In this study, oxidized alginate/gelatin/biphase calcium phosphate (BCP)- based hydrogel composites were fabricated. Alginate sodium was oxidized by periodate. The oxidized product was confirmed by using $^1H$ and $^{13}C$ NMR spectra. The number average molecular weight ($M_n$), the average molecular weight ($M_w$) of the oxidized alginate were determined by Gel Permeation Chromatography (GPC). The hydrogel was formed from the oxidized alginate and gelatin solution via Schift-base reaction. The hydrogel showed a highly porosity by a Scanning Electron Microscope (SEM) and Mercury Intrusion Porosimetry (MIP). Crosslinked density of the gel matrix were assessd by trinitrobenzene sulfonic acid (TNBS) assay that shows a high effect on swelling ratio. Increment of the crosslinked desity resulted in enhancing compressive strength of the hydrogel composite. The cytotoxity of hydrogel was assessed with osteoblast MG-63. The hydrogel composites show a high compatibility. The obtained results showed a potential application for bone regeneration in future.
The alginate-based hydrogel was prepared as a pH-sensitive drug delivery system. To enhance the drug loading capacity, activated carbon was introduced as a drug absorbent. The iron oxide was incorporated into the alginate matrix for the magnetic transferring to the target organ. The activated carbon and iron-oxide were dispersed uniformly in the alginate hydrogel. The drug release from the alginate/activated carbon composite hydrogel was carried out in various pH conditions with vitamin B12 and Lactobacillus lamnosers as model drugs. The fast and sustainable release of drug was observed in the basic condition due to the pH-sensitive solubility of alginate. The novel drug delivery system having pH-sensitive release property and magnetic movement to target place was developed by using the alginate/activated carbon composite magnetic hydrogels.
In this study, a co-axial flow induced microfluidic chip to fabricate pure collagen type I microfiber via the control of collagen type I and Na-alginate gelation process. The pure collagen type I microfiber was generated by selective degradation of Ca-alginate from 'Core-Shell' structured hydrogel microfiber. To make 'Core-Shell' structure, collagen type I solution was introduced into core channel and 1.5% Na-alginate solution was injected into side channel in microfluidic chip. To evaluatethe 'Core-Shell' structure, the red and green fluorescence substances were mixed into collagen type I and Na-alginate solution, respectively. The fluorescence substances were uniformly loaded into each fiber, and the different fluorescence images were dependent on their location. By immoblizing EpH4-Ras and C6 cells within collagen type I and Na-alginate solution, we sucessfully demonstrated the co-culture of EpH4-Ras and C6 cells with 'Core-Shell' like hydrogel microfiber for 5 days. Only to produce pure collagen type I hydrogel fiber, tri-sodium citrate solution was used to dissolve the shell-like Ca-alginate hydrogel fiber from 'Core-Shell' structured hydrogel microfiber, which is an excellent advantage when the fiber is employed in three-dimensional scaffold. This novel method could apply various application in tissue engineering and biomedical engineering.

Natural cellulose hydrogel membrane cannot be directly used for cell encapsulation because it has many large pores on the surface that immune biomolecules are able to penetrate into easily. For the reason, alginate was used for the control of pore size of the cellulose hydrogel membrane. The surface morphology of cellulose/alginate nanocomposite confirmed the successful control of the porosity of the membrane. The permeability of the cellulose/alginate nanocomposite was decreased but mechanical properties were increased compared with the bacterial cellulose membrane. The cellulose/alginate nanocomposite could be used for the functional membrane as a promising biomedical material in the future.

Initial burn wound care is one of the important factors in the outcome of burn treatment. In this study, we tried to develop spray-type alginate hydrogel dressing with advantages such as promoting wound healing, reducing pain, and increasing ease of use for emergency burn treatment. Spray implementation, physical properties, and cytotoxicity of the newly developed spray-type alginate hydrogel dressing were evaluated. As a result, a new functional spray-type hydrogel dressing with excellent physical properties and biocompatibility was developed along with the development of spray able containers, and it was confirmed that it could be applied as a treatment for skin regeneration in the future.
Comb-type graft hydrogels composed of alginate and poly(N-isopropylacrylamide) (PNIPAAm) were prepared to manifest rapid temperature and pH sensitivity. To appear the electro-sensitivity, the polyaniline, conducting polymer, was added into the matrix. The swelling kinetics and ratios were compared under the various compositions of polyaniline. The swelling behaviors revealed that conducting polymer/hydrogel composites could control the swelling ratio and kinetics. The addition of polyaniline in the matrix improved the thermal stability in comparison with that of the hydrogel without polyaniline. In temperature sensitivity, the adding the polyaniline into the matrix decreased the degree of change in the swelling ratio. The swelling ratios continuously increased with increasing pH values. The drug release rate from the hydrogel increased with the adding the polyaniline and the applying the direct voltage to the hydrogels.
Biocompatible hydrogels from 2-hydroxyethyl methacrylate (HEMA) monomer containing various amount of alginate in the presence and absence of hydrophilic methacrylic acid (MAA) were synthesized in order for biomedical application. The antimicrobial effect and interaction with proteins for hydrogels were investigated in this study. MAA was introduced because it was expected to increase the amount of water content in the polymer which is an important factor for biocompatibility, and alginate was expected to enhance the antimicrobial activity. The antimicrobial effect against S. aureus and E. coli increased for all hydrogels as the amount of alginate and MAA contained. Presence of MAA further enhances the antimicrobial effect. Amount of adsorption of bovine serum albumin (BSA) increased with increasing concentration of alginate whether MAA was present or not. Contrarily, the amount of lysozyme was not affected with increasing alginate concentration in the absence of MAA, while it decreased in the presence of MAA.
In this study, a series of highly swelling hydrogels based on sodium alginate (NaAlg) and polymethacrylamide (PMAM) was prepared through free radical polymerization. The graft copolymerization reaction was performed in a homogeneous medium and in the presence of ammonium persulfate (APS) as an initiator and N,N'-methylenebisacrylamide (MBA) as a crosslinker. The crosslinked graft copolymer, alginate-graft-polymethacrylamide (Alg-gPMAM), was then partially hydrolyzed by NaOH solution to yield a hydrogel, hydrolyzed alginate-graft-polymethacrylamide (H-Alg-g-PMAM). During alkaline hydrolysis, the carboxamide groups of Alg-g-PMAM were converted into hydrophilic carboxylate anions. Either the Alg-g-PMAM or the H-Alg-g-PMAM was characterized by FTIR spectroscopy. The effects of the grafting variables (i.e., concentration of MBA, MAM, and APS) and the alkaline hydrolysis conditions (i.e., NaOH concentration, hydrolysis time, and temperature) were optimized systematically to achieve a hydrogel having the maximum swelling capacity. Measurements of the absorbency in various aqueous salt solutions indicated that the swelling capacity decreased upon increasing the ionic strength of the swelling medium. This behavior could be attributed to a charge screening effect for monovalent cations, as well as ionic cross-linking for multivalent cations. Because of the high swelling capacity in salt solutions, however, the hydrogels might be considered as anti-salt superabsorbents. The swelling behavior of the superabsorbing hydrogels was also measured in solutions having values of pH ranging from 1 to 13. Furthermore, the pH reversibility and on/off switching behavior, measured at pH 2.0 and 8.0, suggested that the synthesized hydrogels were excellent candidates for the controlled delivery of bioactive agents. Finally, we performed preliminary investigations of the swelling kinetics of the synthesized hydrogels at various particle sizes.
Polyvinyl alcohol/alginate hydrogel beads containing Mg-Al layered double hydroxide (LDH-PVA/alginate beads) were synthesized for phosphate removal. Results showed that blending PVA with the LDH-alginate beads significantly improved their stability in a phosphate solution. The kinetic reaction in LDH-PVA/alginate beads reached equilibrium at 12 hr-post reaction with 99.2% removal. The amount of phosphate removed at equilibrium ($q_e$) was determined to be 0.389 mgP/g. The equilibrium data were described well by the Freundlich isotherm with the distribution coefficient ($K_F$, 0.638) and the constant (n, 0.396). Phosphate removal in LDH-PVA/alginate beads was not sensitive to solution pH. Also, the removal capacity of LDH-PVA/alginate beads ($q_e$, 1.543 mgP/g) was two orders of magnitude greater than that of PVA/alginate beads ($q_e$, 0.016 mgP/g) in column experiments. This study demonstrates that LDH-PVA/alginate beads with a higher chemical stability against phosphate compared to LDH-alginate beads have the potential for phosphate removal as adsorptive media.
Purpose: Wogonin is an active component isolated from scutellariae radix. This study was conducted to evaluate the inhibitory effect of wogonin mixed with hydrogel on osteoclast differentiation. Methods: It was performed to estimate cytotoxicity of Wogonin alginate hydrogel disk(WHD) in BMMs stimulated with RANKL, M-CSF. ROS synthesis and actin ring formation were analysed to observe the effect of WHD. Results: WHD has no cytotoxicity at the concentration of 0.1 ${\mu}g/ml$ or lower. 0.1 ${\mu}g/ml$, 1 ${\mu}g/ml$ WHD restrained the synthesis of ROS and 0.1 ${\mu}g/ml$, 1 ${\mu}g/ml$ WHD restrained the formation of actin ring. Conclusions: WHD has the inhibitory effect of osteoclast differentiation and bone resorption. Further studies are needed to treat osteoporosis by herbal medicine.
