35종 해조류 추출물의 in-vitro 항혈전 활성 평가 (Evaluation of In-vitro Anticoagulation Activity of 35 Different Seaweed Extracts)

  • 혈전성 질환을 예방, 개선할 수 있는 안전한 항혈전제를 개발하기 위해, 일반적으로 항혈전 효과가 있다고 알려진 식용 해조류 35종(갈조류 17종, 홍조류 11종 및 녹조류 7종)을 대상으로 메탄올 추출물을 조제하고, 이들의 thrombin time (TT), prothrombin time (PT), activated partial thromboplastin time (aPTT)을 평가하였다. 트롬빈 저해(TT)의 경우 35종 중에서 감태, 곰피, 대황, 넓패, 패, 알송이모자반 및 야마다모자반에서 강력한 활성을, 프로트롬빈 저해(PT)의 경우 트롬빈 저해활성이 나타난 7종 중 야마다모자반을 제외한 6종에서 강력한 활성을 확인하였으며, 혈액응고인자 저해(aPTT)에서는 감태, 곰피, 대황, 넓패, 패, 알송이모자반 및 톳에서 강력한 활성이 나타났다. 선별된 8종의 시료를 대상으로 다양한 농도에서 항혈전 활성을 평가한 결과 알송이모자반에서 가장 강력한 활성을 확인하였으며, 곰피, 대황 및 넓패가 효과적이었다. 항혈전 활성과 해조류 추출물의 총 flavonoid, 총 polyphenol, 총당 및 환원당 함량과의 상관 관계를 검토한 결과, 활성물질은 flavonoid성 물질로 추측되었으며, 선별된 8종은 총 flavonoid 함량 및 총 polyphenol 함량이 매우 높은 특징을 나타내었다. 본 연구 결과는 경제적이면서 대량공급이 가능한 식용 해조류로부터 신규의 안전한 항혈전제 개발이 가능함을 제시하며, 야마다모자반과 톳은 트롬빈 또는 혈장내 응고 인자에 대한 특이 저해제로 개발 가능함을 제시하고 있다.

식용 꽃차 추출물의 항혈전 및 항산화 활성 (Anti-thrombosis and Anti-oxidant Activities of Edible Flower Teas)

  • 꽃은 특유의 모양, 색, 향, 맛으로 인한 시각적, 후각적 관능성이 우수하며, 최근에는 식용 꽃을 이용한 다류, 음료 제품이 사회적 관심을 받고 있다. 본 연구에서는 23종의 국내 시판 꽃차를 대상으로 메탄올 추출물을 조제한 후, 이들의 명도, 황색도, 적색도 및 식용 타르색소 함유 여부를 조사하여, 개별 꽃차 특유의 색차를 비교 분석하고 타르색소를 포함하지 않음을 확인하였다. 23종 꽃차 추출물의 총 폴리페놀 평균 함량은 80.2±50.92 mg/g이었으며, 진달래, 매화, 메밀, 목련 및 살구꽃 추출물에서 150 mg/g 이상의 매우 높은 함량을 보였다. 총당 함량 분석결과 케모마일(610.6), 국화(550.2), 동백(417.0) 및 원추리꽃(401.0) 추출물에서 400 mg/g 이상의 높은 함량을 확인하였으며, 23종 꽃차 추출물의 평균 함량은 187.4±166.5 mg/g으로 매우 큰 편차를 보였다. 항혈전 활성 평가결과, 장미꽃 추출물이 가장 우수한 항응고 활성을 나타내었으며, 달맞이, 동백, 히비스커스 및 메밀꽃 추출물에서는 강력한 트롬빈 저해 및 혈액응고인자 저해를 동시에 나타냄을 확인하였다. 또한 장미, 금잔화, 메밀, 목련, 동백 및 진달래꽃에서 강력한 항산화 활성을 확인하였다. 따라서 장미, 동백 및 메밀꽃은 새로운 항혈전 기능성 식품 소재로 개발 가능함을 확인하였다.

고지방 섭취 흰쥐에서 양파 껍질 추출물의 보충 섭취가 혈중 지질농도와 혈행 개선에 미치는 효과 (Effect of Onion Peel Extracts on Blood Lipid Profile and Blood Coagulation in High Fat Fed SD Rats)

  • 1. 본 연구는 고지방 섭취 동물 모델에서 양파 추출물을 용량별(0.01%, 0.02%, 0.05%, 0.1%)로 8주간 보충 섭취시킨 후 혈중 지질 농도와 혈행 장애 개선에 미치는 영향을 살펴보는 것을 주된 목적으로 하였다. 2. 양파 껍질 추출물 섭취에 따라 total cholesterol과 HDLcholesterol 및 LDL-cholesterol은 그룹간의 유의한 차이를 보였다. 사후분석 결과 LDL-cholesterol은 정상 대조군에 비해 고지방 대조군 및 고지방+양파 껍질 추출물 그룹이 유의하게 높았고, 고지방 대조군 대비 0.1% 양파 껍질 추출물 섭취군의 농도가 유의하게 감소되었다. HDL-cholesterol은 정상대조군 대비 고지방 식이 후 유의하게 감소하였다가 양파 껍질 추출물 섭취에 따라 용량 의존적으로 유의하게 증가하는 결과를 보였다. 3. Hematocrit, WBC, RBC, platelet count, MPV, MCV, MCH, MCHC 등 혈액학적 지표는 처치에 따른 유의적인 차이를 나타내지 않았다. Hemoglobin의 경우, ANOVA 분석 시 군간 유의한 평균값의 차이를 보였으나, Bonferroni correction을 통한 사후분석 결과 각 군간 유의적인 차이가 없었다. 4. 혈액 응고 지표로 분석된 PT, aPTT, TT와 fibrinogen은 ANOVA 분석 시 모든 군간 유의적인 차이가 없었다. 그러나 t-test 결과 고지방 대조군에 비해 0.05% 양파 추출물 첨가군에서 aPTT가 유의적으로 연장되었다. 5. 체중 증가의 cut-off에 따라 low gainer와 high gainer로 나누어 항혈전 지표를 비교한 결과, high gainer에서는 0.01%, 0.02%, 0.05% 양파 추출물 섭취군이 고지방 대조군에 비해 PT와 aPTT가 전반적으로 길어지는 경향을 보여주었다. 6. 이상의 결과에서 고지방 섭취 동물 모델에서 용량별 양파 추출물 첨가는 혈중 지질 농도에 영향을 주는 한편, 혈액응고시스템에 현저한 변화를 유발하지는 않는 것으로 보인다. 하지만 고지방 대조군에 비해 0.05% 양파 추출물 첨가 시aPTT의 유의적인 증가는 양파 추출물이 내인성 혈액 응고시스템에 부분적으로 영향을 줄 수 있는 가능성을 시사하므로 향후 비만 동물 모델을 대상으로 다양한 농도 범위의 양파 보충의 효과를 규명하는 후속 연구가 요구된다.

Allithiamine Exerts Therapeutic Effects on Sepsis by Modulating Metabolic Flux during Dendritic Cell Activation

  • Recent studies have highlighted that early enhancement of the glycolytic pathway is a mode of maintaining the proinflammatory status of immune cells. Thiamine, a wellknown co-activator of pyruvate dehydrogenase complex, a gatekeeping enzyme, shifts energy utilization of glucose from glycolysis to oxidative phosphorylation. Thus, we hypothesized that thiamine may modulate inflammation by alleviating metabolic shifts during immune cell activation. First, using allithiamine, which showed the most potent anti-inflammatory capacity among thiamine derivatives, we confirmed the inhibitory effects of allithiamine on the lipopolysaccharide (LPS)-induced pro-inflammatory cytokine production and maturation process in dendritic cells. We applied the LPS-induced sepsis model to examine whether allithiamine has a protective role in hyper-inflammatory status. We observed that allithiamine attenuated tissue damage and organ dysfunction during endotoxemia, even when the treatment was given after the early cytokine release. We assessed the changes in glucose metabolites during LPS-induced dendritic cell activation and found that allithiamine significantly inhibited glucose-driven citrate accumulation. We then examined the clinical implication of regulating metabolites during sepsis by performing a tail bleeding assay upon allithiamine treatment, which expands its capacity to hamper the coagulation process. Finally, we confirmed that the role of allithiamine in metabolic regulation is critical in exerting anti-inflammatory action by demonstrating its inhibitory effect upon mitochondrial citrate transporter activity. In conclusion, thiamine could be used as an alternative approach for controlling the immune response in patients with sepsis.

Roles of Fucoidan, an Anionic Sulfated Polysaccharide on BSA-Stabilized Oil-in-Water Emulsion

  • Fucoidan, a sulfated polysaccharide derived from brown seaweed, is an important material valued for its various biological functions, including anti-coagulation, anti-aging, and immune system support. In this study, we examined the potential of fucoidan as a novel emulsifying agent in BSA (bovine serum albumin)-stabilized emulsion at a neutral pH. We measured the dispersed oil-droplet size, surface zeta-potential and creaming formation of 0.5 wt% BSA emulsion (20 wt% oil traction) in the absence and presence of fucoidan. The average particle size and zeta-potential value were 625.4 nm and -30.91 mV in only BSA-stabilized emulsion and 745.2 nm and -44.2 mV in 1.0 wt% fucoidan-added BSA emulsion, respectively. This result suggested that some positive charges of the BSA molecules interacted with the negative charges of fucoidan to inhibit the flocculation among the oil droplets. The creaming rate calculated from the backscattering data measured by Turbiscan dramatically decreased in 1.0 wt% fucoidan-added BSA emulsion during storage. Accordingly, the repulsion forces induced among the oil particles coated with 1.0 wt% fucoidan in emulsion solution resulted in significantly increased emulsion stability. The turbidity of the BSA-stabilized emulsion at 500 nm decreased during five days of storage. However, the fucoidan-added BSA emulsion exhibited a higher value of turbidity than the BSA-stabilized emulsion did. In conclusion, an anionic sulfated fucoidan lowered the surface zeta-potential of BSA-coated oil droplets via the electrostatic interaction, and subsequently inhibited the flocculation among the oil droplets, thereby clearly minimizing the creaming and phase separation of the emulsion.

In silica Prediction of Angiogenesis-related Genes in Human Hepatocellular Carcinoma

  • Hepatocellular carcinoma (HCC) is one of the most common malignancies worldwide and a typical hypervascular tumor. Therefore, it is important to find factors related to angiogenesis in the process of HCC malignancy. In order to find angiogenesis-related factors in HCC, we used combined methods of in silico prediction and an experimental assay. We analyzed 1457 genes extracted from cDNA microarray of HCC patients by text-mining, sequence similarity search and domain analysis. As a result, we predicted that 16 genes were likely to be involved in angiogenesis and then the effects of these genes were confirmed by hypoxia response element(HRE)-luciferase assay. For instant, we classified osteopontin into a potent angiogenic factor and coagulation factor XII into a significant anti­angiogenic factor. Collectively, we suggest that using a combination of in silico prediction and experimental approaches, we can identify HCC-specific angiogenesis­related factors effectively and rapidly.

Emergent Endovascular Embolization for Iatrogenic Vertebral Artery Injury during Cervical Discectomy and Fusion

  • Injury to the vertebral artery during anterior cervical discectomy is rare but potentially fatal. We report a case of cerebellar infarction after endovascular embolization for iatrogenic vertebral artery injury at C5-C6 during an anterior cervical discectomy and fusion. A 61-year-old man had an intraoperative injury of the right vertebral artery that occurred during anterior cervical discectomy and fusion at C5-C6. Hemorrhage was not controlled successfully by packing with surgical hemostatic agents. While the patient was still intubated, an emergency angiogram was performed. The patient underwent endovascular occlusion of the right V2 segment with coils. After the procedure, his course was uneventful and he did not show any neurologic deficits. Brain computed tomographic scans taken 3 days after the operation revealed a right cerebellar infarction. Anti-coagulation medication was administered, and at 3-month follow-up examination, he had no neurologic sequelae in spite of the cerebellar infarction.

Deoxyribonucleic Acid Was Responsible for the Anticoagulatory Effect of an Earthworm, Lumbricus rubellus

  • Earthworm extracts are known for anti-inflammatory, analgesic. antipyretic, and anticancer effects but can also influence blood circulation. It was previously shown that an earthworm, Lumbricus rubelius. contained a water-extractable anticoagulant which was a heat- and acid-stable molecule with hydrophilic property. In order to uncover the biochemical nature of this molecule, the anticoagulant was processed with various hydrolases such as trypsin, DNase, RNase. and lysozome. When the digested samples were analyzed with an in vitro coagulation test measuring activated partial thromboplastin time (APTT) and agarose gel electrophoresis, the anticoagulant proved to be a relatively homogeneous DNA fragment with relative molecular size around 72 base pairs. Interestingly, the activity was further stimulated with a trypsin digestion. RNA. on the other hand, did not prolong the APTT. It was also demonstrated that the DNA accelerated the antithrombin III (AT-III) inhibition of thrombin from $IC_{50}$ of 0.34 to 0.16 unit determined with S-2238 as a substrate, whereas heparin, a popular anticoagulant. shifted the value to 0.05. Therefore, it is suggested that the DNA could be considered as an alternative antithrombotic agent to heparin, which would exhibits bleeding side effects.

Severe dapsone hypersensitivity syndrome in a child

  • Dapsone (4,4'-diaminodiphenylsulfone, DDS), a potent anti-inflammatory agent, is widely used in the treatment of leprosy and several chronic inflammatory skin diseases. Dapsone therapy rarely results in development of dapsone hypersensitivity syndrome, which is characterized by fever, hepatitis, generalized exfoliative dermatitis, and lymphadenopathy. Here, we describe the case of an 11-year-old Korean boy who initially presented with high fever, a morbilliform skin rash, generalized lymphadenopathy, hepatosplenomegaly, and leukopenia after 6 weeks of dapsone intake. Subsequently, he exhibited cholecystitis, gingivitis, colitis, sepsis, aseptic meningitis, disseminated intravascular coagulation, syndrome of inappropriate antidiuretic hormone secretion, pneumonia, pleural effusions, peritonitis, bronchiectatic changes, exfoliative dermatitis, and acute renal failure. After 2 months of supportive therapy, and prednisolone and antibiotic administration, most of the systemic symptoms resolved, with the exception of exfoliative dermatitis and erythema, which ameliorated over the following 4 months. Agranulocytosis, atypical lymphocytosis, aseptic meningitis, and bronchiectatic changes along with prolonged systemic symptoms with exfoliative dermatitis were the most peculiar features of the present case.

Spectroscopic and Electrochemical Detection of Thrombin/5'-SH or 3'-SH Aptamer Immobilized on (porous) Gold Substrates

  • Thrombin is a serine protease that catalyzes the conversion of soluble fibrinogen to insoluble fibrin, and thus induces physiological and pathological blood coagulation. Therefore, it is important to detect thrombin in blood serum for purposes of diagnosis. To achieve this goal, it has been suggested that a 15-mer aptamer strongly binds with thrombin to form a G-quartet structure of the aptamer. Generally, 5'-end thiol-functionalized aptamer has been used as an anti-thrombin binder. Herein, we evaluate the possibility of utilizing a 3'-SH aptasensor for thrombin detection using SPR spectroscopy, and compare the enhancement of the electrochemical signal of the thrombin-aptamer bound on a porous gold substrate. Although the two aptamers have similar configurations, in SPR analysis, the 3'-SH aptamer was a effective aptasensor as well as 5'-SH aptamer. Results from electrochemical analysis showed that the porous gold substrate acted as a good substrate for an aptasensor and demonstrated 5-fold enhancement of current change, as compared to gold thin film.
