Gene Transformation of Ailanthus altissima Swingle by Agrobacterium tumefaciens (외래유전자(外來遺傳子)에 의(依)한 가중나무의 형질전환(形質轉換))

  • An efficient transformation system was established for Ailanthus altissima utilizing the binary system of A. tumefaciens strain LBA4404. Callus was initiated from small portions of cambium tissue of A. altissima in vitro. Optimum regeneration was achieved with Murashige and Skoog(MS) medium containing 0.01mg/${\ell}$ 2, 4-D, 0.5mg/${\ell}$ BAP, 3%(w/v) sucrose and 0.75% agar. The multiplication of explants remarkably showed up on medium containing 1.0mg/${\ell}$ BAP. Leaf discs or internodal stem segments were inoculated with A. tumefaciens strain LBA 4404 containing the binary vector pPMB 101, which has both ${\beta}$-glucuronidase (GUS) marker gene and neomycin phosphotransferase II (NPT II) gene. Shoots had been regenerated from 24 lines out of inoculative 50 lines. Transformants were selected by their ability to grow on medium containing kanamycin sulphate (100mg/${\ell}$). Putative transformation was confirmed by GUS assays. Five GUS-positive plantlets were obtained which confirmed that this marker gene has been transferred into A. altissima.

Flavonoids from the Leaves of Ailanthus altissima Swingle and their Antioxidant Activity

  • Phytochemical studies on the leaves of Ailanthus altissima (Simaroubaceae) have not been reported previously. Thus, the authors isolated and identified secondary metabolites from A. altissima. Dried and powdered leaves were extracted with 80% aqueous methanol, and the concentrated extract was successively partitioned with ethyl acetate, n-butanol, and water. Four flavonoids were isolated from the ethyl acetate fraction through repeated silica gel and octadecyl silica gel column chromatography. Spectroscopic data including NMR, MS, and IR allowed for identification of the chemical structures as quercetin (1), afzelin (2), quercitrin (3), and isoquercitrin (4). This is the first report of the isolation of these compounds from A. altissima. The four isolated flavonoids 1-4 as well as solvent fractions (ethyl acetate, n-butanol, and water), were evaluated for DPPH radical scavenging activity.

Capillary Flow in Different Cells of Ginkgo Biloba, Diospyros Kaki and Ailanthus Altissima (은행나무, 감나무, 가중나무 세포내강의 액체이동)

  • A study was carried out to observe the 1% aqueous safranine solution flow speed in longitudinal and radial directions of softwood G. biloba, ring-porous wood A. altissima, and diffuse- porouswood D. kaki. In radial direction, ray cells and in longitudinal direction tracheids, vessel and wood fiber were considered for the measurement of liquid penetration speed at less than 12% moisture contents (MC). The length, lumen diameter, pit diameter, end wall pit diameter and the numbers of end wall pits determined for the flow rate. The liquid flow in the those cells was captured via video and the capillary flow rate in the ones were measured. Vessel in hardwood species and tracheids in softwood was found to facilitate prime role in longitudinal penetration. Radial flow speed was found highest in ray parenchyma of G. biloba. Anatomical features like the length and diameter, end-wall pit numbers of ray parenchyma were found also responsible fluid flow differences. On the other hand, vessel and fiber structure affected the longitudinal flow of liquids. Therefore, the average liquid penetration depth in longitudinal tracheids of G. biloba was found the highest among all cells considered in D. kaki and A. altissima.

Plant Regeneration from Cambium Callus of Ailanthus altissima Swingle (가중나무의 형성층(形成層) Callus에서 식물체(植物體) 재분화(再分化))

  • The stem segments of Ailanthus altissima were cultured on the Murashige & Skoog's medium(1962) supplemented with 0.1 mg/l BAP and 1.0 mg/l 2, 4-D for callus induction and proliferation, Shoot primordia were observed as greenish regions on the surface of yellow-brown calli about 8 weeks after culture. Shoot primordia were selected and transferred to the MS media containing various combination of BAP and 2, 4-D. Among these combinations the shoot primordia cell clusters on the medium added to 0.5mg/l BAP and 0.01mg/l 2, 4-D exhibited the highest number of shoot formation. These shoots were successfully transferred on the solid MS medium with no growth regulators for the rootings.

Physiological Activities of Hot Water Extract from Ailanthus altissima (가죽나무(Ailanthus altissima) 열수 추출물의 생리활성)

  • In this study, extract of Ailanthus altissimawere prepared using hot water under high pressure. The extract were examined for election donating ability (EDA), superoxide dismuase (SOD)-like activity, nitrite scavenging ability (NSA), xanthine oxidase inhibition levels, and tyrosinase inhibition ability. The EDA, using the 1,1-diphenyl-2-picrylhydrazyl method of root extract was 91.25% at 1.0mg/mL. The SOD-like activity of leaf extract was highest at 49.07% and the NSA was 93.33% at pH 1.2, and 85.40% at pH 3.0. The xanthine oxidase inhibitory levels of extracts of A. altissima roots, stems, and leaves were 92.09 97.44% when the extract were tested at 2.0mg/mL. The highest tyrosinase inhibition levels obtained from loot extract were 67.38% at 2.0mg/mL and 63.97% at 0.1mg/mL.

Allelopathic Effects on Seed Germination and Fungus Growth from the Secreting Substances of Some Plants (수종식물의 분비물질이 종자 발아와 균류 생장에 미치는 알레로파시 효과)

  • Phenolic compounds from 7 species of naturalized invader species and Korean wild plants were analyzed by high performance liquid chromatography. Eleven phenolic compounds including benzoic acid were identified. The extract of naturalized plants was significantly more inhibitory to seed germination and seedling growth of the both naturalized and Korean wild plants. The content of total phenolic compounds in each extract were 43.5 mg/l in Ailanthus altissima as the maximum amount and 25.5 mg/l in Phytolacca americana as the minimum. Phytotoxic substances of ethanol extracts was investigated for antifungal activity against 23 selected fungus species. The antifungal activity of Phytolacca americana showed the greatest clear zone of 23 mm in Aspergillus awamori and its activity had an effect against 6 fungus species. Ailanthus altissima formed the greatest clear zone of 26 mm in Erwinia carotovora sub. sp. carotovora and had an effect agsinst 2 fungus species.

  • During the serial attempts to identify the chemical and biological characteristics of Ailanthi Cortex Radicis, the root bark of Ailanthus altissima (Simaroubaceae), we find out the serious toxic effect on kidney by chloroform fraction of the methanolic extract of the herb drug. The toxicities were revealed as the increase of urea nitrogen amount in blood and lactate dehydrogenase and ${\gamma}-glutamyltransferase$ activities in urine and the decrease of the concentration of glutathione and both of protein bound and non-protein bound -SH in kidney tissue.

  • For the biological survey, effects of Ailanthi Cortex Radicis, the root bark of Ailanthus altissima(Simaroubaceae) on epoxide hydrolyzing enzymes were checked. The methanolic extract and its chloroform fraction were shown to activate the liver metabolizing enzyme system including epoxide hydrolase system which was monitored by activities of transaminase, lactate dehydrogenase, alkaline phosphatase and epoxide hydrolase system in bromobenzene treated rats. But they showed no effect on glutathione S-transferase activity.

Photosynthetic Inhibition in Leaves of Ailanthus altissima under O3 Fumigation

  • We investigated the enect of $O_3$ on the photosynthetic characteristics of tree of heaven (Ailanthus altissima) that is naturalized plant and used as restoration plant for contaminated area. Two-year-old seedlings were planted to pots and transferred into closed $O_3$ chamber. Photosynthetic pigments contents and photosynthetic characteristics were measured every three weeks under 100 pub $O_3$ fumigation. There was no visible foliar injury by $O_3$ exposure and contents of photosynthetic pigments did not show significant differences between control and $O_3$-treated seedlings. Also there were no significant differences in stomatal conductance, and water use efficiency. But photosynthetic rate and apparent quantum yield (AQY) of $O_3$ treated seedlings were reduced after nine weeks of ozone fumigation. In addition, the reduction of carboxylation efficiency and photorespiration were observed in the leave of $O_3$ treated seedlings after six weeks. In accordance with our result, carbon fixation system of A. altissima was most sensitive to $O_3$ stress to evaluate physiological damage induced by $O_3$.
