Study on the effects of Juguieum and Juguieum-gamibang on the Anti-allergic effect (주귀음(酒歸飮) 및 주귀음가미방(酒歸飮加味方)의 항(抗)알레르기에 관(關)한 실험적(實驗的) 효과(?果))

  • Experimental studies were done to research the clinical effects of Juguieum and Juguieum-gamibang(Juguieum with Lonicerae Flos and Forsythiae Fructus added) on the Antiallergic effect. The results obtained as follows; 1. In the effects of Jugieum and Jugieum-gamibang on vascular permeability responses to intradermal histamine, Juguieum group revealed none significant effect, but Jugieum-gamibang group revealed significant effect. 2. In the effects of Juguieum and Juguieum-gamibang on vascular permeability responses to intradermal serotonin, Juguieum and Juguieum-gamibang groups revealed significant effect. 3. In the 48hrs homologous passive cutaneous amaphylaxis provoked by the IgE-like antibody against egg albumin, Juguieum and Juguieum-gamibang groups revealed significant effect. 4. In the delayed type hypersensitivity responses to Picryl Chloride, Juguieum and Juguieum-gamibang groups revealed significant effect. 5. In the delayed type hypersensitivity responses to SRBC, Juguieum group revealed significant effect, but Jugieum-gamibang group revealed none significant effect. According to above-stated results, Juguieum and Juguieum-gamibang are concluded to be effective as anti-allergic regimen and recommended to be used for the treatment of type I and type IV allergic disease.

  • Allergic inflammation results from stimulation of ${\beta}$-hexosaminidase secretion, increased calcium influx, and activation of MAPK pathways. Some fractions of Chamaecyparis obtusa decreased secretion of ${\beta}$-hexosaminidase, calcium influx, ROS, and phosphorylation of ERK. These results suggest that Chamaecyparis obtusa would be valuable for development of allergy therapeutics.

  • Gamipaidok-san(GPDS) is the Hyungbangpaidok-san prescription fortified with the additional ingredients known to be effective for dermatitis. So it has been used for atopic dermatitis in the clinic work actually. In this study, we investigated the effects of GPDS on in vitro and in vivo anti-allergic effect on RBL-2H3 rat basophilic leukemia Cells and on IgE-induced passive cutaneous anaphylaxis (PCA) in mice. The in vitro anti-inflammatory activity of GPDS in RAW 264.7 cells was investigated. GPDS potently inhibited $\beta$-hexosaminidase release from RBL-2H3 and the IgE-mediated PCA reaction in mice. GPDS inhibited LPS-induced NO and PGE2 production in a dose-dependent manner Furthermore, It also inhibited inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) protein expression in RAW 264.7 cells, and the activation of the transcription factor, NF-kB, in nuclear fraction. The antiallergic action of GPDS may originate from anti-inflammatory activities, and can improve the inflammation caused by allergies.

Anti-allergic Effect sof Bee Venom on IgE-mediated Type I hypersensitivity Response in vivo (봉독이 IgE가 매개하는 제1형 과민반응 동물모델에 미치는 항알레르기 효과)

  • Bee venom (BV), well known as a traditional Oriental medicine, has been widely used in the treatment of some immune-related diseases. However, the anti-allergic effect of BV have not been reported. In this study, we investigated the antiallergic effect of BV on triphasic cutaneous reaction (TpCR) and passive cutaneous anaphylaxis (PCA). Our results indicated that BV suppress ear swelling and vascular permeability on IgE mediated type I hypersensitivity response. Increase in ear thickness was significantly inhibited by BV in this model. BV also blocked the infiltration of immune cells into the ear. Moreover, BV suppressed expression of HDAC3, Tryptase, MCP-1 in ear tissue. These results demonstrated that BV has a suppressive effect on allergic reaction.

Inhibitory Effect of Brassica rapa in Ovalbumin-Stimulated Experimental Asthmic Mice

  • To evaluate antiallergic effect of Brassica rapa Metzg (BR, Family Cruciferae), which is widely used as a food source, its anti-asthmic effect was investigated. BR reduced IgE level in the blood of ovalbumin (OVA)-induced asthmic mice. BR also inhibited IgE and ploinflammatory cytokine IL-6 productions in trachea of OVA-induced mice, but did not inhibit IL-4 production. BR did not inhibit mRNA expression of proinflammatory cytokines IL-6 and IL-4 in IgE-induced RBL-2H3 cells. BR and its isolated components 10-undecenoic acid 2-methoxy methyl ester and galactosyl diglyceride potently inhibited the NO production of RAW264.7cells induced by lipopolysaccharide. These findings suggest that the anti-asthmic effect of BR may be due to the inhibition of the biosynthesis of proinflarnmatory cytokine IL-6 in macrophage and BR can improve IgE-induced allergic disease asthma.

Effects of Senggang Gamcho-tang (SG), ZingberisRhizoma (ZR) and Glycyrrihizae Radix (GR) extracts in Senggang Gamcho-tang (SG) on Activity of Murine Splenocytes (생강감초탕(生薑甘草湯) 및 생강(生薑), 감초(甘草)가 생쥐 비장세포의 활성에 미치는 영향)

  • Objectives : Zingberis Rhizoma (ZR) and Glycyrrihizae Radix (GR) have been widely used to prevent or treat allergic diseases. However, relatively little research has been conducted on the immune response to Senggang Gamcho-tang (SG) in a mixture of ZR and GR. The purpose of this study was to determine antiallergic effects of Senggang Gamcho-tang (SG) extracts ZR and GR on activity of murine splenocytes. Methods : This study was performed to investigate the effect of SG in mice, using in vitro experiments. Cells were treated with SG extract (1 ${\mu}g$/ml, 10 ${\mu}g$/ml and 100 ${\mu}g$/ml) plus Con A (2 ${\mu}g$/ml) and ZR (6.6 ${\mu}g$/ml and 66 ${\mu}g$/ml), GR (3.3 ${\mu}g$/ml and 33 ${\mu}g$/ml) respectively for 48 hrs. The production of IFN-${\gamma}$ and IL-4 were determined by ELISA. Results : No toxicity was found in splenocytes treated with SG extract for 48 hrs at the concentration of 0 ${\mu}g$/ml, 1 ${\mu}g$/ml, 10 ${\mu}g$/ml and 100 ${\mu}g$/ml, respectively. The production of IFN-${\gamma}$ increased to 23,968.7 pg/ml (p<0.001) while that of IL-4 significantly decreased to 14.1 pg/m l(p<0.05) only at the SG concentration of 100 ${\mu}g$/ml. With the GR concentration of 33 ${\mu}g$/ml in SG extract, the production of IFN-${\gamma}$ rose to 32,102.2 pg/ml (p<0.001) while that of IL-4 (p<0.05) declined to 34.2 pg/ml. However, there was no significant observation with regard to the effect of ZR on production of IFN-${\gamma}$ and IL-4. Conclusions : This study suggests an antiallergic effect of Senggang Gamcho-tang, which leads to the implication that SG extract may be used to enhance human immune function.

  • Effects of the aqueous extract of Aquilariae Lignum (Thymelaeaceae) on the allergic reactions were investigated. Oral administration of this extract (50, 250, and 500mg/kg) exhi bited a dose-dependent inhibition on passive cutaneous anaphylactic reactions in rats. Administrations of this extract (500mg/kg, i.p.) at 60 min before and 5, 10 min after the compound 48/80 treatment (8mg/kg, i.p.) decreased the mortality rates to 0, 0, and 14.2%, respectively. The aqueous extract of Aquilariae Lignum (0.05 ~ 1.6mg/ml) showed a dose-related inhibition on histamine release from rat peritoneal mast cells. The morphological examination also clearly showed that the aqueous extract of Aquilariae Lignum prevented the degranulation of mast cells in rats.

  • In the present study, we investigated the effect of the water extract of Perilla frutescens var. acuta (Labiatae; WEPF) on the mast cell-mediated allergic reactions. WEPF was anally administered to mice for high and fast absorption. WEPF inhibited compound 48/80-induced systemic allergic reaction. WEPF attenuated immunoglobulin E (IgE)-mediated local allergic reaction. In addition, WEPF decreased the gene expression of pro-inflammatory cytokines in phorbol 12-myristate 13-acetate plus calcium ionophore A23187 (PMACI)-stimulated HMC-1 cells. These results indicate that WEPF inhibits mast cell-mediated allergic reactions in vivo and in vitro.

  • The antiallergic drug, cetirizine, inhibits the histamine release from a rat basophilic leukemia (RBL-2H3) cell line, which is frequently used as a mast cell model. By investigating inhibitory activities of (+)- and (-)-cetirizine in RBL-2H3 cells on the histamine release, we aimed to evaluate the effect of their structual characteristics on the antihistamine activity. The study on RBL-2H3 cell has clearly demonstrated that the (-)-cetirizine is significantly more potent than the (+)- or the racemic cetirizine, although there was no difference in pharmacokinetics between (+)- and (-)-cetirizine in rats.

  • We studied the effects of sulfasalazine(SSZ) on anaphylaxis. SSZ was found to exhibit a inhibitory activity on the compound 48/80-induced anaphylaxis. SSZ also inhibited local a naphylaxis activated by anti-dinitrophenyl(DNP) IgE. Moreover, SSZ dose-dependently inhibited histamine ralease in rat peritoneal mast cells activated by compound 48/80 or anti DNP IgE. We investigated the effects of SSZ on cAMP of rat peritoneal mast cell. The level of cAMP in rat peritoneal mast cells, when SSZ was added, transiently and significantly increased approximately 16-fold compared with that of basal cells. These results suggest that the antianaphylactic action of SSZ may be associated with an increase in the intracellular cAMP content of the mast cells as the result of an inhibition of the cAMP phosphodiesterase.
