Effect of Ripe Fruits of Rubus coreanus on Anaphylactic Allergic Reaction and Production of Cytokines

  • The anaphylactic allergic reaction is involved in many allergic diseases such as asthma and allergic rhinitis. In this report, I investigated the effect of ripe fruits of Rubus coreanus Miq. (Rosaceae) (RFRC) on the anaphylactic allergic reaction and studied its possible mechanisms of action. RFRC inhibited compound 48/80-induced systemic anaphylactic reaction in mice. RFRC dose-dependently decreased the IgE-mediated passive cutaneous anaphylaxis. In addition, RFRC decreased the gene expression and production of tumor necrosis factor-$\alpha$ (TNF-$\alpha$) and interleukin (IL)-6 in phorbol 12-myristate 13-acetate (PMA) plus calcium ionophore A23187 (A23187)-stimulated human mast cells. These findings provide evidence that RFRC could be a candidate as an anti-allergic agent.

  • Objectives NJ is being used to treat inflammatory diseases in Korea. In this study, we attempted to evaluate the effects of NJ on atopic dermatitis (AD)-like lesions and mast cell-mediated allergy inflammation in vivo and in vitro. Methods and Results We investigated to ascertain the pharmacological effects of NJ on 2,4-dinitrofluorobenzene (DNFB)-induced allergic reactions under in vivo conditions. Additionally, to find possible explanations for the anti-inflammatory mechanisms of NJ, we evaluated the regulatory effects of NJ on the level of inflammatory mediators in phorbol 12-myristate 13-acetate plus calcium ionophore A23187 (PMACI)-stimulated human mast cells (HMC-1). Conclusions NJ inhibited the production of the inflammatory cytokines (IgE, IL-6, IL-8 and TNF-${\alpha}$) significantly in vivo and in vitro.

Adenine Inhibits B16-F10 Melanoma Cell Proliferation

  • Adenine, a purine base, is a structural component of essential biomolecules such as nucleic acids and adenine nucleotides. Its physiological roles have been uncovered. Adenine suppresses IgE-mediated allergy and LPS-induced inflammation. Although adenine is known to inhibit lymphocyte proliferation, the effect of adenine to melamoma cells is not reported. Here, we investigated the growth inhibitory effects of adenine on B16-F10 mouse melanoma cells. Adenine suppressed the proliferation of B16-F10 cells in dose-dependent manner with the maximal inhibitory dose of 2 mM. Adenine treatment induced cell death molecular markers such as PARP and caspase 3 cleavages. Pan-caspase inhibitor z-VAD dramatically rescued the cell death molecular markers, cell proliferation recovered marginally. These results provide the possibility of adenine to be used as an anti-tumor agent.

Effect of Gamiokyaek-tang on Immediate Type Hypersensitivity (가미옥액탕이 즉시형 과민반응에 미치는 영향)

  • Object : We investigated the effect of Gamiokyaek-tang (GOYT) on immediate type hypersensitivity. Methods : We investigated anti-dinitrophenyl (DNP) IgE-mediated passive cutaneous anaphylaxis and acetic acid-induced vascular permeability in rodents. Also perfonned MTT assay and b-hexosaminidase activity is measured in RBL-2H3. Results : GOYT inhibited passive cutaneous anaphylaxis and acetic acid-induced vascular permeability by oral administration. All the concentrations of GOYT from 0.1 to 5 mg/ml didn't have an effect on cell viability and cytotoxicity. b-hexosaminidase release in RBL-2H3 was significantly reduced by 2 and 5 mg/ml of GOYT. Conclusion : These results indicate that GOYT have inhibition effects on immediate type hypersensitivity .

Antiallergic Activity of Menthae Herba (박하의 항알레르기 활성)

  • Antiallergic activity of the aqueous extract of Menthae herba (MHAE) was investigated in mice and rats. MHAE inhibited systemic anaphylaxis induced by compound 48/80 in mice. Especially, MHAE inhibited compound 48/80 induced systemic anaphylaxis 100% with a dose of 500 mg/kg body weight. MHAE significantly inhibited serum histamine levels induced by compound 48/80. MHAE inhibited histamine release from the rat peritoneal mast cells activated by compound 48/80 or anti-DNP IgE. Our studies provide evidence that MHAE will be beneficial in the treatment of anaphylaxis.

  • Objectives : We studied the effects of Radix Asteris herbal acupuncture solution (RAHAS) on the type I hypersensitivity. Methods : In vivo, we measured compound 48/80 induced active systemic anaphylactic shock, anti-DNP IgE induced passive cutaneous anaphylaxis (PCA) and acetic acid induced microvascular permeability using ICR mice. In vitro, we showed effects on cytotoxicity and ${\beta}$-hexosaminidase release from RBL-2H3 cells. Results : In vivo, RAHAS pretreatments at $BL_{13}$ and optional points inhibited active systemic anaphylactic shock induced by compound 48/80 and microvascular permeability increased by acetic acid. PCA was only inhibited by RAHAS pretreatments at $BL_{13}$. In vitro, RAHAS treatments inhibited ${\beta}$-hexosaminidase release. Conclusion : These results suggest that RAHAS may be beneficial in the prevention of type I hypersensitive inflammatory response.

Conclusion : These results indicate that GOYT have inhibition effects on immediate type hypersensitivity.

  • Objectives : We studied the effects of Scutellariae Radix pharmacopuncture solution (SRHAS) on the type 1 hypersensitivity. Methods : In vivo, we measured compound 48/80-induced active systemic anaphylactic shock using ICR mice and anti-DNP IgE-induced passive cutaneous anaphylaxis (PCA) using Sprague Dawley rats. In vitro, we showed effects on cytotoxicity and ${\beta}-hexosaminidase$ release from RBL-2H3 cells. Results : In vivo, SRHAS pretreatments (100% or 50%) at BL13 inhibited active systemic anaphylactic shock induced by compound 48/80. PCA was only inhibited by pretreatments of SRHAS at optional points. In vitro, $0.1{\sim}2%$ SRHAS treatments did not affect cell viability while ${\beta}$-hexosaminidase release was significantly inhibited. Conclusions : These results suggest that SRHAS may be beneficial in the inhibition of type I hypersensitive inflammatory response.

  • Thia report deals with an imported case of calamar swelling suspective of loiasis, who had been in Nigeria, Africa for 2 years. This 33-year-old Korean technician was admitted to Severance Hospital, Yonsei University Medical Center, because of erythematous swellings on left hand and foot. His peripheral blood showed persistent eosinophilia (over 30% of WBC), increased IgE(1,000unit/ml) and significantly high antialarial antibody titer with ensyme immunoassay, although no microfilaria was detected on the periplleral blood films. Under the impression of Loa lea infection diethylcarbamasine was administered for a month. Four months later mobile swellings and eosinophilia disappeared, and anti-filarial antibody titers were normalized. It is assumed that the patient had suffered from Loa lea infection, which is the first report on loiasis in Korea.

Oldenlandia diffusa Ameliorates on Atopic Dermatitis in Mice

  • Oldenlandia diffusa (OD), a member of the Rubiaceae family, has been used for treating inflammatory and infectious diseases. However, the anti-atopic effects of OD are not completely understood yet. Thus, the aim of the present study was to elucidate beneficial effects of OD on atopic dermatitis (AD) in vivo. We examined the pharmacological effects of OD on compound 48/80- or histamine-induced scratching behaviors and 2, 4-dinitrochlrobenzene (DNCB)-induced AD-like skin lesions in mice. Additionally, we evaluated regulatory effects of OD on the expression of interleukin (IL)-6 and tumor necrosis factor (TNF)-α in DNCB-induced AD-like skin lesions. Results showed that OD inhibited the scratching behavior and AD symptoms. It also decreased serum levels of IgE and histamine in mice. Moreover, OD significantly ameliorated the expression of IL-6 and TNF-α in AD-like skin lesion. These finding suggest experimental evidence of that OD's potential for treating AD.
