This study was performed to determine the antimutagenic and cytotoxic effects of ethanol extract, ethylacetate fraction, water fraction and water soluble and insoluble polysaccharides I and II isolated from Inonotus obliquus using Ames test and SRB assay. Each sample itself did not show mutagenic effect. Among samples, the water unsoluble polysaccharides II in the presence of 50 $\mu\textrm{g}$/plate showed the strongest antimutagenic effect with over 90% against MNNG, 4NQO, B(a)P and Trp-P-l. However, ethyl acetate fraction (1 mg/mL) which had 90.8%, 94.3% and 83.5% inhibitory effect against on MCF-7, A549, AGS showed the strong cytotoxic effect compared to other samples.
In order to investigate the antimutagenicity of the sweet potato enzymatic browning reaction products (SPEBRP) were studied the DNA breaking action, spore rec assay and Ames test. In the DNA breaking action of reaction mixture of SPEBRP and polyphenol compounds with an agarose horizonal electrophoresis, catechol (CAT)-SPEBRP and hydroxyhydroquinone (HHQ)SPEBRP inhibited DNA breaking effect in the presence of $Fe^{2+}$. In the spore ree assay using Bacillus subtilis H17(rec+) and M45(rec-), 3,4-dihydroxytoluene (DHT)-SPEBRP showed strong antimutagenic effects on MNNG. In the Ames test using Salmonella tYPhimurium TA 98 and TA 100, pyrogallol(PYR)-, 3,4-dihydroxytoluene (DHT)- and hydroxyhydroquinone (HHQ)-SPEBRPs suppressed about 67%, 71% and 63% in the mutagenesis induced by Benzo($\alpha$)Pyrene(B($\alpha$)p).
The antimutagenic effects of 27 kinds of plant flavonoids on the mutagenicity of aflatoxin $B_1(AFB_1)$ and N-methyl-N'-nitro-N-nitrosoguanidine(MANG) in Salmonella typhimurium TA 100 were investigated. In the mixed applications of $AFB_1\;(1\;\mu{g/plate)}$ with the flavonoids $(300\;\mu{g/plate)}$ in the presence of a mammalian metabolic activation system (S9 mix), chrysin, apigenin, luteolin and its glucoside, kaempferol, fisetin, morin, naringenin, hesperetin, persicogenin, (+)-catechin and (-)epicatechin showed the antimutagenic effect against $AFB_1$ with more than 70% inhibition rate. A little or no antimutagenicities except flavone against MNNG $(0.5\;\mu{g/plate)}$ were observed. For the antimutagencity of the flavonoids on $AFB_1$, the flavonoid structure that contains the free 5, 7-hydroxyl gorup seemed to be essential. However, saturation of the 2, 3-double bond of elimination of the 4-keto group did not affect the activity.

The methanol extracts of perilla leaves reduced the mutagenicities mediated by of aflatoxin B$_1$(AFB$_1$), 3-amino-l-methyl-5H-pyrido (4,3-b) indole (Trp-P-2) and Benzo(a)pyrene (B(a)p) in Salmonella typhimurium TA98 and TA100. The methanol extracts were more fractionated, and the fractions of hexane and butanol revealed the antimutagenic activities against AFB$_1$ and B(a)p. The production of malondialdehyde (MDA) was decreased when the methanol extracts of perilla leaves were added to the system. The significantly higher antioxidative activity was observed in the butanol fraction. 2-Propyl furan, ethanedioic acid, dibutyl ester, benzaldehyde, 2-methyl-2-ethyl-3-hydroxy-propanoic acid and octahydro-3a-methyl-2H-inden-2-one were identified tentatively as major compounds from the butanol fraction.

This study was performed to determine the antimutagenic and cytotoxic effect of the Phellinus linteus methanol extract on Salmonella typhimurium TA98, TA100 and human cancer cell lines. In the Ames test, methanol extract of P. linteus alone did not exhibit any mutagenicity but showed substantial inhibitory effects against mutation induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG), 4-nitroquinoline-nitroquinoline-1-oxide(4NQO), 3-amino-1,4-dimethyl-5H-pyrdo[4,3-blindol(Trp-P-1) and benzo(α)pyrene(B(α)P). The methanol extracts of P. linteus(200㎍/plate) showed approximately 78.3%, 78.7% and 88.1% inhibitory effect on the mutagenesis induced by 4NQO, Trp-P-1 and B(α)P. The anticancer effects of P. linteus extract against human breast adenocarcinoma(MCF7), human lung carcinoma (A549), human fibrosarcoma (HT1080), human hepatocelular carcinoma (Hep3B) and human epitheloid carcinoma (HeLa) were investigated. The treatment of 1mg/mL P. linteus extracts had the highest cytotoxicity against MCF7 (92.0%), followed by Hep3B (84.9%), A549 (84.2%) and HT1080 (82.9%). In contrast 1mg/mL treatment of P. linteus extracts had only 10∼40% cytotoxicity on normal human liver cell (WRL68).
The antimutagenic mechanism of the fraction III(RG III)separated from the water extract of Rehmannia glutionosa was investigated by Escherichia. coli GW and B/r strains. RG-III treatment did not affect the ${\beta}$-galactosidase activity E. coli GW-1060, 1106, 1107 and 1105. These results indicated that RG-III did not induce RecA protein amplification and did not also prevent the proteolytic cleavage of LexA. The bio-antimutagenicity and survival effect of RG-III on 4-nitroquinoline 1-oxide(4NQO), N-methyl-N-nitor-N\`-nitrosoguanidine(MNING) were investigate by E. coli B/r strains with have different pathway of DNA repai. RG-III slightly increased the survival of 4NQO-treated WP2, WP2s, WP67, CM561, CM611 cells, but the reactivation of survival cannot ve explained by the repair mode. RG-III caused the decrease of mutagenicity and lethality treated with MNNG in ZA159 despite of the increase in WP2, WP2s, WP67, CW561, CM611. Compared with bio-antimutagenic effects of RG-III on 4NQO, greatly increased antimutagenic effects of RG-III were observed with all the E. coli B/r strains tested, but less active in ZA159. These results suggest that RG-III was identified as a blocking agent for preventing the 4NQO induced mutagenesis, and may act as chl-products.
The antimutagenic activities of ethanol extracts of Korean and American propolis were tested using Salmonella Typhimurium TA98 with two indirect mutagens of 3-amino-1,4-dimethyl-5H-pyrido [4,3-b]indole (Trp-P-1) and 2-aminoanthracene (2-AA) with S9 mix. Additionally, their antimicrobial activities against acne-related pathogenic strains of Propionibacterium acnes, Staphylococcus Epidermidis, Staphylococcus aureus and Pseudomonas aeruginosa were evaluated using both paper disk method and agar dilution method. Ethanol extracts of Korean and American propolis showed strong inhibitory effects, in a dose dependant manner, against the mutagenicities induced by Trp-P-1 and 2-AA. The antimutagenic effect of ethanol extracts of Korean propolis showed significantly higher protective activity than that of American propolis against the Trp-P-1 induced mutagenicity of S. Typhimurium TA98 at the lower concentration ($1-10\;{\mu}g$), but significantly lower protective activity at the higher concentration ($50-200\;{\mu}g$). The antimutagenic effect of ethanol extract of Korean propolis showed significantly higher protective activity than that of American propolis against the 2-AA induced mutagenicity at the concentration of $1\;{\mu}g$, but significantly lower protective activity than that of the American at the higher concentration ($50-200\;{\mu}g$). Both extracts showed strong antimicrobial activities against all the acne-related pathogens tested, with minimal inhibitory concentration (MIC) values in the range $1,500-5,000\;{\mu}g/mL$.
This study was undertaken to investigate the mutagenicity and antimutagenicity of Acanthopanax koreanum Nakai. Antimutagenic study on extract of A. koreanum was studied using the test with Salmonella typhimurium TA100, TA98. And mutagenicity study was studied using the test with S. typhimurium TA100, TA98, TA1535, TA1537 and Escherichia coli WP2 uvr A. A. koreanum was negative in Ames test with S. typhimurium and E. coli with or without S-9 mixture. Test substances of $5000\;{\mu}g/{\mu}l$, $2500\;{\mu}g/{\mu}l$ and $600\;{\mu}g/{\mu}l$ of A. koreanum extracts were chosen via toxicity test. Ames test was performed on positive control group, experimental group and negative control group in the presence of the metabolic activation system and metabolic non-activation system. As a result, there was no coherent increase and reverse mutation in all concentrations. Therefore, A. koreanum does not cause reverse mutation. In addition, A. koreanum showed strong antimutagenic activities in S. typhimurim TA100 and TA98. In conclusion, A. koreanum root may be an excellent antimutagenic agent.
Antimutagenic effects of extracts from curry powder and its individual fourteen kinds of spices, were investigated by Ames test. The antimutagenic effects against a direct mutagen, 2-nitrofluorene(2 -NF) and two indirect mutagens, 2-anthramine(2-AT) and 2-acetamidofluorene (2-AE) in the S. typhimurium TA98 were tested. For the 2-NF, the antimutagenicity of cinnamon, fenugreek, fennel, ginger, clove, turmeric and celery seed were determined as 42, 38, 32, 28, 24, 23 and 20%, respectively. The antimutagenicity of clove against the 2-AT was the highest (116%), and followed by the order of celery seed(103%), cardamon(100%), red pepper(99%), cinnamon(92%), cumin(83%), ginger(82%), fennel(82%), coriander (71%), nutmeg(68%) and turmeric (55%). The results also showed that the antimutagenic effect of clove against the 2-AF was superior to other spices. In case of curry powder among more than 10 kinds of spices, the antimutagenenicity against the 2-AT and 2-AF showed 23% and 6%, respectively, but no effect was observed against the 2-NF.
Antimutagenic effects og Eleutherococcus senticosus Maxim. on the mutagenicty induced by mutagens, 4-NQO, MNNG, $\textrm{NaN}_{3}$, 2-NF, and 1-NP was studied by the Ames test with Salmonella typhimurium TA98 and TA100. The methanol extract ($500\mu\textrm{g}$/plate) of E. senticosus Maxim. showed inhibitory effect on the mutagenicty induced by 1-NP only among the tested mutagens. In S. typhimurium TA98, the methanol extracts of the root, stem and leaf showed inhibitory effects of 54.9, 29.5, and 32.9% inhibition on 1-NP mutagenicity, respectively. In S typhimurium TA100, the methanol extracts of the root, stem, and leaf showed inhibitory effects of 593, 30.2, and 43.6%, respectively. The methanol extract were further fractionated by a subsequent liquid-liquid partition technique with chloroform, butanol, and water. The chloroform ($300\mu\textrm{g}$/plate) fraction of the root, stem and leaf showed the strong antimutagenic effects on the mutagenicity induced by 1-NP in S typhimurium TA98 and TA100. But none or weak antimutagenicities were observed in the butanol and aqueous fraction. The chloroform fractions of root, stem and leaf showed the antimutagenic effects of 61.6~88.6% in a dose-dependent manner. In the antimutagenic mode test, the inhibition effect of root was mainly bio-antimutagenic, whereas stem and leaf were desmutagenic.
