Anabaena koreana sp. nov. (Cyanophyceae), a new species, and new record of fresh-water blue-green algae from Korea

  • The present study summarizes the occurrence, distribution and autecology of 12 taxa of the class Cyanophyceae collected from several swamps, reservoir and highland wetlands in South Korea from 2009 to 2012. A new species, Anabaena koreana sp. nov. and 11 taxa of blue-green algae newly recorded are described and illustrated. Anabaena koreana is similar to A. oumiana, A. spiroides and A. crassa in that the trichomes form regular coils. However, A. koreana is distinguished from these three species by the morphological characteristics of the vegetative cell, heterocyst, and akinet shape and size. This study considers 12 blue-green algal species, including a new species, Anabaena koreana sp. nov. and 11 species that are recorded for the first time in the Korean freshwater algal flora. Among them, the genus Nostochopsis Wood ex Bornet et Flahault 1886 had not previously been recorded in Korea.

Temperature effects on the growth and morphology of Anabaena sp.: lab-scale investigation and onsite validation

  • This study presents the characteristics of growth and morphology of Anabaena sp., a representative filamentous cyanobacterium, depending on temperature variation from 10 to 30 ℃. Both the filament density (or number) and its length of Anabaena were highly affected by temperature, as well as growth stage. Rapid growth at a higher temperature led to an increase in Anabaena filament density, as well as optical density at 680 nm (OD680). However, the number of vegetative cells within a single filament of Anabaena grown at 30 ℃ was smaller than those grown at lower temperatures, due to the intercalary division of the filament. Of the three different cells comprising a single Anabaena filament, the vegetative cell marginally affects the growth of Anabaena. The main dimensions of the vegetative cell, i.e., length and width, depend on the temperature and growth stage. The length-to-width (L/W) ratios of vegetative cells and akinetes were relatively consistent regardless of the temperature. However, in vegetative cells with dichotomous growth, the L/W ratio shows clear differences depending on their growth stage. It has been demonstrated that the L/W ratio could be used as an indicator to indirectly predict the growth stage of on-sit Anabaena samples.

대청호에서 사상 남조류 포식성 Asterocaelum sp.(Protozoan)의 발생에 관한 연구 (A study on Asterocaelum sp., a grazer of filamentous cyanobacteria in the Lake Daecheong)

  • A milky gray scum was observed at some water area of the lake Daecheong in the summer of 1997 and 1998. To identify a causing organism of scum and affecting factors, we observed the scum material by a phase contrast microscope and surveyed the phsico-chemical water quality during the outbreak of scum. The scum was found out to be clogging cysts of amoeboid protozoan, Asterocaelum sp.(Protozoea Sarcodina Aconchulinida), grazer of filamentous cyanobacteria, Anabaena genus. The protozoan scum appeared during Anabaena bloom period, which continued for a while. This protozoan was presumed acting as a regulator of Anabaena bloom in the lake Daecheong during the summer season. Moreover this is the first report on Asterocaelum sp. a grazing filamentous cyanobacteria occurred in Korean freshwater.

소규모 농업용 전대저수지의 수환경 변화와 조류발생 특성 (Characteristics of Aquatic Environment and Algal Bloom in a Small-scaled Agricultural Reservoir (Jundae Reservoir))

  • 축산과 농경배수가 주요 오염원인 소규모 농업용 저수지, 전대저수지에서 2011년 3월부터 2013년 10월까지의 모니터링 결과를 바탕으로 수환경 변화와 조류발생 특성을 분석함으로써, 부영양화된 소형저수지에서의 수환경과 조류발생 (Algal bloom)과의 관계를 규명하고자 하였다. 2013년의 경우 전년에 비해 강우가 적은 사실을 제외하면 기상의 큰 변화는 없었다. 전대저수지의 TN, TP등 영양염류농도는 농업용수 수질기준을 초과하고 있으며 발생원인이 상이하였다. 조류발생과 수환경요인간의 상관관계가 많은 항목에서 중상관($r{\geq}0.25$)의 관계를 보여주어 부영양 호소에서 COD 발생과 조류발생이 매우 복합적으로 이루어짐을 짐작케 했다. 전대저수지에서 출현한 식물플랑크톤은 총 6과 40속 62종이었으며, 현존량은 $1.3{\times}10^4{\sim}2.8{\times}10^6$ cells $mL^{-1}$로 TN, TP의 농도 감소와 함께 식물플랑크톤의 현존량도 연차별로 점차 감소하였다. 주요 우점종은 남조류인 Anabaena sp., Oscillatoria sp., Microcystis sp. 등이었으며, Oscillatoria sp.의 우점빈도가 가장 높았다. 수온이 증가하면서 Anabaena sp., Microcystis sp., Oscillatoria sp. 순으로 우점종이 변화하였으며, TN이 높은 경우 Anabaena sp., TP가 높은 경우 Microcystis sp.가, TN과 TP가 낮은 경우 Oscillatoria sp.가 우점하였다. N/P ratio는 Anabaena sp. 우점시 가장 높았고, Oscillatoria sp. 우점시 가장 낮았다. Chl-a 농도는 Oscillatoria sp. 우점시 높게 나타나고, 식물플랑크톤 현존량은 Anabaena sp. 우점시 높게 나타났으며, Microcystis sp. 우점시에는 Chl-a와 현존량 모두 낮았다. 이를 통해, 오염이 심각한 소형저수지에서는 Microcystis sp.보다 Anabaena sp.와 Oscillatoria sp.에 의한 대발생이 주로 문제가 됨을 알 수 있었다. Anabaena sp.가 우점할 때는 수환경 요인간의 상관계수가 높아 예측과 관리에 활용이 가능한 반면 Oscillatoria sp. 우점시에는 Chl-a 상관성이 낮게 나타나는 등, 조류대발생을 신속하게 예측하고, 예방, 관리하기 위해서는 조류발생 환경과 원인종의 관계를 규명하고 이를 활용하는 방안이 필요할 것으로 사료된다.

Effects of Environmental Factors on Cyanobacterial Production of Odorous Compounds: Geosmin and 2-Methylisoborneol

  • Geosmin and 2-methylisoborneol (2-MIB), responsible for earthy or musty smell, are a major concern for safe drinking water supplies. This study investigated the effects of environmental factors on odorous compound production and cell growth in cyanobacterial strains. Anabaena sp. FACHB-1384, a 2-MIB producer, was sensitive to low temperature (<$20^{\circ}C$). However, geosmin producers, Anabaena sp. Chusori and Anabaena sp. NIER, were sensitive to high light intensity (>$100{\mu}mol/m^2/sec$), but not to low temperature. Geosmin concentrations increased under higher nitrate concentrations, being linearly proportional to cell density. A P-limited chemostat showed that P-stress decreased the geosmin productivity and extracellular geosmin amount per cell in Anabaena sp. NIER. However, only 2-MIB productivity was reduced in Planktothrix sp. FACHB-1374 under P-limitation. The extracellular 2-MIB amount per cell remained constant at all dilution rates. In conclusion, high light intensity and P-stress can contribute to the lower incidence of geosmin, whereas 2-MIB reduction could be attainable at a lower temperature.

Anabaena cylindrica 분해세균의 분리 및 동정 (Isolation and Identification of Bacteria Lysing Anabaena cylindrica)

  • 남조류 분해세균을 분리하기 위하여 도창, 팔당 저수지와 석촌 호수로부터 저층시료를 채취하였다. Anabaena cylindrica lawn에 각각의 시료를 도말한 후 11 일간 배양하였고 석촌 시료를 접중한 Anabaena cylindrica lawn에서 Anabaena cylindrica에 분해능을 가지는 세균을 분리하였다. 분해능 화인은 남조류와 분해세균을 혼합한 후 chlorophyll a간의 측정과 분리세균의 cell counting방법으로 확인하였고, 세포외 물질이 Anabaena cylindrica를 분해하는 것을 확인하였다. 분리균주는 16S rDNA 염기서열 분석과 형태학적, 생리학적 특징을 기초로 하여 동정하였다. 분리균주의 165 rDNA염기서열을 기초로한 분자계통학적 분석을 통하여 Bacillus속에 포함하는 계통학적 그룹에 속한다는 것을 확인하였고, Bacillus sp. CHSl으로 명하였다.

Moraxella sp. CK-1의 세포외 Autolysin의 분리 정제 및 활성도 측정 (Purification and Assay of Extracellular Autolysin from Moraxella sp. CK-l)

  • Moraxella sp. CK-1는 남조류 Anabaena cylindrica의 생장을 억제한다고 알려져 있다. Moraxella sp. CK-1의 세포외 autolysin의 분리는 다음과 같은 방법으로 분리를 시도하였다. 흡광도 660 nm에서 0.7~0.8이 되도록 BGC-11세포배양액에서 키우고, 원심분리로 균주인 Moraxella sp. CK-1를 제거한 뒤 세포배양액을 Amicon ultrafiltration으로 농축을 하였다. 농축한 세포배양액을 $(NH_{4})_{2}SO_{4}$ 로 0~20%, 20~40%, 40~60%, 60~80%로 분획하여 단백질을 14,000${\times}$g로 침전 시켰다. 침전된 단백질을 20 mM Tris-HC1, pH 8.0완충용액으로 현탁시킨 뒤, 동일 완충용액을 이용하여 투석을 하였다. $(NH_{4})_{2}SO_{4}$로 분획한 뒤 활성확인을 위해 Anabaena Cylindrica가 도포된 평판배지에서 활성을 확인한 결과, 40~60%, 60~80%에서 활성이 있음을 확인하였다. 이들을 각각 Mono-$Q^{TM}$ HR 5/5 (column volume 1 ml) column을 이용하여 FPLC에서 단백질을 분리하였다. $(NH_{4})_{2}SO_{4}$ 로 분획한 40~60%에서는 major peak 3개, 60~80% 분획에서는 2개의 major peak가 분리되었다. Mono-$Q^{TM}$ HR 5/5 column에서 분리되어 나온 major peak 5개를 투석한 뒤 Anabaena cylindrica lawn에서 활성을 확인하였으나 확인이 되지 않았다. PVDF membrane을 이용하여 transfer하여 40~60% 침전에서 17 kDa의 단백질을 얻어냈다. 이를 N-terminal amino acid sequence를 하여 serine pretense의 계열임을 확인하였다.

Biotransformation of Aldrin and Chlorpyrifos-methyl by Anabaena sp. PCC 7120

  • A cyanobacteria species, Anabaena sp. PCC 7120, was tested to assess its biotransformation ability on two widely used insecticides, aldrin and chlorpyrifos-methyl, in the culture medium. The blue-green alga metabolized aldrin mainly to dieldrin by an epoxidation reaction with the participation of cytochrome P450-dependent monooxygenase in the cyanobacteria. The blue-green alga also produced chlorpyrifosmethyl oxon as a primary metabolite from chlorpyrifos-methyl via a desulfuration reaction, presumably conducted by cytochrome P450-dependent monooxygenase. Therefore, two insecticides might be possibly dissipated by cytochrome P450-dependent monooxygenases in the blue-green algae in the contaminated environments.

Enzyme Profiles of Alga-Lytic Bacterial Strain AK-13 Related with Elimination of Cyanobacterium Anabaena cylindrica

  • To investigate bacteria with algalytic activities against Anabaena cylindrica when water blooming occurs and to study enzyme profiles associated with alga-lytic activity, various bacterial strains were isolated from surface waters and sediments in eutrophic lakes or reservoirs in Korea. Among 178 isolates, only nine isolates exhibited lytic abilities against A cylindrica on the agar plates, and then the isolate AK-13 was selected as the strongest in lysing the cyanobacterium A. cytindrica. The strain AK-13 was characterized and identified as Sinorhizobium sp. based on fatty acid methyl ether profiles and 16S rDNA sequence. According to the results of the enzyme assays, in the strain An-13 of Sinorhizobium sp., alginase, amylase, proteinase (caseinase and gelatinase), carboxymethyl-cellulase (CMCase), laminarinase, and lipase was produced, namely CMCase, laminarinase and protease were highly active. None of glycosidase was produced. Therefore, enzyme systems of Sinorhizobium sp. AK-13 were very complex to degrade cell walls of A. cylindrica. The peptidoglycans of A. cylindrica mat be hydrolyzed and metabolized to a range of easily utilizable monosaccharides or other low molecular weight organic substances by Sinorhizobium sp. AK-13.

Iron Starvation-Induced Proteomic Changes in Anabaena (Nostoc) sp. PCC 7120: Exploring Survival Strategy

  • This study provides first-hand proteomic data on the survival strategy of Anabaena sp. PCC 7120 when subjected to long-term iron-starvation conditions. 2D-gel electrophoresis followed by MALDI-TOF/MS analysis of iron-deficient Anabaena revealed significant and reproducible alterations in ten proteins, of which six are associated with photosynthesis and respiration, three with the antioxidative defense system, and the last, hypothetical protein all1861, conceivably connected with iron homeostasis. Iron-starved Anabaena registered a reduction in growth, photosynthetic pigments, PSI, PSII, whole-chain electron transport, carbon and nitrogen fixation, and ATP and NADPH content. The kinetics of hypothetical protein all1861 expression, with no change in expression until day 3, maximum expression on the $7^{th}$ day, and a decline in expression from the $15^{th}$ day onward, coupled with in silico analysis, suggested its role in iron sequestration and homeostasis. Interestingly, the up-regulated FBP-aldolase, Mn/Fe-SOD, and all1861 all appear to assist the survival of Anabeana subjected to iron-starvation conditions. Furthermore, the $N_2$-fixation capabilities of the iron-starved Anabaena encourage us to recommend its application as a biofertilizer, particularly in iron-limited paddy soils.
