전복(Haliotis discus hannai) 추출물의 혈압강하, 항산화능 및 항혈전능에 대한 in vitro 효과 (In vitro Anti-hypertensive, Antioxidant and Anticoagulant Activities of Extracts from Haliotis discus hannai)

  • 본 연구에서는 전복육질과 내장의 추출방법에 따른 추출물의 혈압강하, 항산화, 항혈전 효과에 대한 in vitro 효과를 구명하고자 하였다. 전복육질(abalone body)과 내장(visceral portion)의 80% ethanol 추출물은 ACE(angiotensin converting enzyme)활성에 대해 높은 억제효과를 나타내었으며, 전복육질 추출물의 경우 농도의 증가에 따라 증가되는 경향을 나타내었다. 그러나 내장추출물은 농도증가에 따라 큰 차이가 없는 것으로 나타났다. 수용성추출물의 ACE활성억제효과는 농도별에 따라 증가되는 경향을 나타내었으며, 육질과 내장 간에 큰 차이가 있는 것으로 나타났다. 아질산염 소거활성으로 평가한 항산활 효과는 80% ethanol 추출물의 경우, 전복육질에서 농도증가에 따라 증가된 수준을 나타내었으나 그 수치는 낮은 값을 나타내었다. 내장의 경우, 낮은 농도에서는 육질과 비슷한 수준을 유지하였으나 농도가 증가함에 따라 높은 항산화활성을 나타내었다. 수용성추출물의 항산화효과는 농도증가에 따라 증가된 수준을 나타내었으나 육질과 내장에 큰 차이는 보이지 않았다. 항혈전 효과는 80% ethanol 추출물에서 육질의 prothrombin time이 상대적으로 내장의 prothrombin time보다 높게 나타났다. 수용성추출물의 경우, 항혈전 효과가 거의 없었으며, 전복의 육질이나 내장에 따른 특별한 차이점 또한 없는 것으로 나타났다. 수용성추출물을 48시간 냉장온도에서 저장한 후의 전복육질과 내장의 ACE 활성에 대한 억제효과는 육질과 내장간에 큰 차이가 있었으나 0 time의 값과 큰 차이를 나타내지 않았다. 항산화 효과는 육질과 내장의 경우 농도증가에 따라 직선적인 증가경향을 나타내었으나, 육질에 비해 내장에서 높은 항산화능이 있는 것으로 나타났다. 전복육질과 내장의 prothrombin time은 큰 차이가 없었으며, activated partial thromboplastin time 또한 큰 차이가 없는 것으로 나타났다.

  • Development of new tissue factor (TF) inhibitor is still needed for improved compositions having anticoagulant activity and which can be administed orally or non-intravenously at low doses. Our studies for the development of new TF inhibitors uncovered that aminoalcohols with C-18 alkenyl group, 9-octadecenyl- or 9,12-octadecadienyl groups exhibits in vitro nanomolar level activities. 파래 추출물의 항응고 활성과 항암 활성에 관한 연구 (The Anticoagulant and Anticancer Activities of Enteromorpha intestinalis Extracts)

  • The study was performed to investigate the biological activity of Enteromorpha intestinalis. In order to examine its blood anti-coagulant effects, Enteromorpha intestinalis was extracted with cold water, methanol, hot water, HCl and NaOH. In general, the alkali extract of Enteromorpha intestinalis was approximately 17 times stronger than the control. The anti-cancer effects of select extracts(methanol, hot water, 0.1 N NaOH, 1 N NaOH) were determined in human melanoma cells(Bl6/F10), fibrosarcoma cells(HTl080) and breast cancer cells(MCF7) by MTT assay. With the treatment of 250 ${\mu}g/m{\ell}$ of methanol extracts. HT1080, B16/F10 and MCF7 cell viabilities significantly decreased to 8.06%, 3.62% and 10.10%, respectively. Thus these results strongly support the possibie use of Enteromorpha intestinalis as a functional materials.

카라기난의 유기산 가수분해물의 기능 특성 (Physiochemical Properties of Carrageenan Hydrolysates by Organic Acids)

  • 유기산으로 분해된 카라기난 가수분해물의 각종 생리활성을 실험한 결과, citrate와 malate를 가수분해 매체로 이용할 경우, $100^{\circ}C,\;110^{\circ}C$$120^{\circ}C$ 가수분해물은 B. cereus와 B. subtilis에 강한 항균활성을 나타내었으며, $120^{\circ}C$에서 180분간 가수분해한 것이 B. subtilis에 대해 특별히 강한 항균활성을 나타내었다. 그러나 충치균인 St. mutans에 대한 항균활성은 없는 것으로 확인되었다. Tyrosinase 활성 저해 효과는 $100^{\circ}C$$120^{\circ}C$에서 처리된 시료 중에서 citrate와 malate 처리구가 97% 이상의 높은 tyrosinase 활성을 저해하는 것으로 확인되었다. Citrate로 $100^{\circ}C$에서 90분 처리된 시료의 경우 97.1%, 180분 처리된 것은 99.4%의 저해 활성을 나타내었고, $120^{\circ}C$에서 90분 처리된 시료의 경우 98.2%의 tyrosinase 활성을 억제하는 것으로 나타났다. 한편, malate 처리구도 처리온도와 처리시간에 따라 약간의 차이는 있지만 전체적으로 citrate 처리구와 비슷한 결과로 $98{\sim}99.5%$의 높은 tyrosinase 활성 저해 효과를 나타내었다. 당질 농도에 따라 tyrosinase 활성 저해 효과가 다르게 나타났으며, 환원당 및 전당의 농도에 비례하는 것을 확인할 수 있었다. 항혈액응고활성은 유기산의 종류에 따라 차이를 나타내고 있는데, 0.3% malate 용액으로 $80^{\circ}C$에서 180분간 처리 시료의 경우 APTT가 $2,451{\pm}18(sec)$로 항혈액응고활성이 가장 높았으며, 동일한 조건에서 얻어진 0.3% citrate 가수분해물이 $1,617{\pm}15(sec)$였으며 lactate, succinate, acetate 가수분해물 순으로 높았다. 가수분해가 진행됨에 따라 분자량이 적어지고 결국은 황산기 함량도 낮아지게 됨으로써 항혈액응고활성이 낮아지는 것으로 여겨졌다.

Isolation and Characterization of Proteoglycan Derived From Human Placenta and its Biological Activities

  • Chondroitin sulfates proteoglycans were isolated from human placenta. For the identification of enzymatic digestion products of isolated proteoglycan, strong anion exchange-high performance liquid chromatography (SAX-HPLC) was performed. By the action of chondroitin ABC and chondroitin B lyase, three unsaturated disaccharides 2-acetamide-2-deoxy-3-O-($\beta$-D-gluco-4-enepyranosyluronic acid)-D-galactose ($\delta$Di-OS), 2-acetamide-2-deoxy-3-O-($\beta$-D-gluco-4-enepyranosyluronic acid)-6-O-su lfo-D-galactose ($\delta$Di-6S) and 2-acetamide-2-deoxy-3-O-($\beta$-D-gl uco-4-enepyranosyluronic acid)-4-O-sulfo-D-galactose ($\delta$Di-4S) were produced from the human placenta proteoglycan. The anticoagulant activity of chondroitin sulfate proteoglycan was evaluated by activated partial thromboplastin time (aPTT) assay and thrombin time (TT) assay. The clotting times of aPTT and TT were increased from 72 to 144 sec and 19 to 27 sec, respectively. The Immune-modulating activity of chondroitin sulfate proteoglycan was examined by cell proliferation assay and these results suggest that it may play a role in suppression of the function of immune-related cells.

Effects of Insect Crude Drugs on Blood Coagulation and Fibrinolysis System

  • The in vitro anticoagulant and fibrinolytic activities of crude extracts from insects were evaluated in order to find effective therapeutic drugs for the treatment of myocardial and cerebral thrombosis. We prepared three types of extracts (water, methanol and ethylacetate) from 28 insects for use as raw materials for the activity assays. The fibrinolytic activity was tested using the fibrin plate method and the activated partial thromboplastin time and thrombin time were measured for blood clotting activity. With regards to the fibrinolytic system, water extracts of six kinds of insects displayed a remarkable level of activity with a plasmin-like action. The water extracts of [Catharsius molossus, Eupolyphaga sinensis, Huechys sanguinea, Mantidis $o\ddot{o}theca$, Mimela splendens, and Polistes mandarinus (Vespae Nidus)] exhibited the activity. On the other hand, the methanol extracts did not display any fibrinolytic activity. In terms of the coagulation system, an aqueous extract of silkworm Tongchunghacho (Paecilomyces japonica), Oxya japonica japonica and Buthus martensi (Scorpion) increased the clotting time significantly longer (181 times) than the control. These results suggest that crude drugs from insects are useful sources for the development of new drugs for use in treatments involving blood coagulation and fibrinolysis.

전통된장의 제조방법 조사 및 혈액응고 저해활성이 높은 된장의 스크리닝 (Manufacturing Method for Traditional Doenjang and Screening of High Fibrin Clotting Inhibitory Samples)

  • 전통된장 제조방법으로서 원료 혼합비, 메주의 건조시간 및 온도, 메주와 된장의 발효 시간 및 온도, 된장 간장의 분리 여부, 된장 숙성(aging) 시간을 조사하였다. 혈액응고 저해 활성(anticoagulant activity)을 fibrin clotting assay법으로 분석하여 혈액응고 저해활성과 제법간의 상관 관계를 검토하였다. 숙성 기간이 길수록 혈액응고 저해활성이 높게 나타나는 경향을 보였으나 선형적인 상관관계보다는 비선형적으로 양의 상관관계를 갖는 것으로 판단되었다. 이상의 결과를 토대로 혈액응고 저해활성이 높은 2종의 된장시료를 선발하였으며 숙성기간이 180일 이상일 경우 전통된장의 혈액응고 저해활성이 높은 것으로 판단되었다.

오매(Mume Fructus) 추출물의 혈행개선 효과 (The Effects of Mume Fructus Extracts on Blood Flow Improvement)

  • Mume Fructus extract is used in folk medicine in Eastern Asian countries including Korea. However, its therapeutic effect on thrombosis is not known. Mume Fructus methanol extract (ME) dose-dependently inhibited ADP-induced platelet aggregation and also exhibited about 130% fibrinolytic activity compared to the natto. Oral administration of ME to mice significantly extended tail-bleeding time. ME prolonged both aPTT and PT in human citrated plasma also. These results suggest that the methanol extract from Mume Fructus have antithrombosis activity.

Food Functionality of Opuntia ficus-indica var. Cultivated in Jeju Island

  • Opuntia ficus-indica, commonly known as prickly pear cactus, is commercially grown as a food and medicinal plant in Jeju Island, Korea. The crushed pads and fruits of Opuntia ficus-indica were dried in a freeze-dryer and ground into flour to be used for food materials. The major components of proximate compositions were part of a nitrogen free extract. The major minerals were Ca (4391.2-2086.9㎎%), K (1932.1-2608.7㎎%), and Mg (800.6-1984.8㎎%). The major amino acid was glutamic acid, comprising 16.3% of total amino acids in fruit and 25.2% in pad. Dihydroflavonols were identified as (+)-trans-dihydrokaempferol and (+)-trans-dihydroquercetin. Citric acid methyl esters extracted from fruits showed inhibitory activities against monoamine oxidase-B. The presence of trimethyl citrate has been reported in other plants, but 1,3-dimethyl citrate and 1-monomethyl citrate have not been previously reported. The results of pharmacological efficacy tests, including serum biochemical and hematological parameters, autonomic nervous system, anti-inflammatory, analgestic activity, anti-diabetic activity, antithrombotic, anticoagulant, dopamine beta-hydroxylase, monoamine oxidase activity, hyperlipidemia, the respiratory system, antigastic, and anti-ulcerative actions indicate that the fruit and pad of the Opuntia ficus-indica are rich sources of food and medicinal materials.

Isolation and Partial Characterization of a Polysaccharide with Antithrombin Activity against Blood Coagulation in Manda®, a Fermented Natural Food

  • A polysaccharide with antithrombin activity in Manda$^{(R)}& (PAM) was purified via procedures comprising three major steps, i.e. fractional precipitation with ethanol, anion exchange chromatography, and gel permeation chromatography. PAM showed a symmetrical peak on size exclusion HPLC, as assessed by refractive index, and behaved as a single band on cellulose acetate electrophoresis. The average molecular mass was estimated to be 222 kDa by gel filtration. PAM was found to be a sulfated heteropolysaccharide that contains sulfate group (20.5%, w/w) and uronic acid moiety (7.1 %, w/w) in addition to neutral sugar consisting of fucose, xylose, mannose, galactose, and glucose in a molar ratio of 1.00 : 0.35 : 0.28: 0.22 : 0.15. This polysaccharide appeared to inhibit blood coagulation via the intrinsic pathway in a dose-dependent pattern. The clotting of fibrinogen by thrombin was also significantly mitigated by the presence of PAM.
