Regulatory roles of NKT cells in Anaplasma phagocytophilum infection

  • Human granulocytic anaplasmosis (HGA) is caused by the obligate intracellular bacterium Anaplasma (A.) phagocytophilum. Natural killer T (NKT) cells are key players in host defense against various microbial infections. We investigated the role of NKT cells in immune response to A. phagocytophilum infection using NKT-knockout ($J\alpha$18-/-) mice. $J\alpha$18-/- and wild-type (WT) mice were infected with low-passage A. phagocytophilum and assayed for hepatic histopathology and cytokine production during 7 days post-infection. Compared to WT controls, the infected $J\alpha$18 -/- mice had much less histopathologic lesions and less apoptosis through day 7, and lower concentrations of ${IFN\gamma}$ and IL- 12, but not of IL-10. This result suggests that NKT cells are major components in the pathogenesis of HGA.

Serological evidence of Anaplasma spp., Borrelia burgdorferi and Ehrlichia canis in dogs from the Republic of Korea by rapid diagnostic test kits

  • Background: Emergent and re-emergent canine tick-borne infections are attracting increasing attention worldwide. The rise in pet ownership and the close relationship between dogs and their owners are the most concerning factors because dogs may act as competent reservoirs for human tick-transmitted infectious agents. Objectives: This study contributes to the epidemiological surveillance of canine tick-transmitted infections with zoonotic risk in the Republic of Korea (ROK) by investigating the seroprevalence of the pathogens, Anaplasma spp., Borrelia burgdorferi, and Ehrlichia canis. Methods: Four hundred and thirty whole blood samples from domestic dogs were collected in seven metropolitan cities and nine provinces in the ROK and tested using SensPERT Ab test kits (VetAll Laboratories®) to detect seroreactive animals. Results: The seroprevalence rates identified were 9.8% (42/430) for Anaplasma spp., 2.8% (12/430) for B. burgdorferi, and 1.4% (6/430) for E. canis. The risk factors evaluated in this study that could be associated with the development of a humoral immune response, such as sex, age, and history of tick exposure, were similar. There was only one exception for dogs seroreactive to Anaplasma spp., where the risk factor "tick exposure" was statistically significant (p = 0.047). Conclusions: This serological survey exhibited the widespread presence of Anaplasma spp., B. burgdorferi, and E. canis throughout the ROK. Hence, dogs may play a key role as the sentinel animals of multiple zoonotic infectious agents in the country.

  • 우리나라에서 사육되고 있는 재래산양과 호주로부터 수입한 산양에서 빈혈, 식욕감퇴, 높은 발병율과 사망율을 나타내는 괴질이 발생하여, 이에 대한 원인을 밝히기 위하여 전자현미경적으로 충체를 관찰하였던 바, 적혈구내 단일막으로 위요되어 있는 기본소체와 이중막으로 둘러싸여 있는 봉입체가 관찰되었기에 이 질병의 병원체를 Anaplasma ovis로서 동정 보고하는 바이다.

  • Anaplasma phagocytophilum의 주요 표면단백질인 Msp2 (p44)는 세균의 표면에 발현되는 주요한 항원 단백질이다. 본 실험에서는 A. phagocytophilum이 주요 숙주세포인 호중구나 HL-60 세포에 침입하는 데 있어, 세균의 주요 표면 단백질인 Msp2와 숙주세포의 표면에 발현되는 PSGL-1과의 반응 여부를 알아보았다. 그 결과, 재조합 단백질인 Msp2나 순수하게 분리된 A. phagocytophilum이 PSGL-1/FucT IV 유전자가 형질전환되어 PSGL-1이 발현되는 BJAB 세포와는 결합하지만, 순수한 BJAB 세포와는 전현 반응하지 않는 것을 관찰할 수 있었다(p<0.01 & p<0.01). 또한, 순수하게 분리된 A. phagocytophilum과 형질전환된(PSGL-1/FucT IV) BJAB 세포와의 결합이 Msp2의 단-클론항체나 Msp2 재조합 단백질의 농도에 따라 억제됨을 관찰할 수 있었다(p<0.05 & p<0.01). 따라서 A. phagocytophilum의 Msp2가 숙주세포인 호중구의 표면에 발현되는 PSGL-1과 직접적으로 결합하는 부착물질임을 알 수 있었다.

경북 북부지역의 한우에 대한 진드기매개성 병원체의 감염실태 조사 (A survey for tick-borne pathogens in Korean native cattle from northern area of Gyeongbuk)

  • This study was carried out to investigate the tick-borne pathogens from one hundred nineteen cattle farms (38 farms of Andong, 41 of Yeongju, 12 of Uiseong, 5 of Cheongsong, 5 of Yoengyang, 18 of Bonghwa) in northern areas of Gyeongbuk province by polymerase chain reaction (PCR). Among 119 cattle farms, the positive ratios against Babesia, Theileria, Anaplasma, Ehrlichia and Rickettsia were 3.4% (4/119), 10.1% (12/119), 6.7% (8/119), 1.7% (2/119) and 16.8% (20/119), respectively. Also, the PCR results revealed that 8 farms were positive for T. sergenti in positive of Theileria and 2 farms were positive for A. phagocytophilum in positive of Anaplasma. Therefore, further studies regarding vectors, environmental condition, interaction between domestic and wild animals and development of control program are needed to reduce the numbers of bovine tick-borne disease in northern areas of Gyeongbuk province.

PCR을 이용한 개의 아나플라즈마병 진단과 장기간 치료 (Diagnosis of Anaplasmosis by PCR and Long Term Medication in a Dog)

  • A 4-year-old female Shunauzer dog was referred to the Veterinary Teaching Hospital of Chungbuk National University due to anorexia and depression. The dog had a history of regular walking on grass fields, weight loss, and hyperthermia $(40.6^{\circ}C)$. In the physical examination, lymph node enlargement was confirmed. Complete blood count result revealed leukocytosis and thrombocytopenia but there was no decreasing of red blood cells. On blood chemistry, serum ALP, GGT, CPK, and LDH were elevated. Abdominal radiograph showed splenomegaly. Anaplasma platys infection was suspected with inclusion body-like substances in platelets on blood smear. Anaplasma platys was confirmed by PCR. On the basis of laboratory examination, final diagnosis was anaplasmosis. Treatment was followed for 3 months with tetracycline and doxycycline. The patient was monitored every week during the treatment. The patient has recovered to normal condition without any clinical signs. We are going to emphasize the need of PCR technique in diagnosis and to report the possibility of long term treatment more than two months in rickettial disease.

Prevalence of Anaplasma and Bartonella spp. in Ticks Collected from Korean Water Deer (Hydropotes inermis argyropus)

  • Deer serve as reservoirs of tick-borne pathogens that impact on medical and veterinary health worldwide. In the Republic of Korea, the population of Korean water deer (KWD, Hydropotes inermis argyropus) has greatly increased from 1982 to 2011, in part, as a result of reforestation programs established following the Korean War when much of the land was barren of trees. Eighty seven Haemaphysalis flava, 228 Haemaphysalis longicornis, 8 Ixodes nipponensis, and 40 Ixodes persulcatus (21 larvae, 114 nymphs, and 228 adults) were collected from 27 out of 70 KWD. A total of 89/363 ticks (266 pools, 24.5% minimum infection rate) and 5 (1.4%) fed ticks were positive for Anaplasma phagocytophilum using nested PCR targeting the 16S rRNA and groEL genes, respectively. The 16S rRNA gene fragment sequences of 88/89 (98.9%) of positive samples for A. phagocytophilum corresponded to previously described gene sequences from KWD spleen tissues. The 16S rRNA gene fragment sequences of 20/363 (5.5%) of the ticks were positive for A. bovis and were identical to previously reported sequences. Using the ITS specific nested PCR, 11/363 (3.0%) of the ticks were positive for Bartonella spp. This is the first report of Anaplasma and Bartonella spp. detected in ticks collected from KWD, suggesting that ticks are vectors of Anaplasma and Bartonella spp. between reservoir hosts in natural surroundings.

한국에서 사육되는 고양이, 말, 소로부터 Anaplasma phagocytophilum과 Ehrlichia chaffeensis에 대한 항체 검출 (Detection of Antibodies Reacting with Anaplasma phagocytophilum and Ehrlichia chaffeensis from Cats, Horses and Cattle in Korea)

  • 국내 전주, 광주 그리고 제주에서 고양이, 더러브렛 말, 홀스테인 소로부터 진드기 매개성 인수공통 병원체인 Anaplasma phagocytophilum과 Ehrlichia chaffeensis에 대한 항체가 조사를 위하여 면역형광항체법을 이용하였다. 본 연구를 위하여 254마리의 혈청(33마리의 애완 및 길거리 고양이, 92마리의 방목장 말 그리고 129마리의 방목장소)을 수집하였다. 33마리의 고양이 중에서 6마리가 A. phagocytophilum(titer $\geq$ 80)에 대한 양성항체가 검출되어 18.2%의 양성율을 나타내었으며, 1마리에 있어서는 E. chaffeensis에 대한 항체가 검출되어 3%의 양성율을 나타내었다. 소에 있어서는 129마리 중에서 1마리에서 그리고 말에 있어서는 92마리 중에서 2마리에서 A. phagocytophilum에 대한 양성항체가 검출되어 각각 0.8%와 2.2%의 양성율을 나타내었으나 E. chaffeensis에 대한 항체는 모두 음성 결과를 나타내었다. 이 결과는 국내 사육 고양이, 말 그리고 소에 있어서 A. phagocytophilum과 E. chaffeensis에 대한 혈청학적 양성결과로서 국내에서 처음 보고되는 자연감염 예이며, 진드기 서식지역의 방목동물에서는 이들 질병의 감염으로 경제성 손실이 우려되며 예방대책이 마련되어야 할 것으로 판단된다.
