strain. The inhibitory effect of LnA shown in the Ames assaying system was 95%, 78% and 73% when the mutagenicities were mediated by AFB$_{1}$, MNNG and 4-NQO, respectively. LnA shows a strong antimutagenic activity against indirect mutagen of AFB$_{1}$, whereas the same concentration of LnA exhibited weaker inhibitory effects on the direct mutagen of MNNG and 4-NQO than that of AFB$_{1}$. However. LnA reduced more than 80% of SOS responses induced by MNNG and 4-NQO when the adding concentration increased to 5%. We conclude that LnA contains in vitro antimutagenic properties and that this finding warrants further investigation both in vitro and in vivo to assess its possible chemotherapeutic potential.
Antigenotoxicity test (SOS chromotest) and antimutagenecity test (Ames test) were carried out using water-soluble and methanolic extracts from Dianthi Herba. Antigenotoxic activity of methanolic extract against mutagens both MNNG and NQO was much more effective than that of water-soluble one. When the extract was added to the certain concentration $(100\;{\mu}l/tube)$, antigenotoxic activities against both mutagens were enhanced. Against the mutagen MNNG with Ames test, antimutagenic activity of the methanolic extract was better than that of water-soluble one. The 74.6% of inhibition ratio for revertant forming CFU/plate was shown at $300\;{\mu}l/plate$ of the methanolic extract.
The effects of various fractions from the aerial parts of Echinosophora koreensis Nakai on the antimutagenic and the immuno-regulating activities were evaluated by in vivo bone-marrow micronucleus test and HA titer reaction. No significant suppressive effects of these extracts and echinoisosophoranone were shown on cyclophosphamide-induced micronuclei, but HA titers were significantly enhanced in ether and BuOH extract treated-group. Tetracosanol and docosanol were isolated from the ether extract of this plant.
To confirm the effects of binlang(Areca catechu L.) and tansen(Salvia miltriorrhiza bung) on the mutagenicity induced by hydrogen peroxide, SOS Chromotest with Escherichia coli PQ37 and Ames test with Salmonella typhimurium TA104 were perlormed. Methanol-soluble parts of their water extracts showed high inhibitory effect against the mutagenicity of hydrogen peroxide in two bacterial mutation assays. Step-wise fractionation of methanol-soluble part from tansen was done using ethyl acetate, butanol and water. Among these fraction was further partitioned by Sephadex LH-20 column chromtography, and 6 subfractions were obtained. The fraction III showed the strongest inhibitory effects against the mutagenic activities induced by hydrogen peroxide. The inhibition rates of fraction III at concentration of 500$\mu\textrm{g}$/assay were 28%, 30% and 15% against 4-NQO, MNNG and B(a)P, respectively. But the mutagenic potency of AFB1 was increased.
The anti-mutagenic effect of Orostachys japonicus (OJ) toward aflatoxin (AFB$_{1}$) and N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) in the Salmonella assay system was studied. The methanol extract of OJ inhibited the mutagenicity induced by AFB$_{1}$ about 97% when 5% of the extract added to the system. Butanol fraction from the methanol extract was the most effective against AFB$_{1}$. However, other fractions of hexane, chloroform, and ethylacetate also showed considerable antimutagenic activity against AFB$_{1}$. Several identified compounds from the fractions of OJ exhibited anti-mutagenic effect. $\beta$-Sitosterol, astragalin and kaempferol-3-rhamnosyl-7-glucoside were selected from the compounds, and these compounds inhibited the mutagenicity dose-dependently. These 3 compounds also decreased the mutagenicity induced by MNNG. From these results, it is suggested that the major compounds such as triterpene, sterol and flavonoid in the OJ were responsible for the inhibition of the AFB$_{1}$ and MNNG-induced mutagenicities.
In order to determine the effectiveness of linoleic acid(LA) to inhibit carcinogens/mutagens-induced mu-tagenesis, Ames test using Salmonella typhimurium TA100 and the SOS chromotest using E. Coli PQ37, were carried out. The inhibitory effect of LA(1%) on the Ames mutagenicity test were 98%, 78%and 69% mediated by aflatoxin B₁(AFB₁), N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) and 4-nitroquinoline-1-oxide(4-NQO), respectively. LA exhibited a strong antimutagenic activity aganist indirect mutagen, AFB₁whereas exhibited the same concentration of LA showed weaker inhibitory effects on direct mutagens of MNNG and 4-NQO than that AFB₁. LA also reduced the SOS responses induced by MNNG and 4-NQO significantly. This result showed a possibility that LA can be a protective agent in early step of cancinogenesis.

The antimutagenicities of methanol extracts from traditional and commercial kochujang (Korean Red Pepper Soybean Paste) and their ingredients were evaluated in Salmonella/mammalian microsome assay system.
