Occupational Exposure Assessment for Benzene Using Exposure Models (ECETOC TRA and Stoffenmanager) and Applicability Evaluation of Exposure Models in K-REACH (노출 모델의 화평법 적용성: ECETOC TRA와 Stoffenmanager Tier 1 노출 모델을 활용한 벤젠의 작업자 노출 평가)

  • Objectives: The objectives of this study are to estimate the inhalation exposure level of benzene for workers using Tier 1 exposure models ECETOC TRA (European Center for Ecotoxicology and Toxicology of Chemicals Target Risk Assessment) and Stoffenmanager, and to investigate their reliability for exposure assessment in K-REACH. Methods: Two exposure scenarios, 'manufacture of benzene' and 'use as solvents,' were developed for assessment of workers' exposure to benzene. The Process Category (PROC) for ECETOC TRA was collected from the European Chemical Agency (ECHA) registration dossier, and the Activity for Stoffenmanager was converted from PROC using translation of exposure models (TREXMO). The information related to exposure, such as working duration, Respiratory Protective Equipment (RPE), Local Exhaust Ventilation (LEV), and Risk Management Measure (RMM) were classified into high, medium, and low exposure conditions. The risk was determined by the ratio of the estimated exposure and occupational exposure limits of benzene. Results: Under high exposure conditions, the worker exposure level calculated from all PROCs and Activities exceeded the risk level, with the exception of PROC 1 and Activity 1. In the medium exposure condition, PROC 8a, 8b, and 9 and Activity 3, 7, and 8 all exceeded the risk, whereas in the low condition, all PROCs and Activities were determined to be safe. As a result, action corresponding with the low exposure condition is required to reduce the risk of exposure among workers in workplaces where benzene is manufactured or used as a solvent. In addition, the predicted exposure levels derived from the exposure models were lower than measured levels. The exposure levels estimated from Stoffenmanager were more conservative than those from ECETOC TRA. Conclusions: This study demonstrates the feasibility of exposure models for exposure assessment through the example of occupational inhalation exposure assessment for benzene. For more active utilization of exposure models in K-REACH, the exact application of collected information and accurate interpretation of obtained results are necessary.

  • Biological nitrogen fixation is an important process for academic and industrial aspects. This review will briefly compare industrial and biological nitrogen fixation and cover the characteristics of biological nitrogen fixation studied in Azotobacter vinelandii. Various organisms can carry out biological nitrogen fixation and recently the researches on the reaction mechanism were concentrated on the free-living microorganism, A. vinelandii. Nitrogen fixation, which transforms atmospheric $N_2$ into ammonia, is chemically a reduction reaction requiring electron donation. Nitrogenase, the biological nitrgen fixer, accepts electrons from biological electron donors, and transfers them to the active site, FeMo-cofactor, through $Fe_4S_4$ cluster in Fe protein and P-cluster in MoFe protein. The electron transport and the proton transport are very important processes in the nitrogenase catalysis to understand its reaction mechanism, and the interactions between FeMo-cofactor and nitrogen molecule are at the center of biological nitrogen fixation mechanism. Spectroscopic studies including protein X-ray crystallography, EPR and $M{\ddot{o}}ssbauer$, biochemical approaches including substrate and inhibitor interactions as well as site-directed mutation study, and chemical approach to synthesize the FeMo-cofactor model compounds were used for biological nitrogen fixation study. Recent research results from these area were presented, and finally, a new nitrogenase reaction mechanism will be proposed based on the various research results.

  • The aerial parts of tile Garland (Chrysanthemum comnarium L.) were extracted in MeOH and solvent fractionated with EtOAc, n-BuOH and water, successively. EtOAc fractions gave three steroid compounds through application of silica gel column chromatographies. The chemical structures of the steroids were determined by the interpretation of several spectral data, including NMR and MS as $stigmast-5-en-3{\beta}-ol\;(1,\;{\beta}-sitosterol),\;stigmast-4-en-6{\beta}-ol-3-one$ (2), $stigmast-4-en-6{\alpha}-ol-3-one$ (3). Compounds 2 and 3 have been so far reported only in the aquatic plants, were isolated for the first time from the land plants.

Chemical Compositions and Nematicidal Activities of Essential Oils on Meloidogyne hapla (Nematoda: Tylenchida) Under Laboratory Conditions (식물정유의 당근뿌리혹선충(Meloidogyne hapla)에 대한 살선충활성 및 방향성성분 분석)

  • To evaluate the efficacy of natural nematicides for the control of root-knot nematode in strawberry greenhouses, commercial essential oils were examined by 24-well culture plate bioassay for their nematicidal activities against second-stage juveniles and eggs of Meloidogyne hapla. Based on the mortality of M. hapla juveniles at a concentration of $125{\mu}g/mL$, the most active essential oil was Alpinia galanga (100%), followed by Carum carbi (22.3%), Eugenia caryophyllata (9.4%), Cinnamonum zeylanicum (7.2%), Mentha pulegium (2.4%), and Foeniculum vulgare (2.1%). Moreover, A. galanga significantly reduced hatching at 7, 14, and 21 days after treatment. The volatile constituents identified in the A. galangal oil were methyl cinnamate (87.4%), 1,8-cineole (4.4%), ${\beta}$-pinene (2.5%), ${\alpha}$-pinene (2.2%), and p-cymene (1.1%), as major constituents. Results of this study show that A. galangal essential oil and its major constituents may serve as an environmental friendly agent of a promising natural nematicide to control Meloidogyne spp.

  • The MeOH extracts obtained from whole plant of Trigonotis peduncularis Benth. were solvent fractionated using EtOAc, n-BuOH and water, successively. The EtOAc and n-BuOH fractions gave four flavonol glycosides through application of silica gel and octadecyl silica gel (ODS) column chromatographies. The chemical structures of the flavonol glycosides were determined by the interpretation of several spectral data including 2D-NMR as $kaempferol-3-O-{\beta}-{D}-glucopyranoside\;(astragalin,\;1),\;kaempferol-3-O-{\alpha}-{L}-rhamnopyranosyl\;(1{\rightarrow}6)-{\beta}-{D}-glucopyranoside\;(nicotiflorin,\;2),\;quercetin-3-O-{\alpha}-{L}-rhamnopyranosyl(1{\rightarrow}6)-{\beta}-{D}-glucopyranoside\;(rutin,\;3),\;quercetin-3-O-{\beta}-{D}-glucopyranoside\;(isoquercitrin,\;4)$. The flavonoids have been first isolated from this plant. Nicotiflorin $(100\;{\mu}g/ml)$ showed $68.3{\pm}1.2%$ of the inhibitory effect on hACAT1(human Acyl CoA: cholesterol transferase 1) activity.

  • This research was carried out to investigate chemical pretreatment using limestone in treating abandoned metallic mine drainage with anaerobic biological treatment, and to estimate application of papermill and livestock sludge as carbon sources for SRB (Sulfate Reducing Bacterial. Capacity of anaerobic limestone bed was steeply decreased. But if limestone was utilized as pretreatment process in treating them with anaerobic biological treatment. it could look forward to stabilize system because it did initally neutralize them. Effluent SCOD in R-4 was lower than R-l~R-3 in inital HRT 5day but its concentration was high in HRT 1day after passed time. Therefore in point of durability and supply of organic matter. it seemed that R-4 was useful became organic matter in R-4 was not consumed by excessive degradation within short period. In all reactors, pH was suitable for SRB growth in whole HRT, but on the evidence of ORP, SRB was active after HRT 2day. Fixation trend of heavy meta s showed high as $SO_4{^{2-}}$ reduction efficiency increased, and $SO_4{^{2-}}$ reduction and fixation of heavy metals were relatively high in HET 2day.

  • The following results are from the test of semi-gel electrolyte to store energy efficiently and use advanced VRLA batteries by photovoltaic and wind power generation. Semi-Gel electrolyte with Silica 5% became Gel after 1 and half hour. It shows it is the most suitable time that the electrolyte can be absorbed into the separator and active material of plate to be gel. The test also says that semi-gel electrolyte shows the much better performance for low-rate discharge and the liquid electrolyte is good for high-rate discharge because the reaction rate of gel electrolyte is slower than liquid one for high-rate discharge performance. The test with DOD10% and DOD100% says that 5% silica electrolyte shows much better performance for life efficiency than liquid one. Because semi-gel electrolyte increase the efficiency of gas recombination at the chemical reaction of VRLA battery and it makes minimizing the reduction of electrolyte. Using the 5% silica electrolyte in order to improve the stroage efficiency and life performance for photovoltic and wind power generation, it causes improving by 4.8% for DOD100% and 20% for DOD10%.

A Study on the Nutrient Release Characteristics from Sediments in the Asan Reservoir (아산호 퇴적물에서 영양염류 용출특성에 관한 연구)

  • To investigate the effects of sediments on the water quality of an estuary reservoir, Asan reservoir, we analyzed physical and chemical characteristics of the sediments and estimated nutrients release rate from the sediments. The sediments characterized as sandy loam are mainly composed of Nonapatite-P(64.7%) which has strong influence on the eutrophication of the reservoir by releasing from the sediments under the condition of increasing pH and anaerobic environment. High nutrient release rates was observed in April. Negative release rates in June show that there is no significant nutrient release from the sediments. The nutrient release was active at the Site B around the confluence of tributary compared to the Site A near the embankment. Based on the information of nutrient release rates and sediments surface area, daily average nutrient release rates of Asan reservoir are estimated; TN 6,609 kg/d, TP 3,877 kg/d. Since the amount of N and P released from the sediments corresponds to the 7.06% of N and 22.04% of P incoming from the watershed, it can be concluded that there is little influence of sediments on the water quality of Asan reservoir.

Mutant p53-Notch1 Signaling Axis Is Involved in Curcumin-Induced Apoptosis of Breast Cancer Cells

  • Notch1 has been reported to be highly expressed in triple-negative and other subtypes of breast cancer. Mutant p53 (R280K) is overexpressed in MDA-MB-231 triple-negative human breast cancer cells. The present study aimed to determine whether the mutant p53 can be a potent transcriptional activator of the Notch1 in MDA-MB-231 cells, and explore the role of this mutant p53-Notch1 axis in curcumin-induced apoptosis. We found that curcumin treatment resulted in an induction of apoptosis in MDA-MB-231 cells, together with downregulation of Notch1 and its downstream target, Hes1. This reduction in Notch1 expression was determined to be due to the decreased activity of endogenous mutant p53. We confirmed the suppressive effect of curcumin on Notch1 transcription by performing a Notch1 promoter-driven reporter assay and identified a putative p53-binding site in the Notch1 promoter by EMSA and chromatin immunoprecipitation analysis. Overexpression of mutant p53 increased Notch1 promoter activity, whereas knockdown of mutant p53 by small interfering RNA suppressed Notch1 expression, leading to the induction of cellular apoptosis. Moreover, curcumin-induced apoptosis was further enhanced by the knockdown of Notch1 or mutant p53, but it was decreased by the overexpression of active Notch1. Taken together, our results demonstrate, for the first time, that Notch1 is a transcriptional target of mutant p53 in breast cancer cells and suggest that the targeting of mutant p53 and/or Notch1 may be combined with a chemotherapeutic strategy to improve the response of breast cancer cells to curcumin.

Environmental Characteristics of the Seawater and Surface Sediment in the vicinity of Pusan Harbor Area in Winter (겨울철 부산항 주변해역의 수질과 표층퇴적물 환경특성)

  • The chemical constituents for the seawater and sediment were measured to evaluate pollution in the sea around Pusan Harbor in winter, n992. The average value of trophic state index (TSI) was 19.4 at the outside of Buk Harbor, 50,4 at the inside of Buk Harbor, 56,3 at the Nam Harbor and 5,0 at the Kamchun Harbor. The high correlation found in salinity-nutrients diagram with AOU suggested that the enrichment of nutrients in Pusan Harbor during winter was mainly due to the influx of terrestrial effluents and partially by regenerated nutrients from suspended organic matters in the water column. The mean values of total ignition loss (TIL), COD and total sulfide in the surface sediments were$12.1\%$, 17.5 mg/g.dry wt. and 1.18 mg/g.dry wt. respectively. The highest level of those parameters was shown mostly at the inside of Buk Harbor. The mean concentration of total organic carbon (TOC), total organic nitrogen (TON), and total phosphorus were 24.9 mg/g.dry wt., 1.3mg/g.dry wt. and 0.69 mg/g. dry wt., respectively, Both of the highest level for TOC and total phosphorus have found at the Nam Harbor. On the other hand, the Highest level for TON was found at the inside of Buk Harbor. The TOC/TON atomic ratio with a range of 10.2-60.2 (mean value of 22.5) strongly indicated the active role of the input from the terrestrial organic pollutants.
