참당귀 초임계 이산화탄소 선택 추출물의 항염 효능 (The Anti-inflammatory Effect of Angelica gigas Nakai Supercritical Carbon Dioxide Selective Extracts)

  • 본 연구에서는 참당귀(Angelica gigas Nakai)의 핵심 유효 성분인 decursin과 decursinol angelate를 높은 수율로 추출한 초임계 이산화탄소 선택 추출물의 항염증 효능을 에탄올 추출물과 비교하여 측정하였다. 항염증 효능을 측정하기 위해 염증 매개 인자인 nitric oxide(NO), 염증성 사이토카인인 interleukin(IL)-6, IL-8 생성량을 측정하였다. NO 생성량은 lipopolysaccharide(LPS)로 염증 반응을 유도한 Raw 264.7 세포를 대상으로 Griess assay를 통해 측정하였고, IL-6 및 IL-8 생성량은 tumor necrosis factor(TNF)-α로 염증 반응을 유도한 human keratinocyte cell line(HaCaT) 세포를 대상으로 enzyme-linked immunoadsorbent assay(ELISA)를 통해 측정하였다. NO 생성량은 초임계 이산화탄소 추출물이 에탄올 추출물에 비해 뛰어나게 억제하였다. IL-6 및 IL-8 생성량 또한 에탄올 추출물은 오히려 증가시키는 반면, 초임계 이산화탄소 추출물은 6.25 ㎍/mL의 농도에서 통계적으로 유의하게 억제하였다(P<0.01). 우리는 이러한 결과를 통해 참당귀 초임계 이산화탄소 선택 추출물이 아토피 피부염을 완화시키는 항염증 코스메슈티컬 소재로써 활용 가능함을 확인하였다.

당분해효소를 이용한 물푸레나무 효소처리물의 항염 및 항산화 효능 연구 (A Study on Anti-inflammatory and Antioxidant Effect of Fraxinus rhynchophylla Using Carbohydrate-Hydrolyzing Enzymes)

  • 물푸레나무(Fraxinus rhynchophylla Hance, F. rhynchophylla)는 동아시아에서 널리 사용되어 온 전통적인 약용 식물로, 만성 기관지염, 세균성 이질과 시력 개선 등을 위해 사용돼 왔다. 물푸레나무에는 esculin, esculetin, fraxin 및 fraxetin과 같은 다양한 형태의 쿠마린이 존재하며, esculetin은 항산화, 항염, 항균, 항암 및 아토피성 피부 염증 완화 등의 효과를 가지고 있다. 지금까지 물푸레나무 내 유효 성분을 증가시키는 공정개발에 관한 연구는 보고되지 않았다. 따라서 본 연구의 목적은 물푸레나무 내 esculetin함량을 증가시키고, 항염 및 항산화 효능을 증대시키기 위한 물푸레나무 생물전환 공정을 개발하는 것이다. 당분해효소 viscozyme L을 이용하여 물푸레나무에 처리한 결과 물푸레나무 효소처리물(FRE-VL)은 물푸레나무 추출물(FRE)대비 esculetin 함량이 3.1배 증가하였으며, 항염 활성 증대와 DPPH 라디칼 소거 활성의 효과를 나타내었다. 본 연구를 통해 화장품 분야에서 생물전환 기술을 적용한 물푸레나무 효소처리물의 신규 화장품 기능성 소재로의 활용 가능성을 확인하였다.

한국 약용식물자원 추출물의 항알레르기 활성 평가 및 선발 (Screening and Evaluation of the Anti-allergic Effect of Korean Medicinal Plant Extracts)

  • Background: Allergic diseases like such as atopic dermatitis, asthma, and rhinitis have recently increased both domestically and globally. The present study was undertaken to select candidates with anti-allergic activity from plant resources. Methods and Results: Fifty-six plant extracts at $20{\mu}g/m{\ell}$ were screened against ${\beta}$-hexosaminidase production and interleukin (IL)-4 release in degranulated rat basophilic leukemia (RBL)-2H3 cells. The anti-allergy activity of three plant extracts selected from the preliminary screening experiment, Polygonatum sibiricum F. Delaroche (root), Pyrus pytifolia var. culta (Makino) Nakai (leaf), and Rehmannia glutinosa (Gaertn.) Libosch. ex Steud (root) were measured at concentrations of $2-250{\mu}g/m{\ell}$ in three cell lines as RBL-2H3, HaCaT and Jurkcat T cells. The assay showed the root extract of R. glutinosa to have an inhibitory activity of 4.2% - 28.6% on ${\beta}$-hexosaminidase production from IgE-sensitized RBL-2H3 cells. Each extract of P. sibiricum and R. glutinosa reduced IL-4 release in IgE-sensitized RBL-2H3 cells, respectively. The leaf extract of P. pyrifolia var. culta showed a significantly potent suppressive effect of 10.2% - 74.7% on the mRNA expression of tumor necrosis factor (TNF)-${\alpha}$ in HaCaT cells sensitized with TNF-a and INF-g, and showed inhibitory effect of -8.6% - 90.9% on the mRNA expression of IL-2 in Jurkat T cells sensitized with PMA and A23187. Conclusions: The results showed that the root of R. glutinosa and leaf of P. pyrifolia var. culta could be useful candidates as antiallergy materials.

비파엽(Eriobotrya japonica Lindl.) 열수추출물의 생리활성 (Physiological Activities of the Hot Water Extract from Eriobotrya japonica Lindl.)

  • 비파엽 열수추출물을 이용하여 항산화성과 항히스타민 및 항염증 효과를 조사하였다. 비파엽 열수추출물의 총 폴리페놀 함량은 28.91$\pm$2.1 mg/g, 총 플라보노이드 함량은 10.54$\pm$4.6 mg/g으로 분석되었다. 전자공여능은 400 $\mu g$/mL 에서 81.04$\pm$1.55%로 가장 높았으며, 아질산염 소거능은 pH 1.2($49.52\pm1.04\sim72.52\pm$0.84%)> pH 4.2($35.28\pm0.78\sim51.20\pm$1.10%)> pH 6.0($27.90\pm0.36\sim32.26\pm$1.20%) 순으로 pH가 낮을수록 비파엽 열수추출물 농도가 높을수록 높았다. 비파엽 열수추출물 100, 200, 300 $\mu g$/mL에서 $\beta$-hexosaminidase 방출 억제능은 각각 14.58$\pm$0.97%, 43.69$\pm$0.96%, 95.58$\pm$0.75%로 농도에 의존적으로 증가하였다. 비파엽 열수추출물을 이용하여 RAW cell을 대상으로 항염증 활성을 조사한 결과, 농도가 증가함에 따라 증가하여 400 $\mu g$/mL에서 44.35$\pm$0.63%를 나타내어 다양한 생리활성을 보여주었다.

참치심장 에탄올 추출물의 항아토피 효과 (Anti-Atopic Activity of Tuna Heart Ethanol Extract)

  • 본 연구는 DNCB(2,4-dinitrochlorobenzene) 도포를 통해 아토피 피부염을 유발시킨 BALB/c mice에 참치심장 에탄올 추출물(THEE)을 2주 동안 지속적으로 처리하여 THEE의 아토피 피부염 완화 효과에 대해 연구하였다. 이를 위해 Th1 및 Th2 cytokines의 분비량, 비장 세포 증식능, 혈청중 총 IgE 함량, 육안 평가 및 skin clinical severity score를 실시하였다. 그 결과 THEE를 지속적으로 도포함으로써 혈청 내 총 IgE의 분비량이 현저히 감소함을 나타내었다. Th1 세포로부터 생성되는 $IFN-{\gamma}$ cytokine은 증가하고 염증매개성 cytokine인 $TNF-{\alpha}$의 분비량은 감소함을 확인하였다. 또한 Th2 cytokine에 속하는 IL-4, IL-5, IL-13 분비량은 감소하였으며 항염증성 cytokine인 IL-10의 분비량이 증가함을 보여 항아토피 효과를 나타내었다. 육안 평가를 통해서 THEE를 2주간 지속적으로 도포 처리하였을 때 그 증상이 눈에 띄게 완화되는 것을 확인할 수 있었으며, skin clinical severity score에서도 DNCB 단독 처리군과 비교시 유의적으로 그 값이 감소하였다. 이상의 결과들로부터 THEE는 Th1과 Th2 cytokine의 생성 및 활성을 조절하고 IgE의 과다 발현을 억제함으로써 아토피 피부염의 개선에 뛰어난 효능을 가지고 있는 것으로 사료된다.

오레고닌의 in vitro 방출 특성에 미치는 연고기제의 영향 (Influence of Ointment Base on In Vitro Release Characteristics of Oregonin)

  • The bark of Alnus japonica has been used for the treatment of fever, hemorrhage and diarrehea in oriental traditional medicine. Recently, it was revealed that the diarylheptanoids from the bark of Alnus japonica possess anti-inflammatory activity and are expected to be applicable for atopic dermatitis. In this study, oregonin, one of major active components in the bark of Alnus japonica, was developed in the form of semisolid formulations for topical delivery. Oregonin was incorporated into four ointment bases: O/W cream, W/O cream, hydrophilic ointment and lipophilic ointment. Oregonin release from all formulation prepared was evaluated. Franz cell method and immersion method were employed to characterize the release patterns of drug from each formulation based on solvent availability. O/W cream showed a better release profile than the other formulations when evaluated with Franz cell method with an order of O/W cream, hydrophilic ointment, W/O cream and lipophilic ointment. In the immersion method, hydrophilic ointment showed the greatest release rate at times 1 hour exceeding compared to other bases with an order of hydrophilic ointment, O/W cream, W/O cream and lipophilic ointment. Hydrophilicity and solvent availability of formulation seems to significantly influence the release rate of oregonin from ointment bases. In this study, we successfully characterized the oregon in ointment and found that o/w cream is a promising formulation for the topical delivery of oregonin.

HaCaT 각질형성세포에서 개똥쑥(Artemisia annua L) 유래 성분인 Artemisinic acid의 Macrophage-derived Chemokine 억제 효과 (Inhibitory Effect of Artemisinic Acid Isolated from Artemisia Annua L on the MDC in HaCaT Keratinocytes)

  • In the present study, we investigated anti-inflammatory activity of artemisinic acid in HaCaT cells and RAW264.7 cells. Artemisinic acid showed inhibitory activity on macrophage-derived chemokines (MDC) expression, a factor related with atopic dermatitis (AD), in interferon (IFN)-${\gamma}$ and tumor necrosis factor (TNF)-${\alpha}$-stimulated HaCaT cells. In the study on action mechanism, pretreated artemisinic acid reduced the phosphorylation of STAT1 and p38 and the degradation of $I{\kappa}B$ by IFN-${\gamma}$ and TNF-${\alpha}$ stimulations. However, artemisinic acid didn't show the inhibitory activity on LPS-induced inflammatory mediators (NO, $PGE_2$, IL-6) in RAW264.7 cell. These results indicate that artemisinic acid inhibits IFN-${\gamma}$ and TNF-${\alpha}$-induced MDC expression through inhibition of signal factors, STAT1, NF-${\kappa}B$, and p38, in HaCaT keratinocytes.

$CD4^+$ Jurkat T 세포주에서 Th1과 Th2 사이토가인 조절에 미치는 황금 유래 Baicalin, Baicalein 및 Wogonin의 효과 (Th1 and Th2 cytokine Modulation by Baicalin, Baicalein and Wogonin from Scutellaria Radex on the $CD4^+$ Jurkat T Cells)

  • In the present study, baicalin, baicalein, and wogonin, a major flavone isolated from Scutellaria Radix were examined for their effects on PMA-induced Interlukin-6 (IL-6), $interferon-\gamma(IFN-\gamma)$, tumor necrosis factor $(TNF)-\alpha$, IL-4, IL-10, and IL-13 productions in the PMA-stimulated $CD4^+$ Jurkat T cells. These three compounds inhibited PMA-induced Th1 cytokine $(IL-6,\;IFN-\gamma,\;TNF-\alpha)$ and Th2 cytokine (IL-4 and IL-13) productions in a concentration-dependent manner. But wogonin, but not baicalin baicalein, increased PMA-induced IL-10 production. These results suggest that baicalin, baicalein, and wogonin, a major flavone modulate Th1 and Th2 cytokine productions in $CD4^+$ Jurkat T cells and these properties may contribute to the anti-atopic dermatitis activity of Scutellaria Radix.

The effect of Korean red ginseng on allergic inflammation in a murine model of allergic rhinitis

  • Korean red ginseng (KRG) is reported to have anti-allergic properties, including beneficial effects on asthma and atopic dermatitis. However, its effect on allergic rhinitis has not been studied extensively. This study examined how KRG affected allergic inflammation of the nasal cavity in an allergic mouse model. A total of 40 Balb/c female mice were divided into four experimental groups according to treatment and allergic state: group 1 (G1), saline only; group 2 (G2), ovalbumin (OVA); group 3 (G3), OVA+KRG; and group 4 (G4), OVA+dexamethasone. Serum IgE levels were significantly lower in the KRG treatment group (G3) than in the allergic group (G2). However, serum IgG1 levels did not differ between G2 and G3. In the nasal lavage fluid, IL-4 and IL-5 levels were significantly lower in G3 than in G2 (p<0.05). H&E and Luna staining revealed that the eosinophil count was lower in G3 and G4 than in G2 (p<0.05). Immunohistochemical staining revealed that there were fewer IL-4-, IL-5-, and MUC5AC-positive cells in G3 and G4 than in G2 (p<0.05). These results indicate that KRG reduces the nasal allergic inflammatory reaction in an allergic murine model by reducing Th2 cytokines.

Ethanolic extract of Red Sweet Pepper (Capsicum annuum L.) regulates the skin inflammation in vitro and in vivo

  • Allergic inflammatory disease has been increased by abnormal lifestyle and food habits. Especially, prevalence of atopic dermatitis (AD) has been elevated and treatment of AD has not been unclear. Red sweet pepper (RSP), named as Capsicum annuum L, has been known as having pharmacological effects such as antioxidant, detoxification and antibacterial effects. However, the beneficial effect of ethanolic extract of RSP on AD has not been partly examined yet. Therefore, the aim of this study was to investigate anti-inflammatory effects of RSP on AD in vitro and in vivo models. The treatment of RSP inhibited the secretion of inflammatory cytokine such as interleukin (IL)-6 and IL-8 in tumor necrosis factor (TNF)-${\alpha}$ and interferon (IFN)-${\gamma}$-stimulated human keratinocyte (HaCaT cell). Also, RSP extract regulated 2,4-dinitroflorobenzene (DNFB)-induced AD-like skin lesions in BALB/c mice. Oral administration of RSP ameliorated DNFB-induced AD-like symptoms. In presented results indicated that RSP inhibited inflammatory cytokines in HaCaT cell and ameliorated AD-like skin lesion through suppression of symptom of DNFB-induced skin inflammation. Thus, RSP might be a potential therapeutic agent for AD.
