아위버섯 추출물의 혈전용해, 트롬빈저해, 항산화 및 항염증 활성 (Fibrinolytic, thrombin inhibitory, anti-oxidative and anti-inflammatory activities of Pleurotus ferulea)

  • 아위버섯을 혈관계질환 예방과 치료를 위한 기능성 식품 개발에 이용하기 위한 기초 자료를 얻기 위해 아위버섯 물추출물과 유기용매 분획물의 혈전용해활성과 트롬빈 저해활성, 항산화활성 및 항염증활성을 확인하였다. 에틸아세테이트 분획물은 1.33 plasmin unit의 높은 혈전용해 활성과, 94.45%의 높은 트롬빈 저해활성을 나타냈으며, 물 추출물은 37.01%의 항산화활성을 나타냈으며, 클로로포름 분획물은 98.13%의 가장 높은 항염증활성을 나타냈다. 실험 결과로부터 아위버섯의 에틸아세테이트 분획물은 높은 혈전용해활성, 트롬빈 저해활성 그리고 클로로포름 분획물은 높은 항염증활성을 나타내 심혈관계 질환 관련 치료나 예방을 위한 기능성식품 개발에 이용 가능할 것으로 기대된다.

The root extract of Paeonia lactiflora Pall inhibits the oxidative damage via its anti-oxidant activity

  • Objectives : Reactive oxygen species (ROS) have been associated with pathogenic processes including carcinogenesis through direct effect on DNA directly and by acting as a tumor promoter. Therefore, it has been regarded that ROS may be a major target for cancer prevention. The root of Paeonia lactiflora pall (PL), a traditional Chinese herb, has been a component of effective prescriptions for treatment of liver disease. Also, there are some reports about the antioxidant activities of the extracts from PL. However, little has been known about the effects of PL against oxidative damage. This work aimed to elucidate the anti-oxidant effects of Paeonia lactiflora pall (PL) in the non-cellular system and cellular system. Methods : Antioxidant activities of PL were evaluated by hydroxyl radical scavenging assay and $Fe^{2+}$ chelating assay. Anti-oxidative effect of PL was evaluated by ${\varphi}X$-174 RF I plasmid DNA cleavage assay in non-cellular system. In addition, DNA migration assay, expression level of phospho-H2AX, MTT assay and lipid peroxidation assay were performed for evaluate the anti-oxidative effect of PL in cellular system. Results : PL had a dose-dependent hydroxyl radical scavenging and $Fe^{2+}$ chelating capacity. In addition, PL inhibited oxidative DNA and cell damage induced by hydroxyl radical in non-cellular system and cellular system. Conclusion : Taken together, P. lactiflora pall may be possible for the application to a potential drug for treating the oxidative diseases such as cancer.

우슬(牛膝), 회우슬(懷牛膝) 및 천우슬(天牛膝)의 산화억제 효과 비교 연구 (Comparative Study on the Anti-Oxidative Effects of Achyranthis Japonicae Radix, Achyranthis Bidentatae Radix and Cyathulae Radix)

  • Objectives : This study was intended to compare genetic differences and anti-oxidative effects of Achyranthis Japonicae Radix, Achyranthis Bidentatae Radix and Cyathulae Radix. Methods : The genetic differences of Achyranthis Japonicae Radix, Achyranthis Bidentatae Radix and Cyathulae Radix were observed with RAPD analysis. Anti-oxidative effects of Japonicae Radix, Achyranthis Bidentatae Radix and Cyathulae Radix were investigated. Result : 1. As the result of RAPD analysis, Achyranthis Japonicae Radix was found to be genetically similar with Achyranthis Bidentatae Radix. 2. There was a little difference in the DPPH free radical scavenging activity, lipid peroxidation inhibiting activity, SOD activity of Achyranthis Japonicae Radix, Achyranthis Bidentatae Radix and Cyathulae Radix. Generally, Achyranthis Japonicae Radix was found to have stronger effect than the others. Conclusions : Achyranthis Japonicae Radix and Achyranthis Bidentatae Radix were genetically similar. Achyranthis Japonicae Radix, Achyranthis Bidentatae Radix and Cyathulae Radix all exhibited anti-oxidative action.

녹용대보탕(鹿茸大補湯)이 AAPH로 유발된 산화병태(酸化病態)모델 흰 쥐의 항산화(抗酸化) 작용(作用)에 미치는 영향 (The Anti-oxidative Effect of Oral Administration of NYD (Nocyongdaebo-tang) in Oxidized Rats induced by AAPH)

  • 1. Objectives This study aimed to investigate the anti-oxidative effect of Nocyongdaebo-tang (NYD) decoction in oxidized rats induced by AAPH. 2. Methods AAPH (2.2-azobis-2 aminodinopropane-hydrochloride) was injected intraperitoneally to induce oxidization and NYD was orally administered to the rats. To observe the anti-oxidative effect of NYD, we performed blood chemistry analysis, histological analysis, and evaluated the levels of SOD, catalase, glutathione, NO and MDA in liver. 3. Results & Conclusions 1) Serum Albumin level was increased significantly in the NYD group as compared with the control group and the saline group. 2) Serum LDL-cholesterol level was reduced significantly in the NYD group as compared with the control group. 3) GPT level in liver was reduced significantly in the NYD group as compared with the control group. 4) SOD, catalase activity and glutathione levels were increased significantly in the NYD group as compared with the control group and the saline group. 5) The levels of NO and MDA were reduced significantly in the NYD group as compared with the control group. 6) The degenerative change of liver tissue of NYD group was decreased in comparison with the control group. In conclusion, NYD decoction is considered to have an anti-oxidative effect in rats.

수종의 한약재 분획물의 생리활성 효과 (The Biological Activities of Extracts and Fractions of Herbal Plants)

  • Objectives : This study was conducted to identify the antimicrobial anti-oxidative and tyrosinase inhibitory effect of three kinds of herbal plants, Polygonum cuspidatum, Chelidonii herba, Pharbitis nil L. naturally grown across the nation. Methods : To investigate in vitro anti-oxidative activity assay, antimicrobial effect and tyrosinase inhibitory activity, MeOH 80% extract, n-hexane, chloroform, ethyl acetate, n-butanol and water fractions of herbal plants were tested by diphenylpicrylhydrazyl(DPPH) method. Results : Bsutanol fractions of each plants showed about 90% on anti-oxidative effect. In case of tyrosinase activity, ethlyacetate fraction of Polygonum cuspidatum showed a potent inhibition effect. In the antimicrobial effect, ethylacetate fraction of Polygonum cuspidatu, hexane fractions of Chelidonii herba and the methanol 80% extracts of Pharbitis nil L. exhibited inhibition effect significantly. Conclusions : In MeOH 80% extracts, we couldn't found any extracts or fractions which have antimicrobial, anti-oxidative and tyrosinase inhibitory activity, however ethylacetate fractions of Polygonum cuspidatum showed effects commonly in these three assay system.

  • The objective of this study is screening the anti-oxidative effects of several plant MeOH extracts against oxidative stress in Neuroblastoma cell. Oxidative stress has been implicated in the pathogenesis of many neurotoxicity, neurodegenerative disorders and cell death. This oxidative stress is generated by ROS (Reactive Oxygen Species) such as nitric oxide, nitrogen dioxide, peroxyl, superoxide ($O_2^-$), hydroxyl, alkoxyl. So, in the present study, we induced oxidative stress by treatment of sodium nitroprusside (2.5 mM) in human neuroblastoma SH-SY5Y cell which was treated samples before 24hr, and cell viability was measured by MTT reduction assay. Of those tested, the extracts of Paeonia japonica (roots), Eucommia ulmoides (炒)(barks), Paeonia japonica (曝乾)(roots), Phyllostachys bambusoides (stems), Polygala tenuifolia (去心, 炒)(roots), Paeonia japonica (roots), Polygala tenuifolia (roots), Machilus thunbergii (barks), Mallotus japonicus (leaves), Poria cocos (whole), Sophora flavescens (roots), Angelica tenuissima (roots), Angelica gigas (當歸尾)(roots) showed anti-oxidative effects[$EC_{50}$<15.20 ${\mu}g$/ml(Carnosine:Positive control)]in dose dependent manner.

Cotoneaster horizontalis Decne 추출물의 항산화 및 항염증 활성 (Anti-Oxidative and Anti-Inflammatory Activities of Cotoneaster horizontalis Decne Extract)

  • Cotoneaster horizontalis Decne 에탄올 추출물(CHEE)의 항산화능과 항염증 생리활성을 분석하였다. CHEE의 항산화능을 DPPH radical scavenging activity로 분석한 결과 radical 소거능의 정도가 양성 대조군으로 사용한 ascorbic acid와 유사한 정도의 높은 활성을 보여 매우 강한 항산화능을 보유함을 확인하였다. 또한 RAW 264.7 세포주를 이용하여 H2O2 유도에 의해 생성된 ROS에 대한 소거능을 분석한 결과에서도 강한 소거능을 보였다. 뿐만 아니라 항산화효소 HO-1 및 그 전사 인자인 Nrf2의 단백질 발현이 CHEE의 처리에 의해 증가되었다. 한편 CHEE가 LPS에 의해 유도된 NO 생성에 미치는 영향을 분석한 결과 농도의존적인 NO 생성 저해능을 보였으며 이는 NO 생성 단백질인 iNOS의 발현 저해에서 기인함을 확인하였다. 이러한 결과를 통해 C. horizontalis의 항산화능과 항염증 활성을 세포 수준에서 처음으로 확인하였으며 향후 기능성 소재로서 유용하게 활용될 수 있을 것으로 판단된다.

  • Objective : This study was carried out to investigate diagnostic difference and anti-oxidative effect of 3 different Anemarrhenae Rhizoma (AR) extracts. Methods : Microscopic examination was used to distinguish histological differences. And eliminative ability of several kinds of free radicals was also measured. Results : In microscopic examination, we can distinguished three different lineage of AR with different structure of vascular bundles and secretory structures. The extract of AR show profitable abilities of elimination of DPPH free radical, ABTS free radical and hydrogen peroxide. Conclusion : So, it can be concluded that AR extract has an anti-oxidative effects. Especially indeciduous lineage was most effective to remove free radicals.

[6]-Gingerol Attenuates Radiation-induced Cytotoxicity and Oxidative Stress in HepG2 Cells

  • [6]-Gingerol, a major polyphenol of ginger (Zingiber officinale), exhibits a variety of biological properties including anti-oxidant, anti-inflammatory and anti-cancer activity. However, the radioprotective effect of [6]-gingerol is still unknown. The aim of this study was to investigate the radioprotective effect of [6]-gingerol against radiation-induced cell cytotoxicity and oxidative stress in HepG2 cells. [6]-Gingerol pretreatment attenuated radiation-induced cell cytotoxicity caused by 5Gy (half lethal dose, $LD_{50}$ of HepG2 cells). The measurements of superoxide dismutase (SOD) and catalase (CAT) activity were also performed. The results showed that [6]-gingerol pretreatment reduced increasing SOD and CAT activity after exposure of IR, indicating that [6]-gingerol protected oxidative stress by regulating cellular antioxidant enzyme (SOD and CAT) activity. These findings suggest that [6]-gingerol acts as a radioprotector by attenuating cell cytotoxicity and oxidative stress.

Anti-inflammatory, Anti-oxidative and Anti-bacterial Activities of the Constituents Extracted from Leaves of Talipariti hamabo

  • Talipariti hamabo is a plant growing around salt marshes in the Lava Coast region of Jeju Island, Korea. In this study, the extract of T. hamabo leaves was investigated for the anti-inflammatory, anti-oxidative and anti-bacterial activities and their active constituents were identified. In the anti-inflammatory tests using lipopolysaccharide-stimulated RAW264.7 cells, the ethyl acetate (EtOAc) fraction inhibited the nitric oxide production without causing cell toxicity. Moreover, the EtOAc fraction reduced pro-inflammatory cytokines (tumor necrosis factor-α, interleukin-6) as well as prostaglandin E2. In the anti-oxidative studies with DPPH and ABTS+ radicals, potent scavenging activities were observed in the EtOAc and n-butanol fractions. Upon the anti-bacterial tests using Staphylococcus epidermidis, EtOAc and n-butanol fractions exhibited good activities. Through the phytochemical studies on EtOAc fraction, three components were isolated by repeated column chromatography; oleic acid (1), p-hydroxyphenethyl-trans-ferulate (2), nicotiflorine (3). Based on these results, the extract of T. hamabo leaves can be developed as natural resources for cosmetic applications.
