신선초 분말의 위생화를 위한 오존처리와 감미선 조사와의 비교 효과 (Comparative Effect of ${\gamma}$-Irradiation and Ozone Treatment for the Improvement of Hygienic Quality of Dried-Angelica Keiskei Koidz Powder)

  • For the purpose of improving hygienic quality of dried-Angelica Keiskei Koidz powder, the effect of ozone treatment and gamma irradiation on the microbial decontamination and physicochemical properties were investigated. Gamma irradiation at 5 to 7.5 kGy resulted in sterilizing total aerobic bacteria, molds and coliforms below detective levels, while ozone treatment for 8 hours up to 18 ppm did not sufficiently eliminate the total aerobic bacteria of the sample. The physicochemical properties of the sample were not changed by gamma irradiation up to 7.5 kGy, whereas, ozone treatment caused remarkable changes in pH, TBA value, chlorophyll, carotenoid and fatty acid compositions. Therefore, this investigation demonstrated conclusively that gamma irradiation was more effective than ozone treatment for decontaminating and sterilizing the dried-Angelica Keiskei Koidz powder, with minimal effect on the physicochemical properties analyzed.

  • For the purpose of improving hygienic quality of health-foods, sterilization effects of ${\gamma}$-ray and ozone on microoganisms associated with food cultured in the media and contaminated in Angelica keiskei powder were investigated. Ozone was immersed in water and sprayed in air, on the concentration of 3 mg liter-1 at an air flow rate of 5 liter min-3. Test strains cultured in the media completely inhibited by ${\gamma}$-ray at irradiation doses of 0.25~2 kGy. In the case of ozone, test bacteria inactivated after treatment of 10~20 minutes, but test mold, Aspergillus flavus was not effective. Strains contaminated in Angelica keiskei powder completely inhibited by ${\gamma}$-ray at irradiation doses of 2.5~7.5 kGy. However, when the powder was sprayed with ozonized air for 10hours, Bacillus subtilis and Staphylococcus aureus among five strains were eliminated.

신선초 김치 숙성 중 Chlorophyll 함량 및 휘발성 향기 성분의 변화 (Changes in Chlorphyll Contents and Volatile Compounds of Angelica keiskei Kimchi during Fermentation)

  • Changes in the contents of volatiles and chlorphylls of Angelica keiskei Kimchi during fermentation were investigated. Total chlorphyll contents decreased gradually during the fermentatin period. Chlorphyll a decreased rapidly from 1.38 to 0.5$\mu\textrm{g}$/cm by the 5th day of fermentation with no further changes at 7th day, while chlorophyll b did not show any change by 3rd day and decomposed rapidly thereafter. Major voltile compounds identified in the kimchi were sabinene, $\alpha$-pinene and $\alpha$-terpinolene, which were gradually decreased during fermentation. There were significant differnce in color, sourness, bitterness and overall acceptability in sernsory evauation of Angelica keiskei Kimchi during fermentation at 2$0^{\circ}C$.

Angelica keiskei가 흰쥐의 지질대사에 미치는 영향 (Effects of Angelica keiskei on Lipid Metabolism in Rats)

  • 흰쥐에 cholesterol(0.5%) 및 A. keiskei(5%)를 급여 한 후 혈장 및 간장 지질대사 및 분변 중의 neutral steroids 및 담즙산 배설에 미치는 영향을 관찰하였다. 혈장 cholesterol 농도는 식이중 cholesterol 첨가에 의해 현저히 증가되었으며, A. keiskei 급여시 유의적으로 감소하였다. 식이중 cholesterol 첨가는 혈장 HDL-cholesterol 함량을 유의적으로 감소시키는 반면 LDL-cholesterol을 크게 증가시켜 동맥경화지수를 높였다. A. keiskei 급여는 혈장 LDL-cholesterol의 유의적으로 감소시켰으며, 동맥경화지수도 cholesterol을 급여하지 않은 군에서 0.21, cholesterol 급여 군에서 0.52를 보여 A. keiskei를 급여하지 않은 각각의 대조군에 비하여 유의적 감소를 보여 A. keiskei가 혈장지질 개선효과를 보였다. A. keiskei 급여는 또한 cholesterol 급여와 관계없이 혈장 triglyceride 함량을 유의적으로 감소시키는 결과를 보였으나, 혈장 인지질 함량에는 영향을 미치지 않았다. A. keiskei 급여는 간조직의 total cholesterol, free cholesterol, cholesteryl ester 및 phospholipid 함량에는 영향을 미치지 않았으나, triglyceride 함량은 식이중 cholesterol 첨가와 관계없이 유의적으로 감소시켰다. A. keiskei는 cholestanol, 총 neutral steroid 및 담즙산의 배설을 증가시켰다. 이와 같은 결과로 보아 A. keiskei에 의한 혈장 cholesterol의 감소는 cholesterol 흡수의 감소 및 cholesterol을 cholestanol으로의 전환과 담즙산으로의 배설이 증가됨에 일부 기인하는 것으로 사료된다.

신선초와 무시레기의 항산화 성분 및 활성 비교 (Antioxidant Activities of Angelica keiskei L. and dried leaves of Raphanus sativus L.)

  • 무시레기와 신선초의 항산화 성분 및 활성을 분석하여 비교하였다. 무시레기와 신선초 추출물의 polyphenol 함량은 각각 100 g당 $2.84{\pm}0.11g$, $2.88{\pm}0.02g$이었으며, flavonoid 함량은 $0.19{\pm}0.00g$, $0.38{\pm}0.01g$ 으로 확인되었다. 항산화 활성 측정 결과, 무시레기 및 신선초 추출물이 DPPH radical 소거활성에서 $IC_{50}$값이 $348.72{\pm}31.65{\mu}g/mL$, $183.37{\pm}48.17{\mu}g/mL$으로 나타났으며, $ABTS^+$ radical 소거활성은 17.8 %, 11.9 %의 활성을, SOD 유사활성에서는 63%, 58%의 활성을 가진 것으로 나타났다. 총 polyphenol 및 flavonoid 함량과 항산화 활성간의 상관관계에서 총 flavonoid 함량과 DPPH radical 소거능이 r=0.9804, $ABTS^+$ radical 소거활성은 r=-0.9863의 상관관계를 보였으며, DPPH radical 소거활성은 $ABTS^+$ radical 소거활성과 r=-0.9609, SOD 유사활성과는 r=0.4865로서 각각 유의한 부와 정의 상관관계를 나타내었다.

전남지역 자생식물들의 소염활성검색 및 활성화합물 (Screening of Medicinal Plants on the Anti-inflammatory Effect and Active Component)

  • The anti-inflammatory effect of the methanolic extracts of some medicinal plants was investigated on the hydrolysis of N-benzoylarginine-p-nitroanilide by trypsin in vitro, the writhing syndrome induced by acetic acid in mice, and the carrageenin-induced paw edema in rats. Among tested medicinal plants, the methanoilc extract of Angelica keiskei, Rosa rugosa and Cudrania tricuspidata showed significant inhibitory effect. And we also found that adenosine, isolated from A. keiskei was identified as one active components.

초고압처리 신선초 녹즙의 저온저장 안정성 및 관능적 특성 변화 (Effect of High Hydrostatic Pressure on the Shelf-life and Sensory Characteristics of Angelica keiskei Juice)

  • 초고압처리한 신선초 녹즙을 $4^{\circ}C$에서 냉장보관 하면서 미생물의 변화 및 관능적 특성의 변화를 살펴보았다. 초고압처리는 전보에서 최적살균조건으로 계산된 $5700\;kg_f/cm^2$, 7 min의 조건으로 처리하였는데 초기 $8.7{\times}10^6\;CFU/mL$ 존재하던 일반 세균수가 초고압처리 후에는 $3.2{\times}10^3\;CFU/mL$로 감소하여 전보에서 계산된 예상 살균치인 3.44 log cycle 감소에 잘 부합하였다. 특히, 저온저장 식품에 문제가 되는 녹농균이 완전히 사멸하였으므로 저온저장에 유리한 특성을 부여하리라 생각되었다. 대장균 및 대장균군도 완전히 사멸되었다. 관능검사 결과, 밝기와 풀냄새를 제외하고는 초고압처리에 의해 관능특성상의 유의적 차이가 생기지 않았으며, $4^{\circ}C$에서 8일간 저장한 후에는 신선감에서 초고압처리한 녹즙이 유의적으로 높은 점수를 얻었다. 하지만 초고압처리 녹즙에서도 효소적 갈색화로 인한 색도의 변화가 일어나 해결해야 할 문제점으로 부각되었다.

Antimutagenic Activity of the Methanol Extract and Compounds of Angelica keiskei in the Salmonella Assay System

  • The methanol extract of aerial part of Angelica keiskei Koidzumi exhibited a strong antimutagenic activity against aflatoxin $B_1\;(AFB_1)$. N-methyl-N'-nitro-N-nitrosoguanidine and 4-nitroquinoline-1-oxide in the Ames test with Salmonella typhimurium TA100. Cynaroside, isolated front ethylacetate fraction of the methanol extract Over silica gel, inhibited the mutagenicity of $AFB_1$ with an inhibition value of 96% at 1.0 mg/plate concentration and 87% at 0.5 mg/plate concentration. Other compounds, hyperoside, sucrose and luteolin-7-rutinoside, isolated from ethylactate or n-butanol fraction, also showed antimutagenic effect.

Xanthoangelol and 4-Hydroxyderricin Are the Major Active Principles of the Inhibitory Activities against Monoamine Oxidases on Angelica keiskei K

  • Monoamine oxidase inhibitors (MAOI) have been widely used as antidepressants. Recently, there has been renewed interest in MAO inhibitors. The activity-guided fractionation of extracts from Angelica keiskei Koidzumi (A. keiskei K.) led to the isolation of two prenylated chalcones, xanthoangelol and 4-hydroxyderricin and a flavonoid, cynaroside. These three isolated compounds are the major active ingredients of A. keiskei K. to inhibit the MAOs and DBH activities. Xanthoangelol is a nonselective MAO inhibitor, and a potent dopamine ${\beta}$-hydroxylase (DBH) inhibitor. $IC_{50}$ values of xanthoangelol to MAO-A and MAO-B were calculated to be 43.4 ${\mu}M$, and 43.9 ${\mu}M$. These values were very similar to iproniazid, which is a nonselective MAO inhibitor used as a drug against depression. The $IC_{50}$ values of iproniazid were 37 ${\mu}M$, and 42.5 ${\mu}M$ in our parallel examination. Moreover, $IC_{50}$ value of xanthoangelol to DBH was calculated 0.52 ${\mu}M$. 4-Hydroxyderricin is a potent selective MAO-B inhibitor and also mildly inhibits DBH activity. The $IC_{50}$ value of 4-hydroxyderricin to MAO-B was calculated to be 3.43 ${\mu}M$ and this value was higher than that of deprenyl (0.046 ${\mu}M$) used as a positive control for selective MAO-B inhibitor in our test. Cynaroside is a most potent DBH inhibitor. The $IC_{50}$ value of cynaroside to DBH was calculated at 0.0410 ${\mu}M$. Results of this study suggest that the two prenylated chalcones, xanthoangelol and 4-hydroxyderricin isolated from A. keiskei K., are expected for potent candidates for development of combined antidepressant drug. A. keiskei K. will be an excellent new bio-functional food material that has the combined antidepressant effect.
