Bacterial β-glucuronidase in the intestine is involved in the conversion of 7-ethyl-10-hydroxycamptochecin glucuronide (derived from irinotecan) to 7-ethyl-10-hydroxycamptothecin, which causes intestinal bleeding and diarrhea (side effects of anti-cancer drugs). Twelve compounds (1-12) from Polygala tenuifolia were evaluated in terms of β-glucuronidase inhibition in vitro. 4-O-Benzoyl-3'-O-(O-methylsinapoyl) sucrose (C3) was highly inhibitory at low concentrations. C3 (an uncompetitive inhibitor) exhibited a ki value of 13.4 μM; inhibitory activity increased as the substrate concentration rose. Molecular simulation revealed that C3 bound principally to the Gln158-Tyr160 enzyme loop. Thus, C3 will serve as a lead compound for development of new β-glucuronidase inhibitors.

Scutellaria Radix, originated from Scutellaria baicalensis Georgi, is one of the most important medicine in traditional Oriental medicine, and possesses anti-bacterial activity and sedative effects, can be applied in the treatment of a range of conditions including diarrhea and hepatitis. It is reported that chronic global ischemia induces neuronal damage in selective, vulnerable regions of the brain, especially the hippocampus and cerebral cortex. In the present study, to investigate the effect of Scutellaria Radix extract on cerebral disease, the changes of regional cerebral blood flow and pial arterial diameter on ischemia/reperfusion state was determinated by Laser-Doppler Flowmetry and some parameters concerned with oxidative stress also measured. When SRe were administered for five days with the concentration of 100 mg/kg, GSH activity significantly increased. But SRe administeration showed no significant change in lipid peroxidation. When the activities of CAT, Cu, Zn-SOD and GSH were measured, CAT and GSH were activated by SRe administration. When 1 and 3 ㎍/㎖ SRe was applied to the neuronal cell cultures, the quantities of LDH was significantly reduced when compared with cultures treated only with NMDA. Through this study, it can be concluded that the ischemia/reperfusion induced brain stress may have contributed to cerebral damage in rats, and the present study provides clear evidence for the beneficial effect of SRe on ischemia induced brain injury.

The purpose of this study was to investigate the anti-inflammatory effect and the antimicrobial activity to skin flora of essential oil from rosemary that naturally grown in Jeju. rosemary essential oil was extracted by water distillation essential oil extraction method. In order to confirm the anti-inflammatory activity of rosemary essential oil, it was confirmed that the production of NO and $PGE_2$ induced by LPS in RAW 264.7 cells was inhibited in a concentration-dependent manner. Western blot analysis showed that the expression of iNOS and COX-2, which are biosynthetic enzymes, decreased in a concentration-dependent manner. In addition, production of $TNF-{\alpha}$ and IL-6 the pro-inflammatory cytokines were inhibited. Antimicrobial activities of three S. epidermidis and three P. acnes strains including two antibiotic resistant strains were observed in paper disc method and MIC and MBC tests showed inhibition of bacterial growth and death. From the results of the experiment, we confirmed that rosemary essential oil has the anti-inflammatory and antimicrobial efficacy and it could be used as a cosmetic and skin care material in the future.

Fraxinus rhynchophylla Hance (F. rhynchophylla) is a traditional medicinal plant that has been widely used in East Asia and has been used for chronic bronchitis, bacterial dysentery and improved eyesight. F. rhynchophylla contains various type of coumarins such as esculin, esculetin, fraxin and fraxetin. Esculetin possesses versatile activities including antioxidant, anti-inflammatory, antimicrobial, anticancer properties and improvement of atopic dermatitis. However, there is no research on the process of increasing active components in F. rhynchophylla. The objectives of the present study were to apply biotransformation technology to F. rhynchophylla for increasing the content of esculetin, and enhancing anti-inflammatory and antioxidant activities. F. rhynchophylla extract (FRE) treated with viscozyme L (FRE-VL) showed 3.1 times higher content of esculetin than FRE, and exhibited effects such as increased anti-inflammatory activity and DPPH radical scavenging activity. Based on the these results, it is concluded that biotransformed FRE-VL could be potentially applicable as a new active ingredient in the cosmetic field.

Silsosangami(SSG) is a formula of treaditional korean medicines as an effective biological response modifier for augmenting host homeostasis of body circulation. Little is known of the biological activity of SSG and previous studies have focused mainly on their anti-thrombosis8). There is a growing interest in the pharmacological potential of the SSG due to the recent finding by our group that SSG and each constituent herbs of SSG were able to inhibit NO and prostaglandin E2 (PGE2) synthesis in murine peritoneal macrophages stimulated with bacterial endotoxin. In this paper, the effects of SSG on platelet aggregation and hemolysis in human blood were studied. SSG provoked remarkable inhibiting effect on platelet aggregation, and APTT were sensitive to the presence of this SSG. Using an in vitro system, APTT was delayed with the increment of the concentrations of these seven compounds. These results suggested that SSG might be used as a novel antithrombotic therapeutic agents in post-myocardial infarction. A SSG reduced NO production in mouse peritoneal macrophages stimulated with lipopolysaccharide, without the influence on the activity of iNOS being observed. SSG significantly reduced mouse paw edema induced by carrageenan. Western blot analysis showed that SSG reduced the expression of iNOS. The results indicate that SSG exerts anti-inflammatory effects related to the inhibition of NO production, which could be due to a decreased expression of iNOS.

We investigated characterization as cosmetic substances of chestnut inner skin extracts with in vitro antioxidant activity. Total phenolics of various extracts from chestnut inner skin were the highest 60% methanol (164.82 mg/g), and ethyl acetate fractions (191.14 mg/g). We found that the both samples from chestnut inner skin dose-dependently increased in vitro antioxidant activities (DPPH radical scavenging activity and inhibition of lipid peroxidation). In addition, the both samples also showed a strong UV absorption in the range of UV-B (290-320 nm). Especially the 60% ethanol extracts presented higher inhibitory effect on elastase (46.40% at $100{\mu}g/mL$) than that of the ethyl acetate fractions, so that it showed in vitro anti-wrinkle activity. Finally, the 60% methanol extracts and ethyl acetate fractions showed anti-bacterial activity against skin pathogenic bacteria. Consequently, these results suggest that the chestnut inner skin can be used for cosmetic industry.
Several growth factors and polypeptides are not commonly yet used for regenerators of bone tissue or alveolar bone because of the insufficiency of studies on their side effects, genetic engineering for mass production and stability for clinical application. Recently, many herbal medicines, which have advantage of less side effects and possibility of long-term use, have been studied for their capacity and effects of anti-bacterial, antiinflammatory and regenerative potential of periodontal tissues. Morindae Radix, Cibotium Barometz (L.), Albizziae Cortex, Cistandhis Herba have been traditionally used as medicines for treatment of bone disease in Eastern medicine. The objective of the present study is to examine the ability of alkaline phosphatase (ALP) activity of human fetal osteoblast (hFOB1) when several natural medicines were supplemented. hFOB1 were cultured with Dulbecuo's Modified Eagle's Medium Nutrient Mixture F-12 HAM ( DMEM/F-12 1:1 Mixture, Sigma, USA) and negative control, dexamethasone (positive control), and each natural medicines for 3 days. And then ALP activity was measured by spectrophotometer for enzyme activity and Alizarin red S staining for morphometry. Among the natural medicines of this study, Morindae Radix, Cibotium Barometz (L.) and Cistanchis Herba induced higher activity of ALP synthesis than negative controls in all experimental group. Albizziae Cortex showed mild increases than negative control group. According to measurement of positively stained area, all of the natural medicines of this study increased compared to negative control. Especially, Cibotium Barometz (L.) and Cistanchis Herba showed statistical significance compared to negative control (p<0.05). These results indicate that Morindae Radix, Cibotium Barometz (L.), Albizziae Cortex, Cistandhis Herba have an inducing ability of ALP synthesis on osteoblast.
The Opuntia ficus-indica var. saboten Makino (Cactus) is a tropical or subtropical plant, which is cultivated or grows naturally in Jeju island. It has been widely used as folk medicine for burned wound, edema and indigestion. In addition, its extract has been claimed to have several biological activities including anti-inflammation in oriental medicine. In this study, we examined the antimicrobial activities of the methanol extract of Opuntia ficus-indica var. saboten Makino. The extract showed a broad spectrum of antimicrobial activity against pathogenic bacteria, including antibiotics resistant bacteria (MRSA, R-P. aeruginosa, VRE) and Propionibacterium acnes, yeast, and fungi. The extract retained the activity after heat treatment for 15 min at $100^{\circ}C$ and $121^{\circ}C$ and after extended storage, up to 10 weeks storage period at $4^{\circ}C$ and $25^{\circ}C$, also stably retained its activity. It showed a better inhibitoring effect to the growth of E. coli than sodium benzoate did it at the same concentration. Addition of various salts or metal ions did not affect on its antimicrobial activity. Therefore, the antimicrobial characteristics of the extract can be applicable as a natural preservative and an antimicrobial agent for bacterial disease.
Recently, there has been a growing demand for natural preservatives because of increased consumer interest in health. In this study, we produced Lactobacillus rhamnosus cell-free supernatant (LCFS) and evaluated and compared its antimicrobial activity with existing natural preservatives against pathogenic microorganisms and in chicken breast meat contaminated with Escherichia coli and Staphylococcus aureus. Lactobacillus rhamnosus cell-free supernatant possessed 30 units of lysozyme activity and contained 18,835 mg/L of lactic acid, 2,051 mg/L of citric acid and 5,060 mg/L of acetic acid. Additionally, LCFS inhibited the growth of fourteen pathogenic bacteria, S. aureus, Bacillus cereus, Listeria monocytogenes, Vibrio parahaemolyticus, Listeria innocua, S. epidermidis, L. ivanovii, E. coli, Pseudomonas aeruginosa, Shigella sonnei, Shi. flexneri, Proteus vulgaris, Pseudomonas fluorescens, and Klebsiella pneumoniae. The antibacterial activity of LCFS was stronger than that of egg white lysozyme (EWL), Durafresh (DF) and grapefruit seed extract (GSE). Additionally, LCFS maintained its antimicrobial activity after heat treatment at $50^{\circ}C{\sim}95^{\circ}C$ and at pH values of 3~9. Moreover, LCFS inhibited the growth of E. coli and S. aureus in chicken breast meat. In conclusion, it is expected that LCFS, which contains both lysozyme and three organic acids, will be useful as a good natural preservative in the food industry.

There have been several studies regarding lichen-associated bacteria obtained from diverse environments. Our screening process identified 49 bacterial species in two lichens from the Himalayas: 17 species of Actinobacteria, 19 species of Firmicutes, and 13 species of Proteobacteria. We discovered five types of strong antimicrobial agent-producing bacteria. Although some strains exhibited weak antimicrobial activity, NP088, NP131, NP132, NP134, and NP160 exhibited strong antimicrobial activity against all multidrug-resistant strains. Polyketide synthase (PKS) fingerprinting revealed results for 69 of 148 strains; these had similar genes, such as fatty acid-related PKS, adenylation domain genes, PfaA, and PksD. Although the association between antimicrobial activity and the PKS fingerprinting results is poorly resolved, NP160 had six types of PKS fingerprinting genes, as well as strong antimicrobial activity. Therefore, we sequenced the draft genome of strain NP160, and predicted its secondary metabolism using antiSMASH version 4.2. NP160 had 46 clusters and was predicted to produce similar secondary metabolites with similarities of 5-100%. Although NP160 had 100% similarity with the alkylresorcinol biosynthetic gene cluster, our results showed low similarity with existing members of this biosynthetic gene cluster, and most have not yet been revealed. In conclusion, we expect that lichen-associated bacteria from the Himalayas can produce new secondary metabolites, and we found several secondary metabolite-related biosynthetic gene clusters to support this hypothesis.
