Identification of Streptomyces sp. Producing Antibiotics Against Phytopathogenic Fungi, and Its Structure

  • In order to develop a biocontrol agent that can effectively control Fusarium wilt on Cymbidium genus, the effectiveness of antagonistic microbes against the cause pathogen was screened. The selected microbe showed a broad spectrum of antifungal activity, and the culture broth of this microbe had better preventive effect on Fusarium wilt than the commercial chemical agent in the pot assay. This isolated strain, GBA-12, was identified as Streptomyces kasugaensis, and the antifungal substance was purified from a broth culture of GBA-12. This purified substance was identified as a polyene macrolide (YS-822A) that was newly discovered from Streptomyces kasugaensis, and it exhibited antifungal activity against several phytopathogenic fungi.

Effect of Enterococcus faecalis strain PL9003 on Adherence and Growth of Helicobacter pylori

  • The purpose of the present study was to examine the antagonistic activities of Enterococcus faecalis strain PL9003 (PL9003) on Helicobacter pylori. This strain was isolated from infant feces and found to inhibit both the growth of H. pylori and its in vitro adherence to the human gastric cell line MKN-45. The binding of PL9003 to MKN-45 was observed under a light microscope after Cram staining and under a scanning electron microscope. When detected with an FITC-conjugate antibody, both viable and nonviable PL9003 were found to decrease the number of H. pylori bound to MKN-45. When detected by an enzyme-linked immunoabsorbent assay, about 70% of the H. pylori bound on MKN-45 disappeared with the four-1314 addition of viable or nonviable PL9003. The spent culture supernatant (SCS) of PL9003 also decreased the viability of H pylori even after neutralization and pepsin treatment. The above results suggest that PL9003 has a potential as a new probiotic for the stomach.

감초 함유 처방의 글리치리진 대사와 몇가지 효소저해효과 (Metabolism of Glycyrrhizin in Polyprescriptions Containing Glycyrrhizae Radix by Human Intestinal Bacteria and Their Inhibitory Effects on Some Enzymes)

  • To analyze scientifically the prescription principle of polyprescriptions (Gamchotang, Daewhanggamchotang, Jakyakgamchotang, Gamchogungangtang and Gilkyungtang) containing Glycyrrhizae Radix, the transforming rate of glycyrrhizin in these polyprescriptions to 18 ${\beta}-glycyrrhetinic$ acid and their inhibitory effect on ${\beta}-glucuronidase$, hyaluronidase, phosphodiesterase and trypsin were investigated. When Glycyrrhizae Radix containing polyprescriptions were extracted with water, the contents of glycyrrhizin in water extract of Glycyrrhizae Radix with Rhei Rhizoma or with Zingiberis Rhizoma were higher than that of Glycyrrhizae Radix only, but that in water extract of Glycyrrhizae Radix with Platicodi Radix was lower than that of Glycyrrhizae Radix only. By human intestinal bacteria, glycyrrhizin was metabolized to 18 ${\beta}-glycyrrhetinic$ acid. These metabolism of glycyrrhizin in polyprescriptions containing Glycyrrhizae Radix was inhibited by Rhei Rhizoma, Paeoniae Radix and Platicodi Radix, but was not affected by Zingiberis Rhizoma. The inhibitory activity of Glycyrrhizae Radix on hyaluronidase and ${\beta}-glucuronidase$, was synergistic with Rhei Rhizoma or Zingiberis Rhizoma, but was antagonistic by Platicodi Radix.

Corrosion Behavior and Oxide Film Formation of T91 Steel under Different Water Chemistry Operation Conditions

  • The corrosion behavior of a ferritic/martensitic steel T91 exposed to an aqueous solution containing chloride and sulfate ions is investigated depending on the stimulated all-volatile treatment (AVT) and under oxygenated treatment (OT) conditions. The corrosion of T91 steel under OT condition is severe, while the corrosion under AVT condition is not. The co-existence of chloride and sulfate ions has antagonistic effect on the corrosion of T91 steel in both AVT and OT conditions. Unlike to corrosion resistance in the aqueous solution, OT pretreatment provides T91 steel lower oxidation-resistance than VAT pretreatment. From scanning electron microscopy/energy dispersive X-ray spectroscopy (SEM/EDS) and X-ray diffraction (XRD) analysis, the lower corrosion resistance in the aqueous solution by VAT conditions possibly is due to the formation of pits. In addition, the lower oxidation resistance of T91 steel pretreated by OT conditions is explained as follows: the cracks formed during the immersion under OT conditions accelerated peeling-off rate of the oxide film.

인삼의 적변을 유발하는 세균에 대하여 항균활성을 가지는 방선균 선발 및 동정 (Screening and Identification of Antibacterial Actinomycetes against Bacteria Causing Rusty Root on Ginseng)

  • Rusty root, the browning disease on ginseng, decreases quality and value. Recent studies indicated that endophytic bacteria could be a possible cause of rusty root. Actinomycetes antagonistic to the rusty-root-causing bacteria were isolated from soil. Twenty nine out of 932-isolates of Actinomycetes from soil showed antibacterial activity against Agrobacterium tumefaciens and Pseudomonas veronii an endophytic isolate in ginseng. The strongest antibacterial strain(ATO4O104) was classified based on 16S rDNA sequence. The Actinomycetes strain, ATO4O104, isolated in soil of USA volcano national park was identified as Streptomyces adephospholyticus. To test plant toxicity, radish seeds were sprouted with the culture of S. adephospholyticus and it did not show any harmful effect. The butanol partition out of n-hexane, ethyl acetate, butanol, and water partions showed the highest antibacterial activity.

Antagonistic Effect of Streptomyces sp. BS062 against Botrytis Diseases

  • The use of microorganisms and their secreted molecules to prevent plant diseases is considered an attractive alternative and way to supplement synthetic fungicides for the management of plant diseases. Strain BS062 was selected based on its ability to inhibit the mycelial growth of Botrytis cinerea, a major causal fungus of postharvest root rot of ginseng and strawberry gray mold disease. Strain BS062 was found to be closely related to Streptomyces hygroscopicus (99% similarity) on the basis of 16S ribosomal DNA sequence analysis. Postharvest root rot of ginseng and strawberry gray mold disease caused by B. cinerea were controlled up to 73.9% and 58%, respectively, upon treatment with culture broth of Streptomyces sp. BS062. These results suggest that strain BS062 may be a potential agent for controlling ginseng postharvest root rot and strawberry gray mold disease.

Screening of Trichoderma Isolates as a Biological Control Agent against Ceratocystis paradoxa Causing Pineapple Disease of Sugarcane

  • In this study, dual culture, poison agar, and direct methods were used to assess the ability of Trichoderma virens IMI-392430, T. pseudokoningii IMI-392431, T. harzianum IMI-392432, T. harzianum IMI-392433, and T. harzianum IMI-392434 to control Ceratocystis paradoxa, which causes the pineapple disease of sugarcane. The highest percentage inhibition of radial growth (PIRG) values were observed with T. harzianum IMI-392432 using two dual culture methods, 63.80% in Method I and 80.82% in Method II. The minimum colony overgrowth time was observed with T. harzianum IMI-392432 and the maximum was observed with T. pseudokoningii IMI-392431. Different concentrations of different day-old metabolites of Trichoderma isolates were tested against mycelial growth of C. paradoxa. The highest PIRG (84.685%) exhibited at 80% concentration of 30-day-old metabolites of T. harzianum IMI-392432 using the modified bilayer poison agar method. In the direct assay method the maximum mycelial growth weight (PIGW) was observed at the same concentration and the same day-old metabolites of T. harzianum IMI-392432. This study showed that Trichoderma isolates have a good antagonistic effect on C. paradoxa mycelial growth and T. harzianum IMI-392432 has the most potential to control the pineapple disease pathogen.

Biocontrol Activity of Aspergillus terreus ANU-301 against Two Distinct Plant Diseases, Tomato Fusarium Wilt and Potato Soft Rot

  • To screen antagonistic fungi against plant pathogens, dual culture assay (DCA) and culture filtrate assay (CFA) were performed with unknown soil-born fungi. Among the different fungi isolated and screened from the soil, fungal isolate ANU-301 successfully inhibited growth of different plant pathogenic fungi, Colletotrichum acutatum, Alternaria alternata, and Fusarium oxysporum, in DCA and CFA. Morphological characteristics and rDNA internal transcribed spacer sequence analysis identified ANU-301 as Aspergillus terreus. Inoculation of tomato plants with Fusarium oxysporum f. sp. lycopersici (FOL) induced severe wilting symptom; however, co-inoculation with ANU-301 significantly enhanced resistance of tomato plants against FOL. In addition, culture filtrate (CF) of ANU-301 not only showed bacterial growth inhibition activity against Dickeya chrysanthemi (Dc), but also demonstrated protective effect in potato tuber against soft rot disease. Gas chromatography-tandem mass spectrometry analysis of CF of ANU-301 identified 2,4-bis(1-methyl-1-phenylethyl)-phenol (MPP) as the most abundant compound. MPP inhibited growth of Dc, but not of FOL, in a dose-dependent manner, and protected potato tuber from the soft rot disease induced by Dc. In conclusion, Aspergillus terreus ANU-301 could be used and further tested as a potential biological control agent.

살균제 및 길항세균 배양액 처리에 의한 백합 저장구근의 부패 발생억제 (Inhibition of Penicillium Bulb Rot by Fungicides and Culture Broth of Antagonistic Bacteria during Storage of Lilium Oriental Hybrids)

  • 'Casablanca'와 'Marco Polo' 품종의 구근 저장중 발생하는 부패병을 방제하기 위해 azoxystrobin, benomyl+thiram 및 prochloraz와 길항세균배양액 등을 처리하여 적정 약제 및 농도를 구명하고, 약제처리된 구근의 생육상 및 절화의 품질을 조사하여 약해의 유무를 판별하고자 수행하였다. 백합구근이 부패병 발생은 azoxystrobin과 prochloraz가 억제 효과가 있으며, Benomyl+thiram는 국내산 구근의 부패병 발병 억제 효과가 없는 것으로 조사되었다. 생리적으로 백합구근의 견고성을 증가시키기 위해 처리한 calcium chloride 혼용처리는 부패병을 증가시켰다. 길항세균 배양액을 1/5로 희석하여 처리하였을 때, 부패병이 발생을 효과적으로 억제하였다. 재배중 'Casablanca' 품종은 benomyl+thiram 처리구에서, 'Marco Polo' 품종은 benomyl+thiram와 calcium 혼용 처리구에서 맹아 억제가 심하였다. 구근의 저장전 약제처리는 백합의 영양생장 과정에서 생육상과 꽃의 품질에는 영향을 주지 않으며, calcium과 benomyl+thiram 처리구에서 잎과 꽃의 길이가 다른 처리구에 비해 작으나 유의차는 없었다.

비병원성 Erwinia carotovora subsp. carotovora 9-3의 건조 및 저장의 과정에서 생존에 미치는 염 효과 (High Concentration of Sodium Chloride Increases on Survival of Non-pathogenic Erwinia carotovora subsp. carotovora 9-3 during Drying and Storage)

  • 길항력이 있는 그람음성균들의 생물적 방제제로 성공적인 개발은 균의 건조와 저장시의 낮은 생존을 때문에 건조제형으로 만드는 것은 제한되어있다. 그람음성균의 배양시 높은 염(NaCl)의 첨가는 삼투압 조절제인 osmolyte의 생성을 유도하여 건조 시 생존율을 높인다고 알려져 있다. 그람음성의 길항력을 내는 비병원성 Erwinia carotovora subsp. carotovora(Ecc) 9-3의 건조와 저장시의 염의 영향에 관해 연구하였다. Ecc 9-3의 생장률은 0.5 M의 염이 첨가되어 있는 배지에서 큰 차이가 없었지만 0.7 M의 염이 첨가된 배지에서는 급격하게 낮아진 것을 확인하였다. 건조와 저장시의 생존율은 0.5 M의 염을 넣은 배지가 염을 넣지 않은 배지보다 상온건조와 동결건조에서 각각 2배 그리고 3배의 높은 생존율을 얻었다. 건조 후 저장에서도 역시 염이 높을수록 Ecc 9-3의 생존율이 더 높았다. 그리고 상온건조와 동결건조 모두 $4^{\circ}C$에 저장하였을 때 생존율에 큰 차이가 없었다. 그러나 생존율은 $27^{\circ}C$, $37^{\circ}C$에 저장하였을 때 염의 농도가 높을수록 생존율이 증가하였다. 첨가제 실험에서는 lactose가 동결건조와 상온건조 모두에 영향을 주었고 dextrin은 상온건조에 큰 영향을 주었다.
