Development of Cool-touch Functional Dancesport Top for Middle-aged Women (중년 여성을 위한 냉감기능성 댄스스포츠 상의 개발)

  • This study develops a cool-touch functional Dancesport top for middle-aged women by using cool-touch materials in areas where surface temperature becomes high after exercise. The post-exercise surface temperature of the developed research product was compared and analyzed. In addition, subjective evaluation of cool-touch as well as appearance and movement evaluations were performed. The results are as follows. In designing the research product, a detachable neckband was made using highly-preferred hydrated polymer crystals. A material with high moisture absorption speed was also used in F1, S1, under arm and B5, while a material with good thermal conductivity was used in other parts of the bodice. Deodorant tape with antibacterial and deodorant effects was incorporated in the armpit for additional comfort. As for wear evaluation of the research product, significant differences were found in 10 areas using a material with high moisture absorption speed to compare and analyze the post-exercise surface temperatures of the clothing. The temperature difference between the compared top and the research product in the neckband area was 9.16℃, demonstrating clear cool-touch function in using cool-touch material. In the subjective evaluation of cool-touch function, the results showed high scores when asked about the efficiency of the detachable neckband and the ease of movement when wearing the product. In the appearance evaluation, significant differences were found in 11 items, including redundant folds, tightfit, fit, and design line. The overall mean score of the movement evaluation was 4.6, indicating excellent function for movement.

Effects of In Vitro Synergism of Eunkyo-san and Rufloxacin against 9 Strains of Aerobic Gram(+) Bacteria (은교산과 Rufloxacin 병용이 호기성 Gram(+) 세균주에 대한 시험관내 항균력에 미치는 영향)

  • In order to evaluate the in vitro synergic effect of Eunkyo-san, with quinolone antibiotics, rufloxacin (RUFX), the minimal inhibitory concentration (MIC), MIC50 and MIC90 of single use of quinolones and concomitant treatment with Eunkyo-san against 9 strains of aerobic gram positive bacteria. The obtained results were as follows : In the case of aerobic gram positive bacteria, the MIC, MIC50 and MIC90 against Staphylococcus aureus, Staphylococcus aureus smith, Staphylococcus epidermidis, Staphylococcus pyogens, Streptococcus pneumoniae Type Ⅰ, Type Ⅱ and Type Ⅲ was significantly decreased in concomitant treated groups with Eunkyo-san compared to those of single treated groups of RUFX, respectively. However, no significant changes were demonstrated against Bacillus subtilis and Enterococcus faecalis. In conclusion, the in vitro antibacterial activity of RUFX were increased against some strains of aerobic gram positive strains, especially, pneumococcus such as Staphylococcus and Streptococcus by concomitant use of Eunkyo-san.

Antibiotic and Insecticidal Activities of Metabolites Derived From an Entomopathogenic Bacterium, Xenorhabdus nematophila, Against The Bean Bug, Riptortus clavatus (곤충병원세균(Xenorhabdus nematophila) 유래물질의 톱다리개미허리노린재(Riptortus clavatus) 장내세균 증식억제 및 살충효과)

  • A bacterial colony was isolated from the gut of the bean bug, Riptortus clavatus. From morphological and biochemical tests, the bacterial isolate showed the highest similarity to Staphylococcus succinus. DNA sequence of 16S rRNA gene of the bacterium supported the identification. Oral administration of penicillin G to adults of R. clavatus gave a dose-dependent mortality of adults of R. clavatus to adults along with significant decrease of the bacterial population in the gut. Similarly, three metabolites (benzylideneacetone, proline-tyrosine, and acetylated phenylalanine-glycine-valine) derived from an entomopathogenic bacterium, Xenorhabdus nematophila, also inhibited growth of the gut bacterial population and gave significant mortalities to R. clavatus. These results suggest that a gut bacterial population classified as Staphylococcus sp. is required for survival of R. clavatus and that the three bacterial metabolites had toxic effects on the bugs due to their antibacterial properties.

Antimicrobial Effects of Photodynamic Therapy Using Blue Light Emitting Diode with Photofrin and Radachlorine against Propionibacterium acnes

  • Photodynamic therapy (PDT) apply photosensitizers and light. The purpose of this study was to evaluate the in vitro efficacy of PDT using blue LED (light emitting diode) with photofrin and radachlorin for Propionibacterium acnes. The colony forming units method was used to assess the antibacterial activity. Suspension (1 mL) containing P. acnes at $1{\times}10^5CFU/mL$ were prepared and then 2 fold serial diluted to $12.5{\mu}g/mL$ from $50{\mu}g/mL$ concentration of photofrin and radachlorin. After 60 minutes incubation, light was irradiated for 10 to 30 minutes using the following light source of wavelength 460 nm, each energy density 36, 72 and $108J/cm^2$. Bacterial growth was evaluated after 72 hours incubation in a Phenylethanol Blood Agar (PEBA) culture. In addition, flow cytometric analysis were performed to measure the live cell after PDT. Also transmission electron microscopy (TEM) was employed to evaluate the effect of pathogens by PDT. The PDT Group was perfectly killed to all kind of photosensitizers dose of $12.5{\mu}g/mL$ with irradiation of 10 minutes. Also other Groups were killed to all kind of photosensitizers dose of $6.25{\mu}g/mL$ with irradiation time of 20 and 30 minutes. The flow cytometry showed a lower number of viable bacteria in the PDT group compared to the control group. The images of the TEM results were showed in cytoplasmic membrane damage and partially deformed to cell morphologies. These results suggest that radachlorin and photofrin combine blue LED PDT can be effectively treated when was proved treatment for acnes therapy.

Isolation of Lactic Acid Bacteria Showing Antioxidative and Probiotic Activities from Kimchi and Infant Feces

  • The purpose of this study was to investigate lactic acid bacteria with antioxidative and probiotic activities isolated from Korean healthy infant feces and kimchi. Isolates A1, A2, S1, S2, and S3 were assigned to Lactobacillus sp. and isolates A3, A4, E1, E2, E3, and E4 were assigned to Leuconostoc sp. on the basis of their physiological properties and 16S ribosomal DNA sequence analysis. Most strains were confirmed as safe bioresources through nonhemolytic activities and non-production of harmful enzymes such as β-glucosidase, β-glucuronidase and tryptophanase. The 11 isolates showed different resistance to acid and bile acids. In addition, they exhibited antibacterial activity against foodborne bacteria, especially Bacillus cereus, Listeria monocytogenes, and Escherichia coli. Furthermore, all strains showed significantly high levels of hydrophobicity. The antioxidant effects of culture filtrates of the 11 strains included 2,2-diphenyl-1-picryl-hydrazyl (DPPH) radical scavenging capacity, 2.2'-azino-bis (2-ethylbenzthiazoline-6-sulfonic acid) (ABTS) radical cation scavenging activity, and superoxide dismutase activity. The results revealed that most of the culture filtrates have effective scavenging activity for DPPH and ABTS radicals. All strains appeared to have effective superoxide dismutase activity. In conclusion, the isolated strains A1, A3, S1, and S3 have significant probiotic activities applicable to the development of functional foods and health-related products. These strains might also contribute to preventing and controlling several diseases associated with oxidative stress, when used as probiotics.

Functional Red Pigment Production in Solid-state Fermentation of Barley by Monascus sp. EBE1. (보리를 이용한 Monascus sp. EBE1 고상발효에 의한 기능성 적색 색소 생산)

  • The time-dependent changes of red pigments production in solid-state plant scale fermentor using barley cultured with Monascus sp., instead of rice which was traditionally used, were investigated in this study. A steady increase in the yield of red pigments in barley occurred between the 3rd and 6th days. The optimized conditions (inoculation volume = 6∼8%, initial pH = 6, air supply = 0.6∼0.8 m) promoted the production of red pigments. Short-time steaming of barley (< 20 min) decreased fungal growth and pigments production due to the insufficient gelatiniza-tion. The optical density of the red pigments under the optimized conditions was 120 at 500 ]nm per gram of barley. In addition, the metabolites from the fermented barley with Monucus sp. showed antibacterial effects against Escherichia coli and Salmonella typhimurium. Barley was shown to be one of the best grain sources for solid-state fermentation with Monascu sp., fur obtaining natural pigments and also functional food materials.

Synergistic Antimicrobial Effect of Lonicera japonica and Magnolia obovata Extracts and Potential as a Plant-Derived Natural Preservative

  • Most people use cosmetics to protect their skin. Preservatives are often used to prevent their contamination upon use. There has been a great demand for natural preservatives due to recent reports on the side effects of parabens. Therefore, we evaluated the antimicrobial activities of Lonicera japonica and Magnolia obovata extracts and determined their potential as natural preservatives. We found that the 50% ethanol extract from L. japonica had antibacterial activity only against S. aureus and P. aeruginosa, while the ethyl acetate fraction showed antimicrobial activity against all six microbial strains tested. On the other hand, the 70% ethanol extract and the ethyl acetate fraction from M. obovata showed antimicrobial activity against all six strains. A synergistic effect against S. aureus, B. subtilis, and C. albicans was confirmed when two ethyl acetate fractions having antimicrobial activity against all six strains were used in combination. Synergistic activity against B. subtilis was also confirmed through kill-time analysis. High-performance liquid chromatography was performed to identify the components of each extract. Based on the minimum inhibitory concentration and the results of a disc diffusion assay, we confirmed that caffeic acid and luteolin influenced the antimicrobial activity of L. japonica and that the antimicrobial activity of M. obovata was influenced by the interaction of magnolol and honokiol with other components. Therefore, this study suggests that the combination of L. japonica and M. obovata extracts may be used as a plant-derived natural preservative.

Chemical Characteristics and Biological Activities of Herbimycin A and Dihydroherbimycin A Produced by a Soil Isolate Streptomyces sp. AO-0511 (Streptomyces sp. AO-0511이 생산하는 Herbimycin A 및 Dihydroherbimycin A의 이화학적 특성 및 생물 활성)

  • A streptomycete strain was isolated from the soil samples from Korea. The chemotaxonomy and 16S rDNA sequencing confirmed that the strain belonged to the genus Streptomyces and we named it Streptomyces sp. AO-0511. Two antibiotics, herbimycin A and dihydroherbimycin A produced by this strain were tested for their physico-chemical and biological characteristics. Both compounds were stable under acidic pH. Dihydroherbimycin A was more heat-stable and polar compared with herbimycin A. Only weak antibacterial activities were detected against Bacillus subtilus ATCC 6633 and Micrococcus luteus ATCC 9341. However, herbimycin A and dihydroherbimycin A showed strong inhibitory activities on lung cancer cells (A549 cells) and leukemia cells (HL-60). The cytotoxicity was determined using L5178Y and P388 cell lines. The results showed that herbimycin A and dihydroherbimycin A had lower toxic effects on the cells compared with the standard compounds, comptothecin and cyclosporin A. Therefore, both compounds could be good candidates for the development of new anticancer drugs.

Immune Stimulating Efficacy of Soluble β-1,3-glucans (수용성 β-1,3-glucans의 면역 활성 효능에 대한 연구)

  • Background: $\beta$-1,3-glucans are well known to enhance the immune reactions, resulting in antitumor, antibacterial, antiviral, anticoagulatory and wound healing activities. $\beta$-1, 3-glucans have various activities depending on molecular weight, degree of branching, conformation, water-solubility and intermolecular association. However, the $\beta$-1,3-glucan linked backbone structure is essential and $\beta$-D-glucopyranosyl units are required for immuno-potentiating activities. Result: In this study, we tested the immunophamacological activities of soluble $\beta$-1,3-glucans and confirmed the following activities: (1) $IFN-{\gamma}$ production in PBMCs in the presence or the absence of PHA, LPS, or IL-18; (2) induction of various cytokines in the spleen and thymus; (3) adjuvant effect on the antibody production; (4) nitrogen oxide synthesis in macrophages; (5) the cytotoxic and antitumor effects on cell lines and ICR mice. Conclusion: These results strongly suggested that $\beta$-1,3-glucans possessed various immuno-pharmacological activities.

EXPERIMENTAL STUDY ON THE EFFECTS OF VARIOUS DISINFECTANTS TO ORAL MICROFLORA (각종 소독제가 구강균총에 미치는 영향에 관한 실험적 연구)

  • Though oral microgranisms were among the first to be observed by humans, the interest in oral microbiology lagged. When it became apparent that the oral microflora did influence systemic disease of the body, interest was aroused in the nature and kinds of the microgranisms. The risk of infection in dental procedures is due to the abundant flora of the mouth. This hazard can be reduced to some extent by the use of a local disinfectant. The present studies were undertaken to evaluate and compare the various disinfectants which are commonly used in clinics and hospitals. The results were as follows. 1. The bactericidal activity of the disinfectants mainly depends upon the kinds of the agents, not upon the kinds of the microorganisms. 2. In H₂O₂(3%), the bactericidal activity was greatly related to the contact time. So, at least 4 minitues of contract time was required to use it as an oral antiseptic. 3. In ethyl alcohol (70%), Pseudomonas aeruginosa and Streptococcus salivarius surived a little after 15 seconds of contact time, but, no other colony was discovered after more than 15 seconds of contact time in any kins of microorganisms. 4. Merthiolate (0.1%) showed low antibacterial activity, more in Gram-positive organisms and less in Gram-negative organisms. 5. Benzalkonium chloride (0.1%) and povidone-iodine (10%) showed the most excellent results, revealing no surviving organisms only after 14 seconds of contact time.
