The Experimental Study on Anti-oxidant and Anti-inflammatory Effect of Jogantanggagambang(JGTG) (조간탕가감방(調肝湯加減方)의 항산화활성(抗酸化活性) 및 항염증작용(抗炎症作用)에 대한 실험적(實驗的) 연구(硏究))

  • Purpose: This study was performed to evaluate anti-inflammatory effects of Jogantanggagambang(JGTG). Methods: In the study of anti-oxidant activities, JGTG was investigated by DPPH radical scavenger activity, superoxide dismutase activity and superoxide anion radical scavenger activity. In the study of anti-inflammatory effects, JGTG was investigated using cultured cells and murine models. As for the parameters of inflammation, levels of several inflammatory cytokines and chemical mediators which are known to be related to inflammation were measured in mouse lung fibroblast cells(mLFCs) and RAW264.7 cells. Results: 1. JGTG showed a safety in cytotoxicity and toxicity of liver. 2. JGTG effected scavenging activity on DPPH free radical, superoxide dismutase and superoxide anion radical. 3. JGTG in RAW 264.7 cell decreased IL-$1{\beta}$ mRNA expression, IL-6 mRNA expression, TNF-${\alpha}$ mRNA expression at 50, $100{\mu}g/m{\ell}$ and also decreased NOS-II mRNA expression at $100{\mu}g/m{\ell}$, and decreased COX-2 mRNA expression at 10, 50, $100{\mu}g/m{\ell}$. 4. JGTG in RAW 264.7 cell decreased significantly IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ at 50, $100{\mu}g/m{\ell}$. 5. JGTG inhibited significantly IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ production in serum of acute inflammation-induced mice. 6. JGTG decreased significantly IL-$1{\beta}$ mRNA production in spleen tissue. Conclusion: These results suggest that JGTG can be used for treating diverse female diseases caused by inflammation

Protective effect of Samultang and its four herbal plants against reactive oxygen species in vitro and cellular system

  • Oxidative stress and overproduction of free radicals have been reported to be a major pathological hallmark of neurodegenerative diseases. Samultang has been known as a beneficial agent to treat liver disease and cardiovascular diseases. However, the anti-oxidant activities and neuro-protective effects of Samultang against oxidative stress still have not been evaluated yet. The aim of the present study was to investigate the anti-oxidant and protective effects of Samultang and its four herbal plants, Paeonia lactiflora (PL), Ligusticum striatum (LS), Rehmannia glutinosa (RG), and Angelica gigas (AG), in vitro system and in SH-SY5Y neuronal cells. The extracts of Samultang strongly increased the radical scavenging activities of 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical (${\cdot}OH$), and nitric oxide (NO) in a concentration-dependent manner. Furthermore, we investigated the protective effects of Samultang on cellular damage against oxidative stress induced by hydrogen peroxide ($H_2O_2$) in SH-SY5Y cells. Treatment with Samultang alleviated the oxidative stress from $H_2O_2$ by increasing the cell viability and decreasing the intracellular reactive oxygen species levels. Based on these results, we further investigated the radical scavenging effects of PL, LS, RG, and AG. In our results, PL had the highest DPPH, ${\cdot}OH$, and NO radical scavenging activities. Thus, PL has a crucial role in Samultang, which has anti-oxidative and neuro-protective effects. The present research suggests that Samultang and PL have protective roles against oxidative stress from $H_2O_2$-induced neuronal cell death.

  • Objective : The present study designed this study to investigate anti-oxidative effects of EF on HaCaT keratinocyte. Method : The present study measured the amount of polyphenoics and flavonoids, and also measured the levels of catalase, Ascorbate peroxidase (APX), SOD like activities and DPPH free radical scavenging activity. Then the effects of SB on viability and prolferation rates, and protective effects against oxidative stress induced by chemicals such as hydrogen peroxide and rotenone were also investigated. Results and conclusion : EF showed protective effect against cell death of HaCaT keratinocyte induced by rotenone and SNP significantly. In conclusion, these results suggest that EF may have anti-oxidantic action in human skin and also suggest that EF can be used as anti-aging agent.

The Cytotoxic Effect of Chaga Mushroom (Inonotus Obliquus) Water Extract on HepG2 Hepatoma Cells

  • Chaga mushroom (Inonotus obliquus) extract has been known to have therapeutic effects, such as anti-inflammatory, hepato-protective, anti-oxidant and anti-tumor effect. In this study, we evaluated the effects of Chaga extract on the cytotoxic actions of cisplatin in HepG2 hepatoma cells. The viability of the HepG2 cells was decreased to 10% at 3 ${\mu}M$ cisplatin and to 20% at 500 ${\mu}g$/ml Chaga extract as measured by the MTT assay. The viability of HepG2 cells co-treated with cisplatin (3 ${\mu}M$) and Chaga extract (500 ${\mu}g$/ml) was decreased to 50% in compared with the control cells. The cytotoxicity of two drugs was revealed as apoptosis characterized by the chromatic condensation, nuclear fragmentation and the cleavage of pro caspase-3 in HepG2 cells. Also, the cells treated with combination of two drugs showed synergistically the loss of mitochondrial membrane potential and increase of intracellular ROS levels. Therefore, these results suggest that the combination treatment of cisplatin and Chaga extract induces apoptotic cell death in HepG2 cells and has more potential anti-tumor effect than cisplatin alone.

Effect of Immature Citrus sunki Peel Extract on Neuronal Cell Death (미성숙 진귤 과피 추출물이 신경세포 사멸에 미치는 영향)

  • The peel of Citrus sunki exhibits multiple biological activities such as anti-oxidant, anti-inflammation and anti-obesity, but little is known about neurodegeneration-related activities. In this study, we investigated the protective effect of ethanolic extract from both immature and mature Citrus sunki peel on neuronal cell death. Treatment of the neuroblastoma cell line SH-SY5Y with $MPP^+$, an inducer of Parkinson disease model, increased cell death in a dose dependent manner. Increased levels of active caspase-3 and cleaved PARP were detected. Treatment with immature Citrus sunki peel extract significantly reduced $MPP^+$-induced neurotoxicity. Cytoprotection with immature Citrus sunki peel extract was associated with a decrease in caspase-3 activation and PARP cleavage. In contrast, mature Citrus sunki peel extract had no significant effects. These data suggest that immature Citrus sunki peel extract may exert anti-apoptotic effect through the inhibition of caspase-3 signaling pathway on $MPP^+$-induced neuronal cell death.

Effect of Moutan Cortex Radicis on Gene Expression Profile of Differentiated PC12 Rat Cells Oxidative-stressed with Hydrogen Peroxide (모단피의 PC12 cell 항산화 효과와 관련 HO, MIF, COMT 유전자 발현에 미치는 영향)

  • Oriental medicine explains aging as the weakening of Kidney-ai, and Kidney-strengthening herbal medicines such as Yukmijihwang-tang have been studied for anti-aging effects. In Western Medicine, the hypothesis that reactive oxidant species(ROS) contribute to the aging process is generally accepted. It has been reported that Moutan Cortex Radicis extract (MCR) was the most effective constituent of Yukmijihwang-tang in decreasing ROS production in oxidative-stressed cells. The purpose of this study is to confirm the anti-oxidant effect of MCR on PC12 cells, the expression of Heme oxygenase (HO), Macrophage migradon inhibitory factor (MIF), Catechol-O-methyltransferase (COMT) using real time RT PCR. PC12 cells were treated without or with hydrogen peroxide in the presence or absence of MCR using MTS assay. Hydrogen peroxide decreased the viability of PC12 cells by 53% and MCR did not influence that of stressed PC 12 cells irrespective of dose or incubation period. However, MCR showed an inhibitory effect on production of ROS in stressed cells, both dose and incubation time dependently. In particular, 1 ㎎/㎖ of MCR for 24 h culture almost returned to normal level. In the quantiation of anti-aging related gene expression, MCR at 1 ㎎/㎖ increased the expression of HO by 370%, MIF by 180% and COMT by 280% through real time RT PCR. In conclusion, MCR treatment protected PC12 cells from hydrogen peroxide and decreased ROS production and enhanced anti-oxidative gene expression such as HO, COMT and MIF, which suggests that MCR is involved in controlling anti-aging of nerve cells through elimination of cytotoxic stimuli.

The Anti-aging effects of Korean Ginseng Berry in the Skin (인삼열매의 피부노화 억제 효과)

  • The root of Korean ginseng (Panax ginseng C.A. Meyer) is a commonly used herbal medicine in China, Korea, Japan. However, the compositions and effects of Korean ginseng berry are not clear to date. In order to investigate the anti-aging effects in the skin, Korean ginseng berry was extracted with 70% ethanol and tested the biological effects. In the results, Korean ginseng berry extract showed an excellent anti-oxidant effect against oxidative stress and decreased MMP-1 over-expression induced by UV irradiation. Especially the main component of Korean ginseng berry extract, ginsenoside Re, increased hyaluronic acid in HaCaT keratinocytes. We improved Korean ginseng berry could be a good material for the anti-aging effect of skin.

Analysis on Research Trend of Studies Related with Scutellariae Radix in Korea (황금(黃芩)에 관한 국내 연구 동향에 관한 소고)

  • Scutellariae Radix has been used as a traditional medicine for anti-oxidant, anti-inflammatory, anti-allergic function. But most study methods were restricted to in vitro and in vivo. Therefore to perform for clinical trials further for a new natural drug development is necessary and this study will be used as a basis for it. The studies selected from domestic academic database included the following key words; '황금', '黃芩', 'skullcap', 'Scutellariae Radix', 'scutellaria baicalensis' and considered were those published from 1990 to July, 2011. All 1080 studies were found to include the keywords related to the study subjects either in their title of contents or abstracts. and 298 studies were finally selected as subjects for this study. 243 studies among 293 studies were published between 2000 to 2011. Classification was proceeded according to study subjects as followed; anti-Inflammatory effect and antiallergic and antihistamin effect(66), antibacterial and antivirus effect(61), antioxidant effect(51), neuronal cell apoptosis and neuronal cell protective effect(22), liver cell protective effect(20). According to method type of study, 194 studies practicing in vitro, 60 studies practicing in vivo, 37 studies practicing in both. and 5 studies on documentary records. Most study methods were restricted in vitro and in vivo. For developmenting of function of anti-inflammatory effect and antiallergic, antihistamin effect & atopic dermatitis effect, antibacterial and antivirus effect, antioxidant effect, case report on various fields and multicenter clinical trials is necessary.

Effect of Cheongungdajosan-gamibang on oxidative damage and inflammation (천궁다조산가미방(川芎茶調散加味方) 주정추출물이 산화적 손상 및 염증에 미치는 영향)

  • Objectives : The purpose of this study was verification of the anti-inflammation and anti-oxidant effect of Cheongungdajosan-gamibang extract (CG) in mouse macrophage, RAW 264.7 cells. Methods : We have basically using LPS-stimulated RAW 264.7 cells. The cell toxicity was determined by MTT assay. To evaluate the anti-inflammatory effect of Cheongungdajosan-gamibang, amount of nitric oxide(NO) was measured using the NO detection kit and the IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ expression was measured by reverse transcriptase polymerase chain reaction (RT-PCR). Also, free radical scavenging assay has tested for DPPH and ABTS radical activity as well as the contents of total polyphenol. Results : In this study, 96.6% or higher cell viability was observed in all tested groups from 1, 10, $100{\mu}/m{\ell}$ in RAW 264.7 cells. The RAW 264.7 cells were induced by lipopolysaccharide (LPS) and CG 1, 10, $100{\mu}/m{\ell}$. The CG decreased nitric oxide (NO) production activity dose dependently, especially at $100{\mu}/m{\ell}$ of 55%. The production of IL-$1{\beta}$, IL-6 and TNF-${\alpha}$ were decreased by 51%, 78% and 35% in CG treated $100{\mu}g/m{\ell}$. CG showed dose-dependent suppression activity of reactive oxygen species (ROS) production, especially at $100{\mu}g/m{\ell}$ of 37%. DPPH radical scavenging activity and ABTS cation decolorization were activated over 86% and 88% in CG at $1,000{\mu}g/m{\ell}$ concentration. Conclusions : According to the results, we thought that CG showed anti-inflammatory and antioxidant activities on the RAW 264.7 cells in mouse macrophage. Therefore, this research is expected to provide the fundamental data about the natural material analysis of relating to the anti-inflammation and antioxidant.

Antioxidant Activity of N-hydroxyethyl Adenosine from Isaria sinclairii

  • The antioxidant activity of Isaria (Paecilomyces) sinclairii was determined by measuring its radical scavenging effect on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radicals. The n- BuOH extract of P. sinclairii showed strong scavenging activity to DPPH. The anti-oxidant potential of the individual fraction was in the order of ethylacetate> n- BuOH>chloroform>n-hexane. The n-BuOH soluble fraction exhibiting strong anti-oxidant activity was further purified by repeated silica gel column chromatography. N-(2-Hydroxyethyl)adenosine (HEA) was isolated as one of the active principles from the n-BuOH layer. The n-BuOH layer, particularly HEA, did not increase the level of nitric oxide (NO) production in vascular endothelial cells that might be related to vasorelaxation such as the action of viagra. In addition, the vascular endothelial growth factor (VEGF) levels showed little or no increase compared with control group with the treatment of I. sinclairii.
