Antibacterial effect of naturally occurring unsaturated fatty acids from Prunus japonica against Propionibacterium acnes

  • The antibacterial activity-guided fractionation of the MeOH extract of seeds of traditional medicinal plant Prunus japonica resulted in the isolation of linoleic acid and cis-11-eicosenoic acids, and their methyl esters. Linoleic acid inhibited the growth of Propionibacterium acnes, the acne-causing anaerobic bacterium, but cis-11-eicosenoic acid, methyl linoleate, and cis-11-eicosenoate were found to be inactive. Together with isolated linoleic acid, authentic saturated and unsaturated fatty acids were also tested against P. acnes with other bacteria and fungi. Most of the unsaturated fatty acids possessed anti-acne (MIC $16-128{\mu}g/mL$) and antimicrobial properties.

  • The term polyketide defines a class of natural products synthesized through the successive condensation of small arboxylic acids, which results in products containing multiple carbonyl or hydroxyl groups, each separated by one arbon atom, as in the structural element CH$_2$C(=0)CH$_2$CH(OE)CH$_2$C(=0)-. Plant flavonoids, fungal aflatoxins, as well as undreds of compounds of different structures that can inhibit the growth of bacteria, viruses, fungi, parasites or human umor cells are included in this diverse group. Some of antifungal polyketides also have immunosuppresive activity. olyketides can vary widely in structure, and the diversity of polyketide structures reflects the wide variety of their iological properties. This review focuses on the biosynthesis of polyketides and recent progress in combinatorial iosynthesis of new hybrid polyketide compounds.

생칠과 화칠의 우루시올 조성 및 생리활성 비교 (Comparison of Urushiol Composition and Biological Activity between Fresh Sap and Fire Distilled Sap of Lacquer Tree)

  • For the comparison of the urushiol composition and biological acitivity between the fresh sap and fire distilled sap of lacquer tree(Rhus vernicifera), we analysed the urushiol composition by HPLC and EI-MS, and investigated the antioxidative , antimicrobial and anticancer acitivities according to solvent fractionations. There was no difference in the urushiol composition between fresh and fire distilled saps of lacquer tree. The hexane frqctionsof two saps showed a strong DPPH radical scaverging activity (RC50 : 7.0-7.5$\mu\textrm{g}$). They also showed a strong antifungal activity onthe spore germination of Cladosporium herbarum(MIC : 8$\mu\textrm{g}$/ml), whereas they have no or low activity against the bacteria(BAcillus subtilis , Escherichia coli). In addition , hexane and butanol fractions of two saps showed a strong inhibitory activity against cultured tumour cell lines (GI50 : 0.35, 12.29$\mu\textrm{g}$/ml) in vitro. These results confirm that the fresh sap and fire distilled sap might have the similar urushiol compositions and biological activities.

키틴과 키토산 분해 미생물 유래 효소의 식품에의 이용 (Food application of enzymes derived from microorganisms degrading chitin and chitosan)

  • Most reports demonstrated the substrate specificity-based kinetic properties of chitin or chitosan degrading enzymes. However, there is virtually less information on the high quality and quantity production of chitin or chitosan hydrolysates having a larger than (GlcN)7 from the hydrolysis of high molecular weight chitosan using specific enzymes and their biological activity. Therefore, the production of such molecules and the discovery of such enzyme sources are very important. Fortunately, the author has established a mass production method of chitosan hydrolysates (GlcN)n, n=2-13 that have been characterized as a potent antioxidant substance, as well as antifungal and antibacterial activities against Penicillium species and highly selective pathogenic bacteria. In addition, preclinical studies using (GlcN)n, n=5-25 demonstrated that these molecules played a very important role in maintaining biometric balance. Collectively, it is implicated that the application of these mixed substances to foods with significant biological activity is very encouraging.

Biological Control Activity of Two Isolates of Pseudomonas fluorescens against Rice Sheath Blight

  • Two isolates of mucous bacteria, mc75 and pc78, were isolated from fungal culture plate as culture contaminants with an interesting swarming motility. Both isolates were identified as Pseudomonas fluorescens based on microscopy, biochemical analysis, Biolog test and DNA sequence analysis of the 16S rRNA gene. Both strains have the exactly the same 16S rRNA gene sequences, and yet their biological control activity were not identical each other. In vitro analysis of antagonistic activity of two isolates against several plant pathogenic fungi indicated that both produced diffusible and volatile antifungal compounds of unknown identities. Treatment of the bacterial culture of P. fluorescens pc78 and its culture filtrate exhibited a strong biological control activity against rice sheath blight in vivo among six plant diseases tested. More effective disease control activity was obtained from treatment of bacterial culture than that of culture filtrate. Therefore, in addition to antifungal compound and siderophore production, other traits such as biofilm formation and swarming motility on plant surface may contribute to the biological control activity of P.fluorescens pc78 and mc75.

Comparison of Chemical Compositions and Antimicrobial Activities of Essential Oils from Three Conifer Trees; Pinus densiflora, Cryptomeria japonica, and Chamaecyparis obtusa

  • The chemical compositions, and antibacterial and antifungal effects of essential oils extracted from three coniferous species, Pinus densiflora, Cryptomeria japonica, and Chamaecyparis obtusa, were investigated. Gas chromatography mass analysis of the essential oils revealed that the major components and the percentage of each essential oil were 16.66% $\beta$-phellandrene and 14.85% $\alpha$-pinene in P. densiflora; 31.45% kaur-16-ene and 11.06% sabinene in C. japonica; and 18.75% bicyclo [2,2,1] heptan-2-ol and 17.41% 2-carene in Ch. obtusa. The antimicrobial assay by agar disc diffusion method showed that $2.2{\mu}g$ of Ch. obtusa oil inhibited most effectively the growth of Escherichia coli ATCC 33312 and Klebsiella oxytoca ATCC 10031, whereas the C. japonica oil gave weak antimicrobial activity. The minimal inhibitory concentration(MIC) values for bacterial strains were in the range of 5.45-21.8 mg/ml depending on essential oils, but most Gram-negative bacteria were resistant even at 21.8 mg oil/ml. P. densiflora oil showed the most effective antifungal activity and the MIC values for Cryptococcus neoformans B42419 and Candida glabrata YFCC 062CCM 11658 were as low as 0.545 and 2.18 mg/ml, respectively. Cryp. neoformans B42419 was the most sensitive to all essential oils in the range of 0.545-2.18 mg/ml. Our data clearly showed that the essential oils from the three conifers had effective antimicrobial activity, especially against fungi.

Antagonistic Bacillus species as a biological control of ginseng root rot caused by Fusarium cf. incarnatum

  • Background: This study aimed to develop a biocontrol system for ginseng root rot caused by Fusarium cf. incarnatum. Methods: In total, 392 bacteria isolated from ginseng roots and various soils were screened for their antifungal activity against the fungal pathogen, and a bacterial isolate (B2-5) was selected as a promising candidate for the biocontrol because of the strong antagonistic activity of the bacterial cell suspension and culture filtrate against pathogen. Results: The bacterial isolate B2-5 displayed an enhanced inhibitory activity against the pathogen mycelial growth with a temperature increase to $25^{\circ}C$, produced no pectinase (related to root rotting) an no critical rot symptoms at low [$10^6$ colony-forming units (CFU)/mL] and high ($10^8CFU/mL$) inoculum concentrations. In pot experiments, pretreatment with the bacterial isolate in the presumed optimal time for disease control reduced disease severity significantly with a higher control efficacy at an inoculum concentration of $10^6CFU/mL$ than at $10^8CFU/mL$. The establishment and colonization ability of the bacterial isolates on the ginseng rhizosphere appeared to be higher when both the bacterial isolate and the pathogen were coinoculated than when the bacterial isolate was inoculated alone, suggesting its target-oriented biocontrol activity against the pathogen. Scanning electron microscopy showed that the pathogen hyphae were twisted and shriveled by the bacterial treatment, which may be a symptom of direct damage by antifungal substances. Conclusion: All of these results suggest that the bacterial isolate has good potential as a microbial agent for the biocontrol of the ginseng root rot caused by F. cf. incarnatum.

Antibiotic and Phytotoxic Activities of Ophiobolins from Helminthosporium Species

  • Twenty isolates of Helminthosporium species were obtained from various grass plants and tested for controlling efficacy on the development of plant diseases. An isolate of Helminthosporium sp. TP-4 was chosen and six antibiotic substances were purified from cultures of the fungus by repeated silica gel column chromatography and preparative thin-layer chromatography. They were identified as ophiobolin a, 6-epiophiobolin A, 3-anhydroophiobolin A, 3-anhydro-6-epiophiobolin A, iphiobolin B, and iphiobolin I mainly by mass spectrometry and nuclear magnetic resonance spectrometry. Ophiobolins inhibited the growth of a grampositive bacterium Streptomyces griseus, but were not active against gram-negative bacteria. They also showed an antifungal activity. In in vivo tests, iphiobolin B exhibited potent controlling activities against rice blast, tomato late blight, and wheat leaf rust with control values more than 90% and 70% at concentration of $500\mu\textrm{m}$/ml and 100 ${\mu}{\textrm}{m}$/ml. Ophiobolin A and 6-epiophiobolin A controlled the development of wheat leaf rust more than 80% at concentrations of 100 /ml and $500\mu\textrm{m}$/ml respectively. 3-Anhydro-6-epiophiobolin A was not active against any plant disease. On the other hand, the A-series ophiobolins other than 3-anhydroophiobolin A showed stronger phytotoxic activity in a leaf-wounding assay using 8 plant species than those of 3-anhydroophiobolin A, ophiobolin B, and ophiobolin I. The results indicate that there is little correlation between antifungal activity and phytotoxicity of ophiobolins.

Pseudomonas aeruginosa 3120으로부터 항생물질의 생산,분리 및 특성 (Production, isolation and characterization of the antibiotic from Pseudomonas aeruginosa 3120)

  • Pellicularia sasakii의 생육을 저해하는 한 균주를 토양으로부터 분리하였으며 Pseudomonas aeruginosa 3120으로 동정하였다. P. aeruginosa 3120의 배양액으로부터 분리 정제된 암갈색의 MRL3120 항생물질은 ethylacetate, chloroform 및 methanol에 용해되었으며 곰팡이뿐만 아니라 gram 양성 및 음성세균에도 활성을 나타내었다. UV, IR, EI-MS spectra 및 다른 물리화학적 특성의 분석결과로부터 MRL3120은 2개의 N-methyl-N-thioformylhydroxylamine과 1개의 Cu 이온으로 구성된 chelate 화합물로서 fluopsin C와 유사한 구조를 가진 것으로 추정되었다. Soybean meal을 포함하는 발효배지에 $CuSO_4$의 첨가는 항진균 활성을 증가시켰으나 EDTA(0.1%)의 존재하에서는 활성이 없었다. Soybean meal 대신 soytone을 포함하는 발효배지에서 MRL3120은 생산되지 않았지만 $CuSO_4$의 첨가에 의해 빠르게 생산되었다.

Antagonistic Activities of Bacillus spp. Strains Isolated from Tidal Flat Sediment Towards Anthracnose Pathogens Colletotrichum acutatum and C. gloeosporioides in South Korea

  • Anthracnose is a fungal disease caused by Colletotrichum species that is detrimental to numerous plant species. Anthracnose control with fungicides has both human health and environmental safety implications. Despite increasing public concerns, fungicide use will continue in the absence of viable alternatives. There have been relatively less efforts to search antagonistic bacteria from mudflats harboring microbial diversity. A total of 420 bacterial strains were isolated from mudflats near the western sea of South Korea. Five bacterial strains, LB01, LB14, HM03, HM17, and LB15, were characterized as having antifungal properties in the presence of C. acutatum and C. gloeosporioides. The three Bacillus atrophaeus strains, LB14, HM03, and HM17, produced large quantities of chitinase and protease enzymes, whereas the B. amyloliquefaciens strain LB01 produced protease and cellulase enzymes. Two important antagonistic traits, siderophore production and solubilization of insoluble phosphate, were observed in the three B. atrophaeus strains. Analyses of disease suppression revealed that LB14 was most effective for suppressing the incidence of anthracnose symptoms on pepper fruits. LB14 produced antagonistic compounds and suppressed conidial germination of C. acutatum and C. gloeosporioides. The results from the present study will provide a basis for developing a reliable alternative to fungicides for anthracnose control.
