The effect of Saururus chinensis Baill against oxidative damage and inflammation

  • Objectives : ROS are involved in a wide spectrum of diseases including chronic inflammation and cancer. S.chinensis Baill, a perennial herb commonly called Chinese lizard's tail or Sam-baek-cho in Korea, is used for the treatment of edema and inflammatory diseases in the Oriental folk medicine. In this study, we investigated the antioxidant activities and anti-inflammatory effects of the two extracts, water(WE) and ethyl acetate(EAE) from S.chinensis Baill. Methods : Anti-oxidant activity was evaluated using Fe2+ chelating and hydroxyl radical scavenging assay. DNA cleavage assay, and western blot and immunostaining for phospho-p65 were performed to evaluate anti-oxidative effect. Anti-inflammatory effect was performed using NO generation assay and western blot in LPS-stimulated RAW264.7 cell. Results : In Fe2+ chelating activity and hydroxyl radical scavenging activity, WE showed more strong scavenging activity for hydroxyl radical than EAE. WE scavenged hydroxyl radical by 12% at 3.2 ${\mu}g/ml$, 21% at 16 ${\mu}g/ml$, 32% at 80 ${\mu}g/ml$, 66% at 400 ${\mu}g/ml$ and 82% at 2000 ${\mu}g/ml$, respectively. In addition, WE showed more strong chelating activity than EAE. WE chelated Fe2+ ion by 1.1% at 3.2 ${\mu}g/ml$, 8.2% at 16 ${\mu}g/ml$, 26.3% at 80 ${\mu}g/ml$, 72% at 400 ${\mu}g/ml$ and 89% at 2000 ${\mu}g/ml$, respectively. Also, WE inhibited oxidative damage via its anti-oxidant activity. In anti-inflammatory effect, EAE inhibited NO production and iNOS expression. In addition EAE suppressed the NF-${\kappa}B$ and MAPK signaling pathway in LPS-stimulated RAW 264.7 cells. Conclusions : Together, these data indicate that S. chinensis Baill, shows anti-oxidant activity and anti-inflammatory effect.

Comparative Study on the Anti-Oxidative Effects of Achyranthis Japonicae Radix, Achyranthis Bidentatae Radix and Cyathulae Radix (우슬(牛膝), 회우슬(懷牛膝) 및 천우슬(天牛膝)의 산화억제 효과 비교 연구)

  • Objectives : This study was intended to compare genetic differences and anti-oxidative effects of Achyranthis Japonicae Radix, Achyranthis Bidentatae Radix and Cyathulae Radix. Methods : The genetic differences of Achyranthis Japonicae Radix, Achyranthis Bidentatae Radix and Cyathulae Radix were observed with RAPD analysis. Anti-oxidative effects of Japonicae Radix, Achyranthis Bidentatae Radix and Cyathulae Radix were investigated. Result : 1. As the result of RAPD analysis, Achyranthis Japonicae Radix was found to be genetically similar with Achyranthis Bidentatae Radix. 2. There was a little difference in the DPPH free radical scavenging activity, lipid peroxidation inhibiting activity, SOD activity of Achyranthis Japonicae Radix, Achyranthis Bidentatae Radix and Cyathulae Radix. Generally, Achyranthis Japonicae Radix was found to have stronger effect than the others. Conclusions : Achyranthis Japonicae Radix and Achyranthis Bidentatae Radix were genetically similar. Achyranthis Japonicae Radix, Achyranthis Bidentatae Radix and Cyathulae Radix all exhibited anti-oxidative action.

The Comparative Study of the Effects of Fructificatio Inonoti Obliqui Aqueous Extract according to the Extraction Temperature(I) -Anti-oxidative effect- (차가버섯 물 추출물의 추출온도에 따른 효능 비교 연구(I) -산화반응 억제효과-)

  • Objectives : The present study purposed to compare the anti-oxidative effect of Fructificatio Inonoti Obliqui aqueous extract according to extraction temperature. Methods : We measured the total phenol content and anti-oxidative activity of Fructificatio Inonoti Obliqui total extract and $50^{\circ}C$ low-temperature leachate. Results : Anti-oxidative activity of Fructificatio Inonoti Obliqui total extract and $50^{\circ}C$ low-temperature leachate was different according to extraction temperature, concentration, and antioxidant effect measuring method. In all experiments, the total extract showed higher antioxidant than n-BuOH fraction. The total phenol content was higher in Fructificatio Inonoti Obliqui total extract than $50^{\circ}C$ low-temperature leachate. Conclusions : Both Fructificatio Inonoti Obliqui total extract and Fructificatio Inonoti Obliqui $50^{\circ}C$ low-temperature leachate have significant anti-oxidative effect.

The Anti-oxidative and Anti-inflammatory Activities of Malus melliana Ethanol Extract (Malus melliana 에탄올 추출물의 항산화 및 항염증 활성)

  • Malus melliana (Hand.-Mazz.) Rehder (M. melliana) is a Chinese plant that belongs to the Rosaceae family. There have been no previous reports regarding its bioactivity. In this study, the anti-oxidative and anti-inflammatory activities of M. melliana ethanol extract (MMEE) were evaluated using a 1,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging activity assay, reactive oxygen species (ROS) scavenging activity assay, nitric oxide (NO) inhibitory activity assay, and the analysis of related protein expressions through Western blot hybridization. MMEE showed potent scavenging activity against DPPH, similar to ascorbic acid, a well-known anti-oxidative agent, which was used as a positive control. MMEE also inhibited hydrogen peroxide-induced ROS in RAW 264.7 cells. Moreover, MMEE induced the expression of an anti-oxidative enzyme, heme oxygenase 1, and its upstream transcription factor, nuclear factor E2-related factor-2, in a dose-dependent manner. On the other hand, MMEE was associated with a reduction in NO production, which was induced by the lipopolysaccharide treatment of RAW 264.7 cells. The expression of inducible nitric oxide synthase, which is the upstream regulator of NO production, was also inhibited. Taken together, these results suggest that MMEE has anti-oxidative and anti-inflammatory properties, thus appearing to be a potential anti-oxidant and anti-inflammatory agent. The further identification of active compounds that confer the biological activities of MMEE may be necessary.

Anti-oxidative and Anti-inflammatory Effect of 7 Herbal Extracts and Methods of Herbal Formula Compositioning (7종 한약재 추출물의 항산화 및 항염증 효과와 이를 이용한 방제 구성에 대한 고찰)

  • Objectives : Herbal formulas are complicated to analyze and difficult to compose because of its mixed, complex features. So we discussed about how to compose herbal formulas effective and efficient by analyzing various effectiveness of each herbal extracts. Methods : To evaluate the effectiveness of herbal formula, anti-oxidative and anti-inflammatory effectiveness were mathematically analyzed. DPPH, superoxide anions, Nitric Oxide scavenging activity was measured to evaluate the effectiveness of 7 herbal extracts. And next, cytotoxic activity of extracts on RAW 264.7 cells were measured using MTS assay. To asses anti-inflammatory effect, nitric oxide and $PGE_2$ production were measured. Based on these anti-oxidative and anti-inflammatory experiment result, mathematical analyzation were carried out with constOptm function, and determined efficacy-maximizing ratio. Results : This mathematical analysis based formula showed significantly outstanding effectiveness than other formulae. And estimated tendency of anti-inflammatory effectiveness was matched with real effectiveness. Conclusions : So, mathematical analysis can be available to evaluate and estimate the effectiveness of herbal formulae.

Anti-oxidative Activity of the Extracts from Houttuynia cordata Thunb. Fermented by Lactic Acid Bacteria (어성초(Houttuynia cordata Thunb) 유산균 발효물의 항산화 활성)

  • This study was performed to evaluate the possibility of application of lactic acid bacteria fermentation to increase the anti-oxidative activity of extracts from Houttuynia cordata Thunb. Houttuynia cordata Thunb. was fermented by two species of lactic acid bacteria, Leuconostoc mesenteroides 4395 and Lactobacillus sakei 383. The anti-oxidative activities of Houttuynia cordata Thunb. extracts were analyzed both before and after fermentation. Anti-oxidative activity was determined by in vitro assays to measure 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging and super oxide dismutase (SOD)-like activities, and by determining total flavonoid and total phenolic compound contents. The extracts of fermented Houttuynia cordata Thunb. had higher anti-oxidative activity than the unfermented control. The DPPH scavenging activity of the extracts after fermentation by Leuconostoc mesenteroides 4395 at 30℃ for 5 days was 71.67±0.52%, and after Lactobacillus sakei 383 fermentation at 35℃ for 5 days was 70.11±0.67%; these activities were both about 20% higher than the control. Increases of about 10 mg GAE/g of total phenolic compounds were found in both fermented extracts and both contained about 6 mg quercetin equivalents/g of total flavonoids, compared with 35.90±0.61 mg/g and 21.69±1.52 mg/g in the control, respectively. These results also suggested that fermentation time and temperature were important factors in determining the anti-oxidative effect of extracts from fermented Houttuynia cordata Thunb. These findings should be valuable for the development of medicines or functional foods with antioxidative activity.

  • Anti-oxidative and anti-inflammatory activities of Cotoneaster horizontalis Decne ethanol extract (CHEE) were evaluated. CHEE possessed a potent scavenging activity against 1,1-diphenyl-2-picryl hydrazyl, which was similar to the activity of ascorbic acid which was used as a positive control. CHEE also effectively suppressed hydrogen peroxide-induced reactive oxygen species on RAW 264.7 cells. Furthermore, CHEE induced the expression of the anti-oxidative enzyme heme oxygenase 1, and its upstream transcription factor, nuclear factor-E2-related factor 2. CHEE inhibited LPS induced nitric oxide (NO) formation as a consequence of inducible NO synthase (iNOS) down regulation. Taken together, these results provide us with an important new insight; that C. horizontalis possesses anti-oxidative and anti-inflammatory activities. Therefore, C. horizontalis may be utilized as a promising material in the field of nutraceuticals.

Anti-oxidative Activity and Trace Component of a Sprout Serum (새싹 추출물의 항산화 작용과 미량 성분)

  • In this study, the anti-oxidative effects and components of sprout serum were investigated. In the buds, high levels of zinc, iron and manganese were analyzed in addition to copper, magnesium, and potassium. The free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity of sprout serum was evaluated with $IC_{50}$. $IC_{50}$ of sprout serum was $58.0\;mgmL^{-1}$, and that of vitamin C was $3.7\;mgmL^{-1}$. In the test of superoxide scavenging activity of sprout serum, the activity was dependent on the concentration of serum. In this case, the $IC_{50}$ was 2.0 wt%. Cell viability was detected by using the MTT method. Cultured human fibroblast was treated with 15 mM $H_2O_2$ and cell viability was 70% in case of control. However, the effect of treating 0.5% of sprout serum was similar to that of 0.0001% of vitamin C.

Anti-Oxidative and Anti-Obesity Effects of Amomum Cardamomum L. Extract (백두구 추출물의 항산화 및 항비만 효과)

  • In this study, the anti-oxidative and anti-obesity activities of Amomum cardamomum L. methanol extract (ACME) were evaluated using DPPH radical scavenging activity assay, pancreatic lipase enzyme inhibition assay, and the cell culture model system. ACME exhibited DPPH radical scavenging activities dose-dependently, with $IC_{50}$ of DPPH radical scavenging activities of ACME being $25.15{\mu}g/ml$. Furthermore, ACME effectively suppressed pancreatic lipase enzyme activity dose-dependently. ACME also significantly suppressed adipocyte differentiation, lipid accumulation, triglyceride (TG) contents, and triggered lipolysis activity on 3T3-L1 preadipocytes in a dose-dependent manner, without cytotoxicity. Their anti-obesity effect was modulated by the cytidine-cytidine-adenosine-adenosine-thymidine (CCAAT)/enhancer binding proteins ${\alpha}$ ($C/EBP{\alpha}$), $C/EBP{\beta}$ and the peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$) gene and protein expressions. Taken together, these results provide an important new insight that A. cardamomum L. possesses anti-oxidative and anti-obesity activities such as pancreatic lipase inhibition, anti-adipogenic, and lipolysis effects. There is therefore potential for its use as a promising component in the field of nutraceuticals and the identification of the active compounds that confer the anti-oxidative and anti-obesity activities of ACME might be an appropriate next step.

Anti-Oxidative Activity of Pectin and Its Stabilizing Effect on Retinyl Palmitate

  • The purpose of this study was to examine the anti-oxidative activity of pectin and other polysaccharides in order to develop a cosmeceutical base having anti-oxidative effects towards retinyl palmitate (RP). The anti-oxidative stabilizing effects of pectin and other polysaccharides on RP were evaluated by DPPH assay and then the stabilizing effect of pectin on RP was examined as a function of time. Among the polysaccharides we examined, pectin exhibited a considerably higher anti-oxidative activity, with an approximately 5-fold greater DPPH radical scavenging effect compared to other polysaccharides. The DPPH radical scavenging effect of pectin increased gradually with increasing concentrations of pectin. At two different RP concentrations, 0.01 and 0.1% in ethanol, addition of pectin improved the stability of RP in a concentration dependent manner. The stabilizing effect of pectin on RP was more effective for the lower concentration of RP (0.01%, v/v). Further, degradation of RP was reduced following the addition of pectin as measured over 8 hours. From the results obtained, it can be suggested that pectin may be a promising ingredient for cosmeceutical bases designed to stabilize RP or other pharmacological agents subject to degradation by oxidation.
