Regulatory roles of ginseng on inflammatory caspases, executioners of inflammasome activation

  • Inflammation is an immune response that protects against pathogens and cellular stress. The hallmark of inflammatory responses is inflammasome activation in response to various stimuli. This subsequently activates downstream effectors, that is, inflammatory caspases such as caspase-1, 4, 5, 11, and 12. Extensive efforts have been made on developing effective and safe anti-inflammatory therapeutics, and ginseng has long been traditionally used as efficacious and safe herbal medicine in treating various inflammatory and inflammation-mediated diseases. Many studies have successfully shown that ginseng plays an anti-inflammatory role by inhibiting inflammasomes and inflammasome-activated inflammatory caspases. This review discusses the regulatory roles of ginseng on inflammatory caspases in inflammatory responses and also suggests new research areas on the anti-inflammatory function of ginseng, which provides a novel insight into the development of ginseng as an effective and safe anti-inflammatory herbal medicine.

  • Luteolin, 3', 4', 5,7-tetrahydroxyflavone, belongs to a group of naturally occurring compounds called flavonoids that are found widely in the plant kingdom. It possesses many beneficial properties including antioxidant, anti-inflammatory, anti-bacterial, anti-diabetic and anti-proliferative actions. Colorectal cancer (CRC) is a leading cause of cancer related deaths worldwide. Many signaling pathways are deregulated during the progression of colon cancer. In this review we aimed to analyze the protection offered by luteolin on colon cancer. During colon cancer genesis, luteolin known to reduce oxidative stress thereby protects the cell to undergo damage in vivo. Wnt/${\beta}$-catenin signaling, deregulated during neoplastic development, is modified by luteolin. Hence, luteolin can be considered as a potential drug to treat CRC.

  • After applying anti-smoking acupuncture to 756 high school students at least 4 times, I surveyed about the effect of the therapy. The result of the survey is as follows. 1. The age to start smoking is getting lower. And the rate of female students' smoking is increasing significantly. 2. The survey shows that students started to smoke mainly because of curiosity, stress, peer pressure, and their misunderstanding which the way to smoke looks cool. 3. The anti-smoking acupuncture therapy is expected to decrease the smoking desire because the applicants usually experience headache, dizziness, gastroenteric trouble after the treatment. 4. Students complained about earache and inconvenience of washing. 5. The result shows that the anti-smoking acupuncture therapy is generally efficacious, but the virtue of the therapy is much better with the students' willingness to stop smoking.

The regulatory role of Korean ginseng in skin cells

  • As the largest organ in our body, the skin acts as a barrier against external stress and damages. There are various cell types of skin, such as keratinocytes, melanocytes, fibroblasts, and skin stem cells. Korean ginseng, which is one of the biggest distributions of ginseng worldwide, is processed into different products, such as functional food, cosmetics, and medical supplies. This review aims to introduce the functional role of Korean ginseng on different dermal cell types, including the impact of Korean ginseng in anti-photodamaging, anti-inflammatory, anti-oxidative, anti-melanogenic, and wound healing activities, etc. We propose that this information could form the basis of future research of ginseng-derived components in skin health.

Comparison of the Interest in Anti-Aging, Need for Anti-Aging Services and the Performance of Health Promotion Behavior by Sex in their 20s (20대 성인에서 성별에 따른 항노화에 대한 관심도 및 건강증진행위 수행도 및 항노화서비스의 필요성 비교)

  • This aim of this study examined the relationship among the interest in anti-aging, health promotion behaviors and the need for anti-aging services by their 20s. Survey was conducted among adults from their 20s in the Changwon City. 228 responses were used for analysis. The overall average score of the interest and effort of anti-aging were 2.97 point and 2.62 point (out of 5), respectively. And those were both higher in female than men (p<0.01~p<0.001). The overall average score of need for anti-aging service was 3.50 point(total score is 5). In The demand for each area of anti-aging service were ≥3.5 point in all 5 areas, and stress management (4.00 point) was the highest, while the beauty management (3.60 point) was the lowest. There were significant differences in all five areas by sex (p<0.01~p<0.001). The overall score of the performance of health promotion behaviors was 2.44 point(total score is 4), and the interpersonal relationship score (2.85 point) was the highest, while the health responsibility score (2.08 point) was the lowest. The interest in anti-aging and performance of health promotion behaviors showed positive relationship to anti-aging services, and their explanation powers were 34.6% (p<0.001). The results of this study suggest be used as data to establish strategies revitalizing various anti-aging service in the twenties.

Antioxidative-activity of Phellinus baumii Pilát in Caenorhabditis elegans (상황버섯의 예쁜꼬마선충 내의 항산화 효과)

  • In order to research the anti-oxidative activity of methanol extract of Phellinus baumii Pilat (Hymenochaetaceae), Caenorhabditis elegans model system was used. Ethyl acetate soluble fraction of the methanol extract showed the most potent DPPH radical scavenging activity. The ethyl acetate fraction was measured on its activities of superoxide dismutase (SOD), catalase, and oxidative stress tolerance with reactive oxygen species (ROS) level in C. elegans. Furthermore, to see if regulation of stress-response gene is responsible for the increased stress tolerance of C. elegans which treated by the ethyl acetate fraction, we checked SOD-3 expression using a transgenic strain. Consequently, the ethyl acetate fraction of P. baumii increased SOD and the catalase activities in a dose-dependent manner in C. elegans, reduced ROS accumulation dose-dependently. Besides, the ethyl acetate fraction-treated CF1553 worms showed higher SOD-3::GFP intensity than the control worms.

Anti-Oxidant Efficiency and Memchanisms of Phytochemicals from Traditional Herbal Medicine (한약재-식물성천연화학물질의 항산화 효능 및 기전)

  • Antioxidants are compounds that protect cells against the damaging effects of reactive oxygen species (ROS). Some ROS, such as superoxide and hydrogen peroxide, are normally produced in cells as by-products of biochemical reactions or as signaling molecules. When ROS-generating reactions are activated excessively, pathological quantities of ROS are released to create an imbalance between antioxidants and ROS, called as oxidative stress. Oxidative stress, which may result in cellular damage, has been linked to cardiovascular disease, diabetes, cancer, and other degenerative conditions. In humans the first line of antioxidant defence are the antioxidant enzymes, especially SOD, glutathione peroxidase (GPX), and to a lesser extent catalase, as well as the tripeptide glutathione(GSH). These enzymes will help destroy ROS(reactive oxygen species) such as hydroxyl radical, $H_2O_2$ and lipid peroxides, while GSH protects against oxidized protein. Many herbal medicines possess antioxidant properties. Herbal antioxidants may protect against these diseases by contributing to the total antioxidant defense system of the human body. Here, many herbal medicines including Ginseng, Licorice, Ligusticum Chuanxiong, Ginkgo biloba and many others was reviewed in terms of anti-oxidant efficiency related to their components.

Anti-stress effects of Sihoueokgantang in the passive avoidance test and the forced swimming test (시호억간탕(柴胡抑肝湯)이 스트레스로 인한 기억저하와 우울행동에 미치는 영향)

  • Objectives: This study is carried out to investigate the anti-stress effects of the sihoueokgantang in rats. Methods: we performed the passive avoidance test and the forced swimming test. The degrees of TH were also measured with immunohistochemical method in the regions of VTA and LC. Results: 1. As the results of the passive avoidance test. the sihoueokgantang showed tendency to improvement in compared with control group. 2. As the results of the forced swimming test, immobility of rats was significantly decreased in sihoueokgantang in compared with control group. 3. As the results of TH expression in the region of the LC, TH-immunoreactive cells were significantly decreased in the sihoueokgantang 100 mg/kg. As the results of TH expression in the region of the VTA. TH-immunoreactive cells were significantly decreased in the sihoueokgantang 400 mg/kg. Conclusion: As the results above, it can be considered that sihoueokgantang has the effects of preventing depression and memory decrease caused by stress.

Autophagy Is Pro-Senescence When Seen in Close-Up, but Anti-Senescence in Long-Shot

  • When mammalian cells and animals face a variety of internal or external stresses, they need to make homeostatic changes so as to cope with various stresses. To this end, mammalian cells are equipped with two critical stress responses, autophagy and cellular senescence. Autophagy and cellular senescence share a number of stimuli including telomere shortening, DNA damage, oncogenic stress and oxidative stress, suggesting their intimate relationship. Autophagy is originally thought to suppress cellular senescence by removing damaged macromolecules or organelles, yet recent studies also indicated that autophagy promotes cellular senescence by facilitating the synthesis of senescence-associated secretory proteins. These seemingly opposite roles of autophagy may reflect a complex picture of autophagic regulation on cellular senescence, including different types of autophagy or a unique spatiotemporal activation of autophagy. Thus, a better understanding of autophagy process will lead us to not only elucidate the conundrum how autophagy plays dual roles in the regulation of cellular senescence but also helps the development of new therapeutic strategies for many human diseases associated with cellular senescence. We address the pro-senescence and anti-senescence roles of autophagy while focusing on the potential mechanistic aspects of this complex relationship between autophagy and cellular senescence.

  • Interfacial surface crack perpendicular to the surface, which is imbedded into bonded quarter planes under single anti-plane shear load is analyzed. The problem is formulated using Mellin transform, form which single Wiener-Hopf equation is derived. By solving the equation stress intensity factor is obtained in closed form. This solution can be used as a Green's function to generate the solutions of other problems with the same geometry but of different loading conditions.
