Anti-aging and Anti-oxidative Effect of Naesohwangryun-tang(NSHRT) (내소황련탕(內疎黃連湯)의 항노화 및 항산화 효능 연구)

  • Objectives : This study is to investigate the anti-aging and antioxidant effects of Naesohwangryeon-tang(NSHRT) extract in Hs68 fibroblasts. Methods : The fibroblast cell line Hs68 cells were used and MTT analysis was performed to measure the cell viability of Naesohwangryeon-tang, and then MMP-1 was induced with the cytokine TNF-𝛼 to confirm the anti-aging effect through real-time PCR and ELISA. In addition, ROS was induced with the TNF-𝛼 to confirm the anti-oxidant effect through DCF-DA. The signaling pathway according to the antioxidant effect was performed by western blot. Results : Naesohwangryeon-tang did not show cytotoxicity at all concentrations(100-800㎍/㎖) treated on Hs68 cells, and the inhibitory effect of MMP-1 was also confirmed. In addition, it was confirmed that the intracellular ROS increased by TNF-α was decreased by Naesohwangryeon-tang, and it was confirmed that the expression of NQO1 was increased in a concentration-dependent manner of Naesohwangryeon-tang. Conclusions : These results suggest that Naesohwangryeon-tang can be used as an anti-aging material by demonstrating the anti-aging and antioxidant effects of Naesohwangryeon-tang.

The Anti-Oxidant Effect of Extracts from the Vaccinium oldhami (정금나무(Vaccinium oldhami) 열매의 항산화 효과)

  • Natural compounds have been studied to substitute synthetic antioxidants. In this study, the anti-oxidant activity of 70% acetone extracts from the Vaccinium oldhami fruit was investigated for utilization as ingredients for the cosmetic and bio-industries. Anti-oxidant activity was determined by determining total polyphenolic content, electron donating ability, nitric oxide (NO) radical scavenging activity, $ABTS{\cdot}^+$ cation radical scavenging activity and hydrogen peroxide scavenging activity. The polyphenolic content of 70% acetone extracts of the Vaccinium oldhami fruit was 55.972 mg TAE/g. In electron donating activity, 70% acetone extracts of the Vaccinium oldhami fruit showed an effect of 93.9%, which was similar to BHA effect at a concentration of 500 ${\mu}g/ml$. In the NO radical scavenging ability, 70% acetone extracts of the Vaccinium oldhami fruit showed 60% at 500 ${\mu}g/ml$. $ABTS{\cdot}^+$ cation radical scavenging activity of the Vaccinium oldhami fruit at a concentration of 1000 ${\mu}g/ml$ was 75.7%. Also, hydrogen peroxide scavenging activity of 70% acetone extracts showed 80.8% at 100 ${\mu}g/ml$, whichwas higher than BHA. In the natural compound market, the most important factors are the ability to obtain high effects of a material in low concentrations and a long-lasting supply. The Vaccinium oldhami fruit can be harvested every year - this fulfills one of the requirements. From these results, we can confirm that the Vaccinium oldhami fruit has anti-oxidant abilities and has potential as a natural anti-oxidant agent to be utilized in the cosmeceutical and bio-industries.

The Anti-inflammatory Mechanism of the Peel of Zanthoxylum piperitum D.C. is by Suppressing NF-κB/Caspase-1 Activation in LPS-Induced RAW264.7 Cells

  • Zanthoxylum piperitum D.C. (ZP) peels has been used as a natural spice and herb medicine for hypertension reduction, for strokes, and for its anti-bacterial and anti-oxidant activity. However, the anti-inflammatory mechanisms employed by ZP have yet to be completely understood. In this study, we elucidate the anti-inflammatory mechanism of ZP in lipopolysaccharide (LPS)-induced RAW264.7 cells. We evaluated the effects of ZP in LPS-induced levels of inflammatory cytokines, prostaglandin E2 (PGE2), and caspase-1 using ELISA. The expression levels of inflammatory-related genes, including cyclooxygenase (COX)-2 and inducible nitric oxide synthase (iNOS), were assayed by Western blot analysis. We elucidated the effect of ZP on nuclear factor (NF)-κB activation by means of a luciferase activity assay. The findings of this study demonstrated that ZP inhibited the production of inflammatory cytokine and PGE2 and inhibited the increased levels of COX-2 and iNOS caused by LPS. Additionally, we showed that the anti-inflammatory effect of ZP arises by suppressing the activation of NF-κB and caspase-1 in LPS- induced RAW264.7 cells. These results provide novel insights into the pharmacological actions of ZP as a potential candidate for development of new drugs to treat inflammatory diseases.

Santalum album Linn wood and its oil: An aromatic Unani traditional medicine with versatile pharmacological activities

  • Santalum album Linn. [Family: Santalaceae] is commonly known as white sandalwood, sandal safaid and safed chandan. It is one of the most valuable trees and second costliest wood in the world. Sandalwood and its oil is extensively used in the Unani and other traditional systems of medicine as it has blood purifier, anti-inflammatory, analgesic, exhilarant, cardiotonic, antiseptic, nervine tonic and expectorant properties. It is used in skin, cardiac, liver, gastrointestinal, respiratory, integument and urogenital disorders. These uses are supported and proven by many in vitro or in vivo studies. The proven pharmacological activities of S. album are antimicrobial, anti-oxidant, anti-inflammatory, antimutagenic and anti-fatigue. The research has proven that sandal oil or its constituents have anti-microbial activity. Sandalwood oil showed skin cancer preventive effect in mice and its constituent alpha santalol showed the anticancer property. The methanolic extract of wood was confirmed for antioxidant, free radical scavenging, analgesic and anti-inflammatory activities. ${\alpha}$ and ${\beta}$ santalols present in sandal oil showed sedative effects. Sandalwood tea had a significant effect on heart muscles of frog and showed increased myocardial contractility. Its oil showed significant changes in hepatic xenobiotic metabolizing enzymes. Sandalwood oil and its major constituents showed less acute oral and dermal toxicity in laboratory animals. Hence, the aforementioned studies justify the uses of sandalwood and its oil mentioned in the classical Unani literature. However, further clinical trials are suggested to confirm its efficacy and safety in humans.

Evaluation of Antioxidant, Anti-cholinesterase, and Anti-inflammatory Effects of Culinary Mushroom Pleurotus pulmonarius

  • Culinary mushroom Pleurotus pulmonarius has been popular in Asian countries. In this study, the anti-oxidant, cholinesterase, and inflammation inhibitory activities of methanol extract (ME) of fruiting bodies of P. pulmonarius were evaluted. The 1,1-diphenyl-2-picryl-hydrazy free radical scavenging activity of ME at 2.0 mg/mL was comparable to that of butylated hydroxytoluene, the standard reference. The ME exhibited significantly higher hydroxyl radical scavenging activity than butylated hydroxytoluene. ME showed slightly lower but moderate inhibitory activity against acetylcholinesterase (AChE) and butyrylcholinesterase than galantamine, a standard AChE inhibitor. It also exhibited protective effect against cytotoxicity to PC-12 cells induced by glutamate ($10{\sim}100{\mu}g/mL$), inhibitory effect on nitric oxide (NO) production and inducible nitric oxide synthase protein expression in lipopolysaccharide-stimulated RAW 264.7 macrophages, and carrageenan-induced paw edema in a rat model. High-performance liquid chromatography analysis revealed the ME of P. pulmonarius contained at least 10 phenolic compounds and some of them were identified by the comparison with known standard phenolics. Taken together, our results demonstrate that fruiting bodies of P. pulmonarius possess antioxidant, anti-cholinesterase, and inflammation inhibitory activities.

The antioxidative and cytoprotective effect of Lonicerae japonicae Flos water extracts on the ultraviolet(UV)B-induced human HaCaT keratinocytes (금은화 물추출물의 항산화 효과와 Ultraviolet(UV)B로 유도된 사람 각질형성세포 손상에 대한 보호효과)

  • Objectives : Lonicerae japonicae Flos(LJF) has been reported to exhibit anti-oxidant, anti-inflammatory, anti-viral, anti-rheumatoid properties. However, it is still largely unknown whether LJF inhibits the ultraviolet(UV)B-induced oxidative damage in human HaCaT keratinocytes. Therefore in this paper, we investigated the anti-oxidative capacity and protective effect of LJF against UVB-induced oxidative demage in human HaCaT keratinocytes. Methods : To evaluate the anti-oxidative activity of LJF extracts, we measured total phenolic contents, total flavonoid contents, antioxidant capacity, and superoxide scavenging activity. To give an oxidative stress to HaCaT cells, UVB was irradiated with $200mJ/cm^2$ to HaCaT cells. To detect the protective effect of LJF against UVB, we measured cell viability, DNA fragmentation and reactive oxygen species (ROS) production. In addition, we performed high-performance liquid chromatography (HPLC) analysis to find a major component of LJF. Results : LJF contained phenolic and flavonoid contents, and showed the anti-oxidant and superoxide scavenging activity. The UVB-induced oxidative conditions led to the cell death, DNA fragmentation and reactive oxygen species (ROS) production. However, pretreatment with LJF reduced oxidative conditions, including inhibition of cell death, DNA fragmentation and ROS production. In addition, we found out chlorogenic acid as major component of LJF. Conclusions : These results could suggest that LJF contained anti-oxidative contents and exhibited protective effects against UVB on human HaCaT keratinocytes. And the effective compound of LJF which could show protective activities against UVB is chlorogenic acid. Thus, LJF and chlorogenic acid would be useful for the development of drug or cosmetics treating skin troubles.

Anti-oxidant, Anti-inflammation and Anti-microbial Effects of Hoangtonogak Plus Extracts (황토노각플러스 추출물의 항산화, 항염 및 항미생물 효능)

  • This study evaluated the possibility of Hoangtonogak Plus extracts as a bioactive ingredients for cosmetic products. Methanol(MN) and hot-water(WN) extracts were analysed by DPPH/ABTS radical scavenging activity, FRAP value for anti-oxidant activity, MTT assay for cell viability, inhibition of NO production and iNOS protein expression for anti-inflammatory effect, paper disc diffusion method for anti-microbial activity against Staphylococcus aureus, Staphylococcus epidermidis and Escherichia coli.. The contents of total polyphenol of MN and WN extracts were 2.92±0.01 mgGAE/g and 1.67±0.02 mgGAE/g, respectively. DPPH, ABTS and FRAP values of MN extracts were higher than WN at each concentration. No significant cytotoxicity was observed in RAW264.7 cells. Furthermore, NO production of MN and WN at 1 mg/mL concentration was measured as 11.69 μM, 20.4 μM, respectively. In addition, MN extracts showed anti-microbial effect only on S. epidermidis. Also MN extracts suppressed iNOS protein level in a concentration-dependent manner. According to our results, the MN extracts demonstrated its potential as a natural source of antioxidant with anti-microbial and anti-inflammatory properties.

Anti-oxidant and anti-adipocyte differentiation of Aster glehni and Aster yomena (섬쑥부쟁이와 쑥부쟁이의 항산화 및 지방세포 분화 억제 효과)

  • Purpose: Aster glehnii (AG) and Aster yomena (AY) are medicinal plants that belong to the family Compositea and grow widely in Korea. Plants in the genus Aster have been used to treat snakebite wounds or bruises in oriental medicine. This study compared the effects of anti-oxidants and anti-adipocyte differentiation according to the species (the aerial parts of AG and AY). Methods: AG and AY were extracted using 70% ethanol (-E) and water (-W) at room temperature. The anti-oxidant activities were measured by total phenol contents (TPC), total flavonoid contents (TFC), DPPH and $ABTS^+$ assay. In addition, correlation analysis was performed for the anti-oxidant compounds and effect. The level of anti-adipocyte differentiation was assessed using an oil red O assay on pre-adipocytes. Results: AG-W showed higher TPC ($6.92{\mu}g/mL$) and AG-E presented higher TFC ($8.22{\mu}g/mL$) than the other extracts. Furthermore, AG-E exhibited higher radical scavenging activity in the DPPH and $ABTS^+$ assay ($IC_{50}$: 104.88 and $30.06{\mu}g/mL$). In the cytotoxicity assay, AG and AY extracts at concentrations less than $100{\mu}g/mL$ were non toxic. AG-W reduced the lipid accumulation of 3T3-L1 cells significantly after differentiation (70.49%) compared to the other extracts. Conclusion: These results show that the water extract of AG has anti-oxidant effects and reduces the differentiation of 3T3-L1 cells. Therefore, AG has utility as a functional food material for its anti-oxidant activities and ability to prevent lipid accumulation.

Anti-Oxidative and Anti-Inflammatory Effects of Cheongajihwang-Tang Extract on RAW264.7 Cells (청아지황탕(靑蛾地黃湯)의 RAW264.7 Cell에서 항산화 및 항염증 효능 연구)

  • Objectives This study was designed to investigate whether the Cheongajihwang-Tang (CT) has an inhibitory effect association with oxidation or inflammation in RAW264.7 cells. Methods Cytotoxic activity of CT extract on RAW264.7 cells was evaluated by using 3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide (MTT) solution. Nitric oxide production was measured using Griess reagent system. The total phenolic contents and 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was measured to evaluate the anti-oxidative effects of CT. Dichlorofluorescin diacetate (DCFH-DA) has been used as a substrate for measuring intracellular oxidant production. Results Cheongajihwang-Tang does not impair the cell viability in tested concentration. CT showed anti-oxidative effects in vitro by decreasing electron donating ability, and also showed anti-inflammatory effects suppressing NO and ROS expression in LPS induced RAW264.7 activation. CT inhibited the generation of intracellular ROS production as dose dependant manner. Conclusions CT has anti-oxidative effects and anti-inflammatory activities. These results indicate that CT extract has an anti-inflammatory activities via anti-oxidative effects.

The comparative study on physiological activity of White ginseng, Red ginseng and Black ginseng extract (백삼, 홍삼, 흑삼 추출물의 생리활성 비교 연구)

  • This study presents a comparative study for differences in efficacy and ingredient of white ginseng(WG), red ginseng(RG), and black ginseng(BG) using anti-oxidative activity test and anti-inflammatory activity test. In the results of cytotoxicity test for WG, RG, and BG, the survival rate of all cells was more than 95%. In the total polyphenol analysis, DPPH, ABTS radical scavenging test, and ROS production test, BG showed higher anti-oxidant activity than WG and RG. RG and WG showed higher inhibition activity of NO production and $PGE_2$ production, respectively. As results of the test for the effect on reduction of inflammatory cytokine production, WG and RG were effective on reduction of IL-$1{\beta}$ production, and BG was effective on reduction of IL-6. In the case of TNF-${\alpha}$ production, there was no difference among samples. This study could be useful basic data for the development of functional food and the fabrication of safe cosmetic.
