Optimum of Gamma Irradiation Dose for Rice-based Infant Foods with Improved Energy-density and Shelf-life (쌀기본 이유식의 에너지 밀도 및 저장성 증대를 위한 최적 감마선 조사 선량)

  • This study was carried out to determine the optimum irradiation dose for infant foods with improved energy-density and extend shelf-life using gamma irradiation. Rice and rice porridge were irradiated at 0, 2.5, 5, 7.5, 10, 15 and 20 kGy, and rheological characteristics and total plate count of porridge were evaluated for 6 weeks. Rice and rice porridge at 2.5 kGy dose reduced the viscosity with 22% and 98%, respectively. The result of total aerobic bacteria of non-irradiated rice porridge was 6 log CFU/g and that of 2.5 kGy irradiated rice porridge on 2 weeks storage was 3 log CFU/g, while that of 7.5 kGy irradiated sample was not detected even at the end of storage. Irradiation on rice-based porridge was proper to enhance the total solid in inverse proportion to considering viscosity and extend shelf-life. Gamma irradiation was effective technology in enhancing calorie value of porridge due to reduction of viscosity.

Microbial Quality Analysis of Foods from Daycare Centers Supported by the Center for Children's Foodservice Management in Daegu (대구지역 어린이급식관리지원센터 지원 일부 어린이집 급식의 미생물적 품질 평가)

  • This study examined the microbiological quality of daycare center meals. Six menu items from five daycare centers in Daegu included uncooked processed foods (seasoned cucumber and lettuce salad), post-preparation after cooking processed foods (rolled omelet and seasoned soybean sprout), and cooking processed foods (panbroiled beef with oyster sauce and seasoned pork roast). Microbiological analyses were performed for the aerobic plate counts (APC), coliforms, and Escherichia coli. The analyses were conducted in July and November 2017. The mean APC and coliform count of seasoned cucumber decreased significantly from 4.71 log colony forming units (CFU)/g and 2.50 log CFU/g in July to 4.07 log CFU/g and 1.78 log CFU/g in November, respectively (P<0.01 and P<0.001). The APC of panbroiled beef with oyster sauce and seasoned pork roast were significantly lower in July (1.84 and 1.79 log CFU/g) than in November (2.41 and 2.28 log CFU/g) (P<0.001). The coliform counts of panbroiled beef with oyster sauce and seasoned pork roast were significantly greater in November (2.11 and 1.62 log CFU/g) (P<0.001). E. coli was not detected. Among the foods prepared using the three preparation processes, the post-preparation after cooking processed foods had the lowest microbial quality. The APC and coliform counts of cooking processed foods were satisfactory in July, with an acceptable rating for pan-broiled beef with oyster sauce in November. Time-temperature control and the prevention of cross-contamination are essential during meal production for food safety, regardless of the season.

Validation of Sanitation Management Standards for Vegetable Preparation with No-Cook Step Based on Microbiological Analysis (미생물 분석에 의한 채소류 비가열 조리공정의 위생관리 기준 유효성 평가)

  • This study evaluated the sanitation management standards for vegetable preparation processes without a cooking stage. The aerobic plate counts (APC), coliform counts, and Escherichia coli of the samples at each production step were analyzed, and microbial growth of the samples stored at different temperatures was measured. The validation was judged in accordance with the microbial criteria stipulated by the British PHLS. After washing and disinfection, the APCs of the two samples decreased to 3~4 log CFU/g in both seasons. Compared to the purchasing stage, the decrease in coliform counts was approximately 0~3 log CFU/g in both seasons; E. coli was not detected. The initial APC and coliform levels of two vegetable samples were 4~5 log CFU/g in both seasons, with an increase of 1 log CFU/g taking more than 6 h at 25℃ and 2 h at 35℃. More than 10 h at 25℃ and 6 h at 35℃ were required to increase the E. coli O157:H7 count by 1~2 log CFU/g for two seasoned samples. In conclusion, washing and disinfection effects and changes in microbial growth during room temperature storage were similar in the two vegetables. Despite the low sanitizing effect of the two vegetables, when cut vegetables were stored under the critical limit within 2 h at two different room temperatures, seasoned lettuce and chicory were at the 'satisfactory' or 'acceptable' levels of PHLS regardless of the storage temperatures. The validation of sanitation management standards applied to vegetable preparation with the no-cook step was approved.

Effect of Seasonal Distribution Temperature on Storability of Modified Atmosphere Packaged Baby Leaf Beet (계절별 수송 온도가 MA 포장한 어린잎 비트의 저장성에 미치는 영향)

  • The effects of distribution temperature due to season all changes on quality and storability of baby leaf beet (Beta vulgaris L.) was examined in modified atmosphere (MA) packages. The beet leaf had been harvested at the 10 cm leaf length stage and packaged with an oxygen transmission rate (OTR) film of $1,300cc{\cdot}m^{-2}{\cdot}day^{-1}{\cdot}atm^{-1}$ and then held at 4 different distribution temperatures which were $-2^{\circ}C$, $4^{\circ}C$, $20^{\circ}C$, or $30^{\circ}C$ for 5 hrs and then stored for 18 days at $8^{\circ}C$. The loss of fresh weight of packged baby leaf beet was lowest at the $4^{\circ}C$ treatment, and below 0.6% in all distribution temperature treatments. The atmosphere composition in packages did not show any significant differences among treatments. The oxygen conc. was the highest at 18.0% after the $4^{\circ}C$ treatment, carbon dioxide conc. showed the maximum value of 4% at the $30^{\circ}C$ and $-2^{\circ}C$ treatments, and ethylene conc. was highest at the $10^{\circ}C$ treatment after 10 days in storage. The hardness was the highest at the $4^{\circ}C$ treatment on the final storage day. The $4^{\circ}C$ treatment showed the highest visual quality and the lowest off-odor and aerobic plate count. Therefore, it is necessary to establish a low-temperature distribution system which is controlled under $4^{\circ}C$, because the baby leaf beet's storability and microbial growth are effected even during a short time of 5 hrs during the distribution process.

Characteristics of Histamine Forming Bacteria from Tuna Fish Waste in Korea (국내 참치 부산물 내 히스타민 생성 주요 세균의 특성 구명)

  • Biogenic amines are generally formed through the decarboxylation of specific free amino acids by exogenous decarboxylases released by microbial species associated with the fish products and fermented feeds. This study was conducted to investigate the properties of e tuna waste regarding the control of degradation of biogenic amines (histamine, tyramine, tryptamine, putrescine, and cadaverine) that might be related with the anti-nutritional factor of the tuna waste that is used for manufacturing domestic fish meal. The values of pH and the salt content were 6.51, 3.35% in tuna waste and 5.58 and 5.83% in tuna fish meal, respectively. The strains and dominant bacteria tested in the tuna waste sample were 9.20, 9.29, 5.67, 7.82 and 7.58 log CFU/g of total bacteria, aerobic plate count (APC), total coliform (TC), Lactobacillus spp. and Bacillus spp., respectively. The main histamine forming-bacteria (HFB) in tuna waste were detected by silica gel thin-layer chromatography (TLC) and 7 histamine-forming bacterial species were isolated among microbes grown in selective medium. The histamine concentration was determined by detection of fluorescence of ο-phthaldialdehyde (OPA) derivatives using HPLC and the date were used to reconfirm the identities of the amine-producing bacteria. The 15 histamine- forming bacteria strains grown in trypicase soy broth (TSB) supplemented with 1% L-histidine (TSBH) were identified as Lactococcus(L.) lactis subsp. lactis, Klebsiella pneummonlae, L. garvieae 36, Vibrio olivaceus, Hafnia alvei and L. garvieae which were main dominant amine - producing strains, and Morganella morganii identified by 16S ribosomal RNA (rRNA) sequencing with PCR amplification. A Phylogenetic tree generated from the 16S rRNA sequencing data showed different phyletic lines that could be readily classified as biogenic amine forming gram-positive and negative bacteria.

A Study on the Sanitation Condition for Products of Powdered Raw Grains and Vegetables (곡류 가공품 등의 위생실태에 관한 조사 연구)

  • Recently, the demand for the powdered products to process raw grains and vegetables has been expanded, and the growth possibility of their fields(markets) in the future will be highly valued with the westernization of living environment and the change of the dietary life. We have bought and analyzed the 111 products of raw grains and vegetables from the large-sized marts, markets and internet orders from March to October 2006. The rate of moisture content was $1.7{\sim}12.5%$. We also found out that the foods over 10% moisture content was 8 of those samples we bought or collected. Their ash rates were averagely $0.3{\sim}8.6%$. The number of those foods that the alien substances were detected was 2. The tar pigments, artificial sweeteners and sulfur dioxides of components that contained food additives were not detected. Each detection range of Cadmium, Lead and Arsenic of the injurious heavy metals was non-detectable $(nd){\sim}0.55,\;nd{\sim}4.52,\;nd{\sim}0.10mg/kg$, while the average detection content were 0.08, 0.48, 0.01 mg/kg. By investigating the contamination degree of the microflora, we discovered that the number of the aerobic plate count, B. cereus over 1,000 cfu/g and C. perfringens over 100 cfu/g was 36 (32.4%), 9 (8.1%) and zero, and that the whole sanitation condition of the products of powdered raw grains and vegetables circulated in the market was not so good.

Effect on Quality Change of Cherry Tomato by $CO_2$ Concentration of Flushed Gas and Storage Period (충전가스의 $CO_2$ 함량 및 노출기간의 변화가 방울토마토의 품질변화에 미치는 영향)

  • The purpose of this study was to investigate the quality changes and contamination of microorganisms such as Escherichia coli, mold and yeast in cherry tomatoes during storage at different temperatures, gas composition and periods(7 and 14 days). This study determined pH, color change and the growth pattern of microorganisms in cherry tomato during storage at $5^{\circ}C$, $10^{\circ}C$ and $15^{\circ}C$. According to the results, pH level was a little raised with storage period. On average, $L^*$, $a^*$ and $b^*$ value of cherry tomato were irregular value of increase and decrease of all gas packaging with storage period. In regard of the types of microorganism, aerobic count plate, coliform count, mold and yeast were detected when cherry tomatoes were stored at $5^{\circ}C$, $10^{\circ}C$ and $15^{\circ}C$ during storage for 14 days. Equally, all microorganisms of cherry tomato were irregular with storage period and complex gas packaging. However, this study determined that packaging with a higher $CO_2$ concentration than $O_2$ concentration can reduce growth of microorganism. These studies can be used as primary data for determining the optimal complex gas to storage enlargement.

Evaluation of Microbiological Safety of Lettuce and Cultivation Area (상추와 생산환경의 미생물 안전성 평가)

  • Produce, including leafy vegetables, has been implicated in several outbreaks of food illness. To evaluate microbiological safety of lettuce and it's cultivation area, a total of 147 samples were collected from lettuce farms and post harvest facility at Icheon, Gyeonggi province. The collected samples were assessed for presence of sanitary indicator microorganisms (Aerobic plate count, coliform count, Escherichia coli) and foodborne pathogens (Escherichia coli O157:H7, Salmonella spp., Staphylococcus aureus, Listeria monocytogenes, Bacillus cereus). The population of APC was over 4.0 log CFU from most of the samples. While the numbers of APC, and coliform of lettuce at 62 days after transplanting were 4.18 log CFU/g, and 1.00 log CFU/g, respectively, those of 10 days after transplanting were 5.37 log CFU/g, and 2.87 log CFU/g, respectively. B. cereus was highly detected from soil and balance which were contaminated with 3.5 log CFU/g, and 2.6 log CFU/100 $cm^2$, respectively. The number of E. coli recovered from gloves was 3.5 log CFU/hand. However, E. coli O157:H7, Salmonella spp., and L. monocytogenes were not detected. These data suggested that risk management system should be introduced to lettuce farms to enhance safety of lettuce.

Investigation of the Level of Microbial Contamination in the Environment for Juice Production (생딸기 주스 제조 환경에서의 미생물학적 오염도 조사)

  • Microbial contamination levels in commercial strawberry juices were examined for sanitary indication bacteria, such as aerobic plate count (APC), coliforms, and Escherchia coli, and pathogenic bacteria such as E. coli O157:H7, Salmonella spp., Staphylococcus aureus, and Listeria monocytogenes. APC and coliform count ranged 0-5.2 and $0-2.8\;log_{10}\;CFU/(mL,\;g,\;100cm^{2},\;hand)$, respectively, and 80% strawberry juices were contaminated with E. coli and S. aureus, detected at 19%, was found in employee's hands, strawberries, and strawberry juices, whereas E. coli O157:H7, Salmonella spp, and L. monocytogenes were not detected. These results will provide microbiological information for introduction of Hazard Analysis Critical Control Point (HACCP) system in juice shops.

The effects of low temperature storage and aging of Jeot-kal on the microbial counts and microflora (젓갈의 숙성 및 저온 저장이 미생물 균수 및 균총에 미치는 영향)

  • The addition of 5% NaCI to standard plate count (SPC) and bromcresol purple (BCP) agar showed the highest viable cell counts for Jeot-kal samples. The use of 15% glycerol as cryoprotectant showed the highest microbial survival rate at both temperatures, $-20^{\circ}C$ and $-170^{\circ}C$, and on both colony count media, SPC and BCP. During the aging, the pH of Bajirak Jogae-Jeot (fermented clam) decreased from 6.8 to 5.0. Crude protein content was 10% for Bajirak Jogae-Jeot and $6{\sim}7%$ for Myeolchi-Jeot (fermented anchovy). Microbial population of Bajirak Jogae-Jeot was $10^9\;CFU/g$ after 4 weeks of aging, but was only $10^{3-5}\;CFU/g$ in the case of Myeolchi-Jeot. The proportion of Gram positive and catalase negative bacteria in Bajirak Jogae-Jeot increased drastically during the 4 weeks of aging, which showed typical lactic bacterial fermentation. After 2 years' storage of Jeot-kal in liquid nitrogen tank, the cell counts of total aerobic or lactic bacteria were decreased, resulting in about 10% survival rate. Microbial floral change of Jeot-kal was also investigated. In the case of Bajirak Jogae-jeot, the ratio of rod to cocci and that of Gram negative to positive increased after liquid nitrogen storage. But, rod to cocci ratio of Myeolchi-jeot decreased after liquid nitrogen storage. The ratio of yeasts decreased in both cases after storage.
