A novel compound named 'kanakugiol' was recently isolated from Lindera erythrocarpa and showed free radical-scavenging and antifungal activities. However, the details of the anti-cancer effect of kanakugiol on breast cancer cells remain unclear. We investigated the effect of kanakugiol on the growth of MCF-7 human breast cancer cells. Kanakugiol affected cell cycle progression, and decreased cell viability in MCF-7 cells in a dose-dependent manner. It also enhanced PARP cleavage (50 kDa), whereas DNA laddering was not induced. FACS analysis with annexin V-FITC/PI staining showed necrosis induction in kanakugiol-treated cells. Caspase-9 cleavage was also induced. Expression of death receptors was not altered. However, Bcl-2 expression was suppressed, and mitochondrial membrane potential collapsed, indicating limited apoptosis induction by kanakugiol. Immunofluorescence analysis using α-tubulin staining revealed mitotic exit without cytokinesis (4N cells with two nuclei) due to kanakugiol treatment, suggesting that mitotic catastrophe may have been induced via microtubule destabilization. Furthermore, cell cycle analysis results also indicated mitotic catastrophe after cell cycle arrest in MCF-7 cells due to kanakugiol treatment. These findings suggest that kanakugiol inhibits cell proliferation and promotes cell death by inducing mitotic catastrophe after cell cycle arrest. Thus, kanakugiol shows potential for use as a drug in the treatment of human breast cancer.
The antibacterial and insecticidal properties of ethanol extracts and volatile components extracted from Eugenia caryophyllata, Boswellia carerii, Agastache rugosa, Aristolochia contorta, and Aquilaria agallocha were evaluated. The ethanol extract and volatile component of E. caryophyllata showed strong antimicrobial effect against all strains (Mucor hiemalis, Aspergillus niger, Penicillium funiculosum, Trichodema viride) and the volatile component of B. carterii showed antimicrobial effect against all strains except T. viride. The ethanol extract of E. caryophyllata and A. contorta showed $100\%\;and\;32\%$ mortality against Reticulitermes spertus kyushuensis Morimoto for 48 hours and 72 hours, respectively. In the case of volatile component, R. aryophyllata showed $100\%\;and\;20\%$ mortality against R. spertus and Lyctus linearis GOZE, respectively. The main constitute, eugenol $(92\%)$ among nine components from volatile component of E. aryophyllata were identified as antibacterial active substance.
The effect of two plant growth-promoting rhizobacteria (PGPR) on plant growth and systemic protection against soft rot disease and stem rot disease of canola (Brassica napus), caused by Erwinia carotovora and Sclerotinia sclerotiorum was investigated in a laboratory and a greenhouse. Selected PGPR strains C4 and D8 were treated to canola seeds by soaking. Strains C4 and D8 significantly not only increased plant height and root length about 74% and 40.3% and also reduced disease severity of soft rot disease by 80% by C4 and D8 respectively, compared to the control. Especially strain C4 showed antifungal activity against 6 fungal pathogens, S. sclerotiorum, Rhizoctonia solani, Botrytis cinerea, Fusarium oxysporum, Phytophthora capsici and Colletotrichum acutatum. In greenhouse experiment, the seed treatment of both of them increased plant height, leaf width and leaf length of canola plant to 19.5% and 24.9%, 11.3% and 15.3%, and 14.1% and 20.7% by C4 and D8, respectively, and reduced disease severity of S. sclerotiorium. These results indicate that these two PGPR strains can decrease disease severity and increased plant growth under greenhouse condition. Therefore, these two bacteria have a potential in controlling Sclerotinia stem rot of canola. These strains have to investigate under field condition to determine their role of antibiosis, induced systemic resistance and plant growth promotion on canola.
To obtain protoplasts of Colletotrichum acutatum JC24, conidia were inoculated onto cellophane membrane placed on PDA and incubated at $25^{\circ}C$ for 20 hrs under the dark condition. Cellophane membranes, where mycelia were incubated, were soaked into 2% lysing enzyme solution prepared with 0.02 M phosphate buffer (pH 7.0) including 1.2 M sorbitiol. After treatment in 2% enzyme solution for 2 - 3 hrs, it could be possible to harvest $2-3\;{\times}\;10^6$ protoplasts/mL. The effect of several fungicides on reversion ratio was determined by using the protoplasts obtained from C. acutatum JC24. Any protoplasts could not be reversed to mycelia on reversion PDA amended with $10\;{\mu}\;g\;mL^{-1}$ of propineb. With tebuconazole, inhibition ratio of protoplast reversion was 100 and 0.9% at 0.5 and $0.1\;{\mu}\;g\;mL^{-1}$, respectively, while inhibitory effect on mycelial growth was 85.1 and 75.7%. The inhibitory tendency of carbendazim on protoplast reversion was as same as mycelial growth. In the case of strobilurins, trifloxystrobin and kresoxim-methyl, they only could inhibit protoplast reversion of C. acutatum JC24, when salicylhydroxamic acid (SHAM) was amended into reversion PDA with strobilurins.

Objectives : Illicium verum Hook. f. has been known to possess antimicrobial, antioxidant, antifungal, anti-inflammatory, insecticidal, analgesic, sedative, convulsive activities, it has been rarely conducted to evaluate the immuno-biological activity. The present study was examined to evaluate the anti-inflammatory effects of the Illicium verum Hook. f. water extracts (IVE) in vivo and in vitro. Methods : Cell viability was measured by MTT assay. The relative levels of NO were measured with Griess reagent. iNOS, COX-2, $NF-{\kappa}B$ and target proteins were detected by immunoblot analysis, and levels of cytokines were analyzed by ELISA kit. Anti-edema effect was determined in the carrageenan (CA)-induced paw edema model in rats. Results : All dosages of IVE used in MTT assay had no significant cytotoxicity. The increases of NO production and iNOS expression were detected in LPS-treated cells compared with control. However, these increases were attenuated by treatment with IVE. Also, IVE reduced the elevated production of $TNF-{\alpha}$, $IL-1{\beta}$ and IL-6 by LPS. IVE inhibited the $p-I{\kappa}B$ and translocation of $NF-{\kappa}B$ to nuclear. Furthermore, IVE significantly inhibited the increases of hind paw swelling, skin thicknesses and inflammatory cell infiltrations induced by CA injection. Therefore, IVE will be favorably inhibited the acute edematous inflammations. Conclusion : These results provide evidences that anti-inflammatory effect of IVE is partly due to the reduction of some inflammatory mediators by suppression of $NF-{\kappa}B$ pathway.
Anthracnose caused by Collectotrichum acutatum is the most devastating disease of pepper plants in Korea. In this study, we evaluated the effect of selected antagonistic bacteria on control of anthracnose and plant growth promotion of pepper plants under field conditions. Four different bacterial isolates used in the current study were isolated from the pepper rhizosphere (GJ01, GJ11) and tidal flat (LB01, LB14) in previous studies. Four bacterial isolates, together with a control strain (EXTN-1), showed antifungal activity against C. acutatum in a dual culture assay. To test for plant growth promotion effect, seedling vigor index and growth parameters of pepper were measured under field condition. As a result, all four bacterial isolates were effective for improving plant growth promotion. The strain GJ01 was the most effective in improving the seedling vigor on pepper, but the strain GJ11 in increasing the pepper fruit yield. The incidence of anthracnose was inhibited in the range of 63.2~72.5% by treatment of four bacterial isolates. The current study indicated that the four bacterial isolates could be used as potential biological control agents of anthracnose disease of pepper.
Total 443 isolates of actinomycetes were isolated from turfgrass rhizosphere as potential biological control agents. The two isolates (S11 and S4) showed highest cellulase activity with compared to the other isolates that exhibited a clear zone of 1.2 mm around the colony on cellulose agar medium. S12 strain appeared the most active chitin degrading, which exhibited a 1.2 mm of clear zone. The highest proteolytic activity on skim milk agar was which exhibited a 7.5 mm of clear zone by S2 strain. S1 strain from the soli showed siderophore production ability, which exhibited a 0.6 mm of large clear zone on chrome azurol S agar. The antifungal activity of the volatile compound producing by 4 selected actinomycetes was investigated that inhibition rate against Rhizoctonia solani AG2-2 and Sclerotinia homoeocarpa. Growth inhibition effect of S8 isolate against S. homoeocarpa was appeared to 94.8%, S2 to 76.9%, S5 to 46.1% and S12 to 43.5%. The significant inhibition effects on mycelial growth of S. homoeocarpa were shown on media with four strains. The inhibition effect was the highest with S8 strain treatment at 94.8%.
Insects constitute the largest and most diverse group of animals in the world. They also serve as the hosts or nutrient sources for an immense assemblage of pathogens, parasites, and predators. More than 700 fungal species from 100 genera have adopted an entomopathogenic lifestyle. Although entomopathogenic fungi were studied as only biocontrol agents against a variety of pests in various countries, it has been recently focused their additional roles in nature. They are antagonists to/against plant pathogens, endophytes, and possibly even plant growth promoting agents. The potential antimicrobial effect against fungal plant pathogens by an isolate of entomopathogenic fungi including Beauveria bassiana, Lecanicillium spp., and Isaria fumosorosea have been reported since late 1990s, but wasn't reported pathogenicity of the isolate against pests. Later, a Canadian Lecanicillium sp. isolate and L. longisporium isolated from Vertalec$^{(R)}$ showed simultaneous control effect against both aphid and cucumber powder mildew. Therefore, the antimicrobial activities of 342 fungi isolates collected from various regions and conditions in Korea were evaluated against plant pathogenic fungus Botrytis cinerea using dual culture technique on agar plate. As a result, 186 isolates (54.4%) shown the antifungal activity against B. cinerea. The culture filtrates of selected fungi completely suppressed the growth of the microorganisms, indicating that suppression was due to the presence of antimicrobial substances in the culture filtrate. Mode of action of these fungi against insect involves the attachment of conidia to the insect cuticle, followed by germination, cuticle penetration, and internal dissemination throughout the insect. During infection process, secreted enzymes, proteinous toxins, and/or secondary metabolites secreted by entomopathogenic fungi can be used to overcome the host immune system, modify host behavior, and defend host resources. Recently, secondary metabolites isolated from entomopathogenic fungi have been reported as potential bioactive substances. Generally, most of bioactive substances produced by entomopathogenic fungi have reported low molecular weight (lower than 1,000 g/mol) as peptide and, in contrast the high molecular weight fungal bioactive substances are rare. Most substances based on entomopathogenic fungi were shown antimicrobial activity with narrow control ranges. In our study we analyzed the antimicrobial substances having antagonistic effects to B. cinerea. Antimicrobial substances in our fungal culture filtrates showed high thermostability, high stability to proteolytic enzymes, and hydrophilicity and their molecular weights were differed from substance. In conclusion, entomopathogenic fungi showed pathogenicity against insect pests and culture filtrate of the fungi also shown to antimicrobial activity. In the future, we can use the entomopathogenic fungi and its secondary metabolites to control both insect pest control and plant pathogenic fungi simultaneously.
The study researched germination of the plants and growth of Fungus according to concentration of aqueous extracts in order to provide basic data for developing natural agricultural resources by using Persicaria longiseta. The seed germination of Amaranthus spinosus was inhibited at 25% P. longiseta extract, while Agrostis stolonifera ssp. palustris was not affected at all concentrations tested. Especially, the seed germination rate and fresh weight of Trifolium incarnatum at 20% P. blumei extract were higher than those of control plot. The early growth of most receptor plant seedlings was promoted at 25% and 50% of P. blumei extracts, but the radicle growth of all receptor plants was significantly inhibited at > 25% of P. longiseta extract. The response of receptor plants to P. longiseta extract was different according to the plant species and the plant parts. The growth of plant pathogenic fungus in PDA medium showed an increasing inhibition tendency with increasing concentrations of P. longiseta extract. Especially, P. longiseta extract showed the greatest antimicrobial activity against Phytophthora infestans, Phythium graminicola, and Pythium venterpoolii. The content of total phenolic compound in P. longiseta was higher in leaves (1082.3 mg/L) but lower in roots (228.6 mg/L) and stems (207.8 mg/L), which is an allelopathic chemical. As these results are summarized, P. longiseta have competitive advantage because they release phenolic compounds with allelopathic effect and affect on germination, growth and fungi growth on underground flora compared to native plants and they have eligibility for natural herbicide and germicide.

Torilis Japonica (TJ) has been used as an anti-allergy, antifungal, and antibacterial agent. Recent studies have reported that it also shows anti-cancer effects. It is report that TJ inhibits melanin synthesis in melanocyte in the skin. However, the effect and mechanism of TJ extract (TJE) on Ultraviolet (UV)B-induced photoaging are unknown. In this study, we investigated the preventive effects of TJE on matrix metalloproteinase (MMP)-1 and MMP-3 expressions and the underlying molecular mechanism in UVB-irradiated primary human dermal fibroblasts (HDFs). The effect of TJE on HDF cell viability was determined using the XTT assay and cell counting. MMP-1 and MMP-3 expressions levels were measured by western blotting and real-time PCR analysis. Activations of mitogen-activated protein kinase (MAPKinase), nuclear $factor-{\kappa}B$ ($NF-{\kappa}B$), and activator protein-1(AP-1) were measured by western blotting. Our results showed that TJE effectively reduced UVB-induced MMP-1 and MMP-3 protein and mRNA levels. Moreover, TJE significantly blocked the UVB-induced activation of MAPK (p38 and JNK) and transcription factors ($NF-{\kappa}B$ and AP-1), but not ERK. Taken together, our results suggest that the TJE inhibits UVB-induced MMP expressions in HDFs and its may be a potential agent for the prevention and treatment of skin photoaging.
