Increased Antioxidants of Agastache rugosa by the Night Interruption Time (야파(night interruption)처리에 의한 배초향의 항산화 물질 증가)

  • The objective of this study was to determine the proper night interruption of photoperiods and dark periods for accumulating bioactive compounds of Agastache rugosa without decreasing plant growth. Five-week-old seedlings were transplanted in a DFT system with white LEDs. A. rugosa was treated with night interruption time treatments of 18:1:2:3, 18:2:2:2, 18:3:2:1 (light:dark:light:dark), and 20:4 (control) for 4 weeks. There were no significant differences except for leaf length, leaf width, and the number of flowers. The content of antioxidants in the shoot of A. rugosa was high in tilianin and acacetin, and the content of rosmarinic acid (RA) was significantly higher in the underground part. The RA content per dry weight of A. rugosa was 47.92 and 51.46% higher than that of the control in 18:1:2:3 and 18:2:2:2, and tilianin and acactin per dry weight were significantly higher in 18:3:2:1. There was no significant difference in growth due to the same day light integral, but 18:2:2:2 showed high total polyphenol contents. Therefore, it is thought that the effect of increasing secondary metabolites of A. rugosa without degradation of growth can be expected through night interruption treatment in plant factory cultivation systems using artificial light.

  • 수증기 증류법으로부터 얻은 배초향(Agastache rugo-sa, Labiatae)의 정유는 부패균을 포함하는 수종의 미생물에 대하여 현저한 항균활성을 나타내었다. 배초향의 유효성분을 확인하기 위하여 정유성분을 GC-MS로 분석한 결과 GC chromatogram에서 얻은 각 피크의 reten-tion time 및 mass spectral data 의표준물질과의 비교에 의해서 isomenthone, methone, dihydrocarvone, anetho-le, vanillin, eugenol, methyleugenol, $\beta$-caryophyllene, $\beta$-caryophyllene oxide 등의 정유성분이 확인되었다. 이 연구에서 밝혀진 여러 성분들의 항균 활성은 잘 알려져 있으므로 이들이 유효성분으로 추측된다. 또한 Korean mint로 세계적으로 알려져 있는 배초향의 정유는 식품 방부제로서 적합할 것으로 생각된다.

HIV Integrase Inhibitory Activity of Agastache regosa

  • We have been screening anti-HIV integrase compounds from Korean medicinal plants by using an in vitro assay system which is mainly composed of recombinant human immunodeficency virus type 1 integrase and radiolabeled oligonucleotides. From the above screening, the aqueous methanolic extract of the roots of Agastache rugosa exhibited a significant activity. Bioactivity-guided chromatographic fractionation of the methanolic extract resulted in the isolation of rosmarinic acid. The structure of the compound was determined by spectroscopic data and by the comparison with the reported values. The $IC_{50}$ of the rosmarinic acid was approximately $10{\mu}g/ml$ against HIV integrase.

  • The essential oil obtained by steam distillation of the herbs of Jeju Agastache rugosa has shown significant DPPH radical scavenging activity. For the elucidation of its volatile components, the essential oil was analyzed by GC-MS. Each component was identified by GC or mass spectral analysis. The compounds identified were as follows: pulegone, L-menthone, DL-limonene, isopulegone, beta-caryophyllene, beta-myrcene, and estragole. Several abundant components with phenylpropanoid-type structures may have contributed to the activity. The results suggest that the essential oil may be suitable for development as a food preservative and alternative antioxidant.

  • The experiment was carried out to study the influence of seeding dates, planting densities and fertilizer application levels on some agronmic characters and yield of fresh weight in Agastache rugosa. The results obtained are summarized as follow; Fresh weight was increased on Apr. 1 seeding date. Therefore, optimum time for sowing was on Apr. 1. Stem length was long in dense planting and short in spacious planting. Stem diameter was thick in spacious planting, and was thin in dense planting. Yield was higher in dense planting densities($10{\times}20cm:50\;plants/m^2,\;20{\times}20cm:25\;plants/m^2,\;(30{\times}20cm:17\;plants/m^2$) Fertilizing ($N:P_2O_5;K_2O=6:6:6kg/10a$) was increased than without fertilizing because stem length and number of leaf were good.

Effect of Forms and Levels of Nitrogen Fertilizer on Plant Growth and Essential Oil Content of Agastache rugosa

  • This study was carried out to investigate the effect of forms and levels of nitrogen fertilizer on plant growth and essential oil production of Agastache rugosa. Calcium nitrate had more influenced on length and width of leaves and lateral branch length than did urea. When nitrogen fertilizer level was increased from 12 kgN/I0a to 24kgN/I0a, plant growth was stimulated and dry matter of leaf and inflorescence were increased. Top dry matter of plant with calcium nitrate treatment (38.4 g) was heavier than that of urea treatment (32.8 g). Interactions among accession and nitrogen form and nitrogen rate were not significantly different for top dry matter. The forms and rate of nitrogen fertilizer did not affect estragole content. The estragole contents was higher in leaf (91.8%) than that of inflorescence (81.3%). While the essential oil content was not affected by different nitrogen forms, nitrogen level affected the essential oil contents positively by increasing dry matter. Essential oil yield was not affected by accession or nitrogen form, but by nitrogen rate. With increasing N application from 12kgN/I0a to 24 kgN/I0a, essential oil yield was increased by 95.8 %.

  • SH2 domains and their associated catalytic or noncatalytic proteins constitute critical signal transduction targets for drug discovery. Grb2 associates with phosphotyrosine sites of the activated receptors or Shc via their SH2 domain to link receptor tyrosine kinases to ras signalling. Blocking of the Grb2-Shc complex may be to intervene the oncogenic signal transduction pathways and to develop a new antitumor drug. In the search for blockers of Grb2 SH2-Shc interaction, Lutein, a family of carotenoids, was isolated from the extract of the leaf of Agastache rugosa O. Kuntze as SH2 domain antagonists. The $IC_{50}$ of Lutein against Grb2-Shc binding was $6.8\;{\mu}M$.

  • "Eating cosmetic" market comes to prominence worldwide. Inner Beauty has established itself as the top trend nowadays and is leading the health functional food market. Another noticeable trend other than beauty-from-within is that products using "plant-based" natural botanical raw material are increasing. Based on these trends above, we have developed a plant extract named Agatri® for skin health using Agastache rugosa. By ingesting Agatri®, it is expected that the formation of skin wrinkles is suppressed, and skin elasticity is increased by suppressing the decrease in collagen content caused by ultraviolet rays. At the same time, it is possible to prevent skin aging by improving skin hydration and trans-epidermal water loss (TEWL). The studies show that it significantly increased production of hyaluronic acid and collagen, and decreased production of matrix metalloproteinases (MMPs). And Agatri® has been approved as an "Individually recognized functional ingredient for skin health by the Ministry of Food and Drug Safety, Korea in 2020. As a result, Agatri® is a good solution for those who are in need of skin protection from exposure to UV and aging.

Analysis of Seed Storage Data and Longevity for Agastache rugosa (배초향 (Agastache rugosa) 종자의 저장 반응과 수명 분석)

  • There is little information about the seed longevity of wild plants, although seed bank storage is an important tool for biodiversity conservation. This study was conducted to predict the seed viability equation of Agastache rugosa. The A. rugosa seeds were stored at moisture contents ranging from 2.7 to 12.5%, and temperatures between 10 and $50^{\circ}C$. Viability data were fitted to the seed viability equation in a one step and two step approach. The A. rugosa seeds showed orthodox seed storage behaviour. The viability constants were $K_E=6.9297$, $C_W=4.2551$ $C_H=0.0329$, and $C_Q=0.00048$. The P85 of A. rugosa seeds was predicted to 152 years under standard seed bank conditions. The P85 predicted by seed viability equation can be used as basic information for optimization of seed storage processes.

Antimicrobial Activity of Essential Oils from Mentha arvensis L. var. piperascens Malivaud and Agastache rugosa O. Kuntze on Escherichia coli and Salmonella typhimurium (대장균과 살모넬라균에 대한 박하와 배초향 정유성분의 항균활성)

  • For developing natural presevatives, essential oils of Mentha arvensis L. var. piperascens Malivaud and Agastache rugosa O. Kuntze were analyzed the composition of two oils and experimented on microorganism survival. Main components of Mentha arvensis oil were isomenthol (26.84%) and menthol (25.48%), and those of Agastache rugosa oil were estragole (79.83%) and limonene (4.13%) from GC-MSD analysis. Inhibition activities of Mentha arvensis oil against growth of Escherichia coli O157 : H7 ATCC 43895 and Salmonella typhimurium ATCC 7988 were observed from their clear zone $(9{\sim}14 mm\;&\;9{\sim}13\;mm)$, and that of Agastache rugosa oil were done from the clear zone $(13{\sim}20 mm\;&\;10{\sim}18\;mm)$ by concentration-dependent manner, respectively. In the inhibition test on CFU/ml of the microorganisms, both of the plant essential oils at concentration of 5 and 10 mg showed potent growth inhibition activities from 9 hour of incubation. Analysis using transmission electron microscope on E. coli also showed antimicrobial activities of the oils as deformation of the cell and loss of the intracellular materials.
