탁리산 용매 분획물의 in vitro 항산화, 주름 억제 및 미백 효과 (In vitro anti-oxidant, anti-wrinkle and whitening effect of solvent fraction from takli-san)

  • 본 연구에서는 탁리산 열수추출물을 용매 분획하여 항산화, 주름억제, 미백효과에 대한 활성을 검증하여 화장품 소재로서 이용가능성을 연구하였다. 용매 분획물로 DPPH radical scavenge, ABTS radical scavenge, elastase, tyrosinase 저해활성을 확인하였고 에틸아세테이트 분획물(TW-EA)이 가장 높은 저해활성을 나타내었다. 세포내에서의 주름억제, 미백효과를 확인하기 위해서 western blot을 실시하였고 비교 검증을 위해 사용한 positive contol보다 TW-EA의 억제효과가 뛰어난 것을 확인하였다. 이러한 결과로 탁리산의 화장품소재로서 적용 가능성을 검증하였다.

Antioxidant, anti-inflammatory, and adaptogenic activity of Asparagus acutifolius extract

  • Although many species of Asparagus have been studied scientifically and shoots are used in the diet of Sardinians, there is very little literature available on the medicinal uses of Asparagus acutifolius Linn. The acetone-ethanol (1:1) extract was screened for antioxidant, anti-inflammatory and adaptogenic activities. The extract showed good anti-oxidant activity in DPPH, hydroxyl radical, and nitric oxide radical assays. The extract also exhibited anti-inflammatory activity in the carrageenan-induced rat paw edema and adaptogenic activity in the milk induced leucocytosis assay in rats. The results of the present study suggest need to investigate other pharmacological activities of Asparagus acutifolius.

Enhanced anti-inflammatory activity of curcumin, a naturally occurring pigment in turmeric via cyclodextrin complexation

  • Curcumin, a dietary pigment responsible for the yellow color of curry, has been used for the treatment of inflammatory diseases and exhibits a variety of pharmacological effects such as anti- inflammatory, anti-tumor, anti-oxidant, and anti-viral activity. In order to examine the potency of the curcumin in inflammation we used carrageenan induced rat hind paw odema model. As curcumin is practically water insoluble, it is hypothesized that pharmacological activity of curcumin could be improved by enhancing its water solubility. Water soluble complexes of curcumin with cyclodextrins were prepared and screened for greater solubility. Pure curcumin 100 mg/kg body weight along with curcumin complexes equivalent to 100 mg/kg body weight of pure curcumin were tested for the anti-inflammatory activity in Wister rats male rats using carrageenan induced hind paw edema model and compared with that of the reference compound diclofenac sodium at a dose level of 10 mg/kg body weight. Results were statistically analyzed using ANOVA. All the treatment groups showed statistically significant anti-inflammatory activity compared with that of vehicle control and positive control.

Anti-Inflammatory Effects of Fermented Products with Avena sativa on RAW264.7 and HT-29 Cells via Inhibition of Inflammatory Mediators

  • This study investigated therapeutic candidates with anti-inflammatory potential among traditional dietary ingredients targeting inflammatory bowel disease (IBD). Both Avena sativa and traditional fermented products, such as Korean soy paste, are popular health foods. We investigated the anti-inflammatory effects of soy paste combined with A. sativa (KDA), compared with soy paste without A. sativa (KD) by evaluating the expression of pro-inflammatory cytokines in lipopolysaccharide-stimulated RAW264.7 mouse macrophages and HT-29 human colon epithelial cells. KDA significantly inhibited the production of nitric oxide (NO) and downregulated the pro-inflammatory cytokines, such as interleukin (IL)-1β, IL-6, and tumor necrosis factor-α in lipopolysaccharide (LPS)-induced RAW264.7 cells. In another in vitro experiment involving LPS-stimulated HT-29 cells, KDA suppressed the levels of IL-8, which is the chemokine elevated in IBD. In addition, KDA exhibited anti-oxidative properties, such as 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) and 2,2'-azino-bis(3-ethylbenz-thiazoline-6-sulfonic acid) (ABTS) radical scavenging activity. Our findings revealed that A. sativa combined with soy paste exhibits a synergistic anti-inflammatory and anti-oxidant effect following fermentation. These results suggest that KDA may be used as a potential anti-inflammatory therapy against IBD.

조간탕가감방(調肝湯加減方)의 항산화활성(抗酸化活性) 및 항염증작용(抗炎症作用)에 대한 실험적(實驗的) 연구(硏究) (The Experimental Study on Anti-oxidant and Anti-inflammatory Effect of Jogantanggagambang(JGTG))

  • Purpose: This study was performed to evaluate anti-inflammatory effects of Jogantanggagambang(JGTG). Methods: In the study of anti-oxidant activities, JGTG was investigated by DPPH radical scavenger activity, superoxide dismutase activity and superoxide anion radical scavenger activity. In the study of anti-inflammatory effects, JGTG was investigated using cultured cells and murine models. As for the parameters of inflammation, levels of several inflammatory cytokines and chemical mediators which are known to be related to inflammation were measured in mouse lung fibroblast cells(mLFCs) and RAW264.7 cells. Results: 1. JGTG showed a safety in cytotoxicity and toxicity of liver. 2. JGTG effected scavenging activity on DPPH free radical, superoxide dismutase and superoxide anion radical. 3. JGTG in RAW 264.7 cell decreased IL-$1{\beta}$ mRNA expression, IL-6 mRNA expression, TNF-${\alpha}$ mRNA expression at 50, $100{\mu}g/m{\ell}$ and also decreased NOS-II mRNA expression at $100{\mu}g/m{\ell}$, and decreased COX-2 mRNA expression at 10, 50, $100{\mu}g/m{\ell}$. 4. JGTG in RAW 264.7 cell decreased significantly IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ at 50, $100{\mu}g/m{\ell}$. 5. JGTG inhibited significantly IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ production in serum of acute inflammation-induced mice. 6. JGTG decreased significantly IL-$1{\beta}$ mRNA production in spleen tissue. Conclusion: These results suggest that JGTG can be used for treating diverse female diseases caused by inflammation

판람근(Isatis tinctoria L., 板藍根)추출물의 항산화활성 및 화장품약리활성에 관한 연구 (Study on anti-oxidant and cosmeceutical activities of Isatis tinctoria L.)

  • Objective : In this study, anti-oxidant and cosmeceutical activities of Isatis tinctoria L. extracted from water, ethanol and supercritical fluid condition were confirmed to investigate cosmeceutical activities for utilization as cosmetic ingredient. Methods: Anti-oxidant and cosmeceutical activities were investigated by using electron donating ability, xanthine oxidase, tyrosinase, astringent effect. Result : Isatis tinctoria L. extracts by supercritical fluid, water and ethanol showed good electron donating ability which were 82.7%, 62.6% and 44.8% at the concentration at 1,000ppm, respectively. Xanthine oxidase activity related with purine metabolism was inhibited by ethanol extract about 52.3% at the concentration at 1,000ppm. Tyrosinase inhibition effects, by supercritical fluid extract, ethanol extract and water extract, were 83.3%, 52.9% and 41.2% respectively at 1,000ppm. In the measurement of astringent effect, supercritical fluid extract at the concentration to 5,000ppm showed 85.7% in related activity. The water extract showed 95.9% nitrite scavenging activity at 5,000ppm. Conclusion : According to these results, it is possible that the extract of Isatis tinctoria L. can be used as a new natural material of cosmetic industry.

황금(黃芩)의 에탄올추출물에 의한 화장품 방부효과 (Preservation of Cosmetics by Ethanol Extract of Scutellaria baicalensis $G_{EORGE}$)

  • Skin toner와 skin lotion의 두 가지 화장품 formulation에 기존의 파라벤류 방부제 대신 황금 (黃芩) 추출물 (1.0 wt%)을 첨가하고 Staphylococcus aureus (ATCC 25923), Pseudomonas aeruginosa (ATCC 9027) 및 Candida albicans (ATCC 10231) 균주를 접종하였을 때 이 균주들은 7일 이내에 99.9% 이상의 사멸율을 보여 황금추출물이 CTFA기준을 만족하는 항균력이 있음을 확인하였다. 다만 skin lotion에 접종된 Candida albicans의 경우 사멸율은 CTFA기준에 약간 미달하는 99.8%이었다. 또한 황금추출물의 농도가 1.0 wt% 이하일 때 균주의 생존율은 그람 양성균인 P. aeruginosa가 가장 높고 그람 음성균인 P. aeruginosa가 가장 낮았다. 전계방출형 주사전자현미경(FE-SEM)을 이용한 세포형태변화 관찰 결과 황금추출물이 P. aeruginosa에 대하여 더 큰 방부력을 갖는 것은 이 균주의 세포막이 황금추출물에 의해 더 심하게 손상되기 때문으로 사료된다. 고농도의 황금추출물이 더 높은 항산화 효과를 나타냈으며 1,000 ppm에서 80%정도의 SOD-유사활성을 나타내었다.

Effect of Maillard Reaction Products on Inhibition of Burdock Polyphenol Oxidase and Their Antioxidant Activities

  • This study was conducted in an effort to investigate the effect of Maillard reaction products (MRPs) on enzymatic browning of burdock and their anti-oxidant activity. The MRPs were prepared by heating glucose and amino acids at $90^{\circ}C$, which served to produce a strong inhibitory effect on burdock polyphenol oxidase. As the reaction time of the solution containing glucose and amino acid increased at $90^{\circ}C$, the production of MRPs increased and intensity of the brown color deepened. When MRPs were prepared by heating at $90^{\circ}C$ for five hours, the absorbance of MRPs from glucose and lysine was 6.44, while those of glucose and glycine was 1.95. The MRPs synthesized from the glucose and lysine also reduced the pH of MRPs from 5.60 to 4.51, but those from glucose and glycine decreased slightly from 5.57 to 5.33. The Michealis-Menten constant value ($K_m$) of burdock PPO with pyrocatechol as a substrate was 16.0 mM, and MRPs were a non-competitive inhibitor against burdock PPO. The anti-oxidant activity of MRPs was measured by evaluating its radical scavenging activities of DPPH radicals, ABTS radicals and reducing power. The color intensity of MRPs produced by lysine and glucose were deeper than that produced by glucose and glycine. It was also found that MRPs produced from glucose and lysine exhibited stronger anti-oxidant properties than those produced by glucose and glycine.

항산화물질 N-acetylcysteine (NAC)이 메기에서 비특이적 면역지표인 화학발광 반응에 미치는 영향 (Effects of N-acetylcysteine (NAC) on chemiluminescence response as a non-specific immune parameter in Far Eastern catfish Silurus asotus)

  • It has been reported that various anti-oxidant substances stimulate non-specific immune responses in fishes. In this study it was examined whether N-acetylcysteine (NAC), a precusor for anti-oxidant glutathione (GSH) synthesis, can modulate non-specific immune function in Far Eastern catfish Silurus asotus. Immune functions were assessed using the respiratory burst activity monitored by chemiluminescence (CL) responses in isolated leucocyte. NAC stimulated CL responses with doses of 10 or 100 mg/kg, but not with 1 mg/kg after 48 hr injection. It was observed with 10 mg/kg NAC that CL activity continued to elevate from 24 hr through 96 hr post-dosing, and returned to the near preinjection level by 10 days. To understand whether NAC can also activate CL activity in vitro, NAC was directly added to isolated catfish leucocytes. It was observed, however, that NAC can not stimulate CL at reasonable concentration ranges in vitro. As NAC is a precursor for the strong anti-oxidant glutathione (GSH), a putative immune stimulator, it was assessed whether GSH can also stimulate CL responses. Observed results show that GSH activated CL both in vivo and in vitro. The data obtained collectively support the proposition that NAC indirectly stimulates non-specific immune functions in catfish by enhancing GSH biosynthesis, but not by direct action of NAC. Such effects may have beneficial significance in aquaculture for practical utilization.

전복내장추출물의 항산화 및 human dermal fibroblasts에 대한 항피부노화 효과 (Anti-oxidant and Anti-skin-aging Effects of Abalone Viscera Extracts in Human Dermal Fibroblasts)

  • 전복가공부산물인 내장의 이용가치를 향상시키고자 열수추출, 에탄올추출 등의 방법으로 유효성분을 추출하고 항산화 및 항피부노화 활성을 측정하였다. DPPH 라디칼소거능 분석결과 Tris-HCl 추출물은 $58.60{\pm}0.88%$으로 높은 라디칼소거능을 보였으며, 다음으로 에탄올추출물은 $55.40{\pm}0.62%$의 라디칼소거능을 보였다. Anti-elastase활성을 분석한 결과 에탄올추출물이 다른 시험구에 비하여 현저하게 높은 활성을 보였다. 전복내장추출물의 세포독성을 확인하기 위하여 human dermal fibroblast 세포에 대하여 MTT 시험결과 세포독성은 전혀 나타나지 않았다. 에탄올 추출물의 농도를 달리하여 human dermal fibroblast 세포에 대해pro-collagen type I 생합성능시험결과 10.0 ${\mu}g/mL$에서 $705.30{\pm}3.06$ ng/mL로 retinoic acid $453.60{\pm}2.82$ ng/mL보다 우수한 결과를 보였다. MMP-1 활성은 10 ${\mu}g/mL$에서 $45.30{\pm}0.80$ ng/mL를 보여 control의 $62.37{\pm}0.56$ ng/mL 보다 감소정도가 낮았다. 본 연구는 전복내장추출물의 항산화와 항노화활성에 관하여 최초로 시도되었으며 향후 전복내장을 이용한 다양한 연구의 기초자료로서 중요한 의미가 있다.
