Cytoprotective effect of Bojungbangam-tang on cisplatin-induced nephrotoxocity (Cisplatin 유도 신장독성에 대한 보정방암탕 에탄올층의 보호효과)

  • Cisplatin, an antitumor agent widely used in the treatment of cancers, has nephrotoxicity. This side effect is closely related to oxidative stress. In the present study, we studied to protective effect of ethanol extract of Bojungbangam-tang (EBJT) on cisplatin-induced nephrotoxicity. Bojungbangam-tang is a new herbal prescription composed of nine crude drugs. Pretreatment of EBJT prevented cisplatin-induced cytotoxicity and generation of ROS. Also, cellular GSH content and gluathione peroxidase activity were recovered by EBJT. EBJT also decreased cisplatin-induced expression of HO-1 via inhibition of ERK activation. Taken together, these results suggest that EBJT has a cytoprotective effect against cisplatin-induced nephrotoxicity through anti-oxidant activity.

Neuroprotective effects of modified Bo-Yang-Hwan-Oh-Tang in N2a neuroblastoma cells (가감보양환오탕(加減補陽還五湯)의 N2a 뇌신경세포에 대한 보호효과)

  • Objectives : To evaluate the neuroprotective effects of modified Bo-Yang-Hwan-O-Tang (BHT), we investigated the neuronal death protection effects to oxidative damages in N2a neuroblastoma cells. Methods : To study the cytotoxic effect of BHT on N2a neuronal cells, the cell viability was determined by MTT assay. To investigate the neuronal death protection of BHT, N2a neuronal cells were induced oxidative damages by H2O2, and assayed the cell viability and DNA fragmentation. We also investigated DPPH free radical scavenging effects of BHT by tube test. Results : In MTT assay, $500{\mu}g/ml$ of BHT was not showed cytotoxic effect on N2a neuronal cells. BHT protected N2a neuronal cells from H2O2-induced cell death in a dose-dependent manner. BHT also protected N2a neuronal cells from H2O2-induced DNA fragmentation. BHT scavenged DPPH free radicals in a dose-dependent manner. Conclusion : These data suggest that BHT may have strong anti-oxidant effects through the free radical scavenging and neuroprotective effects in neuronal cells.

Anti-oxidant and Anti-inflammatory Potentials of Sasa quelpaertensis Leaf Residue Extracts (제주조릿대 잎 잔사 추출물의 항산화 및 항염 활성)

  • Sasa quelpaertensis Nakai is a native Korean plant that grows only on Mt. Halla of Jeju Island. Its leaf is used for a popular bamboo tea due to its various health-promoting properties, and it has been increasingly used as food and cosmetic ingredients. To utilize the S. quelpaertensis leaf efficiently, the preparation method for phytochemical-rich extract (PRE) using the leaf's residue was previously reported, which was produced after hot water extraction. This study was undertaken to evaluate the anti-oxidant and anti-inflammatory potential of PRE and its solvent fractions. The ethyl acetate fraction of PRE (EPRE) showed higher DPPH, ABTS, and superoxide radical scavenging activities, and it effectively inhibited intracellular reactive oxygen species (ROS) and nitric oxide (NO) production in lipopolysaccharides (LPS)-stimulated RAW 264.7 cells. EPRE also induced the expression of heme oxygenase-1 (HO-1) by increasing the level of nuclear factor E2-related factor 2 (Nrf2) in a nuclear fraction. The inhibiting effect of EPRE on LPS-induced NO production was partially reversed by the HO-1 inhibitor (zinc protoporphyrin, ZPP), suggesting that HO-1 is involved in suppressing NO production. Taken together, the results suggest that EPRE has potential as a promising anti- oxidant and anti-inflammatory agent.

Antioxidant Activity and Melanin Inhibitory Effects of Yambean (Pachyrhizus erosus) Extract (얌빈 추출물의 항산화 효능과 멜라닌 생성 억제효과)

  • Objectives : Yam bean (Pachyrhizus erosus) possess various nutrients, it has been widely used as traditional cosmetic material in Indonesia. The aim of this study was to investigate the anti-oxidant activity and the anti-melanogenic effect of Yambean (Pachyrhizus erosus) extract and its fractions. Methods : The anti-oxidant activity of yam bean extract assessed based on total polyphenol, flavonoid contents, DPPH and ABTS radical scavenging assay. To evaluate anti-melanogenic effects and cytotoxicity of Yambean extract and its fractions, B16F10 melanoma cell was used. Results : In results, total polyphenol content of yam bean water extract (YW) and Yambean 70% ethanol extract (YE) were $1.18{\pm}0.03mg/g$ (mg of gallic acid/g of sample), $1.16{\pm}0.01mg/g$. Total flavonoid contents of YW, YE were $3.55{\pm}0.06mg/g$ (mg of naringin/g of sample), $1.78{\pm}0.03mg/g$. Moreover, YE scavenged DPPH and ABTS effectively in $4mg/m{\ell}$ compared to YW. Cytotoxicity of YE and its fractions in B16F10 melanoma cell was measured using MTT assays. It had no cytotoxicity up to $500{\mu}g/m{\ell}$. Melanin accumulation in B16F10 melanoma cell was induced using alpha-melanocyte stimulating hormone (${\alpha}-MSH$) and 3-isobutyl-1-methylxanthine (IBMX). B16F10 melanoma cell treated with $10-500{\mu}g/m{\ell}$ YE and hexane, ethyl acetate, butanol, $H_2O$ fractions for 24h. Non treated B16F10 melanoma cell (Control) markedly increased melanin contents. In contrast, YE ethylacetate fraction effectively suppressed melanin accumulation in a dose-dependent manner. Conclusion : In conclusion, these results suggest that Yambean extract has the potential as a cosmetic material which possess anti-oxidant and anti-melanogenic activities.

Dual effect of curcumin on viability and motility of bovine sperm exposed to oxidative stress (산화스트레스에 노출된 정자의 생존성 및 운동성에 있어서 커큐민의 이중효과)

  • Although cryopreservation of sperm is routinely used for clinical requirement, it has some problems, such as high generation of reactive oxygen species (ROS) and cold-shock. To reduce the detrimental damage in sperm, anti-oxidants were added to cryoprotectant for sperm. Curcumin is one of anti-oxidants, which are added in cryoprotectants. However, recent studies have demonstrated that curcumin decreases sperm viability and motility. This study was performed to identify the effect of curcumin on hydrogen peroxide ($H_2O_2$)-exposed bovine sperm, which were cryopreserved-thawed. In $H_2O_2$-exposed bovine sperm, reactive oxygen species (ROS) were significantly reduced by treatment with curcumin in a dose-dependent manner (p < 0.05). Among tested concentrations of curcumin (1 to $50{\mu}M$), 30 and $50{\mu}M$ curcumin showed anti-oxidant effect on $H_2O_2$-induced ROS generation. On the other hand, combination of 30 or $50{\mu}M$ curcumin with anti-oxidant $H_2O_2$ increased the percentage of apoptotic sperm compared to only $H_2O_2$ treatment. Sperm viability was also decreased in the combination of 30 or $50{\mu}M$ curcumin with $H_2O_2$ as judged by FDA/PI staining. $H_2O_2$-induced decrease in sperm progressive motility was recovered by treatment with $1{\mu}M$ curcumin. These results show that high concentration of curcumin has anti-oxidant effect, but it has also cytotoxic effect on bovine sperm. Sperm viability and motility might be more affected by cytotoxic signals of curcumin compared to antioxidant signals.

Study on Anti-oxidant and Anti-inflammatory Activity of the Tissue-cultured Shoot Clumps from Raoulia australis (라울리아 신초 추출물의 항산화 및 항염 효능에 관한 연구)

  • In this study, the shoot clumps extract of tissue-cultured Raoulia australis using the bioreactor culture system was tested for use a natural cosmetic ingredient. Tissue-cultured R. australis shoot clumps extract was tested anti-oxidant and anti-inflammatory activity for a cosmetic application. R. australis is a wild herbaceous plant of the asteraceae growing in New Zealand and Australia. Previous studies have reported anti-viral activity of the inhibitory effects for the growth of viruses induced meningitis, bronchitis and respiratory diseases but other biological effects are unknown. The shoot clumps extract of tissue-cultured R. australis showed higher anti-oxidant effect and anti-inflammatory effect than the natural R. australis extract. In DPPH, NBT and ABTS assay, the shoot clumps extract of tissue-cultured R. australis enhanced radical scavenging activity (up to 10~25% at $50{\mu}L/mL$) more than the natural R. australis extract. Also, the shoot clumps extract of tissue-cultured R. australis inhibited expression of iNOS and COX-2 protein in LPS-stimulated Raw 264.7 macrophages more than the natural R. australis extract. From this study, the shoot clumps extract of tissue-cultured R. australis displayed strong possibility as a new natural cosmetic ingredient for skin-care products.

Anti-oxidant Effect of the Ethyl Acetate Soluble Fraction of Sophorae Radix in H9c2 Cells (심근세포에서 고삼 에틸 아세테이트 분획의 항산화 효과)

  • To test the cytoprotective effect of sophorae radix (SR) against hydrogen peroxide (H₂O₂)-induced cytotoxicity, we investigated the cell viability using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay in the presence of ethyl acetate subfractions of SR water extracts In H9c2 cells. And to clarify the cytoprotective mechanism of SR extracts, we evaluated the cellular glutathione (GSH) contents in the presence of subfraction 1, 2, 3, and 4 of SR ethyl acetate soluble fractions. Among 1 -12 subfractions of SR ethyl acetate soluble fractions, 1, 2, 3 and 4 subfractions have an efficacy inhibiting the cytotoxicity induced by H₂O₂ in H9c2 cells. Also, the protective effects of 1, 2, 3 and 4 subfractions of SR ethyl acetate soluble fractions resulted from the anti-oxidant effects. These results suggest that ethyl acetate soluble fractions of SR water extracts is effective in the prevention of H₂O₂-induced cytotoxicity and 1, 2, 3 and 4 subfraction of ethyl acetate soluble fractions possess the anti-oxidant component.

Study on anti-oxidant and cosmeceutical activities of Isatis tinctoria L. (판람근(Isatis tinctoria L., 板藍根)추출물의 항산화활성 및 화장품약리활성에 관한 연구)

  • Objective : In this study, anti-oxidant and cosmeceutical activities of Isatis tinctoria L. extracted from water, ethanol and supercritical fluid condition were confirmed to investigate cosmeceutical activities for utilization as cosmetic ingredient. Methods: Anti-oxidant and cosmeceutical activities were investigated by using electron donating ability, xanthine oxidase, tyrosinase, astringent effect. Result : Isatis tinctoria L. extracts by supercritical fluid, water and ethanol showed good electron donating ability which were 82.7%, 62.6% and 44.8% at the concentration at 1,000ppm, respectively. Xanthine oxidase activity related with purine metabolism was inhibited by ethanol extract about 52.3% at the concentration at 1,000ppm. Tyrosinase inhibition effects, by supercritical fluid extract, ethanol extract and water extract, were 83.3%, 52.9% and 41.2% respectively at 1,000ppm. In the measurement of astringent effect, supercritical fluid extract at the concentration to 5,000ppm showed 85.7% in related activity. The water extract showed 95.9% nitrite scavenging activity at 5,000ppm. Conclusion : According to these results, it is possible that the extract of Isatis tinctoria L. can be used as a new natural material of cosmetic industry.

Anti-oxidant Effects of Samultang-Gami on MEF Cells (사물탕가미방(四物湯加味方)의 항산화 활성에 대한 실험적 연구)

  • Purpose: This experiment is designed to find out anti-oxidant effects of Samultang-Gami which was composed of Rehmanniae Radix(RR), Angelicae Gigantis Radix, Cnidii Rhizoma(CR), Paeoniae Radix(PR), Cortex Moutan Radicis, Hedyotis Diffusa(HD) and Caesalpinia Sappan on MEF cells. Methods: In vitro antioxidant effects were measured by MTT assay, DPPH assay, cell cycle analysis, AnnexinV-FITC/PI assay and DAPI staining using MEF cells treated with various concentrations of 70% ethanol extract of Samultang-Gami. Results: 1. In the scavenging for DPPH radical, the each treated groups of PR, CR and HD showed positive effects. RR and CR increased the viability of oxidative damaged MEF cells in a dose-dependent manner. 2. 70% ethanol extract of Samultang-Gami was shown best antioxidative effect in the concentration of $0.5mg/m{\ell}$. 3. The treatment of Samultang-Gami in oxidative damaged MEF cells didn't have any effect on cell cycle restoration. but it could lower late apoptosis rate a little and be observed the protection of nucleus. Conclusion: It can be concluded that Samultang-Gami, RR and CR have antioxidant effects on MEF cells.

Preservation of Cosmetics by Ethanol Extract of Scutellaria baicalensis $G_{EORGE}$ (황금(黃芩)의 에탄올추출물에 의한 화장품 방부효과)

  • Ethanol extract (1.0 wt%) of Scutellaria baicalensis $G_{EORGE}$ practically satisfied CTFA (The cosmetics, Toiletry, and Fragrance Association) standard in its antimicrobial activity against Staphylococcus aureus, Pseudomonas aeruginosa and Candida albicans which were inoculated in skin toner and skin lotion. At concentrations of the extract less than 1.0 wt% the survival rate was the best for S. aureus and the worst for P. aeruginosa. The antimicrobial effect seems to be due to the damage to the bacterial cell wall as evidenced by the images of a Field Emission-Scanning Electron Microscope. The extract also showed a high anti-oxidant effect, and superoxide dismutase (SOD)-like activity reached 80% at 1,000 ppm. These findings suggest that the extract of Scutellaria baicalensis $G_{EORGE}$ is applicable to cosmetics as a natural preservative and an anti-oxidant.
