Tyrosinase Inhibition and Mutagenicity of Phenolic Compounds from Mulberry Leaves - Research Note -

  • The tyrosinase inhibition activity and mutagenicity as assessed by the Ames test on phenolic antioxidants (5-Caffeoyl quinic acid, 3,4-Dihydroxy cinnamic acid, Quercetin 3-O-${\beta}$-D-glucopyranose, Kaempferol 3-O-${\beta}$- D-glucopyranose) and the ethyl acetate fraction isolated from mulberry leaves were examined. The ethyl acetate fraction and chlorogenic acid exhibited weaker tyrosinase inhibitory activities than kojic acid. In addition, the ethyl acetate fraction from mulberry leaves, containing phenolic antioxidants, showed no mutagenicity by the Ames test.

  • Mutagenic acivity of essential oils was tested using Salmonella typhimurium TA100 in the presence or absence of 59 fraction prepared from the mouse liver. Growth inhibitory effect of the oils on the bacteria was measured to warrant the mutagenic effect. Most oils were (round to be very strongly toxic against the bacteria at a high dose $(2,000{\mu}g/plate)$. At lower doses than this concentration, the Curcuma longa oil was found to be the most mutagenic with S9 fraction whereas it was not mutagenic without the fraction suggesting that this oil could undergo activation for the mutagenicity by cytochrome P45O. However, the mutagenicity of the Eugenia caryohpylata oil was disappeared under S9 fraction. Other oils obtained from Cinnamomum cassia, Chrysanthemum sibiricum, Paeonia moutan the flower of Artemisia princeps var. Orientalis, Allium sativum, were not mutagenic. This result suggested that antimutagenicity assay on the essential oil is necessary for the biological available substances.

야생버섯 추출물의 항돌연변이 효과 (Antimutagenic Effect of Extracts from Wild Mushrooms)

  • 천연물질로부터 항돌연변이원성 물질 및 항암 물질 탐색을 위한 연구의 일환으로 11종의 국내산 야생 버섯류로 부터 메탄올 추출물을 제조한 후 Ames test를 이용하여 돌연변이 억제효과를 검토하였다. 실험에 사용한 농도에서 털이버섯(Lactarius piperatus), 노란다발독버섯(Naematolona fasciculare) 및 금빛시루뻔버섯(Innotus xeran- ticus)의 메탄올 추출물은 Salminella typhimurium Ta98 및 TA100 균주에서 추출물의 농도에 관계없이 간접변이원인 B(a)P에 대하여 40~80% 정도의 항돌연변이 효과를 나타내었으며, 직접변이원인 MNNG에 대해서는 60~80% 정도의 비교적 높은 돌연변이 억제효과를 나타내었다.

음용수의 변이원성에 관한 조사연구 (Study on the Mutagenicity of Drinking Water)

  • This is a study on the risk assessment of drinking water using mutagenicity testing. The tests have been carried with the raw water, treated water, and drinking water (tap water) in Kwangju and Mokpo areas. The Ames preincubation test was carried concentrating samples using by Sep-Pak PLUS cartriges in Salmonella typhimurium TA100 and TA98. The samples were tested with several chemical water quality analysis. The THMs have not been measured in raw water, but measured treated water and tap water at a value of 7.135-12.473 $\mu$g/l. It was observed that the number of revertants colonies increased in treated water and tap water on TA100 without S9 and showed weak mutagenicity on TA98 without S9. Indirect mutation was not seen in TA100 and TA98 with S9. The results indicated that formed substances of treatment process's of water that increased mutagenicity.

  • The three medicinal herbs-Curcuma longa Linne, Paeonia japonica Miyabe, Scutellaria baikalensis George-irradiated with gamma rays have been tested for their possible genotoxicity. The methanol-soluble and water-soluble fractions of the 10kGy gamma-iradiated herbs were examined in the Salmonella typhimurium histidine reversion assy(Ames test) using S. typhimurium TA98, TA100 and TA102 as tester strains. No mutabenicity was detected in this assay with or without metabolic activation. The safety of the herbs irradiated with gamma rays at practical doses needs to be evaluated in further tests of genotoxicity in vivo and chronic and reproductive toxicity.

  • Antimutagenic effect of chungkookjangs prepared with the different variety of soybean and starter were studied against aflatoxin B$_1$ (AFB1) using Ames test and Ν-methyl-Ν'-nitro-Ν- nitrosoguanidine (MNNG) using SOS chromotest. 게르마늄 복합물인 STB-HO-BM에 대한 유전독성에 관한 연구 (Genotoxicity Studies of STB-HO-BM, a Germanium Complex)

  • We have investigated the genotoxicity of STB-HO-BM using in vitro and in vivo system such as Ames reverse mutation test, chromosomal aberration test and micronucleus test. in Ames reverse mutation test, STB-HO-BM treatment at the dose range up to 5,000 ug/plate did not induce mutagenicity in Salmonella typhimurium TA98, TA100, TA102, TA1535, TA 1537 and in Escherichia coli WP2 uvrA with and without metabolic activation. Any significant aberration wasn't observed in chinese hamster lung (CHL) fibroblast cells treated with STB-HO-BM at the concentration of 12.5, 2.5, 5 mg/ml both in the absense and presence of metabolic activation system. In mouse micrnucleus test, no significant increase in the occurrence of micronucleated polychromatic erythrocytes was observed in ICR male mice orally administered with STB-HO-BM at the doses of 0.5, 1.0, 2.0 g/kg. These results indicate that STB-HO-BM has no mutagenic potential under the condition in this study.

  • AG6O, the complex of acriflavine and guanosine, has been shown to possess the synergistic antitumorigenic activity in the previous paper (J. Pharm. Pharmacol. 1997, 49:216). In this study, we have investigated the genotoxic properties of AG60 using in vitro and in vivo system such as Ames bacterial reversion test, chromosomal aberration assay and micronucleus assay. In Ames reverse mutation test, AG60 treatment at the dose range up to 250 $\mu\textrm{g}$/plate caused the dose-independent random induction of the mutagenic colony formation in S. typhimurium TA98, TA100, TA1537, and E. coli WP2uvrA, while any mutagenic effect of AG60 wasn't observed in S. typhimurium TA1535. Any significant chromosomal aberration wasn't observed in chinese hamster lung (CHL) fibroblast cells incubated with PBS or AG60 at the concentrations of 2.5, 5, 10 $\mu\textrm{g}$/$m\ell$ for 24 hours without but even with 59 metabolic activation system for 6 hours. In vivo ICR mice, the intramuscular injection of AG60 at the doses of 7.15, 14.3, and 28.6 mg/kg did not induce the frequency of micronucleus formation. However, mitomycin C, as one of the positive controls at the dose of 2 mg/kg caused the 8.4% induction in the frequency of micronucleus and 24% increase in the chromosomal aberration.

새로운 플라보노이드 유도체인 DA-6034에 대한 유전독성에 관한 연구 (Genotoxicity Studies of DA-6034, a New Flavonoid Derivative)

  • Inflammatory bowel disease (IBD) is a multifactorial disorder with unknown etiology and pathogenesis. Eupatilin, a kind of flavonoids, has been known to be effective for chronic diarrhea in Korea. In this study, we have investigated the genotoxicity of DA-6034, a new synthetic derivative of Eupatilin, wing in vitro and in viuo system such as Ames reverse mutation test, chromosomal aberration test and micronucleus test. in Ames reverse mutation test, DA-6034 treatment at the dose range up to 5,000 $\mu\textrm{g}$/ plate did not induced mutagenicity in Salmonella typhimurium TA98, TA100, TA102, TA1535, TA1537 with and without metabolic activation. Any significant aberration wasn't observed in chinese hamster lung(CHL) fibroblast cells treated with DA-6034 at the concentration of 5, 2.5, 1.25 mg/ml both in the absence and presence of metabolic activation system. In mouse micronucleus test, no significant increase in the occurrence of micronucleated polychromatic erythrocytes was observed in ICR male mice orally administered with DA-6034 of the doses of 2.0, 1.0, 0.5 g/kg. These results indicate that DA-6034 has no mutagenic potential under the condition in this study.

Lack of Mutagenicity Potential of Periploca sepium Bge. in Bacterial Reverse Mutation (Ames) Test, Chromosomal Aberration and Micronucleus Test in Mice

  • Objectives: The root barks of Periploca sepium Bge. (P. sepium) has been used in traditional Chinese medicine for healing wounds and treating rheumatoid arthritis. However, toxicity in high-doses was often diagnosed by the presence of many glycosides. The potential mutagenicity of P. sepium was investigated both in vitro and in vivo. Methods: This was examined by the bacterial reverse mutation (Ames) test using Escherichia coli WP2uvrA and Salmonella typhimurium strains, such as TA98, TA100, TA1535, and TA1537. Chromosomal aberrations were investigated using Chinese hamster lung cells, and the micronucleus test using mice. Results: P. sepium did not induce mutagenicity in the bacterial test or chromosomal aberrations in Chinese hamster lung cells, although metabolic activation and micronucleated polychromatic erythrocytes were seen in the mice bone marrow cells. Conclusions: Considering these results, it is suggested that P. sepium does not have mutagenic potential under the conditions examined in each study.
