Algicidal Effects of a Newly Developed Thiazolidinedione Derivative, TD49, on Dinoflagellate Akashiwo sanguinea (Thiazolidinedione 유도체(TD49) 물질을 이용한 적조생물 Akashiwo sanguinea의 제어)

  • To evaluate the algicidal impact of a newly developed algicide thiazolidinedione derivative, TD49, on dinophyceae Akashiwo sanguinea in aquatic ecosystems, tentative culture experiments for the target species were conducted in small (SS), middle (MS), and large scale (LS) culture vessels. When TD49 was introduced at the final concentration of $2{\mu}M$ in SS and MS, as well as $1{\mu}M$ in LS, the abundance of A. sanguinea decreased significantly in all the treatments. On the other hand, total phytoplankton abundance, except A. sanguinea in the TD49 treatments, gradually increased with culture time, which implies that a cell destruction of A. sanguinea by TD49 is a major cause of the population growth by other phytoplankton species. Also, A. sanguinea was easily destroyed, which was likely to be a source of extracellular substances. In particular, a pH decrease was significant in the treatments than in the control, which indicates that the water in the treatments has been acidified, due to an increase in the heterotrophic metabolisms of bacteria and degradation of A. sanguinea cells. Our results indicate that the TD49 substance is the potential agents for the control of A. sanguinea in the enclosed and eutrophic water bodies.

  • This is the first recorded observation of Akashiwo sanguinea excreting mucilaginous substances from pores on the cell surface. Observations were from samples collected in the Caloosahatchee Estuary, Florida, USA during a bloom event, with densities of 672 cells $mL^{-1}$ of A. sanguinea, including 51 cells surrounded by mucous. Scanning electron microscopy observations revealed that the mucous was secreted from thecal pores on the cell surface. The potential significance of mucous production is discussed.

Growth Response of the Dinoflagellate Akashiwo sanguinea in Relation to Temperature, Salinity and Irradiance, and its Advantage in Species Succession (무각 와편모조류 Akashiwo sanguinea의 수온, 염분 그리고 광 조건에 따른 생장반응과 그에 따른 종 천이의 이점)

  • The effects of temperature, salinity and irradiance on the growth of the dinoflagellate Akashiwo sanguinea isolated from Jaran Bay were examined in the laboratory. Maximum specific growth rate($0.28day^{-1}$) was observed with combination of $25^{\circ}C$ and 30 psu. Optimal growth (${\geq}80%$ of maximum specific growth rate) was obtained at $25^{\circ}C$ with salinities of 15~35 psu. This results indicated that A. sanguinea is a stenothermal of the high water temperature and euryhaline species. The irradiance-growth curve was described as ${\mu}=0.31(I-16.87)/(I+51.19)$. The compensation photon flux density ($I_0$) and half-saturation photon flux density ($K_I$) were $16.87{\mu}mol\;m^{-2}s^{-1}$ and $84.93{\mu}mol\;m^{-2}s^{-1}$, respectively. In conclustion, A. sanguinea has advantage physiological characteristics for the species succession at the coastal areas in summer with sufficient irradiance, high water temperature and large salinity gradient.

Effect of Water Temperature on Infectivity of the Parasitoid Amoebophrya sp. Infecting the Harmful Bloom-forming Dinoflagellate Akashiwo sanguinea (유해 적조생물 Akashiwo sanguinea를 감염시키는 포식성 기생생물 Amoebophrya sp.의 감염력에 대한 수온의 영향)

  • Marine parasitoid Amoebophrya infects and kills various bloom-forming dinoflagellates and strongly influences the harmful algal bloom dynamics. We investigated the effect of temperature on survival, infectivity, generation time of the parasite from the parasitoid Amoebophrya sp. and the harmful dinoflagellate host Akashiwo sanguinea system. Temperature had a significant effect on the parasite generation time and infectivity. While the lower temperature ($15^{\circ}C$) arrested parasite intracellular development and infectivity, resulting in the longer generation time ($115{\pm}0.1h$), the higher temperatures ($25^{\circ}C$ and $20^{\circ}C$) accelerated the parasite development, with the generation times of $58{\pm}0.1h$ and $83{\pm}0.1h$, respectively. Parasite prevalence (percent of host infected) was $71.5{\pm}0.30%$, $54.3{\pm}1.68%$, and $29.6{\pm}1.42%$ at $25^{\circ}C$, $20^{\circ}C$, and $15^{\circ}C$, respectively. These results suggest that biological control by parasitism on A. sanguinea bloom would not be highly effective during low water temperature season. Further, water temperature would be an important factor of bottom-up controls for the host-parasite population dynamics.

  • The morphological characteristics of the bloom-forming dinoflagellate Akashiwo sanguinea isolated from Jangmok Harbour, Geoje in Korea was examined using light and scanning electron microscope (SEM), and its large subunit (LSU) rDNA was sequenced. Additionally, investigation was done on the effects of temperature and salinity on the growth of A. sanguinea. The cells were dorso-ventrally compressed up to 54.7-70.3 ㎛ long and 31.5-48.5 ㎛ wide. The epicone was conical while the hypocone was separated into two lobes. The nucleus was positioned at the center of the cell. The yellow-brown chloroplasts radiated close to the cell center. SEM observation indicated that A. sanguinea has an e-shaped apical groove. Molecular phylogeny based on LSU rDNA gene sequences revealed that the A. sanguinea strains isolated from Jangmok Harbor were classified in the clade of ribotype A. The maximum growth rate (0.50 day-1) was observed at 20℃ and 20 psu, while the maximum cell density (1,372 cells mL-1) was observed at 25℃ and 30 psu. This indicates that the blooms of A. sanguinea ribotype A in Korean coastal area are affected by water temperature, rather than the salinity.

식물플랑크론 5종에 대한 benzo[a]Pyene의 독성에 관한 연구

  • 본 연구는 해양 식물플랑크톤에 지속성 유기오염물질의 영향을 이해하기 위한 목적으로, 연안역에서 우점하는 식물플랑크톤 주요 5종 즉, 규조류 Skeletonema costatum, 침편모조류 Heterosigma akashiwo, 와편모류 prorecentrum dentatum, Prorocentrum minimum, Akashiwo sanguinea를 이용하여을 benzo[a]pyrene (PAHs)에 72시간동안 노출시킨 다음 각 종의 성장 및 저해, 회복능력 등을 조사하였고, benzo[a]Pyrene의 농도 0.1, 1, 5, 10 $\mu\textrm{g}$/l에 Heterosigma akashiwo(Raphidophyceae)를 노출시켜 시간에 따른 광합성률의 변화를 측정하였다. benzo[a]Pyrene에 72시간 노출시킨 후 S. costatum, P. minimum, P. dentatum, akashiwo의 세포수는 1-10 $\mu\textrm{g}$/l의 농도범위에서 극적인 감소를 보인 반면, A. sanguinea는 0.1-1 $\mu\textrm{g}$/l의 낮은 농도범위에서 지수함수적인 감소를 나타냈다. 성장저해 농도 ($IC_{50}$/)는 A. sanguinea가 0.43 $\mu\textrm{g}$/l로 가장 낮았고, H akashiwo(1.17 $\mu\textrm{g}$/l) S. costatum (3.34 $\mu\textrm{g}$/l), p dentatum (3.97 $\mu\textrm{g}$/l), p minimum (7.24 $\mu\textrm{g}$/l)의 순서로 증가하였다. BenEo[a]pyrene의 낮은 농도(1 $\mu\textrm{g}$/l)에 노출되었던 세포들은 5종 모두 시간이 경과함에 따라 회복하는 경향을 나타냈으나 고농도(10, 100 $\mu\textrm{g}$/l)에 노출시에는 P. minimum을 제외하고는 회복되지 않았다. 이러한 결과들은 조사된 5종 중에서 유각 와편모류 P. minimum이 benzo[a]pyrene에 가장 내성이 강하며, 무각 와편모류 A. sangulinea가 가장 약함을 의미한다. benzo[a]pyrene의 여러 농도수준에 노출시켰을 때 H. akashiwo의 광합성 능력은 낮은 농도들에서는 대조구와 유사하였으나, 5 $\mu\textrm{g}$/l의 높은 농도에서는 초기에 매우 낮은 광합성 능력을 보이다가 시간이 경과하면서 대조군보다 더 높은 경향을 나타냈다. 이러한 결과는 식물플랑크톤이 benso[a]pyrene의 낮은 농도에서 노출될 때는 이 물질을 탄소원으로 사용할 가능성이 있음을 시사한다. 본 연구의 결과들은 연안해역에 benso[a]pyrene과 같은 지속성 유기오염물질이 유입되었을 때 내정여부에 따라 식물플랑크톤 군집내 종 천이와 일차생산력에 크게 영향을 미칠 수 있음을 시사한다.

Effects of Benzo〔a〕pyrene on Growth and Photosynthesis of Phytoplankton (식물플랑크톤의 성장과 광합성에 대한 benzo〔a〕pyrene의 영향)

  • We examined the impacts of anthyopogenic pollutant (benzo〔a〕pyrene) on the growth and photosynthesis of five marine phytoplankton species (Skeletonema costatum, Heterosigma akashiwo, Prorocentrum dentatum, P. minimum, Aknshiwo sanguinea), which are dominant in Korean coastal water. After the 72 h exposure to benzo〔a〕pyrene, the dramatic decrease in cell numbers was observed in the range of 1 to 10 $\mu\textrm{g}$ L$^{-1}$ for S. costatum, P. minimum, P. dentatum, whereas for A. sanguinea and H. akashiwo at the low concentrations 0.1 to 1 $\mu\textrm{g}$ L$^{-1}$ . Among the 5 phytoplankton species, the highest growth inhibition concentration ($IC_{50}$/) was 6.20 $\mu\textrm{g}$ L$^{-1}$ for P. minimum, followed by 2.14 $\mu\textrm{g}$ L$^{-1}$ for P. dentatum, 1.68 $\mu\textrm{g}$ L$^{-1}$ for S. costatum, 0.74 $\mu\textrm{g}$ L$^{-1}$ for H. akashiwo, 0.10 $\mu\textrm{g}$ L$^{-1}$ for A. sanguinea. The five species exposed to the low concentration of 1 $\mu\textrm{g}$ L$^{-1}$ were recovered after transferring to new media, but the species exposed to the high concentrations of 10 and 100 $\mu\textrm{g}$ L$^{-1}$ were not recovered, with the exception of P. minimum. Those results indicate that the thecate dinoflagellate P. minimum is most tolerant to the chemical and the athecate dinoflagellate A. sanguinea is not. Geneyally, the cell-specific photosynthetic capacity of H. akashiwo exposed to the low concentrations of 0.1 and 1 $\mu\textrm{g}$ L$^{-1}$ was higher than that of the cells in the control, whereas the cells exposed to the high concentrations of 5 and 10 $\mu\textrm{g}$ L$^{-1}$ showed the negligible photosynthetic level by the first few days of the experiment. In the case of the cells exposed to the concentration of 5 $\mu\textrm{g}$ L$^{-1}$ , after 12 days of the experiment the photosynthetic capacity was increased toward the end of the experiment. This indicates that H. akashiwo may utilize the benzo〔a〕pyrene as a carton source for its growth when exposed to low concentrations. Results suggest that anthropogenic pollutants such as benzo〔a〕pyrene may have significant influence on the succession of phytoplankton species composition and the primary production in coastal marine environments.

Extraction of anti-microalgal material from Laminaria spp. and effect of oligo-alginate derivatives on membrane potential (다시마 유래 항미세조류 물질 추출 및 알긴산 올리고 유도체의 막전위에 대한 영향)

  • Oligo-alginate derivatives were extracted from brown algae and its antimicroalgal effects and reaction mechanism were investigated. Oligo-alginate derivatives were produced from sequential hydrolysis of high molecular weight alginate by treatment of 2 N HCl and 1% $H_2O_2$. Antimicroalgal activity of extracts was proportional to reaction time and activity was highest at 4 hrs. When oligo-alginate derivatives were treated to Akashiwo sanguinea and Cochlodinium polykrikoides, mobilities of cells were ceased. A. sanguinea cells were crushed and plasmolysis was induced in C. polykrikoides cells. To investigate the action mechanism of oligo-alginate derivatives, changes of intracellular (pHi) and extracellular pH (pHe) were determined in the microalgal cells exposed to 0.05% of oligo-alginate derivatives. pHi was decreased about 0.3 unit and pHe was increased about 0.9 unit. These results suggested that change of membrane potential by oligo-alginate derivatives could led to microalgal cell death.

Impacts of Temperature, Salinity and Irradiance on the Growth of Ten Harmful Algal Bloom-forming Microalgae Isolated in Korean Coastal Waters (한국연안에서 분리한 적조형성 미세조류 10종의 성장에 미치는 온도, 염분, 광도의 영향)

  • In order to understand growth characteristics of ten major species of microalgae responsible for frequent harmful algal blooms in Korean coastal waters, the growth rates of the isolates were examined in relation with the impacts of water temperature, salinity and irradiance. In addition, their bloom events since 1990 as well as monthly abundance of vegetative cells were analyzed. Heterocapsa triquetra, Eutreptiella gymnastica and Alexandrium tamarense were considered as relatively mid temperature adapted species in that growth rates were comparatively high at low water temperatures of $10{\sim}16^{\circ}C$ and drastically decreased at above $22^{\circ}C$. Prorocentrum micans and Pyramimonas sp. were categorized as relatively high temperature adapted species by showing comparatively better growths at high water temperatures above $25^{\circ}C$. Akashiwo sanguinea, Heterosigma akashiwo, Prorocentrum minimum and Scrippsiella trochoidea were eurythermal species with relative high growth rates in a broad ranges of water temperature, $16{\sim}25^{\circ}C$ were slightly halophobic, showing better growths at low salinities of $10{\sim}30$ psu than at above 35 psu. H. akashiwo, P. minimum and H. triquetra were euryhaline species with remarkable growths in a broad ranges of salinity, 15-40 psu. Frequent algal blooms by these three species at extremely low salinities below 25 psu after rainfall were attributed to their euryhaline and slightly halophobic physiological characteristics. Growth rates of H. akashiwo, P. minimum and Pyraminonas sp. increased with the increase of irradiance within the experimental ranges of $2{\sim}150\;{\mu}E{\cdot}m^{-2}{\cdot}s^{-1}$. However, A. sanguinea, A. tamarense and H. triquetra showed better growths at comparatively low irradiance of $50{\sim}100\;{\mu}E{\cdot}m^{-2}{\cdot}s^{-1}$ and drastic decreases in growth rates above $150\;{\mu}E{\cdot}m^{-2}{\cdot}s^{-1}$ of irradiance. Overall, relatively high temperature adapted species make blooms frequently in high water temperature season with strong natural irradiance, and relatively low temperature adapted species grow better at low water temperature with relatively weak natural irradiance.

Relationships between Cell Bio-volume and Growth Rate of Dominant Red Tide Organisms in the Coastal Water (연안내만해역에서 우점하는 주요적조생물의 성장과 세포체적의 관계)

  • To understand growth characteristics of eight dominant red tide species ($Prorocentrum$ $minimum$, $Heterocapsa$ $triquetra$, $Scrippsiella$ $trochoidea$, $Akashiwo$ $sanguinea$, $Chattonella$ $marina$, $Heterosigma$ $akashiwo$, $Amphidinium$ $carterae$ and $Rhodomonas$ $salina$) in the Korean coastal water, the growth rates were examined in relation with the impacts of water temperature and bio-volume. Of these, $P.$ $minimum$, $C.$ $marina$, $H.$ $akashiwo$, $A.$ $carterae$ and $R.$ $salina$ were eurythermal species with relatively high growth rates in a borad ranges (15 to $25^{\circ}C$) of water temperature. On the other hand, the growth rate of $H.$ $triquetra$, $S.$ $trochoidea$ and $A.$ $sanguinea$ were high in relatively mid temperature (optimum: $25^{\circ}C$) condition. In particular, $H.$ $triquetra$ was well adapted in low temperature of 5 to $15^{\circ}C$, implying that the species can survive and grows even at very low temperature. Based on results of our experiment, the growth characterestics of five eurythermal species and three mid temperature species may have dominated in Korean coastal water during summer season and fall season, respectively. Contrastively, the growth characteristics of $H.$ $triquetra$ make a consistently dominant during the cold winter season. In addition, the growth rates of large bio-volume species were lower than those of small bio-volume species, indicates that growth of single cells of several flagellates might be depended on the cells sizes.
