Enhanced Solubility and In vitro Skin Permeation of Lovastatin Using Some Vehicles and Penetration Enhancers (수종 용제와 투과 촉진제를 이용한 로바스타틴의 용해성 및 피부 투과 증진)

  • To enhance the in vitro permeation of lovastatin through excised hairless mouse and human cadaver skins, solubility was determined in various hydrophilic and lipophilic vehicles, and the effects of vehicles and penetration enhancers on the skin permeation from solution formulations were investigated. Solubility of lovastatin was highest in N-methyl-2-pyrrolidone (NMP) ($278.2{\pm}10.1$ mg/ml) and dimethyl sulfoxide (DMSO) ($162.2{\pm}9.7$ mg/ml). Among different pure vehicles used, NMP, DMSO, propylene glycol and isopropyl myristate provided some drug permeation ($6.9{\pm}1.1$, $5.9{\pm}1.6$, $3.0{\pm}0.5$ and $2.2{\pm}0.3{\mu}g/cm^2$ at 24 hr, respectively) through hairless mouse skin. The addition of oleic acid, linoleic acid and oleyl alcohol to DMSO showed the maximum permeation at around 5 v/v%, however, capric acid and caprylic acid had no enhancing effect. The increase of enhancer concentrations showed bell-shaped permeation rate, suggesting the presence of optimal concentration in lovastatin penetration. Increasing donor concentration from 10 mg/ml to 80 mg/ml in DMSO and a cosolvent of DMSO, NMP and DGME (3 : 3 : 4 v/v) did not show significant dose dependent permeation in both hairless mouse and human cadaver skins. The maximum lovastatin flux through human cadaver skin was found to be $0.87{\pm}0.46{\mu}g/cm^2$/hr with 5 v/v% linoleic acid and donor dose of 4 mg/0.64 $cm^2$ in the cosolvent. These results suggest that transdermal delivery of lovastatin would be feasible by establishing the optimal concentrations of donor dose and unsaturated fatty acids in appropriate vehicles.

Effects of Passivation Thin Films on the Optical Properties of the Green Organic Light Emitting Diodes (페시베이션 박막이 녹색 유기발광다이오드의 광학특성에 미치는 영향)

  • The organic light emitting diodes (OLEDs) have been studied as large flexible displays, light source and hard wares of internet of things. However, OLEDs show some drawbacks in terms of external environments due to the low work function of the metals and the reactive organic materials. In particular, the operation functions of the OLEDs tend to deteriorate rapidly by exposing the oxygen and moisture. So as to prevent it, domestic and overseas studies underway in various method such as ALD, PVD, CVD. But it has complex process and high cost. Therefore In order to protect devices from the external environments, it is important to develop the passivation thin films of low-cost and simple process which can prevent the devices from the penetration of the oxygen and moistures. In this study, to improve the reliability, passivation thin films were coated onto the green OLEDs by spin coating method and investigated the changes of the optical properties of the prepared devices at various doping concentrations of sodium alginate (SA). The passivation solutions were synthesized by using polyvinyl alcohol (PVA) host material with a dopant of SA which were added with the amounts of 10, 20 and 40 wt% into the PVA. As a result, the best barrier properties of the OLEDs were obtained for the samples with 40 wt% SA. Finally, the passivation films can be optimized by using the mixture solution of PVA and SA materials.

  • Research trends in the measurement of foliar uptake of pesticides and the recently proposed action mechanism of the surfactant-induced uptake of pesticides were reviewed with the related reports and studies. Major techniques used in those fields are bioassay, radiotracer techniques with leaves or cuticular membrane. Recently, a new method using Congo Red as a tracer was proposed. The limiting factor in the pesticides uptake into leaves is the waxy layer which consists of the epicuticular and cuticular wax. Physico-chemical parameters such as molar volume, water solubility and partition coefficient of pesticides have limited influences on the pesticide uptake into leaves. Polydisperse ethoxylated fatty alcohol surfactants are well known as the good activator for many pesticides. It is now generally agreed that uptake activation is not related to the intrinsic surface active properties of surfactants such as surface activity, solvent property, humectancy and critical micelle concentration. Recent studies using ESR-spectroscopy revealed that the surfactants have an unspecific plasticising effect on the molecular structure of the wax and cuticular matrix, leading to increased mobilities of pesticides. Penetration of surfactants into waxy layer altered the pesticide mobility in wax and the partition coefficient of pesticide, and then the pesticides penetration into leaves was enhanced temporally. The enhancing effect of surfactant could be significantly different depending on the carbon number of aliphatic moiety and the number of ethoxy group in polyoxyethylene chain of surfactants. It is suggested that the rate of penetration of surfactants should have a significant relationship with the rate of penetration of pesticides.

COMPARISON OF CANAL SHAPING METHODS WITH GTTM ROTARY FILE AND CONDENSATION METHODS (GT rotary file을 이용한 근관성형법과 충전방법의 비교)

  • The purpose of this study was to compare the shaping time of two shaping methods and the leakage of three different obturation techniques. Ninty three canaled human molar teeth were used, which were randomly divided into two groups of forty teeth each and ten control teeth. After working length determination, the one group was prepared crown-down technique using rotary root canal instruments of GT rotary files .12/20, .10/20, .08/20 and .06/20 taper(Maillefer Instrument SA. Switzerland). The other group was instrumented with Gates Glidden burs(#1, #2, and #3) to coronal preparation and GT rotary files .08/20 and .06/30 taper to apical preparation. Shaping time was measured. After root canals were instrumented, they were divided to three subgroups and obturated as follows : Subgroup 1, obturated with single cone method Subgroup 2, obturated with lateral condensation : Subgroup 3, obturated with continuous wave technique. Three subgroups were obturated using non-standardized gutta-percha cone(Diadent, Korea, .06 or .08 taper) and AH-26(Dentsply DeTrey, Germany) as a root canal cement. Ten unobturated teeth served as positive and negative controls. After immersion in 2% methylene blue solution for 1 month, the teeth were washed during 24h. The teeth were demineralized in 10% nitric acid and dehydrated by immersion in 80, 90 and 100% ethyl alcohol. The teeth were finally cleared and stored in 100% methylsalicylate, and apical dye penetration was evaluated under stereomicroscope(Leica M420, LC, U.S.A)at $\times$8.75 magnification. Liner measurement of dye penetration was assessed with the use of digitalized image analysing system (analySIS, GmbH, Germany) The data were analysed statistically using independent T-test and Two-way ANOVA and Tukey test. The result were as follows 1. In canal prepared with GT$^{TM}$ rotary file, shaphing time taked more than the group of using Gates Glidden drill to coronal preparation without statistical significance (p>0.05) 2. The group of single cone obturation using canal preparation of GT$^{TM}$ rotary files showed significantly more apical leakage than those of lateral condensation and continuous wave technique regardless of shaping method (p<0.05). 3 The group of single cone obturation using canal preparation of GT$^{TM}$ rotary files and Gates Glidden drill showed significantly more apical leakage than those of continuous wave technique regardless of shaping method (p<0.05). 4. Regardless of shaping method, The group of continuous wave obturation showed less apical leakage than those of lateral condensation without statistical significance (p>0.05). 5. The group of single cone obturation using canal preparation of GT$^{TM}$ rotary files and Gates Glidden drill showed more apical leakage than the group of lateral condensation using same shaping method with-out statistical significance (p>0.05).

Effect of Fiber Types on Fundamental Properties of Pavement Concrete (섬유 종류가 도로포장용 콘크리트의 기초적 특성에 미치는 영향)

  • The objective of the paper is to experimentally investigate the effect of commercially avaliable fiber types such as polypropylene (PP), nylon (NY), polyvinyl alcohol (PVA) and cellulose (CL) on the engineering properties of concrete for pavement application. The results, showed the fluidity tends to decrease with fibers addition compared to that of plain concrete. As for the effect of fiber types on fluidity loss, use of NY appear to give the most favorable results among all of the fiber types investigated in this study while the effect of the fibers on air content was negligible. For the properties of hardened concrete, compressive and flexural strengths increased with fibers compared to plain concrete. The contribution of NY fibers to strength was the highest followed in the order by NY, PVA, PP, and CL. However, in the case of the splitting tensile strength, its values were increased with NY and PP only. For porosity based on MIP(mercury intrusion penetration) method, the number of around 1 was observed when NY was mixed resulting in increased cumulated amounts of porosity compared with that of plain mix. Thus, based on the consideration of fluidity and strength it was found that the addition of NY fiber showed the optimal results under the conditions applied in this study.

Preparation and Characteristics of Multilayer Lamellar Vesicle Using Phosphate Ester Surfactant (Phosphate ester 계면활성제를 이용한 다중층 라멜라 베시클의 제조 및 특성)

  • Various vehicles have been studied in order to protect skin ageing and sustain constantly moisturization. Recently, in relation to maintain of moisture, absorption and penetration of active materials, there has been introducing many preparing methods such as liposome, liquid crystal and multilamellar emulsion. We developed multilayer lamellar vesicle using cetearyl alcohol/ceteth-20 phosphate/dicetyl phosphate as analogy of phospholipid according to variation of shear rate and pH. These multilayer lamellar vesicles were confirmed by cross polarizing microscope. As results, morphologies of lamellar vesicle were not uniformed at low shear rate and pH. Also, stabilities for encapsulation of retinol were observed at $42^{\circ}C$ during two months. As a result, quantitative content of retinol decreased at low pH. Multilayer lamellar vesicle decreased 14% of transepidermal water loss compared with O/W emulsion. We compared multilayer lamellar sun cream to O/W sun cream using in vitro SPF test of water resistance and concluded that multilayer lamellar sun cream is similar to O/W sun cream in water resistance.

A Study on Applicability of Hydrofluoroethers as CFC-Alternative Cleaning Agents (CFC 대체 산업세정제로의 HFEs의 적용가능성 연구)

  • Fluoride-type cleaning agents such as 2,2,2-trifluoroethanol (TFEA) and hydrofluoroethers (HFEs) do not destroy ozone in the stratosphere and have low global warming potential compared to hydrofluorocarbons(HFCs) and hydrochlorofluorocarbons (HCFCs). Especially, HFEs which have no flash point are paid attention as next generation type of cleaning agents for chlorofluorocarbons (CFCs) since they are safe in handling and have excellent penetration ability compared to hydrocarbon cleaning agents with low flash point. Here, the physical properties and cleaning abilities of fluoride-type cleaning agents such as TFEA, HFE-7100, HFE-7200, HFE-476mec, HFE-449mec-f, AE-3000 and AE-3100E and silicide-type cleaning agents such as trifluoroetoxytrimethylsilane (TFES) and hexamethyldisilazane (HMDS) were measured and compared with those of ozone destruction substances such as CFC-113 and 1,1,1-trichloroethane. They were also compared with toxic methylene chloride (MC) and isopropyl alcohol (IPA) which are now being used as an alternative cleaning agents. As a result, TFEA and HFEs had lower cleaning ability for removal of various soils compared to chloride-type cleaning agents, but they showed excellent cleaning ability fur fluoride-type soils. TFES and HMDS also showed excellent cleaning ability for silicide-type soils.

Cosmetic Application Using Skin Whitening and Anti-microbial effects of Ethyl Acetate and n-Butanol Fractions from Eruca sativa (Eruca sativa 에칠아세테이트와 부탄올 분획물의 미백 및 항균효과를 이용한 화장품 응용연구)

  • Eruca sativa, called arugula, is a perennial plant in the Brassicaceae family, an edible plant commonly used in Italian cuisine. To study as a cosmetic material application E. sativa was extracted with 70% ethanol (ES). Then ES was fractionated using n-hexane, chloroform, ethyl acetate, n-butyl alcohol and water (EHex, EEA, ECHCl3, EBuOH and EDW). EEA showed mushroom tyrosinase inhibitory activity. ES, EEA and EBuOH showed inhibition of tyrosinase activity. As a result, ES is expected to have skin whitening efficacy. ES was applied to 0.05, 0.1% the toner and emulsion formulation to test the stability. The anti-microbial activity of eight bacteria and fungi including Staphylococcus aureus and Propionibacterium acnes which cause dermatitis and acne was evaluated. EEA showed effects in all of microorganisms. The toner and emulsion containing ES with 0.05, 0.1% were passed in the challenge test. At -20, 4, 25, 55 ℃ and daylight, there was no significant change on pH, viscosity for 4 months. However, emulsions had phase separation phenomenon at 55 ℃, so the base formulation needs improvement. In addition, through the skin penetration test, EEA penetrated 0.058% in 6 hr, predicting the clinical efficacy. This means that E. sativa can contribute whitening agent and the synergistic effect of preservatives.

Preparation of Liquid Crystal Emulsion for Transdermal Delivery of Glycyrrhizic Acid and Physical Characteristics and In Vitro Skin Permeation Studies (글리시리직애씨드의 경피 전달을 위한 액정 에멀젼의 제조와 물리적 특성 및 In Vitro 피부투과 연구)

  • In this study, we prepared liquid crystal emulsion composed of amphiphilic substance $C_{14-22}$ alcohol, $C_{12-20}$ alkyl glucoside, behenyl alcohol and studied liquid crystal emulsion of properties and in vitro skin permeation. The results of formulation experiments, the clear liquid crystalline structure was observed in the ratio of $C_{14-22}$ alcohol 0.8%, $C_{12-20}$ alkyl glucoside 3.2%, behenyl alcohol 4% in the formulation. The results of physical property measurements, the viscosity of liquid crystal emulsion and O/W emulsion applied as a control group was respectively $1871.26{\sim}1.15Pa{\cdot}s$, $1768.69{\sim}1.14Pa{\cdot}s$ and the shear stress of O/W emulsion was 178.68 ~ 909.18 Pa, that of liquid crystal emulsion was 190.45 ~ 919.38 Pa. The storage modulus of O/W emulsion was 3428.53 ~ 9157.45 Pa, that of liquid crystal emulsion was 4487.82 ~ 8195.59 Pa. The tan (delta) value of O/W emulsion which means a ratio of viscosity to elasticity was 0.43 ~ 0.19, and that of liquid crystal emulsion was 0.23 ~ 0.25. The water content value on the skin for liquid crystal emulsion was significantly higher from 1 h to 6 h compared with that of O/W emulsion and the transepidermal water loss on the skin was significantly superior in skin moisture loss suppression from 30 min to 4 h compared with that of O/W emulsion. The results of skin permeation using glycyrrhizic acid, the result of skin permeation amount of liquid crystal emulsion for 24 h was $64.58{\mu}g/cm^2$, that of O/W emulsion was $37.07{\mu}g/cm^2$, that of butylene glycol solution was $41.05{\mu}g/cm^2$. Hourly permeability results, it is showed that skin penetration effect of the liquid crystal emulsion increases after 8 h. These results suggest that liquid crystal emulsions are effective for skin moisturizing effect and function as potential efficacy ingredient delivery system for the transdermal delivery.

Diet Patterns and Risk of Squamous Cell Oesophageal Carcinoma: A Case-control Study in Uruguay

  • Background: Oesophageal cancer presents high incidence rates in the so-called Brazilian-Uruguayan belt. Materials and Methods: The present study included 1,170 participants (234 cases and 936 controls) which were analyzed by unconditional multiple logistic regression in order to examine risk of oesophageal squamous cell carcinoma (OESCC) associated with several food groups. Results: Boiled red meat (OR 2.59, 95%CI 1.69-3.97), lamb meat (OR 1.64, 95%CI 1.07-2.51), processed meat (OR 1.49, 95%CI 1.01-2.21), whole milk (OR 1.78, 1.19-1.68), fresh vegetables and fruits (OR 0.42, 95%CI 0.27-0.63), mate consumption (OR 2.04, 95%CI 1.32-3.16), and black tea (OR 0.10, 95%CI 0.04-0.28) were significantly associated with risk of OESCC. Conclusions: Hot beverages (mate) and hot foods (boiled meat) appear to be important determinants in the risk of OESCC, allowing the penetration of carcinogens in tobacco and alcohol into the oesophageal mucosa.
